Abstract
본 발명의 목적은 HLA-A2에 결합하여 사람 세포상해성(killer) T세포를 활성화시키는 글리피칸(glypican) 3유래의 펩티드를 동정함으로써, 일본인에 있어서 GPC-3을 과발현하는 다양한 암환자의 40%를 대상으로 하는 면역요법을 가능하게 하는 수단을 제공하는 것이다. 본 발명에 의하면, 이하의 어느 하나의 펩티드가 제공된다: (A) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열로 구성된 펩티드; 및, (B) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열에 있어서 1개 또는 2개의 아미노산이 치환 또는 부가된 아미노산으로 구성되고, 세포상해성(killer) T세포의 유도능력을 가지는 펩티드.

Description
본 발명은 간세포암, 악성흑색종(melanoma) 등, 글리피칸-3(GPC-3)을 과발현하는 암에 대한 백신으로 유효한 신규한 펩티드 및 상기 펩티드를 포함하는 종양치료 및 예방제에 관한 것이다.
원발성 간세포암은 세계각국에서 발생빈도가 높은 악성질환 중 하나이다. B형 간염과 C형 간염의 세계적인 유행에 따라, 아시아 및 유럽각국에서의 간세포암의 발생률은 급격히 상승하고 있고, 간염 바이러스의 감염으로부터 발병하기까지의 긴 잠복기를 고려하면, 향후 50년에 걸쳐 이러한 경향은 지속될 것으로 예상된다. 병이 진행된 간세포암의 예후는 불량하여, 새로운 치료전략의 개발이 긴급히 필요한 상태이다.
한편, 최근의 분자생물학 및 종양면역학의 개발에 의해, 세포상해 성(killer) T세포 및 헬퍼 T세포가, 암세포 또는 항원제시세포의 표면에 HLA분자를 개재하여 제시되는, 암세포에 특이적으로 과발현하는 단백질이 분해되어 생긴 펩티드를 인식하여 암세포를 파괴하는 면역반응을 보이는 것이 밝혀졌다. 게다가, 이와 같이 암을 공격하는 면역반응을 자격하는 종양항원 단백질 및 그들로 부터 유래하는 펩티드가 동정됨에 따라, 항원특이적 종양면역요법의 임상적 응용이 행해지고 있다.
HLA 클래스Ⅰ분자는 신체의 모든 유핵세포 표면에서 발현되어, 세포질이나 핵에서 생산되는 단백질이 세포내에서 분해되어 생긴 펩티드와 결합해서 세포표면에서 발현된다. 정상적인 세포의 표면에는 정상의 자기 단백질로부터 유래되는 펩티드가 HLA 클래스Ⅰ분자에 결합하여, 이것을 면역계의 T세포가 인식해서 파괴하는 것은 아니다. 한편, 암세포는 암이 되는 과정에서, 정상세포에서는 거의 발현하지 않거나, 또는 극히 적은 수밖에 발현되지 않는 단백질을 대량으로 발현되는 경우가 있다. 이와 같은 암세포에 특이적으로 과발현하는 단백질이 세포질 내에서 분해되어 생긴 펩티드가 HLA 클래스Ⅰ분자에 결합해서 암세포의 표면에 발현하면, 세포상해성 T세포가 이것을 인식해서 암세포만을 파괴한다. 또한, 이와 같은 암 특이항원과 펩티드를 개체에 투여하여, 정상세포는 위해하지 않으면서, 암세포만을 파괴하여 암의 증식을 억제할 수 있다. 이것을 암 특이항원을 이용한 암 면역요법이라 한다. 또한, HLA 클래스Ⅱ분자는 주로 항원제시세포의 표면에서 발현되어, 항원제시세포가 세포외부로부터 들어와서 세포 내부에서 분해되어 생긴 암특이항원 유래의 펩티드와 결합해서 세포 표면에서 발현한다. 이것을 인식한 헬퍼 T세포는 활성 화된 다른 면역담당 세포를 활성화하는 여러 사이토카인을 생산하여, 종양에 대한 면역반응을 유도 또는 증강시킨다.
여기서, 이들 암에 특이적으로 과발현하고 있는 항원을 표적으로 한 면역요법을 개발할 수 있다면, 자기의 정상 장기에 유해현상을 미치지 않으면서 암만을 유효하게 배제하는 치료법이 될 가능성이 있다. 또한, 다른 치료를 행할 수 없는, 말기의 암환자에게도 사용가능한 치료법이 될 수 있을 것이다. 그리고, 앞서 이와 같은 암을 발생하는 위험성이 높은 사람에 대해서, 암 특이항원 및 펩티드를 백신으로 해서 투여하여 암 발생을 예방할 수 있는 가능성이 있다.
α-페토단백질(α-fetoprotein, AFP)은 정상조직에서는 태생기에만 발현하지만, 많은 간세포암에 있어서 발현이 재활성화되는 이른바 암 태아성 단백질이라는 것이 보고되었다. 또한, 마우스나 사람의 T세포 중에는 HLA 클래스Ⅰ분자에 의해 제시된 AFP 유래 펩티드 에피토프를 인식하는 것이 존재한다. 태아는 발달단계에 있어서 혈장내에 높은 수준으로 존재하는 AFP에 노출되어 있음에도 불구하고, 성숙 T세포는 AFP에 대해서 완전한 면역관용(tolerance)을 획득하지 않고, AFP 특이적인 T세포가 말초혈 중에 검출된다. 즉, 암태아성 단백질은 면역치료의 표적이 될 수 있다.
