発明の要約
嚢胞性線維症(CF)のための遺伝子治療は、嚢胞性線維症の機能的膜横断伝導
レギュレーター(CFTR)のための遺伝子を罹患した上皮細胞に配送すること
から成る。CFのような劣性な形質について期待されるように正常なCFTR遺
伝子の単一コピーの配送と発現によってCF細胞中に存在するCI!−チャンネ
ルの調節欠陥か軽減される。機能的CFTRの欠損、またはCFTR遺伝子にお
ける欠陥から生ずる生理学的に容認できるレベルより低いCFTR機能の存在に
よって起るCFは、このように本発明の原理にしたがって治療することができる
。
本発明の“正常なCFTR遺伝子は、単に機能性CFTRをコードする核酸配列
のすへてである。したがって機能性CFTRが発現できることを条件として、本
遺伝子の実際の配列における変動は、思容され得る。例えば、遺伝子のクローニ
ングを安定化させるためにサイレント突然変異を導入することができる。本発明
の実際において使用されるCFTR遺伝子は、DNAクローニング、人工的構築
、もしくは他の手段のような常法によって得ることかできる。具体例において利
用された4゜6kbcDNAは、CAMPによって刺戟されたCZ−の電流の分
析によって測定されるごとく、機能性CFTR蛋白質をコードするのに必要な配
列すべてを持っている。
・ 本発明によるCFTR遺伝子のトランスファーは、カルシウム ホスフェー
ト共沈澱を用いるトランスフェクション、標的細胞と、CFTR遺伝子を担って
いるリポソーム、赤血球ゴースト、またはスフェロプラストの融合、プラスミド
及びウィルスのベクター媒介トランスファー、及びDNA蛋白複合体媒介シーン
トランスファーを含む多くの手段によって達成することかできる。
現在では、選択された配送車はヒト上皮細胞に感染することができる組換え体レ
トロウィルスである。これは、CFTR遺伝子が比較的大きいことを考慮すると
、幾分驚くべきことである。本発明のレトロウィルスベクターは、一般的に、少
くとも、感染に必要なレトロウィルスの部分のDNAとそれに対して有効に結合
された正常なCFTR遺伝子を含んている。その上、ベクターの構築において使
用されるレトロウィルスのゲノムの部分は、標的細胞上のウィルス複製の存寄作
用を除くため複製欠陥とすることができる。
膵臓や汗腺細胞のようなCF−罹患上皮細胞はすべて本発明のシーントランスフ
ァ法とベクターで標的とすることができるが、CFの最も重篤な併発症は通常原
性なので、気道上皮細胞は本発明の遺伝子治療にとって最も好ましい標的である
。さらに、気道上皮細胞は組換え体レトロウィルスによって容易に感染されるこ
とが分ったという事実を考慮すると、本発明によるこれらの細胞に対するシーン
トランスファーは、極めて実現可能である。
キャリヤーの診断とスクリーニングもCFTR欠陥細胞と細胞系のトランスダク
ションによって達成することかできる。例えば、本発明の相補性スキームを用い
て他の推定上のCF変異の有効性を決定することができ、また位置特異的な突然
変異生成、またはこの大きなABC遺伝子ファミリーの他のメンバーとのドメイ
ン交換によってCFTRの機能を研究する道具として有用である。
したかって本発明は、CF患者に対して機能的CFTR遺伝子の配送と発現を通
じての嚢胞性線維症のための遺伝子治療にむけられている。組換え体レトロウィ
ルスベクター、ならびに他のCFTRシーントランスファースキームを本発明の
実際において使用することができる。本発明は、その原理にしたがってその中に
トランスデユースされたか、トランスファーされた正常なCFTR遺伝子を担っ
ているCF上皮細胞と細胞系の両者をさらに包含する。他の推定上のCF突然変
異のためのスクリーニング及び相補性検定も、本発明の範囲に入れて考えられる
。

本発明のより好ましい態様の詳細な記述機能的CFTRの不在、または生理学的
に容認し得るレベルにないCFTR遺伝子の欠陥から生ずるCFTRの機能か、
正常なCFTR遺伝子をCFTR欠陥細胞にトランスファーすることによって治
療される。
“CFTRI能の生理学的に容認できるレベルと細胞集団、または患者に”、正
常な量のCFTRの存在か認められ正常な生理学的効果を示すということである
。
CFTRtl能不全の例には、嚢胞性線維症において示されたような上皮細胞に
おけるCI!−チャンネルの調節異常か含まれるが、これに限られるわけではな
い。
本発明の組換え体ウィルスベクターは、少くとも標的細胞に感染することができ
るレトロウィルスのゲノムの部分とそれに有効に連結された正常なCFTR遺伝
子から成っている。感染とは、一般的には、ウィルスがその宿主、または標的細
胞へ遺伝子物質をトランスファーする過程のことである。本発明のベクターの構
築において使用されるしトロウィルスはまた、望ましくは、標的細胞上のウィル
スの複製の効果を除去するために複製欠陥にされる。この様な場合には、複製欠
陥ウィルスゲノムは、在来技術にしたがってヘルパーウィルスと組み合せてパッ
ケージすることかできる。一般に、感染性とCFTR遺伝子のトランスファーの
能力の上記の基準を満たすレトロウィルスは、すべて、本発明の実際において使
用することかできる。本発明の実施のための適切なレトロウィルスには例えば、
この分野の技術に熟達している人々にとって良く知られているPLJ、pZip
。
p W e 、及びpEMかある。複製−欠陥レトロウィルスのための適切なパ
ッケージングウィルス系には、例えば、IFCr ip、WCre及びW2、な
らびに曹Amかある。
CFTR)ランスファーにウィルスベクタースキームが用いられるとき、毒性が
減弱されたか、または無毒性のウィルスがやはり望ましいことは評価される。発
明の実際において適用可能な場合においては、CFTR遺伝子の増幅も、正常C
FTRの発現のレベルを増強するために利用することができる。
