Description

【発明の詳細な説明】 本Ｂ　明に’ｗ　ヒドロコルチゾンを基剤とする組成物
の長期使用により一般的に見られる禁忌徴候を避けるこ
とのできる、可溶化されたヒドロコルチゾンを高濃度で
含有する皮膚治療用の新規の組成物に関する〇 ヒドロコルチゾンを基剤とするローション、ポマード、
クリーム又は軟膏等を不適切して、特？−長期にわたっ
て使用すると必然的に皮膚の真正の非可逆的萎縮を惹起
し得るある種の副作用が生ずる。
現実の組成物は一般にヒドロコルチゾンヲ約２慢未満、
望まじくは約１％程度含有している・約０．１％程度の
かなり低いヒドロコルチゾン濃度の組成物は高濃度の組
成物の不都合を特に回避する目的で使用できるが、この
場合には勿論処置期間をかなり長くする必要があり、し
たがって絶えず医学的監視を行なうことが必要となる・
処置の継続期間を短縮しかつしたがって副作用を回避す
る目的で分散液の形の組成物も同様に提案された。これ
らの組成物はヒドロコルチゾンのより高濃度を達成し得
るが、それにも拘わらず処置すべき皮膚帯域上での浸透
の不足から有効性が劣っている。
さらに、これらの組成物は貯蔵中の保存性が悪く、長期
間の間にはヒドロコルチゾンが分解し、それだけ組成物
の活性が低下する・ 良好な浸透を保証しそれによって処置期間を短縮し、し
たがってＦ！ｉ、ｌ１Ｓｉの非可逆的萎縮を避けること
のできる高濃度のヒドロコルチゾン含有局所皮膚治療用
組成物を提供することは従来不可能であった〇 本発明によれば、この問題はヒドロコルチゾンを２．５
％又はそれ以上の濃度で可溶化できる溶剤混合物の使用
によって解決される。
さらに、本発明による組成物の保存試験はきわめて満足
すべき結果を示し、常温において２ケ月後にＨＰＬＣに
よって測定した分解皐は５％未満であった。
したがって本発明は、炭素数１〜３個のアルコール、フ
ロピレンゲリコール及び２−ヒドロキシエチルサリチレ
ート、ジプロピルケトン及ヒジメチルココベンジルアン
モニウムクロライドからなる群から選んだ第三〇〇Ｔ溶
化剤からなる溶剤混合物中にヒドロコルチゾンを可溶化
状態で含有することを特徴とするヒドロコルチゾンを有
効成分とする局所皮膚治療用組成物を提供するものであ
る〇本発明において使用される脂肪族アルコールはメタ
／−ル、エタノール及ヒイソプロビルアルコールでアリ
、特にエタノールが好ましい。
１）２−ヒドロキシエチルサリチレートは沸点＋６９−
＋７２°Ｃ／　ｌ　２　ｍｍＨｇ　　の無色の液体であ
る。
２）ジプロピルケトン（すなゎちヘプタノン−４）は無
色の液体であって下記の性質を示す：比　　麿（ｄ４　
）、　　　０．８２１肺　点：１４４°Ｃ 屈折率（２２°ｃ）　：　　１．４０７３及び ３）：）メチルココベンジルアンモニウム・クロライド
は式： （式中Ｒは炭素数６〜１８個のアルキル基又はアルケニ
ル基、特に炭素数１２〜１４個のアルシル基の混合物で
あるココ（又はコプラ）基を表わす）（７）ｉ４級アン
モニウム塩である。
本発明に従う溶剤混合物は、好ましくはＣ１−０３の脂
肪族アルコール　　　　１５〜６０％プロピレンクリコ
ール　　　　　１５〜６０％及び上記の可溶化剤　　　
　　　　　５〜６０俤からなる〇 上記規定したとおりの溶剤混合物はまた、特に可溶化剤
がジメチルココベンジルアンモニウム。
クロライドである場合に、ある比率、一般に１０〜５０
％の水を含むこともできる。
溶剤混合物は一般に本発明の組成物中に該組成物の合計
重量に基づいて４０〜９９．９９重量％の範囲の量で存
在する。
本発明に従う局所皮膚治療用組成物のヒドロコルチゾン
母度は一般に組成物全重量に対して１２％を超えず、使
用する可溶化剤によるが、好ましくは０．０１〜５重量
％、特に０．５〜４重量％であるＯ 本発明による組成物は種々の形態、特にローション、シ
ャンプー、ポマード又はゲルの形態とすることができ、
局所皮膚治療に屈するあらゆる疾病の処置に使用される
。
ローションは本質的には溶剤混合物からなり、場合によ
ってはこの型の調剤用の慣用の添加物を添加し得る。
ゲルは組成物の全重量に対して０．５〜１５重量−の濃
度で用いられるシリカ、セルロース誘導体、カルボキシ
ビニルポリマー（Ｃａｒｂｏｐｏｌｇ　）−天然又は合
成ゴムのようなゲル化剤を用いて得られる。
ポマードはたとえばワセリン、ノぐラフイン油又はワッ
クスを基剤とする無水組成物である。
本発明による組成物は特に湿疹、湿疹性又は乾廖性紅皮
症、ル痒性病害、慢性紅斑性狼癒、斑状の乾癖及び類乾
癖、拡張性毎痕、日焼による又は放射線治療による紅疹
の治療に用いられる＠これらの処置は平均して１日２回
、場合によっては浸透を容易にするためｉツサージしな
がらの適用を必要とする。
以下例証・のため、ただし何ら限定的意図なしに、本発
明による組成物の数例を示す。

良好な浸透及び良好な耐薬品性が観察された。
ヒドロコルチゾン基剤の通常の使用榮件と比較して・本
発明の組成物を使用した場合には処理時間を著しく短縮
でき、かくして長期使用及び皮膚萎縮の危険が回避され
た。

Claims (1)

  1. 【特許請求の範囲】 １、炭素数１〜３個のアルコール、プロピレングリコー
    ル及び２−ヒドロキシエチルサリチレート、：）プロピ
    ルケトン及びジメチルココベンジルアンモニウムクロラ
    イドからなる群から選んだ第三の可溶化剤からなる溶剤
    混合物中にヒドロコルチゾンを可溶化状態で含有するこ
    とを特徴とするヒドロコルチゾンを有効成分とする局所
    皮膚治療用組成物＠ ２・　溶剤混合物が ｃ、　−ｃ３ｔ７１肪族アルコール　　１５ないし６０
    ％プロピレングリコール　　　　１５ないし６０％及び
    ＯＴ　　溶　化　剤　　　　　　　　５ないし６０条か
    らなる特許請求の範囲第１項記載の組成物０８、水１０
    乃至５０重ｆｉｔ％を含有する特許請求の範囲第１項又
    は第２項記載の組成物。 ４、ヒドロコルチゾンの濃度が組成物の全重量に対して
    １２％を超えず、好ましくは０．０１乃至５重量係、特
    に０．５乃至４重量−である特許請求の範囲第１項ない
    し第３項のいずれかに記載の組成物。 ５、ローション、シャン−Ｐ−１！マート９又はケルの