간세포암의 치료법에는 여러가지가 있음에도 불구하고, 다른 암에 비해 예후가 좋지 않아서 난치성 암의 하나로 알려져 있다. 그 원인으로서, 간세포암의 베이스에 간경변이 있고, 환자의 간기능이 좋지 않은 경우와, 하나의 암을 치료해도 또 다른 곳에서 암이 발생하는 특징을 들 수 있어서, 가능한 빨리 새로운 치료 전략의 개발이 요구되고 있다. 간세포암에 특이적으로 과발현하고 있는 항원을 표적으로 한 면역요법을 개발한다면, 자기의 정상장기에 유해를 끼치지 않으면서 암만을 유효하게 배제하는 치료법이 될 가능성이 있다. 또한, 모든 말기의 암환자 뿐만 아니라, 간기능이 매우 안좋아서 다른 치료가 불가능한 암환자에게도 사용가능한 치료법이 될 수 있을 것이다. 그리고, 현재 일본에서는 간세포 암의 예비군이라 하는 C형 간염 감염자가 200만명 이상 존재한다고 한다. 이들 감염자에게도 간세포암의 예방에 관련해서 이와 같은 면역치료가 적용될 가능성이 있다.
멜라노머(melanoma)는 악성흑색종이라 불리는 피부암의 일종이다. 피부암에도 여러 종류가 있지만, 가장 악성도가 높아 매우 무서운 암이다. 피부를 구성하고 있는 세포 중에서 멜라닌 색소를 생산하는 세포가 있어서, 이것을 색소세포(멜라이사이트)라 부르는데, 이 세포가 암화된 것이 멜라노머이다.
일본에서 멜라노머의 발생빈도는 인구 10만명당 1.5~2명정도이고, 연간 1500명~2000명정도가 발병한다고 추측되고 있다. 구미에서는 인구 10만명당 10명 이상이 발병하고, 호주에서는 인구 10만명당 20명 이상이 발병해서 세계에서 발병률이 가장 높은 것으로 알려져 있다. 따라서, 구미와 호주 사람들은 멜라노머에 관심을 가지고 멜라노머 발생에 주의를 기울이고 있다. 또한, 멜라노머는 환경파괴에 의한 대기 중의 오존층 감소에 따른 자외선 노출의 증가 때문에, 백인에게 발생빈도가 증가하고 있으며, 일본에서도 멜라노머의 발생이 매년 증가하고 있다. 최근의 조사에서는, 일본에 있어서 멜라노머에 의한 연간 사망자수는 450명 전후로 증가하고 있다. 멜라노머는 연령과 상관없이 발생하지만, 특히 40세 이상이 되면 발생률 이 높아져서, 60~70대가 가장 높다. 유아의 발생률은 극히 적으나 발생하지 않는다고는 말할 수 없고, 최근 20~30대의 젊은층의 발생률이 높아지고 있으며, 남녀 성별에 관계없이 발생한다. 일본인에게 멜라노머가 잘 발생하는 부위는 발바닥이 가장 많고, 약 3할 정도를 차지하고 있다. 손톱과 발톱에도 많이 나타나는 것이 일본인의 특징이다. 그 외에 구미인과 마찬가지로, 몸, 손, 발, 머리 등 피부 어디에도 발생한다.
본 발명자들은 먼저 cDNA 마이크로어레이 해석을 이용한 23,040종의 인간 유전자를 갖는 게놈와이드의 유전자 발현 해석을 통하여 20가지 예의 원발성 간세포 및 태생기를 갖는 여러 정상장기에 있어서, 이들의 유전자의 발현 프로필을 검토하고, 글리피칸-3(glypican-3, GPC-3)이 태생기의 간, 신장, 폐에 발현하고 성인의 정상장기에서는 태반이외에는 거의 발현하지 않는데 반하여, 많은 간세포암으로 과발현하는 것을 발견하였다. 게다가, 이 GPC-3이 분비단백질이고 ELISA법을 이용하여 40%의 간세포암 환자의 혈청 중에서 GPC-3을 검출할 수 있어서, 이것이 간세포암의 새로운 종양마커로서 유용하다는 것이 보고되었다(참조: Nakatsura, T. 등, Biochem. Biophys. Res. Commun. 306, 16-25(2003)). 게다가, GPC-3는 멜라노머 환자의 혈청중에서도 검출되어, 멜라노머의 종양마커로서도 유용하다는 것이 보고되었다(참조: Nakatsura, T. 등, Clin. Cancer Res. 10: 6612-6621(2004)).
본 발명자들은 이미 HLA-A24 양성 간세포암 또는 멜라노머 환자를 대상으로 한 면역요법에 유용한 HLA-A24에 결합해서 인간· 세포상해성 T세포에 제시되는 GPC-3 펩티드를 동정하였다. 이 펩티드를 이용한 면역치료가 유효한 것을 HLA-A24 에 결합하는 펩티드와 같은 모양의 구조를 갖는 펩티드를 결합하는 마우스 Kd분자를 발현하는 BALB/C 마우스를 이용한 동물실험에서도 증명되어 이미 보고하였다(국제출원 제 PCT/JP2004/016374호: 국제출원일 2004년 10월 28일). GPC-3는 정상장기에서는 태반과 태생기의 간에서 밖에 발현하지 않기 때문에, GPC-3를 표적으로 한 면역치료법을 통해 종양의 증식을 억제해도 자기면역병 등의 유해현상이 일어나지 않는다는 것도 마우스 실험에서 확인하고 있다.