レトロウィルスと組換えを行うべき、または本発明の他の方法によってトランス
ファーされるへき、遺伝物質は、在来のクローニング法、すなわちcDNA、オ
ーバーラッピングオリゴヌクレオチド配列による、または望ましい配列が得られ
る他の適切な方法すべてによって提供されることがより好ましい。診断的、また
はスクリーニング検定に使用されるときは、遺伝物質は、通常、患者のDNAの
クローニングによって、または、代替法として患者のゲノムDNAの使用によっ
て提供される。既に述べた如く、正常CFTR遺伝子とは、機能性CFTRをコ
ードする核酸配列のすへてのことである。
本発明にしたかうトランスダクション、またはシーントランスファーのための標
的となる細胞はCFTRの配送か望ましいすへての細胞を含む。一般的に言って
、この細胞は、CF細胞のようなCFTR遺伝子の欠陥を持った細胞である。C
Fの場合では、標的とされる細胞は、膵臓、汗腺、肝臓、小腸、腎臓そして肺の
細胞のような、もっと好ましくさえある上皮気道細胞を含む、上皮細胞であるこ
とがより好ましい。
ましくは生物学的に適合性の溶液、または医薬的に受容できる配送車において、
摂取、注射、吸入、もしくは他のいかなる方法によっても患者に投与することが
できる。
投薬量は患者によって異なり、危険、あるいは存寄な副作用とバランスをとった
CFTR機能の強化のレベルによって決定される。トランスダクションのレベル
、CFTRの発現、及びまたは正常CFTRの存在、またはレベルをモニターす
ると、投薬量を選択し、調節するのに役立つ。in vltro hランスダク
ションも本発明中に考察されている。欠陥CFTR遺伝子を有する細胞集団は患
者から取除くことかできる。または、本発明の原理にしたかって正常なCFTR
遺伝子でトランスデユースし、次に患者に導入することかできる。
形質転換を受けたCF細胞系のようなCFTR欠陥細胞系も本発明にしたかって
トランスデユースすることかできる。このような細胞系は、例えば、CF突然変
異を評価してCFを診断し、キャリヤーをスクリーニングするための相補性検定
に役立つ。例えば患者のCFTRcDNAをCF細胞にトランスファーすること
がてき、その細胞を相補性について、すなわちCFTR機能についてスクリーニ
ングし、CF、または、CFTR遺伝子欠陥を確認、もしくは除外することかで
きる。
以下に続く具体例の最初のセットにおいては、レトロウィルス媒介の遺伝子トラ
ンスファーを用いてCF患者の上皮細胞でのC17−の調節における嚢胞性線維
症(CF)欠陥を補った。両種指向性レトロウィルスを使用して、嚢胞性線維症
機能性膜横断伝導レギュレーター(CFTR)のcDNAをCFの患者起源の膵
臓腺癌細胞系へトランスデユースした。この細胞系は、CF欠陥の特徴である電
解質輸送における異常、すなわちc A M Pによって刺戟されたC1−輸送
が欠けているという異常を安定して発現した。CFPAC−1細胞を対照ウィル
スに(PLJ)とCFTR−発現ウィルス(PLJ−CFTR)に暴露した。ウ
ィルスでトランスデユースされたクローンを単離し、分子解析と生理学的解析に
かけた。アガロースゲルプロット分析によってIOこのPLJクローンのうち1
0こにおいて転位していないプロウィルス配列か明らかになった。RNA分析に
よりPLJ−CFTRクローンのすへてにおいてウィルス起源のCFTR転写物
か検出された。RNA分析により転位していないプロウィルス排列を含んていた
PLJ−CFTRクローンのすへてにおいてウィルス起源のCFTR転位物か検
出された。
PLJとPLJ−CFTRクローンをcAMP、及びカルシウムによって刺戟さ
れる陰イオン輸送について検へるために陰イオン(+2J)の流出を用いた、細
胞内cAMPを増加する作用体はPLJ−CFTRクローンにおいて+26 ■
流出を刺戟したがPLJては刺戟しなかった。2この応答性のクローンについて
行われた全細胞パッチ−クランプによって陰イオン流出応答はCI!−伝導のc
AMPによる促進のためであることが示された。
カルシウムのイオノフすアは、すへてのPLJとPLJ−CFTRのクローンに
おいて115I流出とCl−の流れを増加した。これらの知見から、正常なCF
TR1!伝子の発現は、CF上皮細胞にcAMP−依存性のCI!−チャンネ
ルの調節を付与することか示唆される。
具体例の2番目のセットには、気道上皮細胞ならびに膵臓細胞へのシーントラン
スファー、CFTR遺伝子治療の実行、及びリポフェクション及びDNA−蛋白
複合体媒介ジーントランスファーを含む代替法としてのシーントランスファー配
送システムか記述されている。
組換え体レトロウィルス
オーバーラッピングクローンからCFTRcDNAの全長を再構成する初期の試
みは不成功であった。これらの困難は今後明確にしなければならないか、我々の
データは、CFTRcDNAの内部配列からの原核細胞性の転写の結果、バクテ
リアに対して毒性がある蛋白質の発現か起る可能性を示していた。エキソン6b
にまたがっているCFTRの制限酵素断片への3つのサイレントミューチージョ
ン(930におけるCに対するT、933におけるGに対するA1及び936に
おけるCに対するT)の導入は、可能性として隠れた原核細胞性プロモーターを
妨害することによってこの毒性効果を完全に除去し、連続したCFTRcDNA
排列の4.6kbの再構築を可能にした。この再構築されたcDNAのヌクレオ
チド配列は再び決定され、上に記した3つのサイレントミューチージョンを例外
として以前に発表された配列と同一であった。上記のRiordanらを参照の
こと。
請求項の前に提示された配列のリストはCF膜横断伝導レギュレーターをコード
するヌクレオチド配列を、それから推定したアミノ酸配列とともに図示している
。
DNAの配列決定は、35S−標識ヌクレオチドについてのディデオキシチェイ
