DNAヌクレオチド配列決定方法およびそれを実施するための装置
発明の分野
本発明は分子生物学に関し、特にDNAヌクレオチド配列決定方法およびそれを
実施するための装置に関する。
発明の背景
現在までに、ハイブリダイゼーション技術によりヌクレオチド配列を分析しそし
て個々の塩基の置換を同定する幾つかの方法が記載されている(Cotton、
R,G、H,Biochem、 J、、 1989. 第263巻、 1−1
0頁)。
最も汎用される技術は、試験DNA断片を膜に結合させ、膜上で標識オリゴヌク
レオチドとハイブリダイズさせるものである(Wallace、 P、B、、

DNAヌクレオチド配列を決定するための方法および装置は当業界において既知
であり(E、 5outhernら、PCT/GB 89100460.198
9)、該方法は、ガラス支持体上でオリゴヌクレオチドを合成し、放射能または
蛍光標識された試験DNAとのハイブリダイゼーションを行い、二本鎖解離条件
において洗浄し、オートラジオグラフパターンまたは個々のドツトの蛍光強度を
分析することによって試験配列中の個々の置換の存在を検出し、゛そしてデータ
分析に基づいてDNAヌクレオチド配列を再構成することを含んで成る。前記方
法を実施するための装置は、支持フィルムまたはガラス板、およびその表面に共
有結合された母材を含んで成り、該母材は所望の長さのオリゴヌクレオチドの全
セットまたは選択されたその一部を含み、この後者のオリゴヌクレオチドはハイ
ブリダイゼーション反応に参加することができるものである。オリゴヌクレオチ
ドが取り付けられる支持体の表面はガラス製である。
しかしながら、上記の方法および装置は感度が低い。個々の母材要素から得られ
る標識DNA (支持体の表面近くの層に陳列される)からのシグナル値は支持
体の表面結合容量(共有結合されたオリゴヌクレオチドに関して)により制限さ
れ、母材要素の面積かまたは標識するマーカーの感度のいずれかを増加させない
とそれを高めることはできない。それらの制限は、前記方法および装置の分解能
を低下させ、母材を小量化するのを困難にし、検出器の感度および分解能に課せ
られる要件を増やし、そして試薬の消費量を増大させる。
試験DNA断片をアッセイする際にも、ハイブリダイゼーションが配列に関する
明瞭な情報を与えなかったドツトそれぞれについて、−文字段階で実施されるも
う一巡のオリゴヌクレオチド母材合成を伴った一連の連続ハイブリダイゼーショ
ンを必要とし、従ってその度ごとに新たな最適ハイブリダイゼーション条件(温
度、試薬濃度、等)を選択しなければならず、更なる実験並びに時間や試薬の相
当な消費を伴うため、その方法はより一層複雑である。
発明の開示
本発明の目的は、前記方法および装置の能率を向上させ、感度、正確度および再
現性を高め、ヌクレオチド配列中の点変異の認識を単純にし、そして試薬の消費
を減らすように前記方法および装置を改変することである。
ヌクレオチドの整列の形成、標識された試験DNAとそれらのハイブリダイゼー
ション、二本鎖解離条件下での洗浄、標識マーカーの分布を分析することによる
試験DNA中のそれぞれの一塩基置換の同定、およびDNAヌクレオチド配列の
最終的コンピューター再構成を含んで成る本発明の方法によれば、上記目的は、
本発明の方法が洗浄段階での二本鎖解離の所望の温度を保証するような濃度にお
けるヌクレオチドの整列の形成を含むという事実によって達成される。
点変異(ミスマツチ)を有する二本鎖と完全に相補的な二本鎖との確かな識別を
達成するために、洗浄段階を固定の温度勾配で実施することが好ましい。
正確度を向上させそしてデータ分析の時間を減らすため、洗浄段階中に、完全な
二重らせんDNAの既知配列についての温度依存性と比較して残存二本鎖の量の