본 발명자들은 지금까지 종양거절 항원인 글리피칸-3에 있어서, 주로 HLA-A24에 의해 세포상해성 T세포에 제시된 펩티드를 동정하였다(참조: 국제출원 제 PCT/JP03/10459호: 국제출원일 2003년 8월 19일). 그러나, HLA-A24에 의해 세포상해성 T세포에 제시된 펩티드만으로는, 일본인 중에서 HLA-A24를 가지는 60%밖에 펩티드백신 투여의 대상이 되지 않는다. 게다가, 일본인의 40%가 양성의 HLA-A24에 의해 세포상해성 T세포에 제시된 펩티드를 동정한다면, 양자를 합치면 일본인의 약 85%를 대상으로 할 수 있을 뿐만 아니라, HLA-A2는 구미의 백인들에게도 양성 환자의 빈도가 높기 때문에 많은 구미의 백인들에게도 응용할 수 있게 된다. 따라서, HLA-A2에 의해 세포상해성 T세포에 제시된 펩티드의 동정은 중요한 과제이다. 특히, 멜라노머는 구미의 백인들에게 많고, 면역요법이 유효한 암이며, 간세포암도 구미에서 급속히 증가하고 있는 암이기 때문에, HLA-A2 결합성 GPC-3 펩티드를 이용한 면역요법을 적응하는 환자도 많을 것으로 추정된다.
발명의 개시
발명이 해결하고자 하는 과제
본 발명은 HLA-A2에 의해 세포상해성 T세포에 제시되는 펩티드를 동정함으로써, 일본인의 GPC-3을 과발현하는 다양한 암환자의 약 40%를 대상으로 할 수 있는 면역요법을 가능하게 하는 수단을 제공하는 것을 해결과제로 한다.
발명을 해결하기 위한 수단
본 발명자들은 우선 cDNA 마이크로어레이 해석에 의하여, 사람의 간세포암에 있어서 특이적으로 과발현하는 신규한 암태아성 단백질로서 글리피칸 3(GPC-3)을 동정하고, 이어 간세포암환자 혈청 중에 가용성 GPC-3 단백질이 검출되어 GPC-3는 간세포암의 새로운 종양 마커임을 밝혀내었다. 본 발명자들은 마우스 멜라노머 세포주 B16에서 GPC-3가 발현하는 것을 발견하고, GPC-3가 멜라노머에 있어서 간세포암과 마찬가지로 과발현하여 유용한 종양 마커가 되는 것이 아닌가 생각하고, 확인을 위하여 실험을 행한 결과, GPC-3가 멜라노머에 있어서 이제까지 불가능하였던 조기진단이 가능한 종양마커인 것을 발견하였다. 본 발명자들은 사람 CD8 양성세포상해성 T세포를, 실험실적 조건(in vitro)에서 HLA-A2 결합 모티브를 가지는 사람 GPC-3 펩티드를 펄스(pulse)한 사람 말초혈 단구유래 수상세포과 함께 배양하여 자격함으로써, GPC-3 펩티드 특이적인 세포상해성 세포를 유도하였다. 각 GPC-3 펩티드에 특이적인 세포상해성 세포를 유도하는지를 HLA-A2에 의해 제시된 펩티드를 인식하여 활성화된 세포상해성 세포를 생산하는 γ-인터페론(IFN-γ)를 ELISPOT법을 이용하여 검출함으로써, 면역치료에 응용할 수 있는 표적항원의 후보가 될 수 있는 신규한 GPC-3 펩티드를 동정하였다.
즉, 본 발명에 의하면, 이하의 발명이 제공된다.
(1) 이하의 어느 하나의 펩티드:
(A) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열로 구성된 펩티드; 및,
(B) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열에 있어서 1개 또는 2개의 아미노산이 치환 또는 부가된 아미노산으로 구성되고, 세포상해성 (killer) T세포의 유도능력을 가지는 펩티드.
(2) (1)에 기재된 펩티드를 최소 1종류 이상을 포함하는, 암에 대한 면역유도제.
(3) (1)에 기재된 펩티드를 최소 1종류 이상을 포함하는, 종양의 치료 및/또는 예방제.
(4) (1)에 기재된 펩티드를 포함하는, 종양반응성 T세포 유도능력이 높은 항원제시세포 유도제.
(5) 이하의 어느 하나의 펩티드를 암호화하는 유전자를 포함하는, 종양반응성 T세포 유도능력이 높은 항원제시세포 유도제:
(A) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열로 구성된 펩티드; 및,
(B) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열에 있어서 1개 또는 2개의 아미노산이 치환 또는 부가된 아미노산으로 구성되고, 세포상해성 (killer) T세포의 유도능력을 가지는 펩티드.
(6) (1)에 기재된 펩티드를 포함하는, 종양반응성 T세포 유도제.
(7) (1)에 기재된 펩티드에 대한 항체.

발명을 실시하기
위한 최선의 실시태양
(1) 본 발명의 펩티드 및 그를 포함하는 암에 대한 면역유도제
본 발명의 펩티드는 이하 어느 하나의 펩티드이다.
(A) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열로 구성된 펩티드; 및,
(B) 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열에 있어서 1개 또는 2개의 아미노산이 치환 또는 부가된 아미노산으로 구성되고, 세포상해성 (killer) T세포의 유도능력을 가지는 펩티드.
