本発明のペプチドは以下の(A)から(D)の何れかに記載のペプチドである。

(B)配列番号1又は2に記載のアミノ酸配列を含むペプチドにおいて、1個、2個または数個のアミノ酸が置換、欠失、挿入及び/又は付加されており、細胞傷害性(キラー)T細胞の誘導活性を示すペプチド。
(C)N末端から2番目のアミノ酸がロイシンまたはメチオニンである、(B)に記載のペプチド。
(D)C末端アミノ酸がバリンまたはロイシンである、(B)に記載のペプチド。
実施例において、本発明のペプチドは、生体内における癌細胞特異的キラーT細胞を誘導することができることが示された。さらに、先の発明では、CDCA1は、肺癌、胆管細胞癌、膀胱癌、腎細胞癌、前立腺癌、慢性骨髄性白血病、悪性リンパ腫、子宮頚癌、骨肉腫、乳癌、軟部肉腫、大腸癌等の大部分の症例で高発現していることが示された。したがって本発明のペプチドを有効成分として1種類以上含有する免疫誘導剤には、癌の治療、及び/又は予防剤としての効果が期待できる。つまり、本発明のペプチドを適当なアジュバントと共に、あるいはペプチドを樹状細胞などの抗原提示細胞に負荷した後に体内に注入することにより、腫瘍攻撃性キラーT細胞を誘導ならびに活性化することが期待できる。さらにその結果として抗腫瘍効果も期待できる。また発明のペプチドをコードする遺伝子を適当なベクターに組み込み、この組換えDNAで形質転換されたヒトの抗原提示細胞(樹状細胞など)、BCG結核菌などの細菌、または本発明のペプチドをコードするDNAをゲノムに組み込まれたワクシニアウイルス等のウイルスは、ヒト癌の治療・予防用生ワクチンとして有効に利用できる。なお、癌ワクチンの投与量及び投与法は通常の種痘やBCGワクチンと同様である。
(1)CDCA1を発現する細胞を含む腫瘍に対するキラーT細胞の誘導、
(2)CDCA1を発現する細胞を含む腫瘍を認識する抗体の誘導、および
(3)抗腫瘍性サイトカイン産生の誘導。
本発明は、上記した本発明のペプチドの一部もしくは全部をエピトープ(抗原)として認識する抗体、ならびに当該蛋白質又はペプチドを用いてインビトロ刺激により誘導されたキラーT細胞にも関する。一般的には、キラーT細胞は抗体よりも強い抗腫瘍活性を示す。
本発明はまた、本発明のペプチドを用いてインビトロ刺激により誘導されたヘルパーT細胞、キラーT細胞、又はこれらを含む免疫細胞集団に関する。例えば、末梢血リンパ球や腫瘍浸潤リンパ球を本発明のペプチドを用いて、インビトロで刺激すると腫瘍反応性活性化T細胞が誘導され、この活性化されたT細胞は養子免疫療法に有効に用いることができる。また本発明のペプチドを強力な抗原提示細胞である樹状細胞に負荷、あるいは遺伝子導入により発現させて、これらを用いてT細胞をインビボあるいはインビトロで刺激することにより、抗腫瘍免疫応答を誘導することができる。
本発明はさらに、本発明のペプチドとHLA抗原の間で形成された複合体をその表面上に提示するエキソソームと呼ばれる細胞内小胞を提供する。エキソソームは、例えば、第平11-510507号および第2000-512161号の公開された日本語翻訳文に詳細に記載された方法を用いることにより調製することができ、好ましくは、治療および/または予防の標的である被検体から得られた抗原提示細胞を用いて調製される。本発明のエキソソームは、本発明のペプチドと同様に、癌ワクチンとして接種することができる。
本発明は、本発明の1つまたは複数のペプチドを用いて抗原提示細胞を誘導する方法を提供する。末梢血単球から誘導した樹状細胞に本発明の1つまたは複数のペプチドを接触させ、刺激することにより抗原提示細胞を誘導することができる。本発明のペプチドを被検体へ投与する場合、本発明のペプチドを細胞表面に提示している抗原提示細胞を、被検体の生体内において誘導することができる。あるいは、本発明のペプチドを抗原提示細胞へ負荷した後、該細胞をワクチンとして被検体に投与するエクスビボ法を用いることができる。例えば、エクスビボ投与は以下の工程を含みうる:
(1) 被検体から抗原提示細胞を収集する工程、および
(2) 工程(1)の抗原提示細胞と本発明のペプチドを接触させる(工程(1)の抗原提示細胞に本発明のペプチドを負荷する)工程。
(1)被検体から抗原提示細胞を収集する工程、
(2)工程(1)の抗原提示細胞と本発明のペプチドを接触させる工程(工程(1)の抗原提示細胞に本発明のペプチドを負荷する工程)、
(3)細胞傷害性T細胞を誘導するために、段階(2)の抗原提示細胞をCD8+T細胞と混合し共培養する工程、および
(4)工程(3)の共培養物からCD8+T細胞を収集する工程。
