물 리터당 기체의 리터로 나타낸 물에 대한 기체의 용해도를 그 기체 분자량의 제곱근으로 나눈 값이 0.003을 초과하지 않는 기체를 충전제로서 사용함으로써 초음파 에코그라피에 있어서, 콘트라스트제로 사용되는 기체-충전 미세소포에 대해 과압하에서의 붕괴 저항성을 현저히 향상시킬 수 있다.

제1도는 흡광도로써 나타낸 기포농도(기포수)와 기포현탁액에 적용된 압력의 관계 그라프이다.
본 발명은 수용액 담체에서 안정한 기체충전 미세소포(microversicle) 조성물 또는 분산체에 관한 것이다. 이들 분산체 액중에 기체를 균질적으로 분산시킬 필요성이 있는 대부분의 적용에 대해 일반적으로 유용하다.
이런 분산체의 가장 주목할만한 적용의 하나는 예컨대 초음파 에코그라피(ultrasonic echography) 및 기타 의학적 적용용으로 생명체내로 주사할 수 있다는 것이다.
본 발명은 또한 예컨대 압력-저항성 기체-충전 마이크로버블(microbubble). 마이크로캡슐(microcapsule) 및 마이크로발룬(microballoon)과 같은 이 제제에 연관된 재료를 포함하는 앞서 말한 조성물을 제조하는 방법에 관한 것이다.
액체에 현탁된 예컨대 마이크로버블 또는 마이크로발룬과 같은 공기나 기체의 미세소구(microbody) 또는 미소체(microglobule)(여기에서는 미세소포라고 정의함)는 매우 효과적인 초음파 반사체라는 것은 잘 알려져 있다.
본 명세서에서, "마이크로버블"이라는 용어는 분할된 형태로 공기나 기체를 그안에 도입함으로써 일반적으로 생기는 액체내 현탁액중에 있는 공기 또는 기체가 채워진 공동(hollow)의 구체 또는 소구를 특히 나타내며, 상기 액체는 바람직하게 기포의 안정성 및 표면특성을 제어하는 계면활성제 또는 텐시드(tenside : 계면활성제)를 또한 함유한다. "마이크로캡슐" 및 "마이크로발룬"의 용어는 바람직하게 유형의 경계 또는 피막, 즉 중합체막 벽을 갖는 공기 또는 기체를 충전한 물체를 나타낸다.
마이크로버블 및 마이크러발룬 양자 모두는 초음파 콘트라스트제(ultrasonic contrast agent)로서 유용하다, 예컨대 생명체의 혈류안으로, 담체액내의 공기-충전 마이크로버블 또는 마이크로발룬(0.5 내지 10μm의 범위로)의 현탁액을 주입한는 것은 초음파 에코그라피 영상을 매우 강화시키며, 따라서 내부기관의 가시화에 도움을 준다. 혈관 및 내부기관을 영상화하는 것은 예컨대 심장혈관 및 기타의 질병을 탐지하는 의학진단에 큰 도움을 줄 수 있다.
에코그라피용으로 적당한 주사가능한 담체액내의 마이크로버블 현탁액의 형성은 가압하에서 이 액체내로 용해 기체를 방출시키는 것, 또는 기체 생산물을 발생하는 화학반응, 또는 그안에 갇혀있거나 흡착된 기체 또는 공기를 함유한 기용성 또는 불용성 고체를 이 액체와 함께 혼합시키는 것에 의해 생산할 수 있다.
예컨대 US-A-4,446,442(Schering)에는 (a) 담체액(수용성) 내의 텐시드 용액 및 (b) 안정화제로 사용한 점성도 증강제 용액을 사용하여 무균처리된 주사가능한 담체액내에 기체 마이크로버블의 현탁액을 제조하는 일련의 난해한 기술이 개시되어 있다. 기포를 발생시키기 위해, 여기에 개시된 기술에는 (a),(b) 및 공기의 혼합물을 높은 점성도로 작은 구멍을 통해 통과시키는 것 : 또는 사용하기전에 생리적으로 수용할 수 있는 기체와 함께 (a)를 (b) 안으로 즉시 주입시키는 것 : 또는 (a)에는 산을 (b)에는 탄산염을 첨가시켜, 사용하기 바로전에 두 성분을 함께 혼합하여 산을 탄산염과 반응시켜 CO2기포를 발생시키는 것 : 또는 저장하에서 (a) 및 (b)의 혼합물에 과가압된(over-pressurized) 기체를 첨가시켜, 상기 기체가 이 혼합물을 주사에 사용할 때 마이크러버블안으로 방출되게 하는 것을 포함하고 있다.
EP-A-131,540(Schering)은 예컨대, 말토스, 덱스트로스, 락토스 또는 갈락토스 같은 당용액 또는 생리식염수 같은 안정화된 주사가능한 담체액을 갇힌 공기를 함유한 동일한 당의 고체 미세입자(0.1 내지 1μm 범위)와 혼합한 마이크로버블 현탁액을 제제하는 것을 개시하고 있다.
담체액내에 기포 현탁액을 발생시키기 위해, 액체 및 고체 두 성분을 수초동안, 무균상태하에서 함께 교반시키고, 일단 만들어진 후에는, 이 현탁액은 즉시 사용해야 한다. 즉, 에코그라피 측정을 위해 5-10분내 주사해야 한다 : 왜냐하면 이들은 소실되어 일정기간후에는 기포의 농도가 실행되기에는 너무 낮게되기 때문이다.
소실 문제점을 치유하기 위한 시도로써, 마이크로발룬 즉 유형의 벽을 갖는 미세소포가 개발되어 왔다. 상기 이전에는 마이크로버블은 단지 무형 또는 소실성 피막을 갖는 것이었다. 즉, 이것들은 이들의 표면장력이 계면활성제의 존재에 의해서 변형되는 액체 벽으로 단지 둘러싸여 있는 것인 반면, 마이크로발룬 또는 마이크로캡슐은 예컨대 명확한 기계적 강도를 갖는 중합체막과 같은 실질적인 물질로 만들어진 유형적인 피막을 갖는다. 비꾸어 말하면, 이들은 공기 또는 기체가 어느정도 단단히 캡슐화되는 물질로 된 미세소포이다.