ンターミネーション法によるか、Dupont Genesis2000自動D
NAシークエンサーによって行われた。図5のカラムの右の数字は塩基の位置を
示す。最初の塩基の位置は、TB2−7よりも1ヌクレオチドだけ大きい5′−
拡張クローンPA3−5の最初の位置に相当する。3′末端とノンコープインク

修飾されたCFTRcDNAは、Korman、 A、J、ら、PNAS(US
A)84:2150−2154 (1987)によって以前に記述されたレトロ
ウィルスベクターにクローニングされた。PLJ−CFTRと呼ばれるこの組換
え体のプロウィルスは図IAに描かれている。図において説明されているこのベ
クターの重要な構造成分は、ロングターミナルリピート配列(LTR)、CFT
RcDNASSV40に由来する配列、0418(Neo)及びpBR322(
pBR)のための複製の起点が含まれている。5’ LTRからの転写は、ウィ
ルスの継代とCFTRの発現の原因である8、5kbゲノム転写物を造り出す。
SV40配列からの転写は、Ne。
セレクタブルマーカーを発現する2番目の転写単位の生成に導かれる。転写開始
部位は、図IAに示されてあり、5’LTRとインターナルSV40配列に矢印
かつけである。制限酵素エンドヌレアーゼKpnIとHindl[[の認識部位
も示されている。DNAとRNAプロット分析において使用されたCFTR遺伝
子(エキノン13)とNeo遺伝子はベクターの下に示されている。
PLJとPLJ−CFTRのベクターのウィルスパッケージング細胞系曹Cr1
pへのトランスフェクションは、複製欠陥ウィルスの一過性の産生へと導かれた
。ウィルスストックの限界希釈法を用いてCFPAC−1細胞に感染させ、トラ
ンスデユースしたクローンを選択するために0418の存在下てその後培養した
。一過性に産生されたPLJ−CFTRウィルスストックは、PLJベクターで
産生されたものよりタイターが低かった(50−100倍)。細胞のIOこの個
々のクローンか各種のウィルス(PLJクローンIからIOまで、及びPLJ−
CFTRクローンlから10まてと名ずけられた)で感染させ、感染体を単離し
、分子分析と生理学的分析にかけた。
トランスデユースされたクローンはレトロウィルスのCFTR配列を発現する。
CFPAC−1のレトロウィルスでトランスデユースされたクローンをプロウィ
ルス配列についてWilson、 J。
M、ら、PNAS (USA)85.4421〜4425(1988)、及びW
ilson、J、M、ら、 、5cience 24 B :1413−141
6 (1990)によって他の細胞種について記述されたように分析された。C
FPAC−1細胞をPLJ、またはPLJ−CFTRて感染させ、各クローンを
単離するために0418の存在下で選抜を行った。高分子量のDNAを各クロー
ンから単離し、図IBに示されているようにゲルプロットハイブリッド法で分析
した。図IBの上のパネルにおいて、DNAをKpnIて消化し、フィルターを
Neo特異性プローブによってハイブリッド形成させた。一方下のパネルにおい
ては、DNAはHindI[[で消化し、フィルターをエキラン13CFTR特
異的プローブに対しハイブリッド形成を行わせた。すべてのレーンにおける4、
3kbバンドは、内性CFTR遺伝子から生ずる。試料には、CFPAC−I
DNA(10μg):レーン“lコピー”CFPAC−I DNA(10μg)
、及びPLJ−CFTRクローンIからIOまでからのレーンズ“PLJ−CF
TRlから10” −DNA (10μg)が含まれる、図の右の境にそって、
分子の大きさのキロベースでの標準が示されている。
ベクターLTRsにおいてユニークな部位を育する高分子量のDNAを制限酵素
Kpnlで消化するとPLJ−CFTRプロウィルスの必須構成系のすへてを共
通な8.5kb断片として放出した。図IBの上のパネルにおいて示されるよう
に、KpnI制限酵素分野を受けたDNAのゲルプロットハイブリッド形成によ
り、転位していないプロウィルス配列が、10/10のPLJクローン、及び9
/lOのPLJ−CFTRクローンにおいて、期待された細胞あたり1コピーの
存在量で明らかとなった。フィルターをNeo−特異的プローブでのハイブリッ
ト形成によりPLJ−CFTRクローン2において著しく転位したプロウィルス
が検出された。このウィルスは明らかにCFTRcDNAの主要な部分を欠失し
ていた(データは示されていない)。
各推定上のPLJ−CFTRクローンの複雑性と、ユニークさを研究するための
ゲルプロット分析の結果は図IBの下のパネル中に示されている。高分子量のD
NAを単離してPLJ−CFTRにおいて2つの内部の部位を持つ制限酵素であ
るHindl[Iて消化し、エキラン13CFTR特異的プローブで分析した。
図IBのパネルにおいて図解されているように、この分析により9/10のPL
J−CFTRクローンにおいて単一でユニークな統合部位が証明された。CFT
R特異的プローブは、PLJ−CFTRクローン2からのDNAにおいては上述
の明らかな欠失のためにプロウィルスを検出てきなかった。
レトロウィルスでトランスデユースされたCFTR遺伝子の発現は、CFTRエ
キソン13プローブを使用するCFPAC−1クローンのRNAプロット分析に
よって研究され、図2に示されている。レトロウィルスでトランスデユースされ
たCFPAC−1細胞のクローンか単離されCFTRの転写物の存在について分
析した。全細胞性RNAを個々のクローンから収穫し、図2の上のパネル中に示
されるようにエキソン13cFTRを使用してフィルターとハイブリッド形成を
行いRNAプロット分析にかけた。図2の下のパネルには、フィルターをはがし
、試料のローディングの変動についてコントロールするためにヒトチーアクチン
cDNAから由来したプローブで再度ハイブリッド形成を行なった。RNAの試