본 명세서에서, 세포상해성 T세포의 유도능력을 가지는 펩티드라 함은, 세포상해성 T세포(killer T cell/CTL)을 자격하는 T세포 유도활성을 가지는 펩티드를 의미한다.
본 발명의 펩티드의 입수 및 제조방법은 특히 한정되지 않고, 화학합성한 단백질이어도 유전자 재조합기술에 의해 작제된 재조합 단백질이어도 가능하다.
화학합성 펩티드를 입수하는 경우에는, 예를 들면, Fmoc(fluorenylmethyloxycarbonyl)법, t-Boc(t-butyloxycarbonyl)법 등의 화학합성법에 의해, 본 발명의 펩티드를 합성할 수 있다. 또한, 시판되는 각종 펩티드 합성기를 이용하여, 본 발명의 펩티드를 합성할 수도 있다.
본 발명의 펩티드를 재조합 단백질로 생산하기 위하여는, 상기 펩티드를 암호화하는 염기서열을 가지는 DNA 또는 그의 변이체 또는 상동체를 입수해서 그를 알맞은 발현 시스템에 도입하여 제조할 수 있다.
바람직한 발현벡터로서는 숙주세포에 있어서 자립복제의 가능성이 있거나, 또는 숙주세포의 염색체내로 넣는 것이 가능하다면, 펩티드를 암호화하는 유전자를 발현할 수 있는 위치에 프로모터를 함유하는 것이 사용된다. 또한, 본 발명의 펩티드를 암호화하는 유전자를 가지는 형질전환체는 상기 발현벡터를 숙주로 도입하여 작제할 수 있다. 숙주는 세균, 효모, 동물세포, 곤충세포 어느 것이라도 무방하고, 숙주로의 발현벡터의 도입은 각각의 숙주에 따른 공지된 방법에 의해 행할 수 있다.
본 발명에 있어서 상술한 바와 같이 작제한 형질전환체를 배양하여, 배양물 내에 본 발명의 펩티드를 생성 축적시켜, 상기 배양물로부터 본 발명의 펩티드를 채취함으로써 재조합 펩티드를 단리할 수 있다.
형질전환체가 대장균 등의 원핵생물, 효모균 등의 진핵생물인 경우, 이들 미생물을 배양하는 배양기는 상기 미생물이 자화해서 얻은 탄소원, 질소원, 무기염류 등을 함유하고, 형질전환체의 배양을 효과적으로 행하는 배지라면 천연배지, 합성배지 등 모두가 가능하다. 또한, 배양조건은 상기 미생물의 배양에 통상적으로 이용할 수 있는 조건에서 행하면 좋다. 배양후, 형질전환체의 배양물로부터 본 발명의 펩티드를 단리정제하기 위해서는 통상의 펩티드의 단리, 정제법을 이용할 수 있다.
아울러, 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열에서 한 개 또는 두 개의 아미노산이 치환, 또는 부가된 아미노산 서열로 구성되는 펩티드는 서열번호 1 내지 3의 어느 하나에 기재된 아미노산 서열을 암호화하는 DNA서열의 정보를 기준으로 하여 당업자라면 적절히 제조 또는 입수할 수 있다. 즉, 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열에서 한 개 또는 두 개의 아미노산이 치환 또는 부가된 아미노산 서열로 구성되어, 세포상해성 T세포의 유도능력을 가지는 펩티드를 암호화하는 유전자는 화학합성, 유전공학기술 또는 돌연변이 유발 등의 당업자에게 알려져 있는 임의의 방법으로 작제할 수 있다. 예를 들면, 유전공학적 방법의 하나인 부위특이적 변이유도법은, 특정의 위치에 특정변이를 도입할 수 있는 방법이 유용하고, Molecular Cloning: A Laboratory Mannual, 2nd Ed., Cold Spring Harbor Laboratory, Cold Spring Harbor, NY., 1989, Current Protocols in Molecular Biology, Supplement 1-38, John Wiley & Sons(1987-1997) 등에 기재된 방법에 준하여 행할 수 있다.
상술한 본 발명의 펩티드는 이하의 실시예에도 개시된 바와 같이 암에 대한 면역을 유도할 수 있다. 따라서, 본 발명에 의하면 본 발명의 펩티드를 포함하는 암에 대한 면역 유도제가 제공된다.
본 발명의 암에 대한 면역유도제는 실험실 조건(in vitro) 또는 생체 조건(in vivo), 바람직하게는 실험실 조건에서 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포 집단을 유도할 수 있고, 이로써 암에 대한 면역을 부여할 수 있다.
(2) 본 발명의 항체
본 발명은 상술한 본 발명의 펩티드의 일부 또는 전부를 에피토프(항원)로서 인식하는 항체 및 상기 단백질 또는 펩티드를 이용하여 실험실 조건에서의 자격에 의해 유도된 세포상해성 T세포에도 관련된다. 일반적으로, 세포상해성 T세포가 항체보다도 강한 항종양 활성을 나타낸다.
본 발명의 항체는 폴리클로날 항체나 모노클로날 항체이어도 무방하고, 당업계의 공지된 방법에 의해 작제될 수 있다.