温度依存性を記録することが好ましい。
分析時間を減らすために、完全に相補的な二本鎖のハイブリダイゼーションと洗
浄とを同一温度で実施できるような濃度でオリゴヌクレオチドの整列を形成させ
ることが有利である。
本発明の方法は、従来の方法に比べて操作を相当簡易化し、その感度、正確度お
よび再現性を向上させることが可能である。当該方法は、時間、労力および使用
する試薬の点で一層経済的にすることによって作業の能率を向上させるのに役立
つ。更に、セルの大きさおよびセル間の隙間は、本発明の装置を現存の技術およ
び測定機器と上手く組み合わせるような形で選択される。
本発明の目的は、更に、DNAヌクレオチド配列決定装置が固体支持体および、
所望の長さのヌクレオチドの整列を含む母材を含んで成り、本発明によれば、該
母材が30μm以下の厚さを育するゲル層によって支持体に取り付けられている
という事実によって達成される。
好ましくは、ゲル層は、母材要素の数に従って一定の間隔があけられた一組の「
ドツト」から成る。このゲル層は、分子層の容量を相当越える容量でのオリゴヌ
クレオチドの三次元的結合に備え、一方間隔があけられた複数のドツトを含むこ
のゲル層は、選択されたゲル容積内に所望の数のヌクレオチドを配置させること
を可能にする。これら全てが、母材を小型化し、全工程の速度を高め、作業時間
を減らし、方法および装置の感度、分解能、正確度および再現性を向上させ、そ
して試薬の消費量を減少させる。
本発明の装置の好ましい態様は、ゲル層の各部分(「セル」)の表面が、25〜
100μmの辺長の正方形で正方形間の隙間が辺長の2倍に等しい形状を有する
装置である。この設計は、ハイブリダイゼーション工程の間に力学的平衡を迅速
に確立させ、試薬の消費量を減らし、感度を高め、そして非毒性で非放射性のマ
ーカーを使用することが可能である。
該装置の全態様において、使用に便利であり、容易に入手でき、且つ再現性のあ
るポリアクリルアミドゲルの層を作製することが好ましい。
本発明の装置は、DNAヌクレオチド配列を決定する作業を単純化し、その時間
を減少させ、感度、正確度および再現性を向上させ、そして試薬の消費量を減ら
すのに役立つ。

発明実施の最良形式
DNAヌクレオチド配列を決定するための本発明の装置は、支持体l (図1お
よび2)、好ましくはガラス板、および、厚さ30μm以下のゲル層によってそ
の表面に取り付けられた母材2を含んで成る。このゲル層は、母材2の中の要素
の数に従って、隙間4によって互いに間隔があけられた複数の部分3を含むこと
ができる。隙間4は異なる寸法を有してもよい。ゲル部分3は異なる形状を有し
てもよい。
好ましい装置は、各部分3が25〜100μmの辺長を有する正方形の形状を有
し、そしてその正方形間の隙間4が辺長の2倍に等しいものである。この層は様
々なゲル、好ましくはポリアクリルアミドゲルから作ることができる。
母材は次のようにして製造することができる:二枚のスライドの一方をバインド
シラン(Bind 5ilane)で前処理し、他方をリベルシラン(Repe
VSilane)で前処理し、後者のスライドをTriton X−100の薄
層で滑らかにする。それらのスライドを厚さ30μmのスペーサーを使って積み
重ね、それらの間に生じた空間をゲル溶液で満たし、ゲル化工程を完了させ、そ
の時点で上側のスライドを取り外す。ゲルコーティングされた下側のスライドを
乾燥し、ゲルの一部を例えば機械的に除去し、そうして隙間により隔てられたゲ
ル部分(セル)をスライド表面上に残す。こうして得られた表面をリペルシラン
(Repel 5ilane)で2〜5分間処理し、最初にアルコールで次に再
蒸留水で洗浄し、乾燥する。3′−末端に3−メチルウリジンを含むオリゴヌク