料(10μg)は以下の細胞由来であった:レーンズ“Ta2”−結腸腫瘍細胞
系T84からの2重の試料;レーン“CFPAC−1”−トランスデユースされ
ていないCFPAC−1細胞:レーン“PLJ6”−PLJ感染からのCFPA
C−1クローン#6;及びレーンズ“PLJ−CFTRlから10”−PLJ−
CFTR感染からのCFPAC−1クローンの#1から#lOまで。
図の左側の境に沿って、分子の大きさのキロベースでの基準が示されている。
図2において図解されているように、前に記述されたヒト結腸腫瘍細胞系T84
からの全細胞性RNAは内生CFTR転写物の高いレベルを示した。まがいの感
染をしたCFPAC−1細胞におけるノーザン分析によってCFTR転写物は検
出されなかった。もしくはPLJクローンズ1からIOまでのCFTRRNAは
、RNA−PCRによってCFPAC−1中に検出することができる。ウィルス
によって指向された期待された大きさくすなわち、8.5キロベース)のCFT
Rの転写物は9/lOのPLJ−CFTRクローンにおいて検出された。
CFTRプローブは、欠失したプロウィルス(PLJ−CFTRクローン2)を
含むクローンからのRNAにおいて転写物を検出できなかった(PLJ−CFT
RクロPLJとPLJ−CFTRクローンズを、cAMPとCa−促進陰イオン
輸送についてスクリーニングを行うためにアイソトープ陰イオン(1si)の流
出を測定した。Venglarip C,J、ら、Am、 J、 Physio
l、259 : C358−C364,(1990)によって記述された流出検
定により、Cl−チャンネル遮断薬の阻害効果と125■流出に対する脱分極膜
電位から判断されるようにCl−分泌上皮細胞におけるアゴニストで刺戟された
Cl−電導経路の定量的推定か与えられる。図3Aには、PLJ 6とPLJ−
CFTR6の2つのクローンにおいてアデニレートシクラーゼを活性化する薬剤
であるforskolinを添加した場合と添加しない場合の125■流出速度
定数(r)の経時変化が示されている。IOμMのforskolinは図3A
に示された時間に加えられた。アゴニストの不在の基礎流出期間の後(示されて
いない)、forskoIinは、PLJ−CFTRクローン6からの1251
流出速度を0.32から0. 70 m1n−’へと増加した:PLJ6は応答
しなかった。forskolin添加の前と、forskolin応答のピーク
を通じて得られたrの値から、 1251流出(すなわちr +a+ik /
rbasal )の相対的促進の推定値か得られる。応答性のPLJ−CFTR
クローンにおいては、陰イオン流出に対するforskolinのピーク効果は
、forskolin添加の後の最初の3つの流束期間を通じて観測された(1
5−45sec ) n=7であったPLJ5を除き、平均±SEMは、すべて
のクローンに対してn=9であった。
20のクローンから誘導されたデータは、図3B中に図解されている。rの値は
forskolinの添加の前後に得られた。PLJ−CFTRクローン2につ
いては、!、0゜未満では同じ度盛か適用される。値は平均±SEMであり;n
=7であったPLJ5を除きすべてのクローンについてn=9であった。図3B
に図解されているようにioのPLJ−CFTRクローンのうち7がforsk
olinに応答して1261流出において有意な増加を示したが、対照PLJク
ローンのいずれもが(0/10)forskolinに応答しなかった。親細胞
系であるCFPAC−1も、Schoumacher、R,Aら、PNAS(U
SA)87:4012−4016 (1990)によって記述された如< fo
rskolinまたはcAMP類縁体に対して応答を示さない。PLJ−CFT
Rは、図IBにおいて示された如くゲル プロット /%イブリ・ソド形成によ
ってそのCFTRcDNAにおいて大きな欠失を示し、forskolinが1
251流出を促進できなかったことを説明した。7つの応答性のPLJ−CFT
Rクローンにおいては、forskolinによる陰イオン流出の相対的促進は
1.8から2,8倍の範囲であった。これは、Venglarik 、上述、に
よって結腸腫瘍細胞系T84について最近報告された流出の3.5倍の促進と同
等である。
我々ノ結果は、CFTRcDNAの発現は、CFPAC−1細胞に、cAMP一
応答性の陰イオン流出を付与することを示している。
PLJ−CFTRクローンのforskolin応答性とそのCFTRmRNA
レヘルの間の相レベ、図2と3Bの比較によって説明されるようにあまり著しく
はなかった。
流出検定における最善の応答者の3つは高いmRNAレヘルレベルわち、PLJ
−CFTRクローンl、6.及びlOを示した。しかし、その他の場合ては相関
はそれ程良くなかった。例えば、クローン7と8は、forskolinに対し
て約2倍の応答を示したが比較的低いm RN Aレベルを有し、クローン3と
9は、容易に検出できるCFTRmRNAの存在にもがかわらず低いforsk
olin応答を示した。
Ca−イオノフオアであるionomycinの添加により、すへての対照とC
FTRクローンズにおいて1251流出か増加した。イ才)/ベーサルの値は各
群においてPLJては平均14+2、PLJ−CFTR(n=20)ては14±
lてあった:個々のクローンの間では官憲な差は検出されなかった。ionom
ycinに対するPLJクローンズの応答の程度は、Schoumacherら
、上述(1990)によって野生型のCFPAC−1において前に観測されたの
と類似しており、Venglarikら上記によって観測されたT84細胞の応
答の約3倍である。Ca++イオノフオアとCa−媒介アゴニストか(1−分泌
を促進できる能力は、健常者とCF患者の両者に由来する気道と汗腺細胞につい

−輸送促進にとって必要ではないことか示唆される。PLJとPLJ−CFTR