예를 들면, 폴리클로날 항체는 본 발명의 펩티드를 항원으로서 포유동물 또는 조류를 면역감작해서, 상기 포유동물 또는 조류로부터 혈액을 채취하여, 채취한 혈액으로부터 항체를 분리, 정제하여 얻을 수 있다. 예를 들면, 마우스, 햄스터, 모르모트, 닭, 랫트, 토끼, 개, 염소, 양, 소 등의 포유동물 또는 조류를 면역화할 수 있다. 면역감작 방법은 당업자에게 공지되어 있고, 항원을 7~30일간격으로 2~3회 투여할 수 있다. 투여량은 1회당, 예를 들면 항원 약 0.05~2mg정도로 할 수 있다. 투여경로는 이들에 특별히 한정되는 것은 아니나, 피하투여, 피내 투여, 복막강내 투여, 정맥내 투여, 근육내 투여 등을 적절히 선택할 수 있다. 또한, 항원은 적당한 완충액, 예를 들면 완전 프로인트 어쥬번트 또는 수산화 알루미늄 등의 통상적으로 사용되는 어쥬번트를 함유하는 적절한 완충액에 용해시켜 사용할 수 있다.
면역감작한 포유동물 또는 조류를 일정기간 사육한 후, 항체가가 상승하면, 예를 들면 100μg 내지 1000μg의 항원을 이용하여 추가면역을 행할 수 있다. 최후의 투여로부터 1~2개월 후에 면역감작한 포유동물 또는 조류로부터 혈액을 채취해서 상기 용액을, 예를 들면 원심분리, 황산암모늄 또는 폴리에틸렌글리콜을 이용한 침전, 겔여과 크로마토그래피, 이온교환 크로마토그래피, 친화성 크로마토그래피 등의 크로마토그래피 등의 통상적인 방법을 이용하여 분리, 정제함으로써, 폴리클로날 항 혈청으로서 본 발명의 펩티드를 인식하는 폴리크로날 항체를 얻을 수 있다.
한편, 모노클로날 항체는 하이브리도마를 조제해서 얻을 수 있다. 예를 들면, 항체생산세포와 미엘로마 세포주와의 세포융합에 의해 하이브리도마를 얻을 수 있다. 본 발명의 모노클로날 항체를 생산하는 하이브리도마는 아래와 같은 세포 융합법에 의해 얻을 수 있다.
항체생산세포로서는 면역된 동물로부터의 비세포, 림프절세포, B림프구 등을 사용한다. 항원으로서는 본 발명의 펩티드를 사용한다. 면역동물로서는 마우스, 랫트 등을 사용할 수 있으며, 이들 동물로의 항원투여는 통상적인 방법에 따라 행한다. 예를 들면, 완전 프로언트 어쥬번트, 불완전 프로언트 어쥬번트 등의 어쥬번트와 항원인 본 발명의 펩티드와의 현탁액 혹은 유화액을 동물의 정맥, 피하, 피내, 복강내 등에 수차례 투여함으로써 동물을 면역화한다. 면역화한 동물로부터 항체생산 세포로서, 예를 들면 비세포를 취득해서 이것을 미엘로마 세포를 공지된 방법에 의해 융합시켜 하이브리도마를 작제할 수 있다(참조: G. Kohler et al., Nature, 256 495(1975)).
세포융합에 사용하는 미엘로마 세포주로서는, 예를 들면, 마우스 같은 경우는 P3X63Ag8, P3U1주, Sp2/0주 등을 들 수 있다. 세포융합을 행할 즈음해서는 폴리에틸렌글리콜, 센다이 바이러스 등의 융합촉진제를 이용하여, 세포융합후의 하이브리도마의 선택에는 히포크산틴·아미노프테린·티미딘(hypoxanthine-aminopterin-thymidine, HAT) 배지를 통상적인 방법에 따라 사용한다. 세포융합에 의해 얻어지는 하이브리도마는 한계희석법 등에 의해 클로닝한다. 또한, 필요에 따라서는 본 발명의 펩티드를 이용한 효소면역 측정법에 의해 스크리닝함으로써, 본 발명의 펩티드를 특이적으로 인식하는 모노크로날 항체를 생산하는 세포주를 얻을 수 있다.
이렇게 해서 수득한 하이브리도마를 목적으로 하는 모노크로날 항체를 제조할 때는 통상의 세포배양법과 복수형성법에 의해 상기 하이브리도마를 배양하고, 배양상청 또는 복수로부터 상기 모노클로날 항체를 정제할 수 있다. 배양상청 혹은 복수로부터의 모노클로날 항체의 정제는 통상적인 방법에 의해 행할 수 있다. 예를 들면, 황산암모늄 분획, 겔여과, 이온교환 크로마토그래피, 친화성 크로마토그래피 등을 적절히 조합해서 사용할 수 있다.
또한, 상술한 항체의 단편도 본 발명의 범주에 속한다. 항체의 단편의 예로는 F(ab')2 단편, Fab' 단편 등을 들 수 있다.
(3) 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포집단
또한, 본 발명은 본 발명의 펩티드를 이용하여 실험실 조건에서 자격하여 유도된 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포집단에 관한 것이다. 예를 들면, 말초혈 림프구와 종양침윤 림프구를 본 발명의 펩티드를 이용하여서 실험실 조건에서 자격하면 종양반응 활성화 T세포가 유도되어, 이 활성화된 T세포는 양자면역요법에 유용하게 이용할 수 있다. 또한, 본 발명의 펩티드를 강력한 항원제시 세포인 수상세포에 실험실 조건 또는 생체조건에서 발현시켜, 그 항원발현 수상세포의 투여에 의해 면역유도를 행할 수 있다.
