【発明の詳細な説明】
細胞サイクル依存性である細胞に特異的な活性化合物を用いる
中枢神経系の疾病の遺伝子療法技術分野
中枢神経系の疾病の遺伝子治療のためのDNA配列が記載されている。
このDNA配列の本質的要素は、活性化配列、プロモーターモジュールおよび
活性物質の遺伝子からなっている。
活性化配列は細胞に特異的な方法で、活性化した内皮細胞またはグリア細胞中
で活性化される。この活性化は細胞サイクルに特異的な方法でプロモーターモジ
ュールによって制御される。活性物質は、神経成長因子、ドパミン代謝酵素、お
よび/または神経細胞の保護因子である。記載されたDNA配列をウイルスまた
は非ウイルスベクターに挿入し、これに、標識細胞に親和性を有するリガンドを
補足する。
1. 中枢神経経および成長因子
個体発生を終了すると、神経細胞は十分に分化し、最早分裂することができな
い細胞からなっている。−般的には、それらは神経細胞本体と神経細胞突起とを
特徴とし、求心性(デントライト(dentrites))および遠心性(神経突起)の突
起に識別される。遠心性の神経突起は、神経細胞当たり通常は1個しか形成され
ないが、シナプスを介してその標的器官(神経細胞または他の種類の体細胞)と
接触する。
神経細胞の解剖学的構造および機能の保持は、ニューロン成長因子の存在下で
行われる。
一般的な意味において、ニューロン成長因子は向神経性因子である理解すべき
である (Massague,Cell 49,437(1987),Pusztai et al.,J.Pathol.169,19
1(1993),Ibanez et al.,PNAS 89,3060(1992),Sonoda et al.,BBRC 185,10
3(1992)の総説)。これらの因子としては、第1表のニューロン成長因子が挙
げられる。
狭義には、神経成長因子(NGF)類は、これらの因子に包含されるべきであ
る。
NGFは、NGFレセプターに結合することによって作用し、これは、特に知
覚神経線維上に形成される。NGFは、細胞内に吸収され、退化的に神経細胞本
体に輸送される(Johnson et al.,J.Neurosci.7,923(1987)。神経細胞本体
では、NGFは、サイクリックアデノシン一リン酸(cAMP)を増加させ、続
いてCa++の流出を増加させ(Schubert et al.,Nature 273,718(1978)、更に
、イノシトール脂質代謝によりジアシルグリセロールの方種およびタンパク質キ
ナーゼCの活性化、およびイノシトール三リン酸放出によるCA++の細胞内放出
をもたらすと思われる(Abdel-Latif,Pharmacol.Rev.38,227(1986))。
特異的、特にシグナル導入タンパク質のリン酸化により、この誘導から、機能
が変化する。これにより、軸索の成長に関与するタンパク質の形成が増加する。
これらのタンパク質としては、チャーティン(chartin)タンパク質(Black et al.
,J.Cell Biol.103,545(1986))、タウタンパク質およびチューブリン(Drubin
et al.,J.Cell Biol.101,1799(1985))が挙げられる。従って、α−チュー
ブリンおよびβ−チューブリン神経フィラメントタンパク質(NF−L、NF−
M、およびNF−H)およびペリフェリン(Portier et al.,Devi Neuroscience
6,215(1983)),Parysek et al.,J.Neurosci.7,781(1987))の合成が増加し
ている。同時に、コリンアセチルトランスフェラーゼ、アセチルコリンエステラ
ーゼ、およびニューロン特異性エノラーゼのような神経系で重要な酵素、の濃度
も増加する(Vinores et al.,J.Neurochemistry 37,597(1981),
Rydel et al.,J.Neurosci.7,3639(1987))。
また、ニューロテンシン(Tischler et al.,Reg.Pept.3,415(1982))および
ニューロペプチドY(Allen et al.,Neurosci.Lett.46,291(1984)のような
神経伝達物質、およびアセチルコリンレセプター(Mitsuka et al.,Brain Res.