예컨대 US-A-4,276,885(Tickner 등)는 초음파 영상을 향상시키기 위해 기체를 함유하는 표면막 마이크로캡슐의 사용을 개시하고 있으며, 이 막은 무독성이고 비항원성인 유기분자를 다수 포함하고 있다. 개시된 실시예에서, 이들 마이크로버블은 합착(coalescence) 저항성인 젤라틴 막을 가지며 이들의 바람직한 크기는 5-10㎛이다. 이들 마이크로버블의 막은 에코그라피 측정을 하는데 충분히 안정적이라고 한다.
젤라틴이 없는 공기-충전 마이크로발룬이 US-A-4,718,433(Feinstein)에 개시되어 있다. 이들 미세소포는 5% 혈청 알부민과 같은 단백질 용액을 음파분쇄(5 내지 30㎑) 시켜 제조하고, 그 직경은 2-20μm 범위, 주로 2-4μm 범위이다. 이 미세소포는 음파분쇄후에 예컨대 포름알데히드 또는 글루타르알데히드와 반응시키는 것같은 화학적 방법 또는 가열방법을 이용하여 막형성 단백질을 변형시킴으로써 안정화된다.
이 기술에 의해 획득되는 안정한 미세소포의 농도는 2-4μm 범위에서는 약 8×106/㎖이고, 4-5μm 범위에서는 약 106/㎖이며 5-6μm 범위에선 5×105/㎖보다 작은 것으로 알려져 있다. 이들 미세소퍼의 안정기간은 48시간 또는 그 이상인 것으로 되어 있어 정맥주사후 좋은 좌심 영상화를 이룰 수 있다. 예컨대 말초정맥안으로 주사된 때, 음파분쇄된 알부민 마이크로버블은 폐를 통과해 지나갈 수 있다.
이것이 심극조직뿐만 아니라 좌심실 공동의 초음파심장그라피 혼탁화를 야기한다.
최근에, 주사 초음파 에코그라피를 위한 보다 더 개량된 마이크로발륜이 EP-A-324,938(Widder)에 보고되었다. 이 기록에는 몇 달 또는 그 이상의 수명을 갖는 10μm보다 작은 고농축물(108/㎖이상인)공기-충전 단백질-경계 미세소포가 개시되어 있다.
이들 마이크로발룬의 수성 현탁액은 사람의 혈청 알부민과 같은 단백질의 열변성 단백질 용액의 초음파 공동화로 제조하며, 이런 조작은 또한 막-형성 단백질의 거품 형성을 어느정도 유도하고 가열에 의해 연이은 경화를 유도한다.
헤모글로빈 및 콜라겐과 같은 다른 단백질도 또한 이런 방법으로 필요에 맞게 할 수 있다. EP-A-324,938에 개시된 마이크로발룬 현탁액의 높은 저장안정성은 이들을 액체 담체상과 같은 것으로 시장에 내놓을 수 있도록 하며, 이것은 사용전 제제가 더 이상 필요하지 않기 때문에 큰 상업적인 이점을 갖는다.
마찬가지의 장점이 수성의 마이크로버블 현탁액 제제에 대해 최근에 발견되었다. 즉 물의 첨가로 장기간 지속하는 기포 현탁액이 만들어지는 저장-안정성 건조분말 조정물이 발견되었다.
이것은 PCT/EP 91/00620 출원에 개시되어 있으며, 여기에는 막-형성 지질로 구성된 리포솜을 동결-건조하여, 일정기간동안 공기나 기체에의 노출후에, 이 동결-건조 지질은 여기에다 수용액 담체를 단순히 첨가하는 것에 의해 장기간-지속되는 기포 현탁액을 생성시킨다는 것이 개시되어 있다.
수성의 마이크로버블 현탁액의 저장상태하에서의 안정성에 대해 많은 진보가 이루어 졌음에도 불구하고, 전구체이거나 최종 제제단계인 이들은 예컨대 환자의 현류에 주사한 후 연속되는 심장박동, 특히 좌심실에서 발생하는 그런 압력변화와 같은 과압에 현탁액이 노출될 소포의 지속성 문제는 지금까지 여전히 남아있다.
실제로, 본 발명자는 예컨대 마취된 토끼에서, 압력변화가 주사후 일정기간동안 가포수를 실질적으로 변화시킬 정도로 충분하지 않다는 것을 관찰했다.
반면, 개 및 사람환자에 있어서 공기, 메탄 또는 CO2와 같은 보통의 기체로 충전된 전형적인 마이크로버블 또는 마이크로발룬은 혈압 영향으로 인해 주사후 대체적으로 수초내에 완전히 붕괴되었다.
이런 관찰은 다른 사람에 의해서도 확인되었다.
즉 예컨대 S, GOTTLIEB 등은 J, Am Soc, of Echocardiography 3(1990) 238에서 음파분쇄 방법으로 제제된 교차-결합 알부민 마이크로발룬은 60토르의 과압에 둔 후에 모든 반향생성특성이 상실된다는 것을 기록하고 있다.
따라서 에코그라피 측정이 생체내에서 안정적이고 반복적으로 수행될 수 있도록 상기 문제를 해결하여 과압하에서의 마이크로버블 및 막경계 마이크로발룬 현탁액의 유용수명을 증가시키는 것이 중요하게 되었다.
공기나 기체를 함유하는 작은 구멍과 같은 다공성 구조를 갖는 판판한 미세소구로 구성된 것으로서 과압에 저항하는 다른 범주의 반향생성 영상 증강제가 제안되어 왔음을 여기에 언급하겠다.
이런 미세소구는 예컨대 WO-A-91/12823(DELTA BIOTECHNOLOGY), EP-A-327 490(SCHER ING) 및 EP-A-458 079(HOECHST)에 개시되어 있다. 판판한 다공성 미세소구의 단점은 캡슐화된 기체-충전 자유공간이 일반적으로 좋은 반향생성반응을 일으키기엔 너무 작고, 이들 소구는 적당한 탄력성이 결여되어 있다는 것이다.
그러므로 일반적으로 공동(hollow) 미세소포에 대한 희망사항은 여전히 남아 있으므로 해서 기포의 붕괴 문제에 대한 해결책을 연구했다.
이런 문제점은 이제 본 발명의 특허청구범위에 개요된 기준에 부합하는 기체 또는 기체 혼합물을 사용하여 해결된다.
요약하면, 반향생성 미세소포가 아래의 식에 부합하는 물리적 특성을 갖는 기체의 존재하에서 만들어져서 적어도 일부분이 이 기체로 각각 채워질 때, 이 미세소포는 주사후 반복성있는 에코그라피 측정을 얻기에 충분한 시간동안 60토르를 초과하는 압력에 대해 현저한 저항을 나타낸다는 것을 발견했다.