바람직하게는, 본 발명의 펩티드와 면역부활제를 이용하여 실험실 조건에서 자격하여, 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포집단을 유도할 수 있다. 이때, 사용하는 면역부활제의 예로서는 세포증식인자 또는 사이토카인 등을 들 수 있다.
이와 같이 해서 얻어진 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포집단을 체내에 이입함으로써, 종양을 억제할 수 있으며, 암의 예방 및 치료가 가능하다.
또한, 본 발명의 펩티드를 이용함으로써 상술한 대로 종양을 억제할 수 있는 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포집단을 만들 수 있다. 따라서, 본 발명에 의하면 본 발명의 펩티드를 포함하는 세포배양액이 제공된다. 이 세포배양액을 사용함으로써 종양을 억제할 수 있다. 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포집단을 작제할 수 있다. 게다가, 본 발명에 의하면, 상기 세포배양액 및 세포배양용기를 포함하는, 헬퍼 T세포, 세포상해성 T세포, 또는 이들을 포함하는 면역세포 집단을 작제하기 위한 세포배양 키트도 제공된다.
(4) 본 발명의 종양의 치료 및/ 또는 예방제(암백신)
본 발명의 펩티드는 암세포 특이적 세포상해성 T세포를 유도할 수 있기 때문에, 암의 치료, 예방제로 기대할 수 있다. 예를 들면, 본 발명의 펩티드를 암호화하는 유전자를 적절한 벡터에 넣고, 이 재조합 DNA로 형질전환된 BCG균의 세균, 또는 본 발명의 펩티드를 암호화하는 DNA를 게놈에 넣은 바시니아 바이러스 등의 바이러스는 인간암의 치료·예방용 생백신으로서 유효하게 이용할 수 있다. 또한, 암백신의 투여량 및 투여법은 통상의 종두와 BCG 백신과 같다.
즉, 본 발명의 펩티드를 암호화하는 DNA(그대로, 혹은 발현벡터에 넣은 플라스미드 DNA의 형태), 상기 DNA를 포함하는 재조합 바이러스 혹은 재조합 세균은 그대로 혹은 어쥬번트에 분산한 상태로 암백신으로 인간을 포함한 포유동물에 투여할 수 있다. 본 발명의 펩티드도 마찬가지로 어쥬번트로 분산한 상태로 암백신으로 투여할 수 있다.
본 발명에서 사용할 수 있는 어쥬번트로서는 프로언트의 불완전 어쥬번트, BCG, 트레할로스디마이클레이트(trehalose dimycolate, TDM), 리포다당(LPS), 묘반 어쥬번트, 실리카 어쥬번트가 있지만, 항체의 유도능력 등의 관계에서 프로언트 불완전 어쥬번트(IFA)를 사용하는 것이 바람직하다.
본 명세서에서 암의 종류는 특별히 한정되는 것은 아니고, 구체적인 예를 들면, 식도암, 유방암, 갑상선암, 대장암, 췌장암, 악성 흑색종(멜라노머), 악성 림프종, 골육종, 갈색세포종, 두경부암, 자궁암, 난소암, 뇌종양, 만성 골수성 백혈병, 급성 골수성 백혈병, 신장암, 전립선암, 폐암, 위암, 간암, 담낭암, 정소암, 갑상선암, 방광암 또는 육종 등이 있다.
본 발명의 펩티드는 T세포 펩티드로서 암세포 특이적인 세포상해성 T세포를 유도할 수 있기 때문에, 암의 예방·치료제로서 유용하다. 또한, 본 발명의 항체도 암항원인 GPC-3의 활성을 저해할 수 있다면, 암의 예방·치료제로서 유용하다. 실제의 사용법으로는 본 발명의 펩티드 또는 항체를 그대로, 또는 의약적으로 허용되는 담체 및/ 혹은 희석제와 같이 필요에 따라 아래의 보조제를 첨가하여 주사제로 투여할 수도 있으며, 분무 등의 방법으로 점막에서 경피흡수 등으로 투여할 수도 있다. 또한, 담체로서는 인간의 혈청 알부민, 희석제로서는 PBS, 증류수 등을 예로 들 수 있다.
투여량은 성인 1인당 본 발명의 펩티드 또는 항체를 예를 들면 1회당 0.01mg~100mg의 범위가 되도록 투여할 수 있지만, 이 범위에서 한정되는 것은 아니다. 제제의 형태도 특별히 한정되지 않고, 동결건조한 것이나, 당 등의 부형제를 첨가한 과립형태도 가능하다.
본 발명의 약제로 첨가가능한 종양반응성 T세포 유도활성을 높이기 위한 보조제로서는 뮤라밀디펩티드(muramyl dipeptide, MDP) 외에 BCG균 등의 균체성분, [Nature, vol. 344, p873(1990)]에 기재된 ISCOM, [J. Immunol. vol. 148, p1438(1992)]에 기재된 사포닌계의 QS-21, 리포좀, 수산화알루미늄 등이 있다. 또한, 렌티난, 시조피란, 피시바닐 등의 면역부활제를 보조제로서 이용가능하다. 또한, IL-2, IL-4, IL-12, IL-1, IL-6, TNF 등의 T세포의 증식, 분화를 증강시키는 사이토카인 및 NKT세포를 활성화하는 α-갈락토실세라미드와 Toll-유사 수용체에 결합해서 자연면역계를 활성화하는 CpG, 리포다당(LPS) 등도 보조제로서 이용가능하다.