314,255(1984))およびエンセファリンレセプター(Inoue et al.,J.Biol.Che
m.257,9238(1982))のような神経伝達物質レセプターの濃度も増加する。同時
に、シナプシン1の濃度が増加する(Romano et al.,J.Neurosci.7,1300(198
7))。
最終的分析では、NGFは神経細胞の機能状態を保持する。同時に、NGFは
、軸索の成長を開始し、促進する。この神経原性およびシナプス原性活性には、
NGFが常に含まれていることが必要である(Smith,Science 242,708(1988),
Mitchison et al.,Neuron 1,761(1988))。
これは、特に毛様の神経親和性因子(CNTF)について報告されている(Lin
et al.,Drugs of the Future 19,557(1994))。
神経成長因子の神経親和性活性は、特に神経細胞の損傷に関して、例えば軸索
の外科的分断に関して、実験的に立証されている。CNTFを切断した神経の基
端部に局所的に投与すると、外科的介入の後に死亡する神経細胞の割合は著しく
減少する(Sendtner et al.,Nature 345,440(1990))。同時に、例えば神経ペプ
チド物質Pの濃度は、CNTFを投与すると、脊髄神経節で著しく上昇する。座
骨神経に損傷を受けたラットは、CNTFを皮下投与すると、運動活性の回復が
促進される(Lin et al.,Drugs of the Future 19,557(1994))。しかしながら
、神経親和性因子の全身投与では、脊髄にありかつ血液−脳関門によって保護さ
れている運動ニューロンが、軸索であって、関門の外側でも機能を示しかつこれ
によって神経親和性因子を吸収することができるものを有する場合にのみ有効で
ある(Apfel et al.,Brain Res.605,1(1993))。
神経細胞が、血液−脳関門の他の側、または反対側まで損傷を受けている場合
には、神経親和性因子を頭蓋内に投与する必要がある。この方法では、視床軸索
の分断の後の基部の視床ニューロンの逆行性変性(retrograde generation)は、
実験により防止することができる(Clatterbuch et al.,PNAS 90,2222(1993))
。しかしながら、再生過程を最良のものとする前提条件は、損傷を受けた神経細
胞の部位に一定の神経親和性因子が存在することである。外科的損傷または損傷
の軽減の時点に局所投与を行うことは可能であるが、これは外科的介入が一旦終
了してしまえば、行うことは困難であるかまたはほとんど不可能である。例えば
、鈍い外傷(blunt trauma)または毒素によるCNSに対する散在性の損傷では、
目標を定めて投与を行う機会は極めて僅かに限定される。
外傷、免疫および毒素による影響を受けた結果として、グリア細胞が刺激され
てTNFαを産生することができる。このTNFαは、その時点では神経細胞お
よびグリア細胞にとって有毒である(Owens et al.,Immunol.Today 15,566(19
94))。
活性化合物をCNSにできるだけ長時間存在させるため、神経親和性の活性化
合物を発現する目的でイン・ビトロで導入した細胞(線維芽腫、内皮細胞および
筋原細胞)を頭蓋内に注射する試みが行われている。この目的は、神経親和性の
活性化合物を用いて、例えばパーキンソン病または痴呆症で外傷によりまたは退
行的に損傷を受けた神経細胞の再生および機能を向上させることである。特に、
パーキンソン病の場合には、イン・ビトロで導入された細胞を注射して、チロシ
ンヒドロキシラーゼおよびドーパデカルボキシラーゼのような神経親和性酵素を
分泌させ(Kopin,Ann.Rev.Pharmacol.Toxicol.32,467(1993),Fisher et a
l.,Physiol.Rev.11,582(1993),Jiao et al.,Nature 362,450(1993))、ま
たはヒト胎児性のドパミン作用性の黒質ニューロンを注射する(Loewenstein,Bi
o/Technology 12,1075(1994))。
しかしながら、この種の細胞は、限定された量でしか利用できない。一方、胎
児性細胞の使用には、重大な倫理上の問題が起きている。
代替法として、ベクターを直接脳に注射して、脳に細胞を形質導入して、所望
な活性化合物を発現させることの可能性が検討されている(During et al.,Scie
nce 266,1399(1994))。しかしながら、これらのベクターは細胞特異性をまった
く示さないので、神経細胞がベクターによる感染またはトランスフェクションに
よって損傷を受ける危険性がかなりある。
2.発明の説明
本発明は、活性化合物であって、医薬品として患者に局所および全身投与する
ことができ、かつこれによって神経特異性因子を神経細胞の損傷部位に比較的長
