Claims (1)
- 【特許請求の範囲】 1.化合物の毒性を測定する、または毒性化合物に対する抗毒素を同定するため の診断用キットであって、該キットが複数の細菌宿主を包含し、該宿主のおのお のがストレスに応答するプロモーターを有し、そして該プロモーターが、アッセ イ可能な産物をコードし、該プロモーターと異なる遺伝子に作動可能なように連 結し、該複数の宿主が、以下のそれぞれに応答する少なくとも1つのプロモータ ーを包含する、キット:酸化還元ストレス、DNAストレス、タンパク質ストレス 、エネルギーストレス、およびpHストレス。 2.請求項1に記載の診断用キットであって、酸化還元ストレスに応答する前記 プロモーターが、sodA、soi28、katG、ahp、rdc、gsh、zwf、およびmicFから選 択される、診断用キット。 3.請求項1に記載の診断用キットであって、DNAストレスに応答する前記プロ モーターが、dinD、ada-alkA、ada、leu-500、gyr、top、mutT、nfoから選択さ れる、診断用キット。 4.請求項1に記載の診断用キットであって、タンパク質ストレスに応答する前 記プロモーターが、rpoD、lon、clpB、merR、fepB-entC、groE、またはmetoから 選択される、診断用 キット。 5.請求項1に記載の診断用キットであって、エネルギーストレスに応答する前 記プロモーターが、sdh、cyo、cyd、またはuncから選択される、診断用キット。 6.請求項1に記載の診断用キットであって、pHストレスに応答する前記プロモ ーターが、hag、micF、aniG、またはkatFから選択される、診断用キット。 7.請求項1に記載の診断用キットであって、前記複数の細菌宿主が全体で、以 下のプロモーターを包含する、診断用キット:soi28、dinD、hag、ada、gyr、ka tG、nfo、clpB、merR、top、cyd、micF、zwf、groE、katF、およびaniG。 8.請求項7に記載の診断用キットであって、rdc、ahp、lon、unc、fepB-entC 、leu-500、cyo、sdh、rpoD、ada-alkA、sodA、mutT、gsh、またはmetoから選択 されるプロモーターをさらに少なくとも1つ包含する、診断用キット。 9.請求項1から8のいずれか1つに記載の診断用キットであって、アッセイ可 能な産物をコードする前記遺伝子がlacZである、診断用キット。 10.化合物の毒性を測定する方法であって、該方法は以下の工程: (a)複数の細菌宿主のそれぞれを別個に培養する工程、ここで、該宿主 のそれぞれがストレスに応答する少なくとも1つのプロモーターを有し、該プロ モーターが検出可能な産物をコードし、そして該プロモーターと異なる遺伝子に 作動可能なように連結し、そして該複数の宿主が全部で、以下のそれぞれに応答 するプロモーターを包含する:酸化還元ストレス、DNAストレス、タンパク質ス トレス、エネルギーストレス、およびpHストレス; (b)該培養物のそれぞれを該化合物とインキュベートする工程;および (c)培養物それぞれの中の検出可能な産物を検出する工程、 を包含する、方法。 11.請求項10に記載の方法であって、酸化還元ストレスに応答する前記プロ モーターが、sodA、katG、ahp、soi28、rdc、gsh、micF、およびzwfから選択さ れる、方法。 12.請求項10に記載の方法であって、DNAストレスに応答する前記プロモー ターが、dinD、ada-alkA、ada、leu-500、gyr、top、mutT、またはnfoから選択 される、方法。 13.請求項10に記載の方法であって、タンパク質ストレスに応答する前記プ ロモーターが、rpoD、lon、clpB、merR、fepB-entC、meto、またはgroEから選択 される、方法。 14.請求項10に記載の方法であって、エネルギーストレスに応答する前記プ ロモーターが、sdh、cyo、cyd、またはuncから選択される、方法。 15.請求項10に記載の方法であって、pHストレスに応答する前記プロモータ ーが、hag、katF、micF、またはaniGから選択される、方法。 16.請求項10に記載の方法であって、前記複数の細菌宿主が全体で、以下の プロモーターを包含する、方法:soi28、dinD、hag、ada、gyr、katG、nfo、clp B、merR、top、cyd、micF、zwf、groE、katF、およびaniG。 17.請求項16に記載の方法であって、前記複数の細菌宿主が、さらに、 rdc、ahp、lon、unc、fepB-entC、leu-500、cyo、sdh、rpoD、 ada-alkA、sodA 、mutT、gsh、またはmetoから選択されたプロモーターを少なくとも1つを包含 する、方法。 18.請求項10から17のいずれか1つに記載の方法であ って、前記化合物を、工程(b)に先立ってS9肝臓抽出物とインキュベートする工 程をさらに包含する、方法。 19.請求項10から18のいずれか1つに記載の方法であって、検出可能な産 物をコードする前記遺伝子がlacZである、方法。 20.毒性化合物に対する抗毒素を測定する方法であって、該方法は以下の工程 : (a)請求項10から19のいずれか1つに記載のプロセスにより、毒性 化合物によって引き起こされるストレスのタイプを測定する工程; (b)請求項10から19のいずれか1つに記載のプロセスにおいて、該 毒性化合物によって引き起こされるストレスに類似のストレスを引き起こす公知 の毒性化合物を同定する工程;および (c)該既知の毒性化合物に対する抗毒性を同定する工程、 を包含する、方法。 21.薬剤の毒性を低減する方法であって、以下の工程: (a)該薬剤により引き起こされるストレスのタイプを請求項10から1 9のいずれか1つに記載のプロセスを用いて測定する工程;および (b)該測定されたストレスを引き起こす該薬剤の部分を改変するまたは 除去する工程を含む、方法。 22.請求項21に記載の方法により産生された改変薬剤。
