스텐트에 기초한 유익한 물질의 국소 전달을 통해서 나타나는 많은 문제들 중에서, 가장 중요한 것 중의 하나가 재협착(restenosis)이다. 재협착은 혈관성형술 및 스텐트의 이식과 같은 혈관 삽입(intervention) 후에 일어날 수 있는 주된 합병증이다. 간단하게 정의하자면, 재협착은 세포외 매트릭스 침착, 신생내막 과증식, 및 혈관 평활근 세포 증식에 의해 혈관 내강 직경을 감소시키는 상처 치유 과정으로서, 결국에는 관강을 더욱 좁히거나 재폐색(reocclusion)을 일으킨다. 향상된 외과수술 기술, 기구, 및 약물의 도입에도 불구하고, 전체적인 재협착율은 혈관성형술 후 6 내지 12 개월 내에 25% 내지 50%의 범위인 것으로 보고되어 있다. 이러한 상태를 치료하기 위해, 추가적인 혈관재생수술이 자주 필요하고, 그럼으로써 환자에게 외상 및 위험을 증가시킨다.

복수의 의료장치, 바람직하게는 하나의 주축 당 11개의 의료 기구를 일련의 주축 상에 배치한다. 각 주축은, 한 가지 이상의 유형의 의료 기구, 의료 기구의 개구부로 로딩될 유익한 물질층, 및 각 주축에 대한 구체적인 정체를 확인할 수 있는 특유의 표시로 바코드화한다. 바 코드 정보 및 위치도 작성 결과는, 스텐트의 로딩을 위해 CPU에 저장된다.
PLGA(poly(latide-co-glycolide))(Brimingham Plymers, Inc.) 및 DMSO와 같은 적절한 용매의 제 1 혼합물을 준비한다. 그 혼합물을 스텐트 내 구멍으로 적상으로 로딩한다. 그리고 나서 스텐트를 55℃의 온도에서 약 60 분간 베이킹하여 용매를 증발시켜서 장벽층을 형성한다. 제 2 층은 개구부 내에 폴리머 용액을 충전하고 용매를 증발시키는 동일한 방법으로, 제 1 층 위에 적층한다. 이 방법은 의료 기구의 개구부 내에 9개의 개별층이 로딩되어 장벽층을 형성할 때까지 계속한다.

그리고 나서, PLGA 및 DMSO의 제 3 혼합물을 개구부 내 치료제 위에 도입하여 덮개층을 형성한다. 용매를 증발시키고, 덮개층이 의료기구에 첨가될 때까지 충전 및 증발 과정을 반복하는데, 본 실시예에서는, 단일의 덮개층을 첨가한다.
원하는 용액을 포함하는 유익한 물질의 복수의 층을 제공하기 위하여는, 저장부를 교체하고, 압전식 미세-분사 디스펜서를 세정한다. 저장부의 교체 및 디스펜서의 세정은 (필요한 경우) 상이한 유익한 층들이 약물, 용매 및 폴리머의 정확한 양을 포함하는 원하는 용액을 확실하게 포함할 수 있도록 해 준다.
생체 내에서 충전된 의료 기구의 주입 후, PLGA 폴리머는 가수분해를 통해 분해되고 파클리탁셀이 방출된다.
본 발명은 그 바람직한 구현예를 참고로 하여 상세하게 기술되었지만, 본 발명의 범위 내에서 그 변형이 가능하며 균등물이 이용될 수 있음은 당업자에게 자명할 것이다.