期間に亙って産生するものに関する。この種の神経特異性因子は、神経細胞をそ
れ以上損傷を受けることから保護しかつ神経細胞の再生をもたらす神経親和性因
子であることができる。しかしながら、神経特異性因子は、チロシンヒドロキシ
ラーゼおよびドーパデカルボキシラーゼのようなチロシンからドパミンの合成に
関与する酵素であることもできる。また、神経特異性因子は、TNFαを阻害し
または中和する物質であることができる。
この活性化合物の中心成分は、下記の要素からなるDNA構築物である。
(本出願明細書の全文において、DNAは、相補性(cDNA)配列およびゲノ
ムDNA配列の両方に共通の用語として用いられる。)
2.1. 活性化配列の選択
活性化配列(UAS=上流活性化配列)は、標的細胞において形成されまたは
活性を有する転写因子と相互作用するヌクレオチド配列(プロモーター配列また
はエンハンサー配列)であって、内皮細胞またはグリア細胞において形成されま
たは活性である転写因子と相互作用する配列と理解すべきである。
CMVエンハンサーまたはCMVプロモーター(EP 0173 177)、
SV40プロモーター、または当業者に知られている任意の他のプロモーター配
列またはエンハンサー配列を、活性化配列として用いることができる。
しかしながら、本発明においては、好ましい活性化配列としては、特に内皮細
胞またはグリア細胞で形成されるタンパク質をコードする遺伝子由来の遺伝子制
御配列または要素が挙げられる。
a) 内皮細胞で活性化される活性化配列
これらのタンパク質の幾つかは、Burrows et al.(Pharmac.Therp.64,155(
1994)およびPlate et al.(Brain Pthol.4,207 (1994))によって記載されてい
る。特に、これらの内皮細胞特異性タンパク質としては、例えば下記のものが挙
げられる。
脳特異性の内皮グルコース−1輸送体
脳の内皮細胞は、D−グルコースの脳内への内皮経由輸送を行う目的でこの
輸送体を極めて強力に発現する(Gerhart et al.,J.Neurosci.Res.22,464(1

エンドグリン
エンドグリンは、シグナル伝達性ではないTGFβであると思われる(Gango
s et al.,J.Biol.Chem.265,8361(1990),Moren et al.,BBRC 189,356(19
92),Cheifetz,J.Biol.Chem.267,19027(1992))。通常の内皮には少量含ま
れているが、増殖内皮では更に強力に発現する(Westphal et al.,J.Invest.D
erm.100,27(1993),Burrows et al.,Pharmac.Ther.64,155(1994))。プロ

2.2 プロモーターモジュールの選択
細胞サイクルによって制御されるプロモーターモジュールは、例えばヌクレオ
チド配列−CDE−CHR−Inrであると理解すべきである。このプロモータ
ーモジュールの本質的機能は、細胞サイクルのGO/G1相における活性化配列
の機能を抑制し、S/G2相および続いて増殖細胞における細胞サイクル特異発
現を確保することである。
プロモーターモジュールCDE−CHR−Inrは、ヒトcdc25Cプロモ
ーターのG2特異性発現の詳細な研究に関連して見いだされた。出発点は、細胞
サイクルのG1相におけるプロモーターの遮断に関与する制御要素(細胞サイク
ル依存性要素;CDE)を見いだしたことであった(Lucibelloら,EMBO U.14,
132(1995))。ゲノムジメチル硫酸(DMS)フットプリント法および機能分析法
(functional analyses)(第1および2図)を用いて、CDEはG1に特異的に
リプレッサー(CDE結合因子;CDF)に結合することによって非増殖(G0
)細胞での転写を阻害することが示された。基底プロモーター(basal promoter)
の領域に配置されているCDEは、その抑制機能において、「上流の活性化配列
」(UAS)によって変化する。
これにより、CDE結合因子は、細胞サイクル依存的に、すなわち非増殖細胞
および細胞サイクルのG1相において、5′に結合した活性化物質タンパク質の
転写活性化作用を阻害するという結論が得られた(第3図)。
この結論を更に実験によって確かめることができ、ウイルス性の非細胞サイク
ルによって制御される初期のSV40エンハンサーをcdc25最小プロモータ
ー(minimum promoter)(CDEおよび3′位の開始部位)と融合することによっ
て、キメラプロモーターの細胞サイクルが明確に制御された(第4図)。次にc
dc25Cエンハンサーについて検討したところ、細胞サイクル依存的にCDF
によって制御される転写因子は、NF−Y(CBF)(Dorn et al.,Cell 50,8
63(1987),van Hijisduijnen et al.,EMBO J.9,3119(1990),Coustry et al.
