Claims (22)
- 1.以下の: −ヒトIFN−Rの細胞外ドメインを認識し、−ヒトI型IFNの生物学的特性 に対する中和能力を有するという特性を特徴とするヒトI型インターフェロン受 容体(IFN−R)に対するモノクローナル抗体。
- 2.ヒト病態I型IFNのIFN−Rとの結合を阻止するその能力を特徴とする 請求項1記載のヒトI型IFN−Rに対するモノクローナル抗体。
- 3.骨髄腫細胞と可溶性形態のヒトIFN−Rで予め免疫化した動物からの脾臓 細胞との融合により調製されるハイプリドーマ細胞から得られる請求項1または 2記載のモノクローナル抗体。
- 4.可溶性形態のヒト細胞性IFN−R又は組換え体IFN−R上のエピトープ を認識することを特徴とする請求項1,2または3のいずれかに記載のモノクロ ーナル抗体。
- 5.100μg/ml以下、好ましくは50μg/ml以下、有益には20μg /ml以下、さらに好ましくは約0.5〜2μg/mlの抗体濃度でhu−IF N−Rを保有する細胞と同時インキュベートされる場合に、ヒトI型IFNのヒ ト細胞性IFN−Rとの結合をinvitroで阻止することを特徴とする請求 項1〜4のいずれかに記載のモノクローナル抗体。
- 6.ヒトI型IFNの抗増殖活性をこのヒトI型IFNに高度に反応する細胞、 例えばDaudi細胞上で1〜10μg/mlの範囲の濃度でinvitroに 中和することを特徴とする請求項1〜5のいずれかに記載のモノクローナル抗体 。
- 7.ヒトI型IFNの抗増殖活性をこのヒトI型IFNにあまり反応しない細胞 、例えばLy28細胞上で50〜100μg/mlの範囲の濃度でinvitr oに中和することを特徴とする請求項1〜6のいずれかに記載のモノクローナル 抗体。
- 8.IFNガンマのヒト受容体と結合しないことを特徴とする請求項1〜7のい ずれかに記載のモノクローナル抗体。
- 9.ヒトIFN−Rのアミノ酸配列27〜427上のエピトープを認識すること を特徴とする請求項1〜8のいずれかに記載のモノクローナル抗体。

- 22.ヒトIFN−Rの細胞外ドメインを認識する能力を有し、そしてヒト細胞 における天然の、非病態性又は病態性のI型IFNの抗ウイルス活性に対する中 和能力を有するモノクローナル抗体の選択方法であって、以下の: −I型IFN及び請求項1〜15のいずれかに記載のモノクローナル抗体を確定 濃度で用いてモノクローナル抗体とヒト細胞のIFN−Rとの間、及び/又はI 型IFNとヒト細胞のIFN−Rとの間に複合体を生成させるのに十分な時間イ ンキュベートし;−上記インキュベート細胞を確定濃度のウイルスに感染させ; 一細胞を洗浄し; 一細胞を培地中に再懸濁し; −ウイルスを複製させるのに十分な時間インキュベートし;−細胞を溶解し; −ウイルス複製を測定するか又は細胞変性作用の阻止を測定する 工程を特徴とする方法。