앞의 식에서. "s"는 "분센"계수를 나타낸 물에 대한 용해도를 나타낸다.
즉, 표준상태(1 bar 25℃), 및 주어진 기체의 1 atm의 부분압력하에서 단위 물 부피에 용해된 기체의 부피로서 나타낸 것이다(기체전문사전, Elsevier 1976 참고).
이런 조건 및 정의하에서 공기의 용해도는 0.0167이고, 공기의 평균 분자량(Mw)의 제곱근은 5.39이므로 상기 관계식은 다음과 같이 간단히 된다.

이후의 실시예에는, 많은 여러 가지 다른 기체들 및 이들의 혼합물로 채운 반향생성 마이크로버블 및 마이크로발룬의 테스트(표들 참고), 및 압력증가에 따른 이들의 상응하는 저항성을 생체내 및 생체외 양자에 대해서 개시하였다.
이들 표에서, 물용해도 계수들은 또한 앞서 언급한 "L'Air Liquide" Elsevier 출반판(1976)의 기체 전문사전으로부터 얻었다.
본 발명에 따른 기체를 함유한 수성 현탁액내의 미세소포는 에코그라피용 콘트라스트제로서 사용하기 위해 지금까지 알려진 대부분의 마이크로버블 및 마이크로발룬을 포함한다. 바람직한 마이크로발룬은 EP-A-324,938, PCT/EP 91/01706 및 EP-A-458,745에 개시된 것들이며 : 바람직한 마이크로버블은 PCT/EP 91/00620에 개시된 것들이다.
이들 마이크로버블은 박막화된 동결-건조 인지질 및 안정화제를 함유하는 건조분말(미세소포 전구체) 및 수용액으로부터 유용히 형성된다.
즉, 이 마이크로버블은 이 분말을 수용액 담체와 혼합하여 교반시킴으로써 발생된다.
EP-A-458,745의 마이크로발룬은 조절된 다공성의 탄력있는 계면침전중합체막(interfacially precipitated polymer membrane)을 갖는다.
이들은 일반적으로 수용액 안에서 중합체 용액을 미세방울(microdroplet)로 에멀션하여 얻어지며, 이 중합체는 이 용액으로부터 뒤에 침전이 야기되어져 소적(droplet)/액체 접촉면에 피막생성막을 형성하며, 이 방법은 먼저 액체-충전 미세소포의 형성을 유도하고, 이것의 액체 핵은 결국 기체에 의해 치환되어진다.
본 발명의 방법을 실행하기 위해, 즉 미세소포를 형성하거나 또는 미세소포를 충전시키기 위해, 수성 담체내의 미세소포 현탁액은 앞서 기술한 관계식에 부합하는 기체를 갖는 희망하는 반향생성 첨가제의 구성요소가 되며, (1) 임의의 적당한 기체의 존재하에서 임의의 적당한 통상적인 기술을 사용하여 적당한 출발물질로부터 미세소포를 제조하는 것, 그리고 (2) 본 발명에 따르는 새로운 기체(제2 기체)를 갖는 미세소포를 제조하기 위해 원래 사용한 기체 (제1 기체)를 대체시키는 것(가스-교환기술)으로 구성된 2단계 방법을 제1 실시태양으로서 사용할 수 있다.
그렇지 않으면, 제2 실시태양에 따라, 본 발명에 따른 새로운 기체의 분위기하에서 적당한 통상적인 방법을 사용하여 희망하는 현탁액을 직접적으로 제제할 수 있다.
2단계방법을 사용하는 경우, 먼저 최초의 기체를 소포로부터 제거하고(예컨대 흡인하에서 배기시키는 것에 의해)다음에 제2 기체를 배기산물과 접촉되도록 하여 대체 시키거나, 또는 선택적으로 제1 기체를 여전히 함유하고 있는 소포를 제2 기체가 소포로부터 제1 기체와 대체되는 조건하에서 제2 기체와 접속시킬 수 있다(가스 치환).
예컨대, 소포 현탁액 또는 바람직하게는 이것의 전구체(여기에서 전구체는 미세소포 피막을 만드는 재료를 의미하거나 또는 수용액 담체와 함께 교반할 때 이 액체내에서 마이크로버블의 형성을 일으키거나 발생시키는 물질을 의미할 수 있다)는 감압에 노출시켜 제거해야할 기체를 배기시키고, 다음에 주위압력을 치환하고자 하는 희망하는 기체로 복원시킬 수 있다. 이 단계는 원래의 기체가 새로운 기체로 완전히 대체되도록 한번 또는 그 이상 반복실시할 수 있다.
이 실시태양은 예컨대 담체액의 일정양과 혼합할 때 반향생성 첨갑물의 기포를 재생 또는 발전시키는 건조분말과 같은 건조상태로 저장하는 전구체 제제물에 대해 특히 적합하다. 그러므로, 미이크로버블이 예컨대 안정화제를 첨가한 탈수된 친액성 리포솜의 분말과 같은 건조 박막 인지질 및 수용액상으로부터 형성되어질 수 있는 하나의 바람직한 경우에 있어서(여기에서 분말은 수용액 담체상에서 교반에 의해 후에 분산되어질 수 있는 것이다)이 건조분말은 본 발명에 따라 선택된 기체의 분위기하에 저장하는 것이 유익하다.
이런 종류의 제제물은 이와 같은 조건하에서 무한히 보전되며, 주사전에 무균의 물에 분산시키면 진단하고자 하는 어느 때라도 사용할 수가 있다.
다른 방법으로, 특히 가스교환이 액체담체상내의 미세소포의 현탁액에 적용되는 경우에, 대체(부분적 또는 완전한 대체)가 원하는 목적에 충분할 때까지, 후자(담체액)를 제2 기체로 플러시 (flush)시킨다.
플러싱(flushing)은 기체 파이프로 기포를 발생시키거나, 또는 어떤 경우에는 새로운 기체의 흐름(계속적이거나 또는 불연속적인 흐름)으로 부드럽게 교반하면서 소포를 함유한 액체표면을 단순히 스위프(sweep)시킴으로써 실행할 수 있다.