本発明はさらに、CDCA1-1ならびにCDCA1-4を認識するT細胞受容体(TCR)のサブユニットを形成するポリペプチドをコードする核酸ならびにその使用法を提供する。TCRは、少なくとも7つの膜貫通タンパク質よりなる。ジスルフィド結合で連結した(α、β)ヘテロ二量体がクロノタイプ抗原認識単位を形成し、一方、ε、γ、δおよびζ、η鎖よりなるCD3の非変異鎖は、シグナル伝達経路にリガンド結合を結び付け、その結果、T細胞活性化および細胞性免疫応答が生じる。このように抗原認識を担うαβサブユニットは、T細胞が特定の標的を認識するような特異性を獲得するために必要である。そこで、特異的に本発明のペプチドを認識するTCRを合成するために、本発明のペプチドを用いて誘導したCTL由来のTCRサブユニットであるαおよびβの核酸配列は、当業者に知られた方法を用いて同定することができる(WO2007/032255およびMorgan et al., J Immunol, 171, 3288 (2003))。同定したTCRサブユニットから形成されたTCRはCDCA1ペプチドを提示する標的細胞へ結合することができるため、殺細胞効果をもつ細胞へ導入することでその有用性を示すことができる。
以下の実施例により本発明を更に説明するが、本発明はこれら実施例に限定されるものではない。
(1)HLA-A2に結合性を示すCDCA1ペプチドレパートリーの選択
ヒトCDCA1のアミノ酸配列をBIMAS system により検索して、推定されたHLA-A2との結合親和性(binding affinity) が高いものから順に41種類を選択した(表1)。
先に報告した方法を用いて(Komori Hら、Clinical Cancer Research 12: 2689-2697, 2006)、まずHLA-A2トランスジェニックマウスの骨髄細胞から樹状細胞(DC)を誘導した。このようにして得られたBM-DCに、CDCA1ペプチドを添加(10μM)し、HLA-A2トランスジェニックマウス一匹あたり5×105個のBM-DC を腹腔内に投与した。1週間の間隔をおいて2回同様に投与しマウスを免疫した後、マウスの脾細胞を回収してキラーT細胞の検出に用いた。CD8+T細胞由来のキラーT細胞の誘導を厳密に検討するために、脾臓の採取後に脾細胞からMACS ビーズを用いてCD4+T細胞を取り除いたものを用いた。
Day-21 (1)HLA-A2トランスジェニックマウスの骨髄細胞にGM-CSFを加え、骨髄由来の樹状細胞(以下BM-DC)の誘導を開始する。
Day-14 (2) 誘導したBM-DCに4種類のCDCA1ペプチドの混合物を添加し、2時間後に1匹あたり5×105 個を腹腔内に投与する。
(1)(2)を1週毎2回繰り返す。
Day 0 免疫したHLA-A2トランスジェニックマウスの脾細胞を回収し、再びBM-DCを CDCA1ペプチドと2時間インキュベートしたものと共培養した後に、6日間培養した。
Day 6 CDCA1ペプチドを特異的に認識するキラーT細胞を検出するために、抗原刺激後にガンマーインターフェロンを産生するT細胞を、ELISPOT法により定量した。ターゲット細胞としてBM-DCに、それぞれのCDCA1ペプチドを負荷したものと、負荷していないものを利用した。
これらの誘導したキラーT細胞の中に、確かにCDCA1に特異的に反応してIFN-γを産生するものが存在するか否かについてELISPOT法にて検出した。IFN-γの検出は、Mouse IFN-γ ELISPOT set (BD社)を用いて行った。刺激細胞(ターゲット)に対して、キラーT細胞(エフェクター)が反応してIFN-γを産生すると、それぞれが赤いスポットとして検出される。標的細胞として、HLA-A2陽性でCDCA1を発現していないT2A2細胞と、CDCA1ペプチドを負荷したT2A2細胞を用いた。T2A2細胞はTAP遺伝子の発現を欠損するマウスT2細胞株に、HLA-A2遺伝子を導入した細胞株であり、理研細胞バンクより購入した。この細胞では、TAP欠損により外部から添加したペプチドがHLA-A2分子に結合する性質を持つ場合にのみ、HLA-A2分子とペプチドの複合体が細胞表面に発現する。まず、抗マウス IFN-γ抗体を、ELISPOTプレート(BD Bioscience社)に18時間コーティングした。その後、10%FCS/RPMIにて2時間ブロッキングを行った。エフェクター細胞(100μL /well)と標的細胞(100μL /well)を混合し、37℃で22時間培養した。エフェクター/ターゲット比(E/T比)は、5:1で実験を行なった。その後、プレートを滅菌水で洗浄し、ビオチン化抗マウスIFN-γ抗体と2時間、さらにストレプトアビジン-HRPと1時間反応させ、基質溶液にてIFN-γ陽性のスポットを検出した。スポットのカウントは、MINERVA TECH社の自動解析ソフトを用いて行った。ペプチドNo.1から20の結果を図1Bに示す。同様な実験により、41のペプチドのうちCDCA1 65-73(No. 1) (配列番号:1)、CDCA1 351-359(No. 4) (配列番号:2)ペプチドで誘導したキラーT細胞において、CDCA1特異的キラーT細胞の免疫応答が観察された(図2)。
