Claims (1)
- 【特許請求の範囲】 1. ヒトパピローマウイルス(HPV)E6 またはE7 タンパク質の変異体 であって、上記変異型が宿主動物中でHPVに対する体液性および/または細胞 性免疫応答を引き出し得るが、上記宿主動物中で細胞形質転換性ではない変異体 。 2. E6 またはE7 タンパク質の完全長配列の少なくとも50%を含むHP V E6 またはE7 タンパク質の欠失変異体である、請求項1記載の変異体。 3. 上記欠失変異体がE6 またはE7 タンパク質の完全長配列の約3分の2 に該当する、請求項2記載の変異体。 4. 上記欠失突然変異体がE6 またはE7 タンパク質の完全長配列のN−末 端3分の2に該当する、請求項3記載の変異体。 5. 上記欠失変変異体がE6 またはE7 タンパク質の完全長配列のC−末端 3分の2に相当する、請求項3記載の変異体。 6. E6 および/またはE7 タンパク質部分、および任意に連鎖部分を含む 融合タンパク質である、請求項1記載の変異体。 7. 上記融合タンパク質が、融合されたE6 および/またはE7 タンパク質 部分、または外来タンパク質もしくはペプチド部分に結合したE6 および/また はE7 タンパク質部分を含む、請求項6記載の変異型。 8. 上記外来タンパク質またはペプチド部分が、上記融合タンパク質の免疫 原性を強化するタンパク質もしくはペプチド部分または融合タンパク質の精製を 援助するタンパク質またはペプチド部分から選択される、請求項7記載の変異体 。 9. E6 および/またはE7 タンパク質部分が、完全長E6 またはE7 タン パク質部分もしくはその非完全長失欠突然変異体から選択される、請求項6ない し8のいずれかに記載の変異型。 10. 上記連鎖部分が、1ないし50、好ましくは1ないし20、および最 も好ましくは1ないし5アミノ酸残基を含む、請求項6ないし9のいずれかに記 載の変異体。 11. 完全長E7 タンパク質部分に融合した完全長E6 タンパク質部分を含 む融合タンパク質を含んでなる、請求項6記載の変異型。 12. E7 タンパク質のC末端3分の2である第2タンパク質部分に融合し たE6 タンパク質のN末端3分の2である第1タンパク質部分を含む融合タンパ ク質を含んでなる、請求項6記載の変異型。 13. E6 タンパク質のC末端3分の2である第2タンパク質部分に融合し たE7 タンパク質のN末端3分の2である第1タンパク質部分を含む融合タンパ ク質を含む融合タンパク質を含む融合タンパク質を含んでなる、請求項6記載の 変異体。 14. 上記E6 またはE7 タンパク質が、HPV−16、HPV−18、H PV−6およびHPV−11遺伝子型から選択される、請求項1ないし13のい ずれかに記載の変異型。 15. 上記E6 またはE7 タンパク質が、HPV−16およびHPV−18 遺伝子型から選択される、請求項14記載の変異型。 16. 宿主動物中のHPVに対する体液性および/または細胞性免疫応答を 引き出すのに使用するワクチン組成物であって、上記組成物が、請求項1ないし 15のいずれかに記載のHPV E6 またはE7 タンパク質の変異型を薬剤的に 許容されるキヤリヤーおよび/または希釈剤と共に含む、ワクチン組成物。 17. アジュバントをさらに含む、請求項16記載のワクチン組成物。 18. 宿主動物中でHPVに対する体液性および/または細胞性応答を引き 出すための方法であって、この方法が、請求項1ないし15のいずれかに記載の HPV E6 またはE7 タンパク質の変異体の有効量を宿主動物に投与すること を特徴とするする方法。 19. HPV E6 またはE7 タンパク質の上記変異型を組成物において薬 剤的に許容されるキヤリヤーおよび/または希釈剤と共に投与する、請求項18 記載の方法。 20. 上記組成物がアジュバントをさらに含む、請求項19記載の方法。 21. 上記宿主動物がヒトである、請求項18ないし20のいずれかに記載 の方法。 22. 宿主動物中でHPVに対する免疫応答を引き出すことにおける、請求 項1ないし15のいずれかに記載のHPV E6 またはE7 タンパク質の変異体 の使用。