이 경우에 있어서, 현탁액을 담고 있는 플라스크안에 대체 기체를 단한번에 첨가시켜 잠시동안 이런 상태를 유지시킬 수 있고, 또는 갱신(기체 교환)의 정도가 대략적으로 완전히 달성되어지도록 확실시 하기 위해 한번 또는 그 이상 반복할 수 있다.
선택적으로, 앞서 언급한 제2 실시태양에 있어서, 상기 선행기술의 통상적인 방법에 따라 조작하고 선행기술에 인용된 것과 같은 통상적인 전구체(출발물질)를 가지고 시작하되, 다만 공기, 질소, CO2등과 같은 일반적으로 인용되는 선행기술의 기체대신에 본 발명에 따른 바람직한 기체 또는 기체의 혼합물의 존재하에서 시작함으로써 방향생성 첨가제 현탁액의 완전한 제제를 실행할 수 있을 것이다.
일반적으로, 미세소포를 제제하기 위한 하나의 제1 형의 기체, 그리고 이어서 상기 미세소포에 따른 반향생성 특성을 부여하도록 의도된 제2 형의 기체로 원래의 기체를 대체하는 것을 포함하는 제제방법은 다음과 같은 잇점이 있다는 것이 주목되어야한다.
즉, 뒤에 있는 실시예의 결과로부터 명확히 알 수 있듯이, 미세소포, 특히 중합체 피막으로 되어 있는 마이크로발룬을 제조하기 위해 사용된 기체의 성질은 상기 미세소포의 일반적인 크기(즉 평균 직경)에 대해 명백한 영향을 미친다 : 예컨대, 엄밀히 규정된 조건의 공기하에서 제제되는 마이크로발룬의 크기는 예컨대, 좌·우심실의 에코그라피에 적당한 범위인 1 내지 10μm와 같이 필요로 하는 범위로 정확하게 조절할 수가 있다. 이것은 다른 기체, 특히 본 발명의 요건에 부합하는 기체에서는 그렇게 쉽지 않다 : 그러므로 주어진 크기 범위내로 미세소포를 얻고자 하나 직접적인 제제가 불가능하거나 또는 매우 어려운 성질의 기체로 충전하고자 하는 경우, 두단계 제제방법 즉, 보다 정확한 직경 및 기포수의 통제가 가능한 기체로 미세소포를 일단 먼저 제제하고, 다음에 가스교환에 의해 제2 기체로 제1 기체를 대체시키는 방법을 따르는 것이 훨씬 유용할 것이다.
뒤에 기술한 실험부분(실시예)에 있어서, 물 또는 다른 수용액내에 현탁시킨 기체-충전 미세소포는 주변압력 이상의 압력조건하에 두었다. 과압이 특정치[이것은 일반적으로 온도, 압축속도, 담체액의 성질 및 담체액의 용해기체 함량 (이들 변수의 상대적 중요성은 뒤에 상세히 기술한 것임), 기체 충전제의 특성, 반향생성물질의 형태 등과 같은 미세소구변수 및 작업조건 세트의 전형임]에 이르렀을 때, 미세소포는 붕괴되기 시작했고, 발생했던 음향 반사체 효과가 완전히 사라질 때까지 압력을 더 증가시킴에 따라서 기포의 수는 점차적으로 감소한다는 것을 알았다.
이런 현상은 이것이 흡광도에 있어서도 상응하는 변화가 병행되어지기 때문에 즉, 매질의 투명도는 기포가 점진적으로 붕괴됨에 따라서 증가하기 때문에, 시각적으로 더 잘 추적할 수 있었다[비탁계 측정(nephelometric measurement)]
이와 같은 이유로, 양(+)의 과수압을 단계적으로 증가하게 적용시키면서, 미세소포의 수용성 현탁액(또는 이것의 적당한 희석액)을 25℃(표준조건)로 유지한 분광광도계셀에 넣고 600 또는 700nm에서 연속적으로 흡광도를 측정했다.
이 압력은 누출되지 않도록 봉인하여 분광광도계 셀에 연결한 높이 가변성 액체 컬럼이 있는 연동펌프(GILSON's Mini-puls)로 발생시켰다.
이 압력은 토르(torr) 단위로 눈금된 수은압력계로 측정했다.
시간에 대한 압축속도는 펌프의 속도(rpm)와 선형상관관계가 있음을 알았다.
앞에서 언급한 범위에서의 흡광도는 담체액내의 미세소포 농도에 비례한다는 것이 발견되었다.
제1도는 앞서 언급한 범위에서의 흡광도로써 나타낸 기포농도(기포수)와 기포 현탁액에 적용된 압력과의 관계 그라프이다.
그라프 작성을 위한 데이터는 실시예 4에 기록된 실험으로부터 얻은 것이다.
제1도는 압력에 대한 흡광도의 변화를 S자형 곡선으로 나타낸 것이다.
특정압력값까지, 이 곡선은 거의 편평하며 이것은 기포가 안정하다는 것을 나타낸다. 다음에, 상대적으로 빠른 흡광도의 강하가 발생하며, 이것은 어떤 압력증가가 기포수에 대해 보다 극적인 영향을 주는 상대적으로 좁은 구역의 임계구역이 존재한다는 것을 나타낸다. 모든 미세소포가 사라지면, 곡선은 다시 편평하게 된다.
이 곡선의 임계점은 흡광도 값의 최고점과 최저점 사이의 중앙에서 선정된다.
즉, 최대-기포(ODmax)와 무-기포(OD min) 측정값의 중간값이며, 이것은 실제적으로 최초에 존재하던 기포의 약 50%가 사라지는 곳에 부합되는 점이며, 즉, 가압된 현탁액의 투명도가 최대인 높이(기선)에 관계되어 그라프에 정해지는 최초 흡광도 값의 약 절반이 되는 흡광도 값이 있는 곳이다.
또한 곡선의 기울기가 최대가 되는 곳의 접선에 있는 점이 임계압력 PC로 정의된다. 주어진 기체에 대한 PC값은 앞서 언급한 변수들에 의존할 뿐만 아니라 또한 담체액에 이미 용해되어 있는 기체(또는 기체들)의 실제 농도에 특히 의존한다는 것을 발견했다. 즉, 기체의 농도가 높을수록 임계압력도 높아진다.
따라서, 이와 같은 관점에서, 소포를 채운 기체와 동일한 기체 또는 다른 기체인 가용성 기체로 담체상을 포화시키는 것에 의해 미세소포의 압력에 대한 붕괴 저장성을 증가시킬 수 있다. 예로서, 공기-충전 미세소포는 담체액으로서 CO2포화용액을 사용함으로써 과압(>120토르)에 대해 매우 저항성있게 만들 수 있었다.