クローンズにおけるca (−ゝにょる促進の程度において有意差か欠けている
ことからCFTRは、c1分泌を支配しているCa++−媒介調節経路の活性を
調節していないことが示唆される。
図3のPLJ−CFTRクローンにおける陰イオン流出のcAMP誘発増加が、
C11ff、 W、H,ら、PNAS(USA)87:4956−4960 (
1990)+1mよって記述されたCI!−伝導経路の促進のためであったがど
うかを決定するために全細胞パス−クランプの記録が使用された。PLJ−CF
TRクローン1の典形的な応答は図4Aに示されており、5μMのforsko
linによる内方への電流の促進を図解されている。膜電位は、−10+nVに
保持され、0と一84mVにパルス修正した。記録中ノキャップは、forsk
olin−誘発電流のイオン選択性を決定するために浴溶液置換か行われた時間
(6分)を表している。I−Vl!If、%を決定するためのパルスプロトコー
ルは、図に示された時間において行われた。C1−電流は、−84mVへの電圧
パルスによって作られた内方への電流として測定された。内方への電流の同様な
増加は、forskolin (51t M) 、またはcAMP(200から
800BM)の添加か、応答性細胞において内方向への電流を200+68PA
から1690±495 PAf:増加したPLJ−CFTRクローン1.6及び
1oがらの13の細胞のうち11において観測された。この応答の大きさはC1
1ffら、上記、にょってT84細胞において観測されたものと比較して優位で
ある。図4Bにおいて示されているように、cAMP (400BM)、または
forskolin (5μM)は、対照クローン、PLJ6(n=6)からの
細胞の膜電位を促進することはできなかった。膜電位は一20mVに保たれ、O
mVと一84mVヘパルスした。同様な結果がPLJクローンの6細胞のうち5
細胞で得られた。 1′I流出がらw1測されたようにionomycin (
2μM)は、PLJ (n=4)とPLJ−CFTR(n−3)のクローンの双
方において内方向への電流を増加させた。
図40において示されたように、NaC1浴、低C1−浴、及びNa”を含まな
い浴におけるforskolin誘発電流の即塵電流−電圧(1−V)関係がP
LJ−CFTRクローンlから得られた。Forskolin−誘発電流は、刺
戟の前後における電流のディジタルな引き算によって得られた。図40に示され
ている値は、電圧パルス開始の6 m5ec後に記録された。これらのデータは
、6分のレコーディングギャップの間に図4Aに示されているPLJ−CFTR
クローン6の細胞の記録から得られた。
図40において図解されている如く、C11ffら、上記、によってT84細胞
において観測されたように、促進された電流のr−’v5!I係は直線のようで
あった。電流は、同じ浴とピペットを使用して決定された。C1−濃度はOmV
のC1平衡電位の近くて逆転された。浴のCf−を6mM(グルタメート置換)
に減少させると、外方への電流を減少させ、電流のための逆転電位を+66mV
へとシフトさせた。この値は、この外方に向けられたCI!−グレーディエンド
についてのC1−平衡電位(+80mV)に近い値であった。浴のNa”のN−
メチル−〇−グルカミン(NMDG)による置換はI−V関係を官憲に変化させ
なかった。これらの知見により、forskolin−刺戟電流はC1−選択性
であり、PLJ−CFTRクローンにおける陰イオン流出の促進はCI!−伝導
経路の活性化のためであるということか示唆される。
実験操作手順
以下の実験操作手順か、上に明らかにされた具体例において使用された。:
CFPAC−1の細胞は、Schoumacherら、上記、(1990)によ
って以前に記述されている如く培養において維持された。、レトロウィルス感染
のために使用された細胞は72回の継代てあった。CFPAC−1細胞の感染集
団を、個々のクローンを単離するためにG418(1■/rlLl)を含む培地
において選択した。トランスデユースされたCFPAC−1の細胞は、選抜を行
い、やがてクローンとして拡大した。これは、プロウィルス配列、またはプロウ
ィルス発現の明かな喪失と結びつかなかった。両種指向性パッケージング細胞系
曹Cr1pは、Danos、 0. ら、PNAS (USA)85 :546
0−6464 (1988)によって記述されている如く10%の修生血清とペ
ニシリン/ストレプトマイシンを補添した0ulbeccosの改変Eagle
の培地中に維持された。
CFTRcDNAの構築
cDNAは、Riordanら、上記、によって記述されたようにオーバーラッ
ピングクローン、10−1.T16−1およびT16−4.5を結合することに
よって構築された。10−1と716−1は、エキソン4におけるユニークなN
ruIの部位において結合され、エキソンlから13までにまたかるその結果生
した構築物を716−4.5に結合させた。これは、エキソン13がらエキソン
24と広かっているT16−4.5のSac I−EcoRI部分消化生成物を
5’13−1キソン構築物のそれぞれの部位に挿入することによって行われた。
これらの操作は、Riordanら、上述、によって以前に記述されたように全
コーディング配列を含む45キロヘースのクローンを生成した。これらの構築の
企てから生じた大抵のクローンでは、転位か大いに起きていた。明らかに無傷の
構築物の配列の決定にあたり、57bpの欠失が、13bpデイレクトリピート
の2つのコピーの間にあるエキソン6bにおいて同定された。吟味したところ、
この間隔は、コンセンサス原核細胞性プロモーター配列を含むことか認められた
。このリピートを破壊する企てにおいて、in vitroで突然変異生成によ
って、3つの単一ヌクレオチドの変更か行われた。
