Claims (11)
- 1.精製破傷風トキシンを0.005〜0.25Mのリジンの存在下で0.2〜 1%(V/V)のホルムアルデヒドとともに、6.0〜8.0のpH、30〜4 5℃の温度で、24〜32日間インキュベートすることからなる破傷風トキソイ ドの調製方法。
- 2.破傷風トキシンが、Clostridiumtetaniの培養物から精製 された請求項1に記載の方法。
- 3.精製トキシンの比活性が、2000〜3000Lf/mgPNである請求項 1、または2に記載の方法。
- 4.精製トキシンが、高圧液体クロマトグラフィー、そして/またはナトリウム ドデシルサルフェートーポリアクルアミドゲル電気泳動で定量したとき、80% 以上の純度を有する請求項1から3のいずれか1つに記載の方法。
- 5.精製トキシンが、リジンの存在下で35〜40℃において、20〜40分間 プレインキュベートされる請求項1から4のいずれかlつに記載の方法。
- 6.精製トキシンが、ホルムアルデヒドとリジンと共に、35〜40℃で約28 日間インキュベートされる請求項1から5のいずれか1つに記載の方法。
- 7.該インキュベーションの工程において、0.25〜0.5%(V/V)のホ ルムアルデヒドが用いられる請求項1から6のいずれか1つに記載の方法。
- 8.0,05〜0.2Mのリジンが該インキュベーションの工程に用いられる請 求項1から7のいずれか1つに記載の方法。
- 9.トキソイド化されたトキシンがワクチン組成物を形成するために、医薬的に 受入れることができる担体、または希釈剤と製剤化されている請求項1から8の いずれか1つに記載の方法。
- 10.37℃で3か月間貯蔵されたときトキシンに復帰せず、2000〜280 00Lf/mgPNの比活性を有し、130〜270Iu/3.5の力価を有す る破傷風トキソイド。
- 11.医薬的に容認できる担体または希釈剤、および活性成分として、請求項1 0に定義されている破傷風トキソイドを含むワクチンの組成物。