그러나 불행히도, 일단 콘트라스트제가 환자의 현류안으로 주입되면 (물론 콘트라스트제의 기체 함량은 외부에서 조정된다). 주입된 샘플내에 원래 용해된 기체의 효과가 무시되어질 정도로 혈류내에서 희석되기 때문에, 이런 발견은 진단분야에 있어서 한정된 가치만을 갖는다.
미세소구 충전제로서의 여러 가지 기체의 성능을 재현성있게 비교하기 위한 또다른 용이하게 접근가능한 변수는 기포의 수 (흡광도로서 나타낸)가 원래 기표수의 75% 및 25%와 같게 되는 압력값으로 한정한 압력간격(△P)의 폭이다.
놀랍게도, 압력차 DP=P25-P75가 약 25-30 토르값을 초과하는 기체들에 대해서, 혈압의 기체-충전 미세소포에 대한 제거효과는 최소화된다는 것을 발견했다.
즉, 기포수의 실질적인 감소는 에코그라피 측정의 반복성, 정확성 및 유의성을 손상하지 않을 정도로 충분히 느리다.
게다가, PC 및 △P의 값은 또한 제1도에 도시한 테스트 실험에 있어서의 압력 증가속도, 즉 압력증가속도의 차이(예컨대, 수십 내지 수백에 이르는 Torr/min의 범위)에 의존한다는 것을 발견했으며, 속도가 높을수록 PC 및 △P의 값은 커진다.
이런 근거로, 표준온도상태하에서 실행된 비교실험은 또한 상수증가속도 100토르/분에서 실행했다. 그러나 PC 및 △P값의 크기에 대한 압력증가속도의 이런 효과는 매우 높은 속도에 대해서는 편평해진다는 것에 주의해야 한다.
즉, 예컨대 수백의 토르/분 속도하에서 측정된 값은 심장박동에 의해 정해지는 조건하에서 측정된 값과 의미적으로 다르지 않다.
비록 왜 어떤 기체는 앞서 언급한 특성에 따르고 반면 다른 기체는 따르지 않는지의 정확한 이유는 완전히 밝혀지지 않지만, 여기에는 분자량 및 물용해도, 용해역학 및 기타 변수뿐만 아니라 연류된 어떤 관계가 존재할 가능성이 있음은 명백하다.
그러나, 기체의 적정성 여부는 앞서 언급한 기준에 따라 용이하게 결정할 수 있기 때문에 본 발명의 실행을 위해 이를 변수를 알 필요는 없다.
본 발명에 특히 적합한 기체 종류는, 예컨대 프레온과 같은 할로겐화된 탄화수소 및 SF6, SeF6와 같은 안정한 플루오르화된 칼코게니드 등이다.
본 발명에 따른 미세소포의 담체로 사용한 액체의 기체포화정도는 압력의 변화 하에서의 소포 안정성에 대해 중요한 것임을 위에서 언급했다.
본 발명의 반향생성 현탁액을 제조하기 위해 미세소포가 분산된 담체액이 기체와 평행상태로 포화될 때, 바람직하게는 미세소포를 채운 기체와 같은 기체로 평형상태로 포화될 때, 압력 변화하에서 미세소포의 붕괴에 대한 저항성은 특히 증가된다.
그러므로, 콘트라스트제로서 사용하기 위한 제품이 사용전에 담체액과 단지 혼합하여 사용하도록 건조물로 판매되는 경우 (예컨대 앞서 인용한 PCT/EP 91/00620에 개시된 제품을 참조), 기체가 포화된 수용성 담체를 분산을 위해 사용하는 것이 매우 유용하다. 선택적으로, 즉시 사용할 수 있는 미세소포 현탁액을 에코그라피용 콘트라스트제로 판매하는 경우, 제제의 담체액으로서 기체가 포화된 수용액을 사용하는 것이 이로울 것이다. 즉, 이 경우 현탁액의 저장 수명은 상당히 증가될 것이며, 이 제품은 장기간동안, 예컨대 수주에서 수달, 특별한 경우에는 1년이상동안 실질적으로 변하지 않고 유지될 수 있다(즉 실질적인 기포수의 변화없이 유지될 수 있다).
기체로 액체를 포화시키는 것은 실온에서 일정기간동안 기체를 단순히 액체안으로 기포발생시키는 것에 의해 손쉽게 실행할 수 있다.
[실시예 1]
공기 또는 여러 가지 기체로 충전한 알부민 미세소포를 스위스 베른에 있는 적십자 기구의 수혈국으로부터 구입한 5%의 사람 형청 알부민(HSA)으로 채운 10㎖로 눈금 되어진 주입기를 사용하여, EP-A-324 938에 기술된 방법에 따라 제제했다.
초음파분쇄기 탐침(Branson Ultrasonic Corp로부터 구입한 Sonifier Model 250)을 주입기의 4㎖ 표시까지 용액안으로 내려 넣어서 25초동안 초음파분쇄를 실행했다(에너지는 8로 맞추었다).
다음에 초음파분쇄기 탐침을 약 6㎖로 표시된 용액 높이로 올려서 초음파분쇄를 40초동안 파동방식(pulse mode) (주기는 0.3)으로 재실시했다.
4℃에 밤새동안 유지시킨 후에, 대부분의 미세소포를 함유하고 있는 상층은 부력에 의해 형성되었고, 사용하지 않는 알부민 부스러기인 변성단백질 및 기타 불용성 물질을 함유하는 바닥층은 폐기했다. 새로운 알부민 용액에 미세소포를 재현탁시킨 후에 이 혼합물을 다시 상온에 유지시키고 마지막으로 상층을 모았다.
전술한 일련의 실행을 대기 분위기하에서 수행했을 때, 공기가 충전된 마이크로발룬이 얻어졌다. 다른 기체들로 채워진 마이크로발룬을 얻기 위해서, 먼저 알부민 용액을 새로운 기체로 씻어내고, 다음에 전술한 일련의 조작을 용액의 표면상으로 이 기체의 흐름을 흘러보내는 조건하에서 실시했다.
다음에 이 조작의 종료시에, 이 현탁액을 밀봉하기에 앞서 원하는 기체로 철저히 씻어낸 유리병에 담았다.