レオチドを1mM過ヨウ素酸ナトリウムで室温にて10分〜1時間酸化し、10
容の2%LIC104/アセトンで沈澱させ、そして水に溶解させる。次いでこ
のオリゴヌクレオチドをゲルに固定化させる。このために、風乾した母材のセル
を、同容量の微量の酸化されたオリゴヌクレオチド(1つのセル中に1種)で満
たす。
オリゴヌクレオチドの整列は、それらの濃度が洗浄段階における所望の二本鎖解
離温度を保証するようにして形成される。オリゴヌクレオチドの整列は、完全に
相補的な二本鎖全てについて同一の解離温度を保証する濃度で、またはその後の
完全に相補的な二本鎖のハイブリダイゼーションと洗浄とを同一温度で行うのを
可能にする濃度で提供することができる。緩衝液中の放射能または蛍光マーカー
でtIIAwI7tされた試験DNA断片を、予め形成させたオリゴヌクレオチ
ドの整列を有する母材に適用する(この工程では、該溶液は固定化されたオリゴ
ヌクレオチドを含む全領域を完全に被覆する)。次いでオリゴヌクレオチドの整
列を、添加した標識試験DNAとハイブリダイズせしめ、そして解離条件下で二
本鎖を洗い流す。マーカーの分布を分析することにより、試験DNA中の一塩基
置換が同定される。
データ分析に基づいて試験DNA配列を再構成する。完全に相補的な二本鎖と燕
麦異体(ミスマツチ)とを確実に識別するために、本法における二本鎖洗浄は固
定の温度勾配で行われる。この分析の時間を減らし且つ正確度を高めるために、
洗浄の間に、温度に対する残存二本鎖の量の依存性を決定し、そして完全二本鎖
の既知DNA配列についての依存性と比較することが好ましい。はとんど常に、
完全に相補的な二本鎖と燕麦異を含む対応二本鎖とから得られるハイブリダイゼ
ーションシグナルの比がそれらを互いに識別できる程十分に高い(少なくとも1
0 : 1.)温度を見つけることができることが確立された。例外は、高い安
定性を有することがある幾つかの末端ミスマツチにより提供される。しかしなが
ら、これは本法の範囲を制限するものではない。実際、既知配列を処理する(例
えば、それらの中に変異体を検出する)時、予想される塩基置換が二本鎖の内部
に存在するであろうオリゴヌクレオチドを固定化のため選択することが常に可能
である。他方、未知のヌクレオチド配列の分析(例えば、DNA配列決定)では
、末端のミスマツチの問題は、幾つかの情報を犠牲にしながらDNA断片とオリ
ゴヌクレオチド母材との間のハイブリダイゼーションのデータをコンピューター
で処理することにより、容易に解決することができる。本発明者らの計算方法は
、多大な情報による高い安定性により特徴づけられる。
本発明のより良い理解のため、それの実際の実現の観点を幾つか下記に例示する
ことにする。
例1
オリゴヌクレオチドの整列を有する母材の製造固相ホスホルアミダイト法によっ
てオリゴヌクレオチドを合成しく保護基は飽和水性アンモニア溶液中で55°C
にて12時間除去する)、ポリアクリルアミドゲル中での電気泳動により精製す
る。オリゴヌクレオチドを5′−末端標識しくT4ポリヌクレオチドキナーゼに
より〔γ−”P)ATPで)、3μCi/μC上ルの比活性にする。
一方をバインドシラン(Bind 5ilane)で前処理し、他方をリペルシ
ラン(Repel 5ilane) (LKD)で前処理しそしてTriton
X−100の薄層で滑らかにした二枚のスライドを、厚さ30μmのスペーサ
ーを使って互いに一定距離を置いて配置する。それらのスライドの間に生じた隙
間を8%アクリルアミド溶液、30:1のN、N’−メチレンビスアクリルアミ
ド、過硫酸アンモニウムおよびTEMEDで満たし、このゲル溶液を1時間重合
