実施例1
SDラット(オス・6週齢)を16時間絶食させた後、グルコース(Gluc)、グリコーゲン(ESG)およびデキストリン(DEX)を2 g/kg体重で経口投与した(各群7匹)。投与前、投与後10、20、30、60分に尾静脈から採血し、遠心分離することで血漿サンプルを得た。血漿サンプル中のグルコース量をグルコースCIIテストワコーを用いて測定し、インスリンの濃度はレビス(登録商標)インスリン-ラットELISAキット(シバヤギ製)を用いて測定した。結果を図1に示す。図1の結果から、グリコーゲンは、投与後の血糖値とインスリン分泌量が低く、食後においても血糖値とインスリン分泌が緩やかに上昇し、インスリン分泌能が低下し、或いはインスリン抵抗性のヒトなどの哺乳動物において、糖尿病の予防あるいはその進行を遅らせ得ることが明らかになった。なお、ESGは、酵素合成グリコーゲン(Enzymatically Synthesized Glycogen)である。
SDラット(オス・6週齢)を表1に示した組成の餌で2週間飼育し、飼育期間中2-3日に一度体重を測定した。体重変化と摂食量を以下の表2に示す。

SDラット(オス・6週齢)を表1に示した組成の餌で2週間飼育し、飼育9日目と10日目に肛門から新鮮な糞を採取し、腸内細菌数を表4に示す培地を用いて培養法にて測定した。結果を表5と図2に示す。表5と図2の結果から、グリコーゲン(ESG)は、腸内細菌のうち有用な細菌である乳酸菌とビフィズス菌、特にビフィズス菌の増殖を促進し、その割合を高め、腸内フローラを顕著に改善することが明らかになった。

SDラット(オス・6週齢)を表1に示した組成の餌で2週間飼育した後屠殺し、心臓から採血後遠心分離することで血漿サンプルを得た。サンプル中の中性脂肪、コレステロール、HDL-コレステロールをそれぞれトリグライセライドE-テストワコー、コレステロール-Eテストワコー、HDL-コレステロールテストワコーで測定した。さらに、屠殺後のラットについて、肝臓と睾丸周辺脂肪の重量を測定した。
方法1
Megazyme International Ireland Ltd.社(Wicklow, Ireland)のResistant Starchアッセイキットを使用し、マニュアルに従って「分解抵抗性成分(RS)」として定量される量によってグリコーゲンを評価した。この方法はRS含量測定のための公定法として採用されている(AOAC Method 2002.02; AACC Method 32-40)。具体的には、以下のスキームに従いRS含量を求めた。

試料100mgを、酵素液(1%パンクレアチン+12Uグルコアミラーゼ)で分解する
↓ 37℃, 16時間
反応液に等量のエタノールを加え、低速で遠心分離(1500×g、10分)
↓ →上清中のグルコースを定量→分解非抵抗性成分量
沈殿をアルカリで可溶化
↓
中和後、直ちに大量のグルコアミラーゼ(330U)で分解(50℃、30分)
↓
グルコースを定量→分解抵抗性成分(RS)量
結果を表9に示す。
試料50mgを、1mLの反応液中でα-アミラーゼ(Sigma社製、ブタすい臓由来 TypeI-A。酵素量300U/g基質)を作用させる。反応液のpHは、10mMリン酸緩衝液により、7に合わせる。

100℃5分加熱することにより、反応を停止し、反応液を0.45μmフィルターによりろ過し、以下の条件でHPLC分析に供する。
本方法で、α-アミラーゼ処理前後のMwの比を求める。
結果を図4に示す。
1.単回脂質投与試験
SDラット(7-9週齢、♂)を18時間絶食させた後、水(コントロール)、20%ESG溶液あるいは20%NG(天然グリコーゲン)溶液を10ml/kg体重で経口投与し、直後にコーンオイルエマルジョン(コーンオイル20%、レシチン1.2%、グリセリン2.3%)を10ml/kg体重で経口投与した。投与前、投与後1、2、3、4時間に尾静脈から採血した後、遠心分離により血漿を分離し、血漿中のトリアシルグリセロール濃度をWAKOのキットで測定した。結果を図5に示す。
2.高脂肪食試験
SDラット(7週齢、♂)を8匹ずつ2群に分け、高脂肪食群と高脂肪食+ESG20%群とし、下表の組成の餌をそれぞれ4週間自由に摂取させた。飼育終了後に各脂肪組織を摘出し重量を測定した。また、血漿と肝臓を回収し、血漿中のアスパラギン酸アミノトランスフェラーゼ(AST)とアラニンアミノトランスフェラーゼ(ALT)の活性を市販のキット(WAKO製)で測定した。肝臓の中性脂肪はFolch法で全脂質を抽出した後、イソプロパノールに溶解し、市販のキット(WAKO製)で定量した。*は高脂肪食群に対する有意差を表す(P < 0.05、t-test)。結果を図6、図7及び表10に示す。