여러 기체들로 채운 마이크로발룬의 여러 가지 현탁액을 공기와 평행상태로 포화된 증류수로 1 : 10으로 희석시켰고, 다음에 앞서 기술한 바와 같이 이것을 광학셀에 넣어서 현탁액에 가하는 압력을 안정적으로 증가하면서 흡광도를 측정했다.
측정하는동안, 현탁액의 온도는 25℃로 유지했다.
이 결과를 아래의 표1에 나타내었으며, 원래의 기포수 및 기포의 평균크기(체적의 평균직경) 뿐만아니라 주어진 기체의 물용해도 및 분자량과 같은 기체의 특징적인 변수, 명치, 화학식으로 규정된 일련의 기체들에 대해, 임계압력 PC값을 나타내었다.
표1의 결과로부터, 임계압력 PC는 보다 낮은 용해도 및 보다 큰 분자량을 갖는 기체에 있어서 증가됨을 볼 수 있다.
그러므로 이런 기체들로 채워진 미세소포가 생체내에서 보다 오래 지속되는 반향생성신호를 제공할 것이라는 것을 기대할 수 있다.
또한 평균기포크기는 일반적으로 기체용해도와 함께 증가한다는 것을 볼 수 있다.
[실시예 2]
실시예 1에서 제제된 약간의 마이크로발룬 현탁액 분취량(1㎖)을 생체내 반향생성을 테스트하기 위해 실험토끼의 경정맥(jugular vein)에 주사했다.
좌·우심실의 영상화는 Acuson 128-XP5 에코그라피장치 및 7.5㎒ 변환기를 사용하여 그레이스케일방식(grey scale mode)으로 실행했다. 좌심실에 있어서의 콘트라스트 증진 지속시간은 일정기간동안 신호를 기록함으로써 측정했다.
이 결과를 아래의 표 2에 나타내었고 또한 사용된 기체의 PC값도 나타냈다.
상기 결과로부터, 실험한 기체의 임계압력과 반향생성신호의 지속시간 사이의 명백한 상호관계의 존재를 알 수 있다.
[실시예 3]
반향생성 공기-충전 갈락톡스 미세입자(SCHERING AG로부터 구입한 Echovist)의 현탁액을 20%갈락토스 용액 8.5㎖에 3g의 상기 고체 미세입자를 넣어 5초동안 흔들어서 얻었다. 다른 제제에 있어서는 Echovist,입자 부분위로 공기를 배기시키고 (0.2 토르), SF6분위기로 대체시킴으로써, 20% 갈락토스 용액을 첨가한 후에, 회합된 육플루오르화황을 함유하는 미세입자 현탁액을 얻었다.
이 현탁액 분취량(1㎖)을 실험토끼에 투여했고(경정맥 주사로), 앞 실시예에서 기술한 바에 따라 심장의 영상화를 실행했다.
이 경우에서, 반향생성 미세입자는 폐의 모세혈관을 통해 통과하지 않으며, 따라서 영상이 우심실에 국한되어 전체의 신호 지속시간은 특별한 의미를 갖지 못한다.
아래 표 3의 결과는 주입 몇초후의 신호피크강도값을 나타낸다.
낮은 물용해도를 갖는 불활성 기체인 육플루오르화황은 공기로 채운 현탁액과 비교할 때 보다 더 강한 반향생성 신호를 발생시키는 반향생성 현탁액을 제공한다는 것을 알 수 있다. 이런 결과는 SF을 포함한 여러 가지 기체로 채운 캐비테이트(cavitate) 및 클라드레이트(clathrate) 화합물을 에코그라피 목적의 미세입자로서 각각 사용하는 것을 개시하고 있는 EP-A-441 468 및 357 163(SCHER ING)에 비추어볼 때 특히 흥미있다.
그러나 이들 기록은 반향생성 반응에 관하여 일반적인 다른 기체를 능가하는 SF의 특별한 이점을 전혀 기록하고 있지 않다.
[실시예 4]
일련의 기체-충전 마이크로버블의 반향생성 현탁액을 아래에 설명하는 일반적인 방법에 따라 제제했다.
수소화된 콩의 레시틴(독일의 Nattermann Phospholipids GmbH로부터 구입) 및 디세틸-포스페이트(DCP)를 몰비 9/1로 혼합한 혼합물 1g을 50㎖의 클로로포름에 용해시키고, 이 용액을 100㎖의 둥근 플라스크에 넣어 로터베이퍼(Rotavapor)장치상에서 증발시켜 건조했다.
다음에, 증류수 20㎖을 첨가하고, 이 혼합물을 75℃에서 한시간동안 천천히 교반시켰다.
그후 75℃에서 연속해서 3μm 및 0.8μm 폴리카르보네이트막(Nuclepore)을 통해 분출시켜 멀티라멜라리포솜(MLV) 현탁액의 형성을 이루었다.
냉각후에, 분출 현탁액 분취량 1㎖를 농축 락토스 용액(83g/ℓ) 9㎖로 희석시키고, 이 희석된 현탁액을 -45℃에서 동결시켰다.
이어서 이 동결 샘플을 아래의 표 4에 나타낸 것과 같은 선정 기체들로부터 선택한 기체 또는 공기로 궁극적으로 채운 용기에서 고진공상태하에서 동결건조하여 자유-흐름 분말(free-flowing powder)로 되게 했다.
다음에 이 분말샘플을 담체액으로 사용하는 10㎖의 물에 재현탁시켰고, 이것은 상기 용기에 채워서 사용했던 기체와 동일한 기체의 흐름하에서 실시했다.
볼텍스 혼합기상에서 1분동안 격렬히 흔들어줌으로써 현탁을 이루었다.
25℃에서 미리 공기와 평행시킨 증류수로 여러 가지 현탁액을 1 : 20으로 희석시켰고. 다음에 모든 기포가 붕괴될 때까지 수압을 점진적으로 증가시키도록 된 분광광도계셀에서 흡광도를 측정하는 것에 의해 실시예 1에서 기술한 것처럼 25℃에서 희석액의 입력 테스트를 했다. 이 결과 아래의 표 4에 나타냈으며, 여기에 추가적으로 임계압력 PC와 △P값을 나타냈다(제1도 참고).
압력증가에 대한 가장 높은 저향성은 대부분 물-불용성 기체들에 의해 제공된다는 것을 앞의 결과로부터 명백히 알 수 있다.
그러므로 마이크로버블의 성질은 이점에 있어서 마이크로발룬의 성질과 같다.
