Abstract
Description
본 발명에서 발명자들은 포유동물의 종양, 바람직하게는 인간 종양, 그리고 가능하다면 신장 세포 암종에서 직접 나온 HLA I형 또는 II형 분자에 결합하는 펩티드를 분리 및 특징지울 수 있음을 증명하고 있다.
RCC 환자로부터 생성된 자가 수지상 세포(DCs)는 이들 IMA-ADF-001 중에서 MUC로부터 유래된 두 개의 TUMAP로 펄스처리되었다. IMA-ADF-001는 접종되지 않았다. 접종은 매 2주 마다 4차례 피하 투여(s.c.) 하였고, 종양 진전될 때까지 매월 반복하였다. 5번째 수지상 세포(DC) 투여 후에 환자들은 저용량 IL-2 (1Mio IE/m2) sc로 주당 3차례 투여 받았다. IFN-감마 ELISPOT을 이용하여 T 세포 전구물질 활성화가 모니터링되었다. 두 개의 접종된 펩티드에 대한 T 세포 유도 외에도, IMA-ADF-001 중에서 TUMAPs로 알려진 여러 다른 것에 대한 T 세포 활동도 검사되었다.
서열번호 1, 2, 3, 4, 5, 6, 7, 8, 9, 10을 갖는 펩티드를 30명의 환자에서 시험하였고, 이중 26명이 10주 간에 걸친 전체 접종 코스를 완료하였다. 높은 환자 비율인 74%가 백신에 포함된 펩티드 서열에 대해 특이적인 면역 반응을 보였다. 이 작은 I상 코호트에서 MHC I형(대립유전자 HLA-A*02)에 결합하는 9개의 펩티드(서열번호 2, 3, 4, 5, 6, 7, 8, 9)중 8개의 생체내 생물학적 기능은 하기 기술된 대로 실시된, 증폭된 ELISPOT 및 사량체(tetramer) 염색 측정 결과에 따라 이미 확인될 수 있었다. 다른 판독이 요구되는 서열번호 1 (IMA-MMP-001)을 갖는 펩티드는 환자 혈액에서 나온 PBMC의 제한된 이용가능성으로 인해 시험될 수 없었다.
"증폭된 ELISPOT" 분석법에서는, 세포들을 각기 다른 시점에서 모으고, 계수하고, 특정 세포가 분할할 수 있도록 약 2주동안 항원으로 미리 자극하였다. 그런 다음, 세포를 배양물로부터 수확하고, 다시 계산한 후, 항원으로 하루 동안 재자극하였을 때의 IFN-γ 점 생산에 대해 상기와 같이 측정힌다. 이런 조건 하에서 활성화된 세포들은 효소 연결 샌드위치 항체 방법으로 감지되는 IFN-γ를 분비한다. 점들은 색상 형성 반응에 의해 시각화되고 높은 해상도의 자동 디지털 카메라(여기서는 "ELISPOT 리더")로 계산된다. 시료 중의 활성화된 항원 특이적 T 림프구의 진짜 빈도와 연관된 점의 수는 ELISPOT 리더에서 획득한 이미지로부터 소프트웨어 알고리즘에 의해 결정된다. 이 분석법은 다양한 제 3자의 임상 시험에서 접종된 종양 항원에 대한 T 세포 반응을 검출하는데 주로 사용되었다. T 세포의 "생체 외 프라이밍"에 기인한 위양성(false-positive) 자료의 가능성은 이 분석법에서 다양한 대조군을 이용함으로써 배제된다. 하단의 사량체(tetramer) 분석법에 비해, 이 분석법은 추가의 기능 정보, 즉 IFN-γ 시토킨 방출을 제공한다.
Claims (36)

- 제 1 항에 있어서,상기 펩티드가 9 내지 30개 아미노산의 전체 길이를 갖는 항종양 면역 반응을 촉진하는 펩티드.
- 제 1 항에 있어서,인간의 주요 조직 적합 복합체(major histocompatibility complex: MHC) I형 또는 II형의 분자에 결합하는 능력을 갖는 항종양 면역 반응을 촉진하는 펩티드.
- 제 4 항에 있어서,인간의 주요 조직 적합 복합체(MHC) II형의 하나 이상의 추가 분자에 결합하는 능력을 갖는 항종양 면역 반응을 촉진하는 펩티드.