CDCA1 65-73(No. 1)(配列番号:1)ペプチドで誘導したキラーT細胞の解析結果を図2Aに、CDCA1 351-359(No. 4)(配列番号:2)ペプチドで誘導したキラーT細胞の解析結果を図2Bに示す。
患者、血液試料および細胞株
NSCLC患者由来の血液試料はインフォームドコンセントを得た後に日常診断より得られた。CDCA1陰性ヒト大腸癌細胞株COLO201は健康科学研究資源バンクから供与された。これらの試料におけるHLA-A2発現はHLA-A2モノクローナル抗体BB7.2(One Lambda Inc.、カノガパーク、カリフォルニア州)を用いてフローサイトメトリーにより確認された。
レンチウィルスベクター経由遺伝子導入はTahara-Hanaoka S, et al. Exp Hematol 2002;30:11-7.に記述された方法を用いた。簡潔には、17 μgのCDCA1 cDNA含有CSII-CMV-RfAとCSIIEF-RfA自己不活性ベクター(Miyoshi H, et al. J Virol 1998;72:8150-7.)と10 μg のpCMV-VSV-G-RSV-Rev およびpHIVgpを293T細胞にLipofectamine 2000(Invitrogen corporation, カリスバッド、カリフォルニア州、USA)を用いて10cmディッシュ上で形質転換させ、60時間後、培養液を回収し、ウィルス粒子は超遠心(50,000 × g、2 時間)によりペレットにした。このペレットを50 μlのRPMI 1640溶液に懸濁し、10 μlのウィルス懸濁液を、5 × 104 個のCOLO201が入っているU底96ウェルプレートの各ウェルへ加えた。形質転換されたCDCA1遺伝子の発現はウェスタンブロットで確認した。
単球由来DC細胞をHLA提示ペプチドに対し応答するCTL誘導のために抗原提示細胞として使用した。DCは以前報告されたin vitroの方法(Suda T, et al. Cancer Sci 2007., Nakahara S, et al. Cancer Res 2003;63:4112-8.)で得られた。簡潔には、HLA-A*0201陽性健常者ボランティアまたはNSCLC患者からフィコールプラーク溶液(GE Healthcare UK, Ltd.、バッキンガムシャー、イギリス)を用いて単離した抹消血単核球(PBMC)からマイクロビーズ(Miltenyi Biotec、ベルギッシュグラートバハ、ドイツ)を用いてCD8陽性CD14陽性集団を選別した。DCを得るため、CD14陽性集団は2%自己血漿含有AIM-V (Invitrogen)中で、100 ng/mLの顆粒球−マクロファージコロニー刺激因子(GM-CSF)と10 ng/mL インターロイキン(IL)-4(PeproTec Inc.、ニュージャージー州、USA)の存在下において培養された。培養4日後、成熟DCを調製するため、OK-432をデッシュに加えた。サイトカイン誘導DCの培養開始5日後、20 μg/mLのHLA-A2結合性ペプチドをAIM-V中に4 μg/mLのβ2-microglobulin (Sigma-Aldrich、セントルイス、ミズーリ州、USA)の存在下で37℃、2時間の条件で負荷させた。これらペプチド負荷DCは放射線照射(3,500 cGy)され、1:50の比で抗CD8マイクロビース(Miltenyi Biotec)を用いてPBMCから得られた自己CD8陽性T細胞と混合した。培養は48ウェルプレートで行われ、各ウェルは0.5 mLの2%自己血漿含有AIM-V 中に1×104 個のペプチド負荷DC、 5 ×105個のCD8陽性T細胞および10 ng/mLのヒトIL-7 (Wako、大阪、日本)の条件で調整された。培養3日後、ヒトIL-2(PeproTec Inc.)を20 IU/mLの濃度に成るよう加えた。12日目と19日目にT細胞はさらにペプチド負荷自己DCで再刺激された。DCは適宜上記の方法で調製した。CTLの抗原特異性応答は、25日目の3度目のペプチド刺激6日後にIFN-γ・ ELISPOT法とクロム(Cr)放出試験により評価された。