또한, 환자로부터 채취한 세포, 또는 일부의 HLA 대립유전자를 공유하는 타인의(allo) 세포에 시험관내에서 상기 항원 펩티드를 첨가하여 항원제시시킨 후, 환자의 혈관내에 투여하여 환자의 체내에서 효과적으로 세포상해성 T세포를 유도할 수 있다. 또한, 환자의 말초혈 림프구에 당해 펩티드를 첨가하고 시험관내에서 배양함으로써, 시험관내에서 세포상해성 T세포를 유도한 후에 환자의 혈관내에 재주입할 수도 있다. 이와 같은 세포이입에 의한 치료는 이미 암치료법으로서 실시되고 있고, 당 업자들에게는 널리 알려진 방법이다.
본 발명의 펩티드를 관내에 주입함으로써 세포상해성 T세포를 유도활성화하고, 그 결과, 항종양효과를 기대할 수 있다. 또한, 림프구를 실험실적 조건에서 본 발명의 펩티드로 자격하면 활성화 T세포가 유도되어, 이 활성화된 T세포를 환부에 주입함으로써 양자 면역치료에 유효하게 이용할 수 있다.
도 1은 GPC-3 펩티드를 특이적으로 인식해서 활성화된 세포상해성 T세포가 생산한 IFN-γ ELISPOT 해석에 의해 검출한 대표적인 결과를 나타낸다. 간세포 암환자의 말초혈 중의 CD8 양성세포를 GPC-3 155-163 펩티드를 부하한 CD14 양성 단구에 유래하는 수상세포에서 자격해서 유도된 세포상해성 T세포는, GPC-3를 발현 하도록 유전자 도입된 SK-Hep-1/GPC-3 세포를 자격세포로 한 경우(그림 좌)는, HLA-A2양성에서 GPC-3를 발현하지 않는 친주의 SK-Hep-1세포를 자격세포로 한 경우(그림 우)와 비교하여 스폿의 수도 스폿의 면적총합도 유의적으로 컸다. 따라서, GPC-3 155-163 펩티드는 GPC-3 특이적 세포상해성 T세포를 유도할 수 있는 펩티드로 판단하였다.
도 2는 ELISPOT 해석과 세포상해성 실험의 결과를 나타낸다. HLA-A2 양성의 간세포 암환자의 말초혈로부터 CD8 양성 T세포를 선별하여, 각 GPC-3 발현세포에 대해 특이적으로 반응해서 IFN-γ를 생산하는지의 여부를 에리스폿어세이(ELISPOT assay)로 검토하고, GPC-3 발현세포를 특이적으로 상해하는지 여부를 세포상해성 실험으로 검토하였다. 표적세포로서 HLA-A2양성으로 GPC-3를 발현하지 않는 친주의 SK-Hep-1/GPC-3세포와 GPC-3를 발현하도록 유전자도입된 SK-Hep-1/GPC-3세포를 이용하였다. 그 결과, GPC-3 44-52, 144-152, 155-163 펩티드로 유도된 세포상해성 T세포는 SK-Hep-1/GPC-3 세포를 GPC-3 특이적으로 식별해서 IFN-γ를 생산하는 한편, 높은 세포상해 활성을 나타내지만 GPC-3 169-177 펩티드로 유도된 세포상해성 T세포에서는 GPC-3 특이적 세포상해성 T세포활성을 나타내지 않았다. 이상의 결과로부터, GPC-3 44-52, 144-152, 155-163 펩티드는 GPC-3 특이적 세포상해성 T세포를 유도할 수 있는 에피토프 펩티드로 판정하였다.
이하의 실시예에 의해 본 발명을 다시 설명하지만, 본 발명은 이들 실시예에 한정되는 것은 아니다.
실시예
1
(1) HLA-A2에 결합성을 나타내는 GPC-3 펩티드의 선택
인간의 GPC-3 아미노산 서열을 BIMAS 시스템에 의해 검색하여, HLA-A2로 추정된 결합력(binding affinity)이 20이상인 것을 4종류 선택하였다.

실시예 2: HLA-A2 결합성 GPC-3 펩티드에 의한 세포상해성 T세포의 유도
(1) 채혈
구마모토대학 의학부 소화기외과 및 국립 암센터의 동병원에서 치료 중인 HLA-A2가 양성의 간세포암 환자로부터 사전 동의를 얻은 후, 혈액시료 30ml를 얻어 이미 보고된 방법(참조: Nakatsura, T. 등, Eur. J. Immunol. 32, 826-836(2002))으로, Ficoll-Conray 밀도구배 원심분리법을 이용하여 말초혈 단핵세포를 단리하였다.
(2) 말초혈 단핵세포로부터 CD8 양성세포와 CD14 양성세포의 분리 및 세포상해성 T세포의 유도
단리된 말초혈 단핵세포로부터 이미 보고된 방법(참조: Monji, M. 등, Clin Cancer Res., 10, 6047-6057, 2004)를 이용하여 세포상해성 T세포를 유도하였다. 우선, MACS를 이용하여 말초혈 단핵세포 중의 CD8 양성세포와 CD14 양성세포를 분리하고, CD14 양성세포를 GM-CSF(100ng/ml)과 IL-4(20ng/ml) 존재하에 5일간 배양하여 수상세포를 분화유도한 후, TNF-α(20ng/ml)을 첨가해서 성숙시키고, 7일마다 각 GPC-3 펩티드를 첨가(10μM)하고 CD8 양성세포와 함께 배양하였다. 이 자기 CD14 양성세포 유래의 수상세포에 의한 항원자격을 1주일마다 3~4회 반복하여, 펩티드 특이적 세포상해성 T세포를 유도하였다. 유도도중 배지는 2일마다 1/2씩 교환하고, IL-2를 10U/ml의 농도로 첨가하였다.