,J.Biol.Chem.270,468(1995))、Sp1(Kadonaga ら,TIBS 11,10(1986)
、および新規でありかつCBS7に結合する転写因子であることが明らかになっ
た。この研究において得られたもう一つの興味ある知見は、cdc25Cエンハ
ンサー内のNF−Yのみが、少なくとも1種類の他のNF−Y複合体またはCI
Fと協同して効率的に転写を活性化することが見られたことである。NF−Yも
Sp1も両方ともグルタミン含量の高い活性化物質のクラスに属し、抑制の機構
(例えば、特定の基底転写因子またはTAFとの相互作用または干渉)に対する
重要な情報を提供する。
cdc25C、サイクリンAおよびcdc2のプロモーター配列を比較したと
ころ、幾つかの領域で相同性が見られた(第5図)。3種類のプロモーター(含
まれている多様性は機能上関連はない)総てに保存されているのは、CDEだけ
でなく、隣接Ycボックスも同様である。予想されるように、これらの領域は総
てイン・ビボでタンパク質結合を示し、このタンパク質結合はCDEの場合には
細胞サイクル依存性であった。更に、3種類のプロモーターは総て、CDEの突
然変異によって制御解除されることも示された(第2表)。cdc25C、サイ
クリンAおよびcdc2配列を比較したところ、直ちにCDEの3′の領域に著
しい類似性のあることも明らかになった(細胞サイクル遺伝子相同領域;CHR
)(第5図)。この領域は機能上はCDEと同様に重要であるが(第2表)、そ
れはイン・ビボでのDMSフットプリント法の実験では明らかではない。
これに対する可能な説明は、この因子とDNAの小さな溝(minor grooves)と
の相互作用である。電気泳動移動度シフト分析(EMSA)実験の結果は、CD
EとCHRとは一緒にタンパク質複合体であるCDFに結合することを示してい
る。これらの観察は、グルタミン含量の高い活性化物質のCDFによって媒介さ
れる抑制は、細胞サイクルによって制御される転写において頻繁に起きる機構で
あることを示している。
しかしながら、cdc25Cプロモーターを制御するのに重要なものは、CD
E−CHR領域だけでなく、基底プロモーターのヌクレオチド配列(位置<−2
0〜>+30、第1図を参照されたい)内の開始部位(位置+1)のものでもあ
ることは明らかである。YY−1のイン・ビトロでの結合部位を含むこの領域で
の突然変異(Seto およびShenk,Nature 354,241(1991),Usheva およびShenk,
Cell 76,1115(1994))により、完全に制御解除される。CDE−CHRが基底プ
ロモーターに近接していることを考慮すれば、その結果としてCDFは基底転写
複合体と相互作用すると思われる。
2.3. 神経特異性因子の選択
a) ニューロン成長因子
本発明において、神経特異性因子は、ニューロン成長因子をコードするD

しかしながら、本発明において、一方では、上記のサイトカインおよび成長因
子、またはレセプターの細胞外残基と、他方ではヒト免疫グロブリンのFc残基
との間に形成した融合タンパク質のDNA配列を、活性物質として用いることも
できる。この種のDNA配列およびその調製は、EP 0 464 633 A
1号明細書に記載されている。
2.4. 数個の神経特異性因子の組合せ
本発明は、活性化合物であって、同一な神経特異性因子(A,A)または異な
る神経特異性因子(A,B)のDNA配列の組合せが含まれているものにも関す
る。2個のDNA配列の発現のためには、「内部リボソームエントリー部位」(
IRES)のcDNAは、制御要素として挿入するのが好ましい。
この種のIRESは、例えばMountford およびSmith(TIG 11,179(1995)、
Kaufman ら,Nucl.Acids Res.19,4485(1991)、Morgan et al.,Nucl.Acids
Res.20,1293(1992)、およびDirks et al.,Gene 129,247(1993)、Pelletie
r およびSonenberg,Nature 334,320(1988)、Sugimoto et al.,BioTech.12
,694(1994)によって記載されている。
従って、ポリオウイルスのIRES配列のcDNA(5′URTの位置<14
0〜>630;Pelletier and Sonenberg,Nature 334,320(1988))を用いて、