させる。結果として二枚のスライド間に厚さ30μmのゲル層が形成し、この層
の寸法はスライドの大きさによって決定される。重合が完了したら、上側のスラ
イドを取り外す。ポリアクリルアミド層がコーティングされた下側のスライドを
室温にて1時間50%ヒドラジンで処理する。
3′−末端に3−メチリルウリジンを含むオリゴデオキシヌクレオチドを1mM
過ヨウ素酸ナトリウムで室温にて1時間酸化し、10倍容の2%LIC104/
アセトンで沈澱させ、水に溶かす。固定化用に準備した風乾した母材を、キャピ
ラリーチップディスペンサーが装備されたミクロマニプレータ−を使って、酸化
オリゴヌクレオチドの0.5μl小滴(10ピコモル/μlの濃度)で滴下被覆
する。次いでプレートを湿潤チャンバー中で4時間暴露し、開放空気中で0.5
時間乾燥し、ハイブリダイゼーション緩衝液(IM NaC1,10mMリン酸
Na、 pH7,0,1mMエチレンジアミン四酢酸酢酸洗浄し、水でフラッシ
ュ洗浄し、そして−20°Cで乾燥保存する。オリゴヌクレオチドは正方形母材
のセル中に固定化される。
リンカ−は、固定化しようとするオリゴヌクレオチドに5’ −3’ヌクレオチ
ド間ホスホジエステ□ル結合によって取り付けられる3−メチルウリジンにより
提供される。3−メチルウリジンは、いずれの天然塩基とも強い水素結合を形成
しないという事実のため選択された。オリゴヌクレオチド固定化の間にポリアク
リルアミドゲル中で起こる化学反応の概要を図3に示す。
Na1Otによるオリゴヌクレオチドの3′−末端リボヌクレオシド1 (図3
)の酸化は、3′末端にジアルデヒド基を有する誘導体2を生じる。他方、ポリ
アクリルアミド3をヒドラジンで処理すると、アミド基の一部がヒドラジド基4
に置き変わり、この基が3′−ジアルデヒドと容易に反応して比較的安定なモル
ホリン誘導体5を生じる。
固定化の過程は、固定化しようとするオリゴヌクレオチド中にキナーゼにより導
入される(5’−”P)マーカ、−により、モニタリングされる。固定化の収率
(即ち、ゲルに不可逆的に結合したオリゴヌクレオチドの比率)は80%である
。同時に、非特異的吸着対照として使った未酸化のオリゴヌクレオチドについて
の収率は2%を越えない。従って、3′−末端を通して特異的に結合した分子の
比率は98%である。
オリゴヌクレオチドとポリアクリルアミドとの間の結合は、該母材がそのハイブ
リダイズ特性の顕著な変化を全く伴わずに少なくとも5〜7回のハイブリダイゼ
ーション/洗浄サイクルに耐えるのに十分な程安定である。オリゴヌクレオチド
−ゲル結合の半減期は60°Cで2時間、25°Cで36時間である。
キャリヤーの結合容量は、未標識のオリゴヌクレオチドによって種々の比活性に
希釈された同量の!″P−標識才リゴヌすレオチドを固定化することにより評価
される。1ドツトあたり(即ち、表面積1mm”またはゲル容積0.03M”あ
たり) 100ピコモルの非放射能オリゴヌクレオチドは該結合を飽和しない。
酸化された〔α−32P〕DT濾過ヨウ素酸塩を使った同様な実験は、ゲルの結
合容量がゲル表面1mm”あたり約1ナノモルに等しく、それは活性基30mM
濃度に相当することを示した(アミド基の濃度は8%アクリルアミドゲル中IM
である)。
例2
17マーのデオキシオリゴヌクレオチドのヌクレオチド配列の再構成ファージM
13配列プライマー: 5’ −d(GTAAAACGACGCCAGT)並び
に1塩基(下線付)が異なるその3つの誘導体:04つのへブタデカヌクレオチ
ドと、ヘプタデカマーの種々の部分に完全にまたは部分的に相補的である固定化
されたオリゴヌクレオチド(7,8,9,12または15モノマー単位を含む)
とのハイブリダイゼーションを、例1で調製したオリゴヌクレオチド母材上で行
う。