CTLは、標的細胞として各癌細胞株またはペプチド負荷もしくは非負荷T2細胞を各エフェクター細胞/標的細胞比で共培養し、51Cr放出試験およびIFN-γ・ELISPOT法を従来の方法(Komori H, et al. Clin Cancer Res 2006;12:2689-97., Makita M, et al. Clin Cancer Res 2002;8:2626-31., Yokomine K, et al. Cancer Sci 2007;98:1930-5.)で行った。簡潔には、標的細胞を3.7 KBq Na2 51Cr4 (Perkin Elmer Life Sciences)を用いて37℃、CO2インキュベーター内で1時間かけて標識した。標識した標的細胞を3度リンスし、3時間、37℃で20 μg/mlのペプチドとインキュベートすることでペプチド負荷標的細胞を調製した。標的細胞は、平底マイクロタイタープレートに200 μlの用量に成るようエフェクター細胞と混合し、インキュベートされた。インキュベート6時間後、その上清50 μlを各ウェルから回収し、放射線活性をガンマカウンターで定量した。特異的細胞傷害活性は、既存の方法(Suda T, et al. Cancer Sci 2007;98:1803-8.)にて特異的な51Crの放出の割合で算出し評価された。ELISPOT法も既存の方法(Komori H, et al. Clin Cancer Res 2006;12:2689-97.)にて行われた。
HLA-A2(A*0201)陽性健常者ドナーよりPBMCを単離し、CD8+ T細胞とCDCA1 65-73 (No. 1) (配列番号:1)ペプチドまたはCDCA1 351-359(No. 4) (配列番号:2)ペプチドを負荷した単球由来DCと共培養し、毎週3回CD8+ T細胞の刺激を行った(図3A)。
自然にプロセシングされ、腫瘍細胞に提示されるTAA誘導ペプチドの同定はペプチドに基づく癌免疫療法の確立に重要である。NSCLCと正常組織を用いたcDNAマイクロアレイ分析を用いて新規癌-精巣抗原であるCDCA1を同定した。CDCA1はNSCLCと正常精巣に強く発現しているが、調べた他の正常組織にはmRNAとタンパク質共に発現していない。精巣は免疫系から隔離された組織であることから、CDCA1応答性CTLはNSCLCのみ攻撃すると考えられる。このため、NSCLC患者の免疫療法におけるTAAとしてCDCA1が選択された。
Claims (23)
- 以下の(A)又は(B)に記載のペプチド;
(A)配列番号:1又は2に記載のアミノ酸配列を含むペプチド、
(B)配列番号:1又は2に記載のアミノ酸配列を含むペプチドにおいて、1個、2個または数個のアミノ酸が置換、欠失、挿入及び/又は付加されており、細胞傷害性(キラー)T細胞の誘導活性を示すペプチド。 - N末端から2番目のアミノ酸がロイシンまたはメチオニンである、請求項1に記載のペプチド。
- C末端アミノ酸がバリンまたはロイシンである、請求項1に記載のペプチド。
- 請求項1に記載のペプチドを有効成分として1種類以上含有する、癌に対する免疫誘導剤。
- 請求項1に記載のペプチドを有効成分として1種類以上含有する、癌の治療及び/又は予防のための薬剤。
- 請求項1に記載のペプチドを有効成分として1種類以上含有する、細胞傷害性(キラー)T細胞の誘導活性を示す抗原提示細胞を誘導するための薬剤。
- 請求項1に記載のペプチドをコードするポリヌクレオチドを有効成分として1種類以上含有する、細胞傷害性(キラー)T細胞の誘導活性を示す抗原提示細胞を誘導するための薬剤。
- 請求項1に記載のペプチドを有効成分として1種類以上含有する、細胞傷害性(キラー)T細胞を誘導するための薬剤。
- 請求項1に記載のペプチドに対する抗体。
- 請求項1に記載のペプチドを用いて誘導される、ヘルパーT細胞、細胞傷害性(キラー)T細胞、又はこれらを含む免疫細胞集団。
- 請求項1に記載のペプチドおよびHLA抗原を含む複合体を提示する抗原提示細胞。
- 請求項6または7に記載の薬剤によって誘導される、請求項11に記載の抗原提示細胞。
- 請求項1に記載のペプチドおよびHLA抗原を含む複合体を提示するエキソソーム。
- HLA抗原がHLA-A2 (HLA-A*0201)である、請求項13に記載のエキソソーム。
- 抗原提示細胞と請求項1に記載のペプチドを接触させる工程を含む、細胞傷害性(キラー)T細胞の誘導活性を示す抗原提示細胞を誘導する方法。
- 請求項1に記載のペプチドをコードするポリヌクレオチドを抗原提示細胞へ導入する工程を含む、細胞傷害性(キラー)T細胞の誘導活性を示す抗原提示細胞を誘導する方法。
