실시예 2: 팔리다자주닭개비 추출물 제조Example 2: Palidajaju chicken gaebi extract preparation
팔리다자주닭개비(T. pallida) 추출물을 제조하기 위해, 제주대학교 생명자원과학대학 유리온실 내에 생육 중인 팔리다자주닭개비의 지상부를 채취하여 건조기에서 건조하였다. 건조시킨 팔리다자주닭개비를 믹서기로 분쇄하고 500 μm 체로 거른 다음 10 g당 100 mL의 에탄올, 메탄올 또는 에틸아세테이트로 24시간씩 2회 반복추출 후 감압농축기로 농축하였다. 완전농축이 안된 시료는 동결건조기로 건조하여 각각 팔리다자주닭개비 에탄올추출물(T. pallida ethanol extract; TPEE), 팔리다자주닭개비 메탄올추출물(T. pallida methanol extract; TPME), 팔리다자주닭개비 에틸아세테이트추출물(T. pallida ethyl acetate extract; TPAE) 시료를 제조하였다. 이후 시료는 DMSO에 녹인 다음 증류수로 1000 μg/mL에서 62.5 μg/mL까지 희석한 후 실험에 사용하였다.To prepare the extract of Palidajaju chicken ( T. pallida ), the above-ground part of Palidajaju chicken gaebi growing in the glass greenhouse of the College of Life Science and Resources at Cheju National University was collected and dried in a dryer. GraphPrism 소프트웨어 Ver. 7(Graphpad Software, lnc., La Jolla, CA, USA)을 사용하여 SC50(50% scavenging concentration, 자유라디칼을 50% 소거하는 농도)을 산출하였다.The electron donating ability was measured by the method of Blois (1958). SC50을 산출한 결과, TPEE, TPME, TPAE 추출물에 대한 SC50은 664.8, 660.8, 277.3 μg/mL이었다.