ハイブリダイゼーション緩衝液(IM NaC1,10mMリン酸Na、 pH
7,0,1mMエチレンジアミン四酢酸酢酸μβ中の標識DNA断片(0,01
μCi、30フ工ムトモル)を、ハイブリダイズ混合物の各小滴が固定化オリゴ
ヌクレオチドのスポットを事実上被覆するように、固定化オリゴヌクレオチドの
母材に適用し、モして0°Cで1時間インキュベートする。母材を0°Cにてハ
イブリダイゼーション緩衝液でフラッシュ洗浄し、次いで各洗浄段階において5
°Cずつ温度を上昇させて20−の同緩衝液で各回1分間ずつ10回洗浄する。
各段階において、母材の各セル中のハイブリダイゼーションシグナルを、パルス
増幅器が装備された放射能計測器(Minimonitor 125. Vic
tore−en、 USA )を使って鉛コリメー゛ターを通して記録する。
与えられた時点における残存放射能対出発放射能の比を、温度に対して対数目盛
り上にプロットする(図4)。
図4は、M13プライマーまたはその類似体と、ゲル中に固定化された高GC(
図4a)および高AT(図4b)オクタヌクレオチド(M13プライマーの異な
る2つの部分に相補的であるが、その誘導体とは不完全二本鎖を生成する)とで
形成された二本鎖の洗浄曲線を示す(二本鎖は全て図4に示されている)。
二本鎖安定性の指標として、洗浄温度(TV)は、対応する点でのハイブリダイ
ゼーションシグナルが出発レベルに比べて1710に減少するように選択される
。二本鎖の洗浄曲線は、ゲル中の固定化オリゴヌクレオチドの異なる容積濃度に
ついて調べた。図5から明らかなように、二本鎖のTVは、与えられたサイズの
スポット中に固定化されたオリゴヌクレオチドの量に強く依存する。実験した濃
度領域では、二本鎖洗浄温度は、実験誤差範囲内で、固定化されたオリゴヌクレ
オチド濃度が数倍高くなると、数°C上昇する。この法則は実験した二本鎖全て
について当てはまる。こうして、各オリゴヌクレオチドの安定性を変化させるこ
とができる。この例では、点あたり5ピコモルの濃度が20〜40″Cの範囲内
で二本鎖を安定に維持する。
図4に示されたオリゴヌクレオチドに加えて、M13プライマーに完全にまたは
部分的に相補的な多数の7.8.9.12および15マーの物質、並びに他のミ
スマツチ型を含む二本鎖を調べた。ヘプタデカデオキシヌクレオチドと固定化オ
クタデオキシヌクレオチドとの二本鎖の洗浄結果を下の表に示す。
図4と下表から明らかなように、完全に相補的な二本鎖と対応する不完全二本鎖
とのハイブリダイゼーションシグナルの比がそれらを確実に識別する程十分に高
い”(少なくとも10)温度が常に存在する。例外は、異常に高い安定性を有す
る幾つかの末端ミスマツチである。
オクタヌクレオチドとのハイブリダイゼーションの場合、7倍過剰の情報が得ら
れる。DNA断片のいずれかのヌクレオチドが8例中6例において固定化オリゴ
ヌクレオチドと内部ペアを形成する一方、2例のみが末端ペアを形成する。完全
に相補的な二本鎖と燕麦異を有する二本鎖とを識別するために洗浄曲線間で比較
を行い、こうしてDNA中の塩基の単置換を検出する。完全に相補的な二本鎖の
重複部分から最初のヘプタデカマーを再構成する。
例3
固定化オリゴヌクレオチドの濃度に対する二本鎖洗浄温度の依存これは次のよう
にして決定することができる:手順は例1と同様である。固定化オリゴヌクレオ
チドの濃度に対する二本鎖洗浄温度T、の依存は、5.0.1.5. O15お
よび0.15ピコモルのオリゴヌクレオチド濃度範囲内で、0.03m”容積の
スポットにおいて測定される。
ゲル中の固定化オリゴヌクレオチドの容積濃度に対する二本鎖洗浄曲線の依存は
図5に示される〔スポット容積0.03mm’ 、オリゴヌクレオチド濃度5.