또한 높은 분자량을 갖는 낮은 물-용해도 기체가 가장 평평한 기포-붕괴/압력곡선을 제공하며(즉△P가 가장 넣다) 이것은 앞서 지적했듯이 생체내에서 반향생성 반응 지속성의 가장 중요한 요인이다.
[실시예 5]
실시예 4의 마이크로버블 현탁액 약간을 실시예 2에서 기술한 바와 같이 실험토끼의 경정맥을 주사했고, 앞서와 같이 좌심실의 영상화를 실행했다. 유용한 반향생성 신호가 검출되는 지속시간을 기록하였고 그 결과를 아래의 표5에 나타내었다.
여기에서 CF은 옥타플루오로시클로부탄을 나타낸다.
이들 결과는 본 발명의 기준에 부합하는 기체는 지금까지 사용해온 대부분의 기체를 보다 훨씬 더 긴 시간동안 초음파 반향신호를 제공한다는 것을 마이크로버블의 경우에서 다시한번 나타내고 있다.
[실시예 6]
마이크로버블 현탁액을 실시예 4에서 기술한 것과 똑같이 여러 가지 기체들을 사용하여 제제했으며, 다만 레시딘 인지질성분 대신 같은 몰수의 디아라키도일포스퍼 티딜코린(diarachidoyl phosphatidylcholine)(C지방산 잔기)을 미국 알라바마 버밍엄의 Avanti Polar Lipids로부터 구입하여 대체 사용했다.
인지질 대 DCP의 몰비율은 여전히 9/1로 했다.
다음에 현탁액을 실시예 4에서 처럼 압력 테스트하여 그 결과를 아래의 표 6A에 나타냈고, 표 4의 결과와 비교할 수 있게 했다.
표 4의 결과와 비교할 때, 상기 결과는 적어도 낮은 용해도를 갖는 기체에 있어서, 인지질 지방산 잔기의 사슬을 길게 하는 것에 의해 압력증가에 대한 반향생성 현탁액의 안정성을 현저히 증가시킬 수 있음을 보여주고 있다.
이것은 인지질성분을 이것의 고급 동족체, 즉 디-베헤노일-포스파티딜코린(C지방산 잔기)으로 단지 대체하여 앞서 행한 실험을 반복해보는 것에 의해 더 명백해졌다. 이때, 마이크로 버블 현탁액의 압력에 대한 붕괴 저항성은 더욱 더 증가되었다.
이 실시예의 마이크로버블 현탁액 약간을 토끼에 대해서 앞에서 기술했던 것과 같이 개에게 테스트했다(5㎖의 샘플을 전뇌정맥(anterior cephalic vein)으로 주사한 후에 심실을 영상화함). 실시예 4에 개시한 제제물의 성질에 비해 생체내애서의 유용한 반향생성 반응의 의미있는 증진이 나타났다. 즉, 인지질 성분에 있는 지방산의 잔기의 사실 길이의 증가가 생체내에서 반향생성제의 유용수명을 증가시킨다.
다음의 아래표는 지방산 잔기가 서로다른 사슬 길이를 갖는 일련의 인지질 현탁액으로부터 제제된 마이크로버블(SF)의 토끼 좌심실에 있어서의 상대적 안정성을 보여주고 있다(주입분량 : 1㎖/토끼).
지금까지 앞에서 언급된 이들 실시예들에 기술된 압력에 대한 저항성의 측정에 있어서, 일정한 압력증가속도 100토르/분을 유지했다고 언급해 왔다.
이것은 여러 가지 기체들의 PC값의 변화가 압력증가속도에 대해 함수관계에 있음을 보여주는 아래에 나타낸 결과로 볼 때 당연한 것이다. 이들 샘플에 사용된 인지질은 DMC였다.
[실시예 7]
물에서 현탁액으로서 사용한 일련의 알부민 마이크로발룬을 실시예 1 에 나타낸 방법을 사용하여 구(sphere) 크기를 조정하는 방식으로 공기하에서 제제했다. 다음에 약간의 샘플중에 있는 공기를 가압분위기에서 가스-교환 스위프 방법(gas-exchange sweep method)에 의해 다른 기체로 대체시켰다. 다음에 통상적인 증류수로 1 : 10 으로 희석시킨 후에 이 샘플을 실시예 1 에서와 같은 테스트 압력조건에 두었다. 아래의 표 7 에 나타낸 결과로부터, 몇몇 경우에 있어서는 두 단계 제제방법이 실시예 1의 한 단계 제제방법보다 더 좋은 압력저항성을 갖는 반향-생성제를 제공한다는 것을 알 수 있다.
[실시예 8]
본 발명의 방법을 예컨대 실시예 1의 WO-92/11873과 같은 선행기술에 개시된 것과 같은 실험에 적용시켰다. 3g의 Pluronic,F68(분자량이 8400인 폴리옥시에틸렌-폴리옥시프로필렌의 공중합체), 1g의 디팔미토일포스파티딜글리세롤(Na 염, AVANTI Polar Lipids) 및 3.6g의 글리세롤을 증류수 80㎖에 첨가했다. 약 80℃로 가열시킨 후, 맑은 균질용액을 얻었다. 이 텐시드(tenside : 계면활성제) 용액을 실온으로 냉각시켜 체적을 100㎖로 맞추었다. 몇몇 실험에서는(표 8 참고) 디팔미토일포스파티딜글리세롤을 디아라키도일포스파티딜코릴(920㎎)과 80㎎의 디팔미토일포스타티드산(Na 염 , AVANTI Polar Lipids)의 혼합물로 대체했다.
셋방향 밸브를 통해 연결된 두 개의 주입기를 사용하여 기포 현탁액을 얻었다. 주입기중 하나는 5㎖의 텐시드용액으로 채웠고 다른 하나는 0.5㎖의 공기나 기체로 채웠다. 이 셋방향 밸브를 기체-함유 주입기에 연결하기에 앞서 텐시드용액으로 채웠다. 두 개의 피스톤을 번갈아 작동시켜, 텐시드용액을 두 주입기 사이에서 앞뒤로 이동시켜(각 방향으로 5번). 우유같은 현탁액을 형성시켰다.
공기와 평형되게 포화시킨 증류수로 희석시킨 후에(1 : 10 내지 1 : 50). 이 제제의 압력 저항성을 실시예 1에 따라 측정했다. 압력증가속도는 240토르/분으로 했다. 아래의 결과를 얻었다.