抗炎症物質AのDNA(3′末端)および抗炎症物質BのDNA(5′末端)を
連結することができる。
組み合わせによっては、この種の活性化合物は本発明の意味において相加的(
A+A,A+B1)または相乗的効果を示す。
2.5. ベクターの構築
新規DNA構築物を、当業者が慣れた方法で完全なベクターとする。このベク
ターはウイルスまたは非ウイルス性のものであることができる。例えば、新規な
DNA構築物をウイルスベクターに挿入し(この場合には、D.Jolly,Cancer
Gene Therapy 1,51(1994)を参照されたい)、またはプラスミドとして用いる
。ウイルスベクターまたはプラスミドをコロイド状分散液、例えばリポソームと
複合体を形成し(Farhood et al.,Annals of the New York Academy of Science
716,23(1994))、またはポリリシン/リガンド抱合体または他の慣用されてい
る補助物質と共に医薬品として処方することもできる(Curiel et al.,Annals o
f the New York Academy of Science 716,36(1994))。
2.6. リガンドの選択
ウイルスおよび非ウイルス性ベクターを、リガンドにより補足することができ
る。増殖する内皮細胞の表面に結合する物質は、例えばポリリシン/リガンド抱
合体におけるリガンドとして好ましい。これらの物質としては、例えばBurrows
et al.(Pharmac.Ther.64,155(1994))またはEP0 408 859 A2
号明細書9)に記載されているように、内皮細胞の膜構造に対する抗体または抗体
断片が挙げられる。これらの物質としては、特にVEGFレセプターに対する抗
体が挙げられる。
ネズミモノクローナル抗体は、人体に適用される形態で用いるのが好ましい。
人体への適用は、Winter et al.(Nature 349,293(1991))およびHoogenbooms e
t al.(Rev.Tr.Transfus.Hemobiol.36,19(1993))によって記載された方法
で行われる。抗体断片は、当該技術分野の状況に従って、例えばWinter et al.
(Nature 349,293(1991))、Hoogenbooms et al.(Rev.Tr.Transfus.Hemobiol
.36,19(1993))、Girol(Mol.Immunol.28,1379(1991)またはHuston et al.
(Int.Rev.Immunol.10,195(1993))によって記載された方法で調製される。
更に、これらの物質としては、内皮細胞上の膜構造または膜レセプターに結合
する総ての物質が挙げられる。活性物質としては、例えば、成長因子、またはそ
れらの断片またはそれらの構成配列であって、内皮細胞によって発現されるレセ
プター、例えばPDGF、bFGF、VEGFおよびTGFβに結合するものが
挙げられる(Pusztain et al.,J.Pathol.169,191(1993))。更に、これらの物
質としては、末端にマンノースを有し、かつナイロン費細胞のマンノース6−リ
ン酸レセプターに結合する物質が挙げられる(Perales et al.,Eur.J.Biochem
.226,225(1994))。
更に、これらの物質としては、活性化したおよび/または増殖する内皮細胞に
結合する付着分子が挙げられる。この種の付着分子、例えばSLex、LFA−
1、MAC−1、LECAM−1またはVLA−4は、既に記載されている(Au
gustin-Voss et al.,J.Cell Biol.119,483(1992),Pauli et al.,Cancer M
etast.Rev.9,175(1990)およびHonn et al.,Cancer Metast.Rev.11,353(
1992)の総説)。
更に、グリア細胞の表面に結合する物質は、リガンドと考えるべきである。
これらの物質としては、例えばMirsky et al.(Cell and Tissue Res.240,7
23(1985))、Coakham et al.(Prog.Exp.Tumor Rex.29,57(1985))およびMcKe
ever et al.(Neurobiol.6,119(1991))によって報告されているように、グリ