0(I)、 1.5(II)、0.5(I)および0.15(IV)ピコモル〕
。図5のグラフから明らかなように、二本鎖のT。
は、与えられたサイズのスポット中に固定化されたオリゴヌクレオチドの量に強
く依存する。調べた濃度範囲では、二本鎖洗浄温度は、実験誤差範囲内で、固定
化オリゴヌクレオチドの濃度が限定数倍増加すると限定数°C増加する。この法
則は調べた二本鎖全てについて当てはまる(図5は一例を示す)。
完全二本鎖と単一ミスマツチを含む二本鎖との熱解離°完全二本鎖に比較した不
完全二本鎖の洗浄温度の減少置注 ミスマツチは太字で示され、Mは母材を表し
、そして記号“d”は簡易化のため省略されている。
例4
完全に相補的な二本鎖の安定性
手順は例2と同じであるが、ただし異なる固定化オリゴヌクレオチドについて濃
度を次のように変える:高ATについては90ピコモル、高GCについては0.
3ピコモル。
ゲル中の固定化オリゴヌクレオチドの濃度に対する二本鎖の安定性(解離温度)
の依存は、異なるGC組成の二本鎖におけるミスマツチを「単一プレート上で」
検出することを可能にする。図4において、示された2つの固定化オリゴヌクレ
オチドはGC含量が非常に異なっており、結果として、高ATオリゴヌクレオチ
ドの完全に相補的な二本鎖(図4a、曲線I)は、ミスマツチを含む高GCオリ
ゴヌクレオチドの二本鎖(図4b、曲線■および■)よりも本質的に安定性が小
さい。この状況では、DNA断片と両方のオリゴヌクレオチドを含む母材との固
定温度でのハイブリダイゼーションによって、この断片がl塩基の正確度でそれ
に相補的な配列を有するかどうかを言うことはできないだろうことは明らかであ
ろう。
固定化オリゴヌクレオチド濃度に対するT、の依存を使って、異なるGC組成の
二本鎖の解離温度を等しくすることができる。図6に示すように、高AT二本鎖
と高GC二本鎖(同濃度でのΔT、=30°C)の洗浄曲線を、それぞれの点で
の固定化オリゴヌクレオチドの濃度を適切に選択することにより、符合させるこ
とができる。
オリゴヌクレオチドの任意の整列の洗浄曲線を平等化し、こうして「標準化され
た」オリゴヌクレオチド母材を得ることができる。
そのような母材の全ての点に対する完全に相補的な(完全)二本鎖の洗浄温度は
互いに近似し、従って、最適温度での一回の洗浄が、DNA断片が分析中に完全
二本鎖を形成した母材の点を明瞭に決定するのに十分であろう。
例5
予め選択された濃度の固定化オリゴヌクレオチドを含む「標準化された」母材上
ではハイブリダイゼーション段階と洗浄段階を同一温度で実施することができる
手順は例4と同様である。
2つのオリゴヌクレオチドの「標準化された」母材(各ヌクレオチドについて3
点)をハイブリダイズせしめ、35°Cで洗浄する。図7の比較図から原理を理
解する゛ことができる。残存シグナルの比較は、たとえ1つの末端ミスマツチG
Tが非常に安定であるとしても、二本鎖が完全に相補的であった点を明瞭に指摘
する。
例6
本発明の方法および装置の感度、正確度および再現性例1に記載した通りにオリ
ゴヌクレオチド母材を調製する。ただし、50%ヒドラジンでの処理の前に、ゲ
ル層がコーティングされたガラス板を風乾し、ゲルの一部を例えば機械的に除去
し、それぞれ50〜200μmの隙間によって互いに間隔があけられた25〜1
00μmの辺を有する正方形を形成させる。
ハイブリダイゼーションは、テトラメチルローダミン蛍光マーカーによって3′
末端が標識された断片を使って実施し、この標識はターミナルポリヌクレオチド
トランスフェラーゼの助けをかりて導入される(各洗浄曲線の比較は、二本鎖安
定性に対するテトラメチルローダミンの影響が全くないことを示した)。
