Claims (1)
- 【特許請求の範囲】 1. そのゲノム中に外来DNA配列が挿入される親ウイルスに由来する組換 えウイルスベクターを原核細胞において作製する方法であって、(i)親ウイル スの上記ゲノムのすべてまたは一部を含んだ第一DNA断片と、(ii)(i)に 相同的なフランキング配列AおよびBにより囲まれた上記外来DNA配列を含ん だ第二DNA断片との分子間組換えにより作製する方法。 2. 親ウイルスがアデノウイルス、レトロウイルス、アデノ随伴ウイルス、 ポックスウイルスおよびヘルペスウイルスからなる群より選択される、請求項1 に記載の方法。 3. 親ウイルスがヒト、イヌ、トリ、ウシ、ネズミ、ヒツジ、ブタまたはサ ル起源のアデノウイルスであるか、あるいはハイブリッドアデノウイルスである 、請求項2に記載の方法。 4. 親ウイルスがイヌ起源のCAV‐2型アデノウイルスである、請求項3 に記載の方法。 5. 親ウイルスがヒト起源の血清型Cアデノウイルスである、請求項3に記 載の方法。 6. 親ウイルスがヒト起源の5型アデノウイルスである、請求項5に記載の 方法。 7. 外来DNA配列が、凝固因子、成長ホルモン、サイトカイン、リンホカ イン、腫瘍抑制ポリペプチド、細胞レセプター、細胞レセプターに対するリガン ド、プロテアーゼ阻害剤、抗体、毒素、免疫毒素、ジストロフィンおよびCFT Rタンパク質のような細胞イオンチャンネルに関与するポリペプチドからなる群 より選択される治療対象のポリペプチドをコードしている、請求項1〜6のいず れか一項に記載の方法。 8. 相同的フランキング配列AおよびBが、長さ10bp〜10kb、有利 には20bp〜5kb、好ましくは30bp〜2kb、絶対的に好ましくは40 bp〜1kbである、請求項1〜7のいずれか一項に記載の方法。 9. 第一DNA断片が外来配列の挿入領域で線状化されている、請求項1〜 8のいずれか一項に記載の方法。 10. 複製に欠陥のある組換えウイルスベクターの作製に関する、請求項1 〜9のいずれか一項に記載の方法。 11. E1、E2およびE4領域から選択される、複製に必須な少くとも1 つの領域のすべてまたは一部を欠いた組換えアデノウイルスベクターの作製に関 する、請求項10に記載の方法。 12. 組換えアデノウイルスベクターがE3領域のすべてまたは一部を更に 欠いている、請求項11に記載の方法。 13. 少くとも20kbの組換えウイルスベクターの作製に関する、請求項 1〜12のいずれか一項に記載の方法。 14. 少くとも30kbの組換えウイルスベクターの作製に関する、請求項 13に記載の方法。 15. (i)親ウイルスのゲノムのすべてまたは一部を含んだ第一DNA断 片、(ii)5′末端にフランキング配列Aを備えた対象DNA配列の第一部分を 含んだ第二DNA断片、および(iii)3′末端にフランキング配列Bを備えた対 象DNA配列の第二部分を含んだ第三DNA断片(上記第二および第三DNA断 片はそれらの各3′および5′末端に相同的配列を含んでいる)の間の分子間組 換えによる、請求項1〜14のいずれか一項に記載の方法。 16. 1以上のヌクレオチドまたは外来DNA配列の欠失、変異および/ま たは置換による修飾をウイルスゲノム中に導入する、請求項1〜15のいずれか 一項に記載の方法。 17. 原核細胞が大腸菌のrecBC sbcBC株に由来している、請求 項1〜16のいずれか一項に記載の方法。 18. 請求項1〜17のいずれか一項に記載の方法を実施することによって 得られた組換えウイルスベクターを含む感染性ウイルス粒子の作製方法であって 、 (a)上記組換えウイルスベクターがトランスフェクトされた哺乳動物細胞を 作るために哺乳動物細胞中に導入され、 (b)上記トランスフェクトされた哺乳動物細胞が上記ウイルス粒子の生産を 行うために適切な条件下で培養され、そして (c)上記ウイルス粒子が工程(b)で得られた細胞培養物から回収される ことを含む方法。 19. 人体の治療または外科処置に関する、請求項18に記載の方法により 作製された感染性ウイルス粒子または請求項1〜17のいずれか一項に記載の方 法により作製された組換えウイルスベクターの使用。 20. 遺伝子治療による人体の治療または外科処置に関する、請求項19に 記載の使用。 21. 請求項18に従い作製された感染性ウイルス粒子または請求項1〜1 7のいずれか一項に記載の方法により作製されたウイルスベクターの治療上の有 効量を、薬学的観点から許容されるビヒクルと一緒に含んでなる、医薬組成物。 22. 細胞系における対象DNA配列の発現に関する、請求項18に記載の 方法により作製された感染性ウイルス粒子または請求項1〜17のいずれか一項 に記載の方法により作製された組換えウイルスベクターの使用。 23. 分子間相同組換えによるプラスミドベクター中へのDNA断片のクロ ーニングに関する、大腸菌のrecBC sbcBC株の使用。
