Description

【発明の詳細な説明】 本発明は、特にアルツハイマー病を含む、初老人件およ
び老人性痴呆の処置に使用する医薬組成物に関する。
痴呆および特にアルツハイマー病は脳機能の漸進的退化
を含み、正常な生活へ導く個人の能力の損失を招く。こ
の病気は５０代またはそれ以前の人々に早くも発生する
ことがあり、そして正常の老化現象の異常に激しい加速
として出現し得る。
現在アルツハイマー病の原因は未知であり、確実な治療
法は存在しない。
われわれの研究は、アルツハイマー病のような痴呆は必
須脂肪酸ＩＪｔ（ＥＦＡｓ）のプロスタグラニシンＩＩ
Ｉ（ＰＧｓ）への変換のインバランスに関係しているこ
とを示した。正常の老化においては、細胞膜流動性が減
少しくすなわち膜が硬くなる）、そしてリンパ球のよう
な細胞によるサイクリックアデノシンモノリン酸（ＡＭ
Ｐ）の生産が減少する。アルツハイマー病における変化
は真向から反対であり、細胞膜の流動性の予期しない増
加と、サイクリックＡＭＰレベルの上昇が起こる。
ＰＧＥＩ　　（ジホモガンマリノレン酸（ＤＧＬＡ）か
ら生成する）、ＰＧ［２（アラキドン酸（Ａ　Ａ）から
生成する）、およびＰＣｌ３　　（エイコサペンクエン
酸（ＥＰＡ）から生成する）のようなある種のＰＧｓは
、膜流動性とサイクリックＡＭＰレベルを上昇し得るこ
とが既知である。このためわれわれはアルツハイマー病
における予想外の変化は、ＥＰＡｓのＰＧｓへの過剰の
変換から発生するものとして説明し得るものと信する。
ＥＦＡｓは生体内のすべての細胞膜の、特にＥＦＡレベ
ルが特に高い脳中のそれらの構造の必須成分である。
そこでは主としてリン脂質の形で見出される細胞膜から
のＥＦＡｓのＰＧｓへの無制限なそして過剰の変換は、
細胞膜機能への構造的損傷および機能的障害へと導くこ
とが予期される。しかしながら、これは可能性ある理論
的説明としてのみ提示され、本発明の有用性は勿論この
説明が正しいかどうかに依存しない。
本発明の目的は、アルツハイマー病のような痴呆を治療
するために患者を処置するための治療剤を提供すること
である。
本発明は、初老人件または老人性痴呆、特にアルツハイ
マー病を治療するための、そして生理学的に許容し得る
リチウム化合物および／または必須脂肪酸もしくは生理
学的に許容し得るその塩を含有する医薬組成物を提供す
る。
リチウム化合物は、そのサイクリックＡＭＰの生産を減
少し、そして一部にはフォスファチジルイノシトールの
代謝を阻止することによってＥＰＡ３のＰＧｓへの代謝
的変換を阻止する能力に鑑み、本発明によって使用され
る。従ってリチウム化合物の投与はＥＦＡｓのＰＧａへ
の変換のインバランスを修正するように働くことができ
、そのため痴呆特にアルツハイマー病の処置に役立ち得
る。
本発明によるリチウム化合物の投与は、便利には非経口
的、または好ましくは経口的であり、それからリチウム
イオンが生理的にアクセスし得る化合物は、好ましくは
炭酸塩、クエン酸塩、塩化物、臭化物、酢酸塩、アセチ
ルサリチル酸塩、安息香酸塩、酒石酸塩、硝酸塩、セレ
ン酸塩、亜セレン酸塩、硫酸塩、アスパラギン酸塩、グ
ルコン酸塩またはテノン酸塩のような塩である。
成人に対するリチウム化合物の１日投与量は、一般にリ
チウムイオンとして１ないし２０００■。
好ましくは２０〜６００■、特に５０〜６００■の範囲
であるが、正確な投与量は患者の体重および状態の重篤
度のようなファクターに依存するであろう、血漿チリウ
ム濃度が一般にモニターされ、そして投与割合は血漿中
例えば０．０５ないし１．６ｍ　Ｍ　／　１の適当なリ
チウム濃度を与えるように調節される。
ＰＣ生合成前駆体として作用することに加え、ＥＦＡｓ
はそれ自体生体学的において重要であり、モしてＥＦＡ
欠乏はアルツハイマー病のような痴呆のある種の効果に
対し責任がある。このため本発明の好ましい具体例にお
いては、もし望むならばリチウム化合物と同時に、ＥＦ
Ａｓが患者へ再び便利には経口的または非経口的に投与
される。
実際、ＥＦＡ欠乏の危険に鑑み、ＥＦＡｓはアルッハイ
マー病患者に対し、リチウム治療なしでも投与すること
ができる。
ここで使用する必須脂肪酸なる用語は、慣用の意味で、
リノレイン酸（ＬＡ）、ガンマリノレン酸（ＧＬＡ）、
ジホモガンマリノレンｆ％ｔ（ＤＧＬＡ）、アラキドン
酸（ＡＡ）、アドレン酸および２２　：　　５ｎ−６と
、そしてアルファリノレンｆ％ｔ（ＡＬＡ）　、　１８
　：　４ｎ−３，２０：　４ｎ−３，エイコサペンクエ
ン酸（Ｅ　Ｐ　Ａ）　、　２２　：　５ｎ−３およびド
コサヘキサエン酸（ＤＨＡ）よりなる二つの脂肪酸シリ
ーズを上杵する。
本発明に従って投与するために特に好ましいＥＦＡｓは
、ＤＧＬＡ、ＡＡ、　　ＥＰＡ、ＤＨＡ、ＬＡ、ＧＬＡ
、ＡＬＡおよび１８　：　４ｎ−３である。本発明の目
的のために、特に適当なＧＬＡおよびＬＡの源は月見油
中に発見された。ＥＦＡをその塩の形で投与する時は、
１種類の塩が両方の活性成分を提供するのでリチウム塩
を使用するのが特に好ましい。ＥＦＡｓの１日当たりの
投与量は、再び患者の体重および症状の重篤度のような
ファクターに依存するが、成人に対し一般に１■ないし
２００ｇの範囲内である。ＬＡおよびＡＬＡに対しては
、１日投与量は好ましくは１００ｍｇないし２００ｇ、
特に好ましくは２〜３０ｇであり、ＧＬＡ、ＤＧＬＡ、
ＡＡ、１８：４ｎ−３，ＥＰＡおよびＤＨＡのような他
の脂肪酸については、１日投与量は好ましくは１ｍｇな
いし５０ｇ、特に好ましくは２０■ないし１０ｇである
。
本発明による医薬組成物は、有利には１００ないし１２
００■のリチウム化合物および／または５０ないし１２
００■のＥＦＡもしくはその塩を含有する単位投与形態
１例えば錠剤、糖衣錠、カプセル等の形を取る。
以下の実施例は本発明の医薬組成物を例証する。
裏脂皿上−鉦五 慣用の錠剤補助剤および以下のリチウム塩を使用して、
慣用方法によって錠剤が製造される。