CFTR翻訳生成物に変化を与えず、その結果安定な構築物を生ずる導入された
変化には、位置930におけるTに対するC1933におけるAに対するG、及
び936におけるTに対するCの置換か含まれる。改変された再構築CFTRプ
ラスミドはCFTR4,6と呼ばれる。
上述した変化は、933におけるG及び936におけるCの存在を除き、正常配
列に対応するオリゴヌクレオチドの合成によって生成した。CFTRcDNAの
切片のアンチセンスストランドを1本鎖M13ファージにクローニングを行い、

ドで突然変異を誘発させた。図6に示されている結果として得られたクローンの
配列を決定し、930の位置に追加の予期されない塩基の変化か起きていること
か見出された。その変化もコートされた蛋白質に変化を与えないサイレントヌク
レオチドの位置にある。
本発明の実際においては、CFTRcDNAの全長を安定化させる他の方法を使
用することかできることも考慮の価値かある。図6に示されている工牛ソン6b
における思いかけないE、coliプロモーターにおける、CFTRのための正
しいアミノ酸配列を保持する一方、そのプロモーターを機能不全にする改変はす
へて同じゴールに到達する。例えば、図6においてアンダーラインを引いたCA
TACT配列の突然変異生成は、アミノ酸配列(例えばCGTATT)を変化さ
せないが原核細胞性プロモーターとしての配列を不活性化し、通常のクローニン
グベクターにおいて安定にするいくつかの方法において達成することができる。
レトロウィルスベクターと組換え体レトロウィルス改変CFTRプラスミドを5
acIで消化すると、4.6kbの制限断片上の改変CFTRcDNAか放出さ
れる。5acIの部位をBcfI部位にオリゴヌクレオチドで変換し、リンカ−
か結合した断片を、Kormanら、上記、によって前に記述されたレトロウィ
ルスベクターのPLJのBam1部位にクローニングした。PLJ−CFTRと
呼ばれる組換え体ベクターは図IAに提示されている。レトロウィルスベクター
PLJとPLJ−CFTRは、両種指向性ウィルスパッケージング細胞系WCr
ipへ記述されているようにトランスファーされた。一過性に産生された両種指
向性ウィルスを収穫するために24時間後にトランスフェクトされたパッケージ
ングを含むプレートから組織培養の培地を取り除いた。
10an2のプレート上にI:5に継代されたCFPAC−1細胞を、ポリブレ
ン(4μg/rnl)を補添したウィルスの上清に12から16時間さらした。
細胞か集密状態に達したとき、細胞は0418(1■/ml)を含む培地に1:
10て継代した。細胞のクローンが、単離され、拡大され、低温保存された。
CFPAC−1クローンのDNAとRNA分析高分子量DNAを記述されたよう
にCFPAC−1細胞から単離し、Wilsonら、上記(1988)、によっ
て記述されたようにゲルプロットハイブリッド形成によって分析した。全細胞性
RNAを精製し、Wi 1sonら、上記(1988)、のRNAプロット分析
にかけた。フィルターは、Feinberg、 A、 P、ら、Anal、 B
iochem、 l 32 :6−13 (1983)のランダムブライミング
法を使用して、高比活性まで標識された様々なりNAプローブでハイブリット形
成を行った。これらのプローブには:l)このエキソン(NT I 900から
2611)の境界に横に接しているオリゴヌクレオチドを使用したクローニング
を行ったcDNAのPCR増幅の後単離された(:FTRのエキソンI 3 ;
2)pSV2NeoのHind■からNcoI断片の960塩基対−hのNeo
−特異的配列、及び3)ヒ]−γアクチンc D N A 。
陰イオン排出の測定
放射同位元素性陰イオン排出は、上記のVenglarikらによっC記述され
たように定量した13手短かに述へれは、細胞の単層は、 +25 iで30分
間プレロードした:2回洗浄した後、流出は、試料−置換操作を使用して15秒
間隔においてモニターした。実験の終りに、細胞の単層に残っているトレーサー
を0.lNHPO3で抽出した。
各サンプリング間隔についての流出速度定数(r)は、以下の如く計算された。

Forskolin 、またはionomycinは、5番目の15秒のサンプ
リング間隔の後に添加された。アゴニストによる促進の程度はr azon+a
+ / r basal として表現される。但し、rag。1.1は、アゴニ
ストの存在において観測された最大値てあり、r basalは、アゴニスト添
加直前の流れ間隔からとられる。
細胞核+2Jの大半の洗い出しは、上記のVenglarikらによって明らか
にされたように最初の60秒のサンプリングを通じて起り、この期間は、速度定
数の計算において無視された。しかし、細胞外隔室の洗出しは、細胞内隔室より
も速いので、60秒後の細胞外空間からの小さい残存流出は、アゴニスト応答の
過少評価に・つながる。
したかって、forskolinに対する流出応答かないときは、forsko
linの添加直後に決定されたrはfarskol inが添加される前に測定
されたよりも僅かに小さい。このことは、r Io+bk / r baAll
Iが図3Bにおいて示されているPLJクローンにおいて0.9と1.0の間に
あるという知見の説明になる。
全細胞電流記録
巨視的電流は、上記のC11ffらによって前に記述された方法によって全細胞
バッチ−クランプを通じて記録された。記録は37°Cて以下の溶液について行
われた(mM) ;浴: 115NaCI!、4ON−メチル−D−グルカミン
(NMDG)−グルタメート、5に一グルターメート、2 M g Cl 2
、 l Ca Cl 2 、l0HEPES(pH7,2); ピペット: l
15KC1135NMDG−グルタメート、0.25EGTA、0.09Ca
CA’t (100nM遊離Ca”) 、2MgC1t、2Na、ATP、0.