본 발명의 방법을 사용하여 예컨대 SF과 같은 다른 기체로 공기를 대체시키므로 해서 알려진 공지의 제제물을 가지고서도 상당한 증진 즉, 압력에 대한 저항성의 증가를 이룰 수 있다는 결론을 얻었다. 음전하를 띤 인지질(예, DPPG) 및 중성과 음전하를 띤 인지질의 혼합물(DAPC/DPPA)의 경우에 있어서도 모두 이런 결론이 해당된다.
마이크로버블 및 마이크로발룬이 압력에 노출되었을 때, 즉 현탁액이 생명체내로 주사되었을때 인지되어 있는 이들의 붕괴에 대한 민감성 문제는 본 발명의 방법에 의해 유리하게 해결된다는 것을 상기 실험이 더욱 입증한다. 붕괴에 대해 보다 좋은 저항성 및 보다 좋은 안정성을 갖는, 마이크로버블 및 마이크로발룬으로 된 현탁액은, 보다 좋은 반복성 및 향상된 안정성을 갖는, 인간이나 동물 신체의 생체내에서 수행되는 에코그라피 측정을 현탁액에 제공하도록 유용하게 제조될 수 있다.

- 수용액 담체상(相)에 현탁액으로서의 기체 또는 기체 혼합물로 충전된 미세소포를 포함하는 초음파 에포그라피용 콘트라스트제로서, 기체 혼합물은 한가지 이상의, 달톤 분자량의 제곱근에 대한 표준상태하에서 물 리터당 기체의 리터로 나타낸 물에 대한 용해도의 비가 0.0027이하인, 생리적으로 수용가능한 할로겐화된 기체를 포함하는 것을 특징으로 하는 콘트라스트제.
- 제1항에 있어서, 상기 할로겐화된 기체는 SF6, SeF6또는 CF4, CBrF3, C4F8, CCIF3, CCI2F2, C2CIF5, CBrCIF2, C2CI2F4, CBr2F2또는 C4F10으로부터 선택되는 프레온인 것을 특징으로 하는 콘트라스트제.
- 제1 또는 제2항에 있어서, 상기 기체 혼합물은 공기, 질소 또는 이산화탄소로부터 선택되는 기체를 함유하는 것을 특징으로 하는 콘트라스트제.
- 동결건조된 인지질 및 안정제를 포함하는 제1항의 기체-충전된 미세소포의 반사성인 현탁액을 형성하도록 수용액 담체내에 분산될 수 있는 콘트라스트제의 건조 분말로서, 상기 분말은 한가지 이상의 달톤, 분자량의 제곱근에 대한 표준상태하에서 물 리터당 기체의 리터로 나타낸 물에 대한 용해도의 비가 0.0027 이하인 생리적으로 수용가능한 할로겐화된 기체를 포함하는 기체 혼합물의 분위기하에 저장되는 것을 특징으로 하는 콘트라스트제의 건조 분말.
- 동결건조된 인지질 및 안정제를 포함하는 제14항의 기체-충전된 미세소포의 반사성인 현탁액을 형성하도록 수용액 담체내에 분산될 수 있는 콘트라스트제의 건조 분말로서, 상기 분말은 달톤 분자량의 제곱근에 대한 표준상태하에서 물 리터당 기체의 리터로 나타낸 물에 대한 용해도의 비가 0.0027 이하인 생리적으로 수용가능한 할로겐화된 기체를 포함하는 기체 혼합물의 분위기하에 저장되는 것을 특징으로 하는 콘트라스트제의 건조 분말.

- 제14항 또는 제15항에 있어서, 상기 혼합물은 공기, 질소 또는 이산화탄소를 포함하는 것을 특징으로 하는 방법.
- 제14항 또는 제15항에 있어서, 상기 수성 담체는 용해된 라멜라화된 인지질 또는 상기 마이크로버블을 안정화하는 안정제를 함유하는 것을 특징으로 하는 방법.
- 제15항에 있어서, 상기 미세소포는 2단계로 형성되는데, 제1단계에서는 상기 미세소포 또는 그것의 건조 분말을 제1 기체의 대기하에서 미리 형성하고, 그리고 나서 제2단계에서는 상기 제1 기체의 일부 또는 전부를 생리학적으로 수용가능한 할로겐화된 기체로 치환하는 것을 특징으로 하는 방법.
- 제18항에 있어서, 상기 기체 혼합물로 미세소포의 형성하는 것은 선택적으로 그것의 건조 분말을 감압하에 두고 상기 기체 혼합물로 압력을 복원시키고, 그리고 마지막으로 건조 분말을 액체 담체내로 분산시키는 것에 의해 실행되는 것을 특징으로 하는 방법.
- 제18항에 있어서, 상기 제1 기체는 공기, 질소 또는 이산화탄소인 것을 특징으로 하는 방법.
- 제14항 또는 제15항에 있어서, 상기 할로겐화된 기체는 SF6, SeF6또는 CF4, CBrF3, C4F8, CCIF3, CCI2F2, C2CIF5, CBrCIF2, C2CI2F4, CBr2F2또는 C4F10으로부터 선택되는 프레온인 것을 특징으로 하는 방법.
- 제20항에 있어서, 상기 제1기체를 SeF6, CF4, CBrF3, C4F8, CCIF3, CCI2F2, C2F6, C2CIF5, CBrCIF2, C2CI2F4, CBr2F2또는 C4F10으로부터 선택되는 제2의 생리적으로 수용가능한 할로겐화된 기체로 완전히 치환하는 것을 특징으로 하는 방법.

- 제4항 또는 제5항에 있어서, 리포솜은 지방산 잔기가 16개 이상의 탄소원자를 갖는 인지질을 포함하는 것을 특징으로 하는 콘트라스트제의 건조 분말.
- 제4항 또는 제5항에 있어서, 상기 할로겐화된 기체는 SF6, SeF6또는 CF4, CBrF3, C4F8, CCIF3, CCI2F2, C2CIF5, CBrCIF2, C2CI2F4, CBr2F2또는 C4F10으로부터 선택되는 프레온인 것을 특징으로 하는 콘트라스트제의 건조 분말.
- 제4항 또는 제5항에 있어서, 상기 혼합물은 공기, 질소 또는 이산화탄소를 함유하는 것을 특징으로 하는 콘트라스트제의 건조 분말.
