Claims (1)
- 【特許請求の範囲】 1.フィルターデバイスであって以下の特徴: ハウジングが、血液、血清、または血漿を該ハウジングを通し流すための液体 流の管腔をその間規定する第1の入口および出口を有する; 該管腔が多孔壁および部分的に閉塞した管腔を有する複数の中空繊維をまた取 り囲む; 該多孔壁が肝細胞よりも小さい小孔を含む; 該中空繊維の該管腔が、肝細胞機能および生存性を支持するために、該管腔を 通して培地を流すための該ハウジング中の第2の入口および出口で液体流の連絡 を有しており;そして 該中空繊維の管腔が、生存肝細胞を含む水性マトリックスで部分的に閉塞され ており、ここで該肝細胞が非凝集細胞および凝集細胞塊の混合物である; その結果該腔と該中空繊維の内側との間の連絡が排他的に多孔繊維壁を通して である、 を含む、フィルターデバイス。 2.前記中空繊維が100〜1000μmの内径を有する、請求項1に記載のデバイス。 3.前記内径が150〜400μmである、請求項2に記載のデバイス。 4.前記内径が200〜250μmである、請求項3に記載のデバイス。 5.前記小孔が100,000kdまでの分子量を有する分子の通過を許容する、請求項 1に記載のデバイス。 6.前記培地が無血清培地である、請求項1に記載のデバイス。 7.前記マトリックスが、コラーゲン、アルギネート、キトサン、およびフィブ リンからなる群から選択される、請求項1に記載のデバイス。 8.前記マトリックスがコラーゲンである、請求項7に記載のデバイス。 9.前記細胞塊が直径30〜300μmである、請求項1に記載のデバイス。 10.前記細胞塊が直径35〜150μmである、請求項9に記載のデバイス。 11.前期細胞塊が直径40〜70μmである、請求項10に記載のデバイス。 12.前記水性マトリックスが、液体として前記繊維中に導入され、そしてイン サイチュでゲル化され、ここで該マトリックスゲルが前記生存肝細胞の存在下で 圧縮する、請求項1に記載のデバイス。 13.前記液体が1ミリリットル当たり5〜40×106細胞の濃度で肝細胞を含む 、請求項12に記載のデバイス。 14.前記濃度が1ミリリットル当たり20〜40×106細胞である、請求項13に 記載のデバイス。 15.前記濃度が1ミリリットル当たり30〜35×106細胞である、請求項14に 記載のデバイス。 16.前記細胞の半分が細胞塊の中にある、請求項13に記載のデバイス。 17.前記細胞の3分の2が細胞塊の中にある、請求項13に記載のデバイス。 18.前記管腔が前記マトリックスにより25〜90%閉塞されている、請求項1に 記載のデバイス。 19.前記管腔が33〜75%閉塞されている、請求項18に記載のデバイス。 20.前記肝細胞がブタ肝細胞である、請求項1に記載のデバイス。