ア細胞の膜構造に対する抗体または抗体断片が挙げられる。これらの膜構造とし
ては、神経付着分子、例えばN−CAM、特にそのポリペプチド鎖C(Nybroe et
al.,J.Cell Biol.101,2310(1985))。
これらの物質としては、グリア細胞の膜構造または膜レセプターに結合する総
ての活性化合物が挙げられる。例えば、これらの物質としては、末端にマンノー
スを有しかつマンノース6−リン酸レセプター(Perales et al.,Eur.J.Bioch
em.226,225(1994))、インシュリンおよびインシュリン様成長因子(Merrill et
al.,J.Clin.Endocrin.Metab.71,199(1990))、PDGF(Ek et al.,Natu
re 295,419(1982))に結合するもの、および関連の膜レセプターに結合するこれ
らの成長因子の断片が挙げられる。
2.7. 活性化合物の調製
新規な活性化合物の調製を、下記の実施例によって更に詳細に説明する。
a) キメラプロモーターエンドセリンICDE−CHR−Inrの構築
ヒトエンドセリン−1プロモーター(位置<−170〜>−10)またはTA
TAボックスを除去することによって切断した変異体(位置<−170〜>−4
0)をその3′末端で、ヒトcdc25C遺伝子のCDE−CHR−Inrモジ
ュール(位置<−20〜>+121)の5′末端に連結する(第6図)。この連
結は、当業者に知られておりかつ市販されている酵素を用いて行われる。
b) 活性化合物の中心成分を含むプラスミドの構築
この方法で調製したキメラエンドセリン−1リプレッサーモジュール転写単位
を、その3′末端で、長さが152アミノ酸のIL−1レセプターアンタゴニス
トの完全コード領域を含むDNAの5′末端に連結する(位置<25〜>557
Eisenberg et al.,Nature 343,341(1990))。このDNAは、分泌に必要なシ
グナル配列(25個のN−末端アミノ酸)をも含んでいる。転写制御単位および
IL−1レセプターアンタゴニストDNAを、直接(Yovandich et al.,Hum.Ge
ne Ther.6,603(1995))またはコロイド分散液系で用いて、イン・ビボでのトラ
ンスファーを行うことができるpUC19/19またはBluescrip由来
のプラスミドにクローンする。
または、一緒に結合した転写制御単位およびIL−1レセプターアンタゴニス
トDNAを、当業者によく知られているウイルスベクターまたは他の非ウイルス
ベクターに移すことができる。
2.8. 活性化合物の活性
例えば腫瘍の部位で局所投与の後、または頭蓋内またはクモ膜下投与、または
全身、好ましくは静脈内または動脈内投与の後、本発明の活性化合物は、組織特
異性エンハンサーおよび基底プロモーターによって、独占的にではないにしても
主に増殖する内皮細胞だけまたは増殖するグリア細胞が、神経特異性因子を分泌
することができるようになる。この種の内皮細胞の増殖またはグリア細胞の増殖
は、神経の損傷をも同時に引き起こした領域および組織損傷に対する反応として
予想されるものである。従って、新規な活性化合物は、神経損傷の部位における
神経特異性因子の濃度を高くすることができる。
活性化合物の細胞特異性およびその細胞サイクル特異性により高度の安全性が
約束されるので、これを高投与量で用い、必要ならば、数日または数週間の間隔
で繰り返し用いて、神経損傷の治療を行うこともできる。

- 【特許請求の範囲】 1. 中枢神経系の疾患の予防または治療のための活性化合物であって、活性 化配列と、細胞サイクルによって制御されるプロモーターモジュールと、神経特 異製因子のDNA配列とからなるDNA構築物を含んでなる、活性化合物。 2. プロモーターモジュールがCDE−CHR−Inr要素を有し、かつc dc25Cプロモーター領域(ヌクレオチド配列:GCTGGCGGAAGGT TTGAATGGTCAACGCCTGCGGCTGTTGATATTCTTG )の位置<−20〜>+30を含んでなり、ここで、CDEが細胞サイクル依存 性要素(ヌクレオチド配列:TGGCGG)からなり、CHRが細胞サイクル遺 伝子相同領域(ヌクレオチド配列:GTTTGAA)からなり、Inrが開始部 位(位置+1)および開始に重要な隣接配列からなるものであり、またはこのプ ロモーターモジュールの機能上活性な変異体および突然変異体である、請求の範 囲第1項に記載の活性化合物。 