Description
Claims (1)
- 【特許請求の範囲】 1.異種遺伝子配列を真核細胞性宿主ゲノムに挿入し、そして該遺伝子配列を発 現させるDNA構築物であって、以下の配列を含む、DNA構築物: 5’ X−A−P−B−Q−C−Y 3’ ここで、 XおよびYは、内因性宿主遺伝子の一部と実質的に相同であり、 Pは、リボゾーム内部エントリー部位(IRES)であり、 Qは、異種遺伝子配列であり、 A、B、およびCは、別々に任意のリンカー配列である。 2.XおよびYが、前記A−P−B−Q−C配列を前記宿主ゲノムに挿入するた めに、該宿主ゲノムとの相同組換えを受けるのに十分な長さである、請求項1に 記載のDNA構築物。 3.XおよびYがそれぞれ少なくとも1000塩基対の長さである、請求項2に記載 のDNA構築物。 4.前記宿主細胞が動物細胞である、請求項1、2、または3に記載のDNA構築 物。 5.XおよびYが、内因性遺伝子の発現を調節する宿主エレメントを含む、請求 項4に記載のDNA構築物。 6.前記リンカー配列A、B、およびCのいずれかまたは全てが存在しない、請 求項2〜5のいずれかに記載のDNA構築物。 7.前記遺伝子コーディング配列の3’末端(下流)にポリアデニル化シグナル をさらに含む、請求項2〜6のいずれかに記載のDNA構築物。 8.前記IRESの5’(上流)に、例えばウサギb-グロビンスプライスアクセプタ ーのようなスプライスアクセプターをさらに含む、請求項2〜7のいずれかに記 載のDNA構築物。 9.前記スプライスアクセプターが、前記遺伝子コーディング配列のイントロン 配列への機能的組み込みを可能にする、請求項8に記載のDNA構築物。 10.Xの5’(上流)に短縮/切断/転写ターミネーター配列をさらに含む、請 求項2〜9のいずれかに記載のDNA構築物。 11.前記短縮/切断/転写ターミネーター配列が、スプライスアクセプターおよ びポリアデニル化シグナルを含む、請求項10に記載のDNA構築物。 12.前記転写ターミネーターが、上流マウス配列またはSV 40のポリアデニル化シグナルのようなポリA配列である、請求項10または11 に記載のDNA構築物。 13.前記遺伝子コーディング配列が抗生物質耐性のような選択性マーカーをコ ードし、該遺伝子コーディング配列を前記宿主ゲノムに挿入した細胞の選択を容 易にする、請求項2〜12のいずれかに記載のDNA構築物。 14.前記IRESが翻訳終結コドンの前にある、請求項2〜13のいずれかに記載 のDNA構築物。 15. XおよびYがプロモーターエレメントを含まない、請求項2〜14のい ずれかに記載のDNA構築物。 16.XおよびYが、特定の発現特性(特性E)に従って第1の細胞(細胞D) 内で内因性遺伝子の発現を調節するエレメントを含むような長さであり、そして 前記構築物が、第2の細胞(細胞T)のDNAへのランダムな組み込みに適応して 特性Eに従って細胞T内で前記遺伝子配列を発現する、請求項1に記載のDNA構 築物。 17.前記遺伝子コーディング配列が選択性マーカーをコードしている、請求項 16に記載のDNA構築物。 18.前記リンカー配列A、B、およびCのいずれかまたは全てが存在しない、 請求項16〜17のいずれかに記載のDNA構築物。 19.前記遺伝子コーディング配列の3’末端(下流)にポリアデニル化シグナ ルを含む、請求項16〜18のいずれかに記載のDNA構築物。 20.前記IRESの5’(上流)に、スプライスアクセプターを含む請求項16〜 19のいずれかに記載のDNA構築物。 21.前記スプライスアクセプターが、前記遺伝子コーディング配列のイントロ ン配列への機能的組み込みを可能にする、請求項20に記載のDNA構築物。 22.遺伝子コーディング配列を宿主細胞または宿主生物内で発現させるための 請求項2〜21のいずれかに記載の使用。 23.前記宿主が、動物細胞、胚幹細胞、および受精卵から選択される、請求項 22に記載のDNA構築物の使用。 24.遺伝子コーディング配列を宿主細胞ゲノム中の標的内因性遺伝子に挿入し 、そして該コーディング配列を発現させる方法であって、該宿主細胞を請求項1 〜15のいずれかに 記載のDNA構築物を含むベクターで形質転換することを包含する、方法。 25.遺伝子コーディング配列を宿主細胞のゲノムに挿入し、該コーディング配 列を発現させる方法であって、該ゲノムに請求項16〜21に記載のDNA構築物 をランダムに組み込むことを包含する、方法。 26.遺伝子コーディング配列を宿主細胞または宿主動物内で発現させる方法で あって、以下の工程: 1.請求項2〜21のいずれかに記載のDNA構築物を作製する工程、 2.該構築物を宿主細胞に挿入する工程、および 3.該遺伝子コーディング配列を発現している細胞または動物を同定する工程 、 を包含する、方法。 27.前記DNA構築物が、前記宿主のゲノムと相同組換えを行うのに適している 、請求項26に記載の方法。 28.前記DNA構築物が、前記宿主のゲノムへのランダムな組み込みに適してい る、請求項26に記載の方法。 29.請求項2〜21のいずれかに記載のDNA構築物を受精卵 に注入する工程を包含するトランスジェニック動物の作成方法。 30.請求項2〜21のいずれかに記載のDNA構築物を用いて挿入された遺伝子 コーディング配列を含む細胞または動物。 31.前記子孫が前記導入された遺伝子コーディング配列を遺伝した、請求項3 0に記載の細胞または動物の子孫。 32.請求項2〜15のいずれかに記載のDNA構築物を含むベクター。 33.請求項16〜21のいずれかに記載のDNA構築物を含むベクター。