(3) ELISPOT법에 의한 GPC-3 특이적 세포상해성 T세포활성의 검토
이들 유도된 세포상해성 T세포 중에서 확실히 GPC-3에 특이적으로 반응해서 IFN-γ를 생산하는 것이 존재하는가의 여부를 ELISPOT법에서 검출하였다. IFN-γ의 검출은 ELISPOT Human IFN-γ ELISPOT set(BD사)를 이용하였다. 자격세포(표적)에 대해 세포상해성 T세포(효과기)가 반응하여 IFN-γ를 생산하면, 각각이 붉은 스폿으로 검출된다. 표적세포로서는 HLA-A2양성에서 GPC-3를 발현하지 않는 친주의 SK-Hep-1세포와 GPC-3를 발현하도록 유전자도입된 SK-Hep-1/GPC-3세포를 이용하였다. 우선, 항 인간 IFN-γ항체를 ELISPOT 플레이트(BD Bioscience사)에 18시간 동안 코팅한 다음, 10% FCS/RPMI로 2시간 블로킹하였다. 효과기 세포(100μL/well)와 표적세포(100μL/well)를 혼합해서 37℃로 22시간 배양하였다. 효과기/표적비율(E/T비)은 5:1로 실험하였다. 그 후, 플레이트를 멸균수로 세정하고, 비오틴화 항 인간 IFN-γ 항체와 2시간, 이어 스트렙트아비딘-HRP와 1시간 반응시켜 기질용액에서 IFN-γ 양성 스폿을 검출하였다. 스폿의 카운트는 MINERVA TECH사의 자동해석 소프트를 이용하였다. 그 결과, GPC-3 44-52, 144-152, 155-163 펩티드로 유도된 세포상해성 T세포에서 GPC-3 특이적 세포상해성 T세포활성에 차이를 보였으나, GPC-3 169-177 펩티드로 유도된 세포상해성 T세포에서는 GPC-3 특이적 세포상해성 T세포활성을 나타내지 않았다(참조: 도 1 및 도 2). 대표적인 GPC-3 155-163 펩티드로 유도된 세포상해성 T세포의 해석결과를 도 1에 나타내었다.
(4) 세포상해성 실험에 의한 세포상해성 T세포의 세포상해활성의 검토
HLA-A2양성에서 GPC-3를 발현하지 않는 친주의 SK-Hep-1세포와, GPC-3를 발현하도록 유전자도입된 SK-Hep-1/GPC-3세포를 자격세포로 하여 유도된 세포상해성 T세포의 세포상해 활성을 세포상해성 실험에 의해 검토하였다. 세포상해성 T세포의 세포상해 활성은 테라스캔 VP에 의한 세포상해성 실험에 의해 평가하였다. 우선, 표적세포를 37℃로 30분간 칼세인 AM 염색액에서 형광표지하였다. 이들 세포를 Coster 96혈 하프·에리아플레이트 위에서 세포상해성 T세포와 같이 배양하여, 경시적인 형광발색 세포를 검출함으로써 세포상해 정도를 측정하였다. 해석은 MINERVA TECH사의 형광법에 의한 세포상해 실험계산처리 소프트 CalCt-96로 행하였다. E/T 비율은 20:1로 실험하였다. 그 결과, GPC-3 44-52, 144-152, 155-163 펩티드로 유도된 세포상해성 T세포에서 GPC-3 특이적인 세포상해성을 나타내었으나, GPC-3 169-177펩티드로 유도된 세포상해성 T세포에서는 GPC-3 특이적인 세포상해 활성을 나타내지 않았다(참조: 도 2).
HLA-A24에 의해 제시된 GPC-3 펩티드를 표적으로 하는 암 면역치료는 마우스 동물실험에서도 유효성이 인정되었으나, HLA-A24에 의해 세포상해성 T세포에 제시된 펩티드만으로는 일본인의 60%밖에 백신투여의 대상자가 되지 않는다. 금번 HLA-A2에 의해 세포상해성 T세포에 제시된 펩티드를 동정함으로써 양자를 합치면 일본인의 85%를 백신투여 대상자가 가능하다. HLA-A24에 의해 세포상해성 T세포에 제시된 펩티드를 이용한 탐색의료에서 유효성을 나타낸다면, 구미의 백인들에게도 임상적으로 응용될 가능성도 높아진다. 또한, 구미의 백인들에서 양성 환자의 빈도가 높은 HLA-A24에 의해 세포상해성 T세포에 제시된 펩티드를 동정함으로써 일본인의 간세포암 및 멜라노머 환자의 40%에 응용가능할 뿐 아니라, 많은 구미의 백 인들에게도 응용할 수 있다.

- 제1항에 기재된 펩티드를 포함하는, 종양의 치료 및/또는 예방제.

- 서열번호 1 내지 3의 어느 하나로 나타내는 아미노산 서열로 구성되고 세포상해성(killer) T세포의 유도능력을 가지는 펩티드를 암호화하는 유전자를 포함하는, 종양의 치료 및/또는 예방제.
