Claims (23)
- 항-IL-13 항체 또는 이의 항원-결합 단편 및 한 가지 이상의 숙주 세포 단백질(HCP)을 포함하는 시료 혼합물로부터 숙주 세포 단백질(HCP) 감소된 항-IL-13 항체 또는 이의 항원-결합 단편을 생산하는 방법으로서,
(a) 상기 시료 혼합물을 친화성 크로마토그래피 수지에 접촉시켜 친화성 크로마토그래피 시료를 수집하는 단계;
(c) 상기 친화성 크로마토그래피 시료를 pH 강하시켜 pH가 약 3 내지 약 4로 강하된 pH 강하된 시료를 생성하는 단계;
(d) 상기 pH 강하된 시료를 약 4.5 내지 약 6.5의 pH로 조정하고 이에 따라 조정된 pH의 시료를 이온교환 수지에 접촉시켜 이온교환 시료를 수집하는 단계;
(e) 상기 이온교환 시료를 소수성 상호작용 크로마토그래피(HIC) 수지에 접촉시켜 HCP-감소된 상기 항체 제제를 포함하는 상기 HIC 시료를 수집하는 단계를 포함하는,
시료 혼합물로부터 HCP 감소된 항-IL-13 항체 또는 이의 항원-결합 단편을 생산하는 방법. - 제1항에 있어서, 상기 pH 강하가 적합한 산을 상기 시료 혼합물과 혼합시킴으로써 달성되고, 상기 적합한 산이 시트르산, 아세트산, 카프릴산 등으로 이루어진 그룹에서 선택되는, 방법.
- 제1항에 있어서, 상기 친화성 크로마토그래피 수지가 단백질 A 수지인, 방법.
- 제3항에 있어서, 상기 단백질 A 수지가 맵셀렉트(MabSelectTM) 수지인, 방법.
- 제1항에 있어서, 상기 이온교환 수지가 음이온 교환 수지 또는 양이온 교환 수지인 방법.
- 제5항에 있어서, 상기 이온교환 수지가 양이온 교환 수지인, 방법.
- 제6항에 있어서, 상기 양이온 교환 수지가 프락토겔(Fractogel), 카르복시메틸(CM), 설포에틸(SE), 설포프로필(SP), 포스페이트(P) 및 설포네이트(S)로 이루어진 그룹에서 선택되는, 방법.
- 제7항에 있어서, 상기 양이온 교환 수지가 프락토겔TM SO3 -인, 방법.
- 제5항에 있어서, 상기 이온교환 수지가 음이온 교환 수지인, 방법.
- 제9항에 있어서, 상기 음이온 교환 수지가 Q 세파로즈(Sepharose), 디에틸아미노에틸(DEAE), 4급 아미노에틸(QAE) 및 4급 아민(Q) 그룹으로 이루어진 그룹에서 선택되는, 방법.
- 제10항에 있어서, 상기 음이온 교환 수지가 Q-세파로즈인, 방법.
- 제1항에 있어서, 상기 이온교환 단계가 일차 이온교환 단계 및 이차 이온교환 단계를 포함하는, 방법.
- 제1항에 있어서, 상기 HIC가 한 가지 이상의 소수성 그룹을 포함하는 컬럼을 사용하여 달성되는, 방법.