3. 内皮細胞、またはグリア細胞で形成される転写因子によって制御される 活性化配列を含んでなる、請求の範囲第1項に記載の活性化合物。 4. 活性化配列として、 CMVプロモーター、CMVエンハンサーまたはSV40プロモーター、また は 脳に特異的な内皮グルコース−1−輸送体、エンドグリン、VEGFレセプタ ー1または2、レセプターチロシンキナーゼtil−1またはtil−2、B6 1レセプター、B61リガンド、エンドセリン、特にエンドセリンBまたはエン ドセリン1、マンノース−6−リン酸レセプター、IL−1αまたはIL−1β 、IL−1レセプター、VCAM−1またはvon Willebrand因子、または GATA−2のような結合部位が5′−TTATCT−3′である内皮細胞中 で優先的または選択的に活性である転写因子のオリゴマー化した結合部位、また は Schwann 細胞に特異的なペリアキシン、グルタミンシンテターゼ、グリア特異 性タンパク質、グリア細胞タンパク質S100b、インターロイキン−6、5− ヒドロキシトリプタミンレセプター、TNFα、IL−10またはインシュリン 様成長因子レセプター、または VEGFのプロモーター配列、またはVEGFのエンハンサー配列、またはc −SRCまたはv−SRCのcDNA配列であってVEGF遺伝子を制御するも の を含んでなる、請求の範囲第2項に記載の活性化合物。 5. 神経特異性因子のDNA配列が、 ニューロン成長因子、特にFGF、NGF、BDNF、NT−3、NT−4ま たはCNTF、または TGFβ、可溶性TNFレセプター、IL−10、可溶性IL−1レセプター 、または可溶性IL−6レセプター、またはL−1レセプターアンタゴニスト、 またはサイトカイン(例えば、TGFβ、IL−10またはIL−1レセプター アンタゴニスト)または可溶性サイトカインレセプター(例えば、TNFレセプ ター、IL−6レセプターまたはIL−1レセプター)から形成された融合タン パク質、または免疫グロブリンのFc残基、または チロシンヒドロキシラーゼまたはドーパデカルボキシラーゼをコードするもの である、請求の範囲第1〜3項のいずれか1項に記載の活性化合物。 6. 2個の同一または2個の異なる神経特異性因子のDNA配列を含み、2 個のDNA配列が互いに内部リボソームエントリー部位のDNA配列を介して接 続してされてなる、請求の範囲第1〜5項のいずれか1項に記載の活性化合物。 7. ベクターに挿入された、請求の範囲第1〜6項のいずれか1項に記載の 活性化合物。 8. ベクターがウイルスである、請求の範囲第7項に記載の活性化合物。 9. ウイルスがレトロウイルス、アデノウイルス、アデノ関連ウイルス、単 純ヘルペスウイルス、またはワクシニアウイルスである、請求の範囲第8項に記 載の活性化合物。 10. プラスミドに挿入された、請求の範囲第1〜6項のいずれか1項に記 載の活性化合物。 11. コロイド分散系で調製された、請求の範囲第7〜10項のいずれか1 項に記載の活性化合物。 12. コロイド分散系がリポソームである、請求の範囲第11項に記載の活 性化合物。 13. コロイド分散系がポリリシンリガンドである、請求の範囲第12項に 記載の活性化合物。 14. 内皮細胞またはグリア細胞の膜構造に結合するリガンドで補足された 、請求の範囲第7〜13項のいずれか1項に記載の活性化合物。 15. リガンドが、 ポリクローナルまたはモノクローナル抗体、またはその抗体断片であって、そ の可変ドメインによって、内皮細胞またはグリア細胞の膜構造に結合するもので あるか、または 末端にマンノースを有する物質、サイトカインまたは成長因子、またはその断 片または構成配列であって、内皮細胞またはグリア細胞上のレセプターに結合す るもの である、請求の範囲第14項に記載の活性化合物。 16. 膜構造が、 成長因子、例えばIL−1、FGF、PDGF、VEGF(特にFIT−1お よびKDR)、TGFβ、インシュリンまたはインシュリン様成長因子(ILG F)のレセプターからなり、または 付着分子、例えばSleX、LFA−1、MAC−1、LECAM−1または VLA−4からなり、または マンノース6−リン酸レセプターからなる、請求の範囲第15項に記載の活性 化合物。 17. 静脈内または動脈内注射、傷害の部位への局所投与、または中枢神経 系の腔部への注射のための製剤とされた、請求の範囲第1〜16項のいずれか1 項に記載の活性化合物。
