Abstract
Description
Claims (21)
- 1.バシラス属徴生物内でグラム陰性リパーゼを発現させる方法であって、a) 各々のプラスミドがバシラス属のプロ配列の1番目から9番目までのアミノ酸の うちの1つに対応するコドンに成熟リパーゼのN末端に相当する遺伝子を融合し てなる複数のプラスミドを構築し、前記バシラス属のプロ配列がそれ自身のシグ ナル配列に結合しており、b)適切な条件下でバシラス属宿主微生物を前記工程 a)で得られたプラスミドにより形質転換して該プラスミドを前記バシラス属宿 主徴生物の染色体に組み込み、 c)必要に応じて、該バシラス属宿主徴生物中に分泌過多突然変異を導入し、 d)前記工程b)または工程c)で得られた形質転換体を前記リパーゼの発現の 有無について検定する、 各工程からなることを特徴とする方法。
- 2.前記バシラス属宿主微生物が枯草菌であることを特徴とする請求の範囲第1 項記載の方法。
- 3.前記バシラス属のプロ配列およびシグナル配列が、枯草菌aprEプロ配列 およびシグナル配列であることを特徴とする請求の範囲第1項記載の方法。
- 4.前記成熟リパーゼ遺伝子がシュードモナス属の微生物由来のものであること を特徴とする請求の範囲第1項記載の方法。
- 5.前記成熟リパーゼ遺伝子がシュードモナスメンドシナ、ATCC53552 由来のものであることを特徴とする請求の範囲第4項記載の方法。
- 6.枯草菌aprEプロモーターの使用を含むことを特徴とする請求の範囲第3 項記載の方法。
- 7.前記工程a)で得られるプラスミドが、枯草菌aprEのプロ配列の3番目 のアミノ酸に対応するコドンに各自のN末端で融合する成熟シュードモナスメン ドシナリパーゼ遺伝子を含み、前記枯草菌aprEのプロ配列が枯草菌aprE シグナル配列に結合していることを特徴とする請求の範囲第1項記載の方法。
- 8.前記工程a)で得られるプラスミドが、枯草菌aprEのプロ配列の6番目 のアミノ酸に対応するコドンに各自のN宋端で融合する成熟シュードモナスメン ドシナリパーゼ遺伝子を含み、前記枯草菌aprEのプロ配列が枯草菌aprE シグナル配列に結合していることを特徴とする請求の範囲第1項記載の方法。
- 9.前記バシラス属宿主徴生物内に導入された分泌過多突然変異がSacU(H y)突然変異であることを特徴とする請求の範囲第1項記載の方法。
- 10.グラム陰性リパーゼをコード化する異種DNAを含む形質転換バシラス属 細胞であって、前記異種DNAがその5′末端でバシラス属のプロ配列の1番目 から9番目までのアミノ酸のうちの1つに対応するコドンに融合し、前記バシラ ス属のプロ配列がそれ自身のシグナル配列に結合していることを特徴とする形質 転換バシラス属細胞。
- 11.枯草菌であることを特徴とする請求の範囲第10項記載の形質転換バシラ ス属細胞。
- 12.前記グラム陰性リパーゼがシュードモナスリパーゼであることを特徴とす る請求の範囲第10項記載の形質転換バシラス属細胞。
- 13.5′末端で枯草菌aprEのプロ配列の1番目から9番目までのアミノ酸 のうちの1つに対応するコドンに融合するシュードモナスメンドシナリパーゼを コード化する異種DNAを含み、前記枯草菌aprEのプロ配列がそれ自身のシ グナル配列に結合していることを特徴とする請求の範囲第10項記載の形質転換 枯草菌属細胞。
- 14.前記シュードモナスメンドシナリパーゼをコード化する異種DNAが前記 枯草菌aprEプロ配列の3番目のアミノ酸に対応するコドンに融合しているこ とを特徴とする請求の範囲第13項記載の形質転換枯草菌属細胞。
- 15.前記シュードモナスメンドシナリパーゼをコード化する異種DNAが前記 枯草菌aprEプロ配列の6番目のアミノ酸に対応するコドンに融合しているこ とを特徴とする請求の範囲第13項記載の形質転換枯草菌属細胞。
- 16.N末端で枯草菌aprEプロ配列の1番目から9番目までのアミノ酸のう ちの1つに対応するコドンに融合する成熟シュードモナスメンドシナリパーゼ遺 伝子を含む組換えDNA構築物。
- 17.請求の範囲第16項記載の組換えDNA構築を含むプラスミド。
- 18.N末端で枯草菌aprEプロ配列の3番目のアミノ酸に対応するコドンに 融合してい成熟シュードモナスメンドシナリパーゼ遺伝子を含む組換えDNA構 築物。
- 19.請求の範囲第18項記載の組換えDNA構築物を含むプラスミド。
- 20.N未端で枯草菌aprEプロ配列の6番目のアミノ酸に対応するコドンに 融合している成熟シュードモナスメンドナリパーゼ遺伝子含む組換えDNA構築 物。
- 21.請求の範囲第20項記載の組換えDNA構築物を含むプラスミド。