ｌカプセル当たり以下の活性成分を使用して、慣用方法
によってカプセルが製造される。

　ガンマリノレン酸リチウム　　１ｏｏｏ■＊ＧＬＡ４
５■とＬＡ３６０＋ｎ＋ｒを含有する。

Claims (9)

    【特許請求の範囲】
  1. （１）生理学的に許容し得るリチウム化合物および／ま
    たは必須脂肪酸もしくは生理学的に許容し得るその塩を
    含有することを特徴とする初老人性または老人性痴呆の
    治療のための医薬組成物。
  2. （２）前記痴呆がアルツハイマー病である第１項の医薬
    組成物。
  3. （３）リチウム化合物がリチウム塩である第１項または
    第２項の医薬組成物。
  4. （４）リチウム塩が、炭酸リチウム、クエン酸リチウム
    、コハク酸リチウム、塩化リチウム、臭化リチウム、酢
    酸リチウム、アセチルサリチル酸リチウム、安息香酸リ
    チウム、酒石酸リチウム、硝酸リチウム、セレン酸リチ
    ウム、亜セレン酸リチウム、硫酸リチウム、アスパラギ
    ン酸リチウム、グルコン酸リチウム、テノン酸リチウム
    より選ばれる第３項の医薬組成物。
  5. （５）必須脂肪酸またはその塩が、ジホモガンマリノレ
    ン酸、アラキドン酸、エイコサペンタエン酸、ドコサヘ
    キサエン酸、リノレイン酸、ガンマリノレン酸、アルフ
    ァリノレン酸、１８：４ｎ−３およびそれらの生理学的
    に許容し得る塩より選ばれる第１項ないし第４項のいず
    れかの医薬組成物。
  6. （６）生理学的に許容し得るリチウム化合物と、必須脂
    肪酸もしくはその生理学的に許容し得る塩とを共に含ん
    でいる第１項ないし第５項のいずれかの医薬組成物。
  7. （７）必須脂肪酸のリチウム塩を含んでいる第１項ない
    し第６項のいずれかの医薬組成物。
  8. （８）各投与単位が前記リチウム化合物１００ないし１
    ２００ｍｇを含有している投与単位の形の第１項ないし
    第７項のいずれかの医薬組成物。
  9. （９）各投与単位が前記必須脂肪酸もしくはその塩５０
    ないし１２００ｍｇを含有している投与単位の形の第１
    項ないし第８項のいずれかの医薬組成物。