2ONaz GTP、l 0HEPES (pH7,2)。膜電位は3つの電圧
において500m5ecの間交互にクランプした。そのうち2つは、CI!(O
mV)、及び(84mV)のための平衡電位に等しくなるように選ばれた。これ
によって上記のC11ffらによって記述されたように、アゴニスト応答の間に
cl−とK“電流がモニターできるようになる。図40に示されているように、
±100mVの間でクランプ電圧を20mVの増加においてステツピングを行う
ことによって電流−電圧の関係を決定するためにパルシングが中断された。
例−■
膵臓及び柿上皮細胞のレトロウィルス媒介トランスダクション
種々の上皮細胞へのレトロウィルス媒介ジーントランスダクションは、E、co
li lからのβ−ガラクトシダーゼを発現する複製欠陥レトロウィルスを使用
して最適化された。ウィルス指令β−ガラクトシダーゼの発現は、トランスデユ
ースされた細胞を青色に染色する組織化学的反応を使用してin 5ituで検
出できるので、これか使用された。前に記述されている、β−ガラクトシダーゼ
発現BAGから作られた両種指向性ウィルスプロデューサー細胞系かウィルスの
源として使用された。このウィルス産生細胞系はBAG5と呼ばれる。BAG5
細胞の集密プレート上の上清を収穫し、濾過して、以下に述べるように種々の上
皮細胞を感染させるのに使用した。
膵臓上皮細胞系
CFPAC−1は、CFの欠陥細胞的特徴(すなわち、cAMPアゴニストの存
在においてCI!−チャンネルは活性化されない)を発現するCF患者の腺癌に
由来した細胞系である。CFPAC−1細胞を種々の希釈(1:2、■、5.1
:101及び1:20)に分け、24時間後にポリブレン(4μg/ml)を補
添した新鮮なウィルスの上清にさらした。12時間後にウィルスを新鮮培地で置
換した。集密状態になったとき、細胞は、記述されているようにウィルス指令β
−ガラクトシダーゼの発現について分析した。最適の感染効率は、感染の前日に
1=5に分けたCFPAC−1細胞で得られた。最適条件下では、ウィルスに対
する1回の暴露によってβ−ガラクシダーゼ遺伝子の細胞の30−40%での安
定なトランスダクションへと導かれた。β−ガラクトシダーゼの発現は培養細胞
において2か月にわたって安定であった。翌日以降の日々にCFPAC−1細胞
を再感染させる試みは、感染効率の増大には、はとんとっながらなか前に述べら
れているようにCFの肺の合併症は、通常、その主要な疾患であり、生命に関わ
るので、気道上皮細胞はシーントランスファーのための最も望ましい標的である
。Taussig 、上記(1984)、気道上皮細胞は、組換え体レトロウィ
ルスで容易に感染されるので、先の及び以下の例において記述されたシーントラ
ンスファーの試みも、肺の場合のような気道上皮細胞に向けられた遺伝子治療に
とって有用である。
CF患者の気道に由来する上皮細胞が、レトロウィルス−媒介シーントランスフ
ァーの育力な標的として使用された。これらの細胞は以前に記述されており、T
43細胞と呼ばれた。収穫された新鮮なりAG5ウィルスにポリブレンを補添し
、24時間前に15に分けておいたT43細胞にさらした。細胞をウィルスに1
2−18時間さらし、集密状態まで培養してから前に述へた生化学的検定を使用
したウィルス指令のβ−ガラクトシダーセの発現について分析した。最適条件下
ては、CFPAC細胞の25%以上か、ウィルスに1回暴露した後、β−ガラク
トシダーゼ遺伝子で安定にトランスデユースさ組換え体しI・ロウイルスの使用
とCFの治療への1つの試みは、気道へレトロウィルスを直接配送することによ
って、機能性CFTR遺伝子をin vivoにおける上皮細胞へ導入すること
である。レトロウィルスの直接配送のためにいくつかのアプローチをとることが
できる。より載皿的なアプローチは、患者の気道に挿管し、CFTRを発現する
レトロウィルスの濃縮溶液で気道を洗浄することである。安定なレトロウィルス
の発現には、プロウィルスか染色体のDNAに組込まれることか必要である。こ
れは、受容細胞か分裂しつつあるならば、最も効率的に起る。ウィルスにさらし
た後、上皮細胞の再生をφり戟することか必要となると思われる。これは、気道
の機械的、または化学的刺戟で達成できるであろう。
より手軽なアプローチは、気道上皮細胞へin vivoで吸入することかでき
る噴霧用製剤によって正常CFTR遺伝子を配送することであろう。多くの異る
薬理学的薬剤は、噴霧剤処理によって、広い気道表面に効率的に配送される。こ
の方法によって達成される有益な作用は一過性である可能性かある。したがって
、薬剤を繰返し投与することか必要であると思われる。直接遺伝子導入に使用さ
れる遺伝子配送システムはウィルスに基ずかなくても良いと思われる。DNA蛋
白質複合体、またはリポソーム中のDNA発現ベクターの直接的吸入は、レトロ
ウィルスよりも安全で、効果的遺伝子配送のシステムであると思われる。
遺伝学的に改変した気道上皮細胞の移植CFの体性遺伝子へのアプローチは、骨
髄指向性遺伝子治療に概念において類似している。我々は、CF患者から気道上
皮細胞を単離し、細胞における欠陥を正し、気道をそれらの細胞で再び満たすこ
とかできるように遺伝学的に改変した細胞を患者に移植することを提案する。
気道において、遺伝子学的に改変した細胞による効率的なりポピユレーションを
達成するためには、機械的、及び化学的刺戟によって内生上皮細胞の打ち内の統
合性を乱すことが必要であると思われる。
代替遺伝子トランスファー配送システムCF欠損の遺伝学的訂正のための他の遺
伝子の配送システムも、本発明の範囲内に入る。これらの実験のためには、プラ
スミドに基いたベクターか使用される。このようなベクターの1例は、図7に指
示されているBA−CFTRBQである。これは、転写かアクチンフランキング
システムから開始され、異質3′ボリアデニレ一ソヨンβ配列から停止された単
純な7762bpl−ランスフゴクンヨンに基いたベクターである。
本発明ベクターは以下の様に構築された。パックホーンはPCl3からの配列(