蛍光マーカーの使用は、ハイブリダイゼーションを大規模で行うのを可能にする
。図8は、間に200μmの隙間を有する100μmの辺の正方形ゲル中にオク
タヌクレオチド5′d(GGCCGTGG)が固定化されている大型母材の略図
を示す。図8のaから明らかなように、この母材とのハイブリダイゼーションは
、配列(図8中)、正方形l (完全二本鎖)中、並びに正方形2,3および4
(それぞれGA。
GTおよびCGミスマツチを有する二本鎖)中の個々の塩基の置換を正確に検出
することを可能にする。
ゲル正方形が25μm、30μm、50μmおよび75μmの辺を有し、正方形
間の隙間がそれぞれ50μm、 60μm、 100μmおよび140μmであ
る大型母材上で同様な実験を行った。
蛍光マーカーの分布を非常に高い感度および空間分解能で検出することができる
ので(例えば、蛍光顕微鏡を使って)、ハイブリダイゼーションシグナルの検出
はシグナル/バックグラウンド比によってのみ制限され、蛍光強度を測定する対
象物の大きさには依存しない。従って、感度は対象物(=オリゴヌクレオチド母
材セル)の面積に反比例する。母材の小型化は該方法の感度を向上させる。例え
ば、図8の正方形“d”は、それぞれ1フ工ムトモル、100アトモルおよび1
0アトモルのデオキシウリジンのテトラメチルローダミン誘導体を含む。正方形
“d3”のシグナル/バックグラウンド比は2に等しく、これは物質の量を確実
に評価するのに十分である。
産業上の利用分野
本発明の方法および装置は、医学、分子生物学および農学において、遺伝子診断
、DNA配列決定およびマツピング、並びに突然変異の検出の目的で利用するこ
とができる。

Claims (9)
- 1.DNAヌクレオチド配列を決定するための方法であって、ヌクレオチドの整 列の形成、標識された試験DNAと前記ヌクレオチド整列とのハイブリダイゼー シヨン、二本鎖解離条件下での洗浄、マーカー分布分析からの試験DNA中の一 塩基置換の検出、および該データ分析に基づいた試験DNAのヌクレオチド配列 の再構成を含んで成り、前記オリゴヌクレオチドの整列が、洗浄工程中に要求さ れる二本鎖解離温度を保証するオリゴヌクレオチド濃度において形成されること を特徴とする方法。
- 2.洗浄が固定の温度勾配において行われることを特徴とする、請求項1に記載 の方法。
- 3.温度に対する残存二本鎖の量の依存性が洗浄の間に決定されそして既知配列 のDNAについての依存性と比較されることを特徴とする、請求項1または2に 記載の方法。
- 4.オリゴヌクレオチドの整列が、完全に相補的な二本鎖全てについて同一の解 離温度を保証するような濃度で形成されることを特徴とする、請求項1に記載の 方法。
- 5.オリゴヌクレオチドの整列が、完全に相補的な二本鎖のハイブリダイぜーシ ョンと洗浄とを同一温度で行えることを保証するような濃度で形成されることを 特徴とする、請求項1に記載の方法。
- 6.DNAヌクレオチド配列を決定するための装置であって、支持体(1)およ び所望の長さのオリゴヌクレオチドの整列を含有しそして前記支持体の表面に取 り付けられた母材(2)を含んで成り、前記母材(2)が30μmを越えない厚 さのゲル層により支持体(1)に取り付けられていることを特徴とする装置。
- 7.前記ゲル層が母材要素(2)の数に従って複数の部分(3)を含んで成り、 該部分が隙間(4)により互いに間隔があけられていることを特徴とする、請求 項6に記載の装置。
- 8.各部分が長さ25〜100μmの辺の正方形であり、その正方形間の隙間が 正方形の辺の二倍であることを特徴とする、請求項7に記載の装置。
- 9.前記ゲル層がポリアクリルアミドゲルで作られていることを特徴とする、請 求項6〜8のいずれか一項に記載の装置。