ヌクレオチド6928から4553)、チキンβアクチン遺伝子の5′フランキ
ング領域(ヌクレオチド6928から7754)及びウシ生長ホルモンポリアゾ
ニレ−ジョンシグナルの3′フランキング配列(ヌクレオチド4827から45
53)を含んていた。全コーディング領域にまたがり、前に論議された3つのヌ
クレオチドの変化を含む全長のCFTR配列か5acIから5ail断片上のベ
クターCFTRから除去され、上に記述したベクターバックボーンへとクローニ
ングした。
このベクターは、いくつかの配送システムにおいて使用されるであろうことは、
この分野の技術に熟達した大人によって評価される。
リボフェクション
前に記述した手順操作はDNAリポソームのエンキャブシデーションに基いてい
る。細胞か、リポソームとともにインキュベートされると、それらはDNAを取
り込み、それを発現する。我々は発現ベクターのDNAとリビッド(DOTMA
)をHepes緩衝化食塩水中1.5−へ希釈し、これらの成分を混合してリビ
ドーDNA複合・ 物を形成することを提案した。それからそのリポソームを使
用し、挿管した患者を溶液を含むリポソームで洗浄するか、吸入によってリポソ
ームを投与することにより標的を定めた遺伝子配送への代替アプローチは、DN
A蛋白質複合体の生成による。この型のシーントランスファー基質は以下のよう
に構築された。呼吸上皮細胞上の受容体のためのポリペプチドリガンドを、記述
されたようにエチリデンジアミノカルボジイミドでポリリジンに結合させる。こ
の蛋白質結合物を、等しい質量の蛋白質結合体と0.25M食塩溶液中のDNA
を混合することによってトランスフェクションベクターのDNAと複合体形成を
行わせる。本DNA/蛋白質複合体は、呼吸系気道細胞によって取込まれ、その
遺伝子が発現される。
これは、リポソームに関して記述されたアプローチを使用して、CFTR遺伝子
をin vivoで気道上皮細胞へ直接配送するのに使用することが出来るであ
ろう。
本発明の多くの改変と変形を、その精神と範囲を逸脱することなく行うことがで
きることは明らかである。本出願において記述された具体的態様は、例を示すた
めだけに与えられており、発明は、添えられた特許請求の範囲によってのみ制限
される。

ーンを単離するために、6418の存在下で選抜した。
高分子量のDNAを各クローンから単離し、ゲルプロット /1イブリッド形成
法で分析した。一番上のパネルにおいて、DNAをKpnIで消化し、フィルタ

Claims (36)
- 1.(a)正常なCFTR遺伝子を提供し;そして(b)トランスファーと相補 に有利な条件下で、正常なCFTR遺伝子を細胞内ヘトランスファーする、工程 から成る 標的細胞におけるCFTRについての遺伝子の欠陥の治療の方法。
- 2.トランスファーの工程がさらにウイルスベクター媒介シーントランスフアー の工程を含む請求の範囲1の方法。
- 3.トランスファーの工程がさらにプラスミドベクター媒介シーントランスフア ーの工程を含む請求の範囲1の方法。
- 4.トランスファーの工程がさらにリポフェクションの工種を含む請求の範囲1 の方法。
- 5.トランスファーの工程がさらにDNA蛋白質複合体−媒介シーントランスフ アーの工程を含む請求の範囲2の方法。
- 6.標的細胞が上皮細胞である請求の範囲1の方法。
- 7.ウイルス媒介シーントランスフアーの工程のウイルスがレトロウイルスから 成る請求の範囲2の方法。
- 8.上皮細胞が気道上皮細胞である請求の範囲6の方法。
- 9.上皮細胞が膵臓細胞である請求の範囲6の方法。
- 10.上皮細胞が汗腺、小腸、肝臓及び腎臓の細胞からなるグループから選ばれ る請求の範囲6の方法。
- 11.レトロウイルスが複製欠陥である請求の範囲7の方法。
- 12.ベクターが (a)その部分が標的細胞を感染することができる少くともウイルスのゲノムの 1部のDNA;及び、(b)ウイルスのゲノムのDNA部分に有効に結合し、i nvivoまたはinvitroで標的細胞において発現ができる正常なCFT R遺伝子を、 含む、標的細胞のCFTRのための遺伝子における欠陥を治療するための組換え 体ウイルスベクター。
- 13.ウイルスがレトロウイルスである請求の範囲12の組換え体ベクター。
- 14.レトロウイルスゲノムが複製欠陥である請求の範囲13の組換え体ベクタ ー。
- 15.さらにPLJを含む請求の範囲13の組換え体ベクター。
- 16.ベクターのプロウイルスが実質的に図1Aに示されている如くである請求 の範囲15の組換え体ベクター。
- 17.治療されている欠陥が嚢胞性線維症の原因である請求の範囲13の組換え 体ベクター。
- 18.標的細胞が上皮細胞である請求の範囲12の組換え体ベクター。
- 19.上皮細胞が膵臓の細胞である請求の範囲17の組換え体ベクター。
- 20.上皮細胞が気道上皮細胞である請求の範囲17の組換え体ベクター。
- 21.上皮細胞が、汗腺、小腸、肝臓、及び腎臓の細胞からなるグループから選 ばれる細胞である請求の範囲17の組換え体ベクター。
- 22.更にPLJを含む請求の範囲18の組換え体ベクター。
- 23.更にPLJを含む請求の範囲19の組換え体ベクター。
- 24.正常CFTR遺伝子が、発現を安定化するサイレント突然変異を含む請求 の範囲15の組換え体ベクター。
- 25.(a)その部分が、上皮細胞を感染することができるレトロウイルスのゲ ノムの少くとも1部のDNAと、それに有効に結合され、標的細胞において発現 することができる正常なCFTRを含む組換え体ウイルスベクターを提供し;そ して (b)患者の上皮細胞集団に、トランスダクションに有利な条件下で、接触され た細胞集団の少くとも1部においてトランスダクションが起るに足る時間接触さ せる、ステップから成る患者の嚢胞性線維症を治療する方法。
- 26.接触がinvivoで行われる請求の範囲25の方法。
- 27.組換え体ウイルスベクターが複製欠陥である請求の範囲25の方法。
- 28.細胞集団が膵臓細胞からなる請求の範囲25の方法。
- 29.細胞集団が気道上皮細胞から成る請求の範囲25の方法。
- 30.細胞集団が汗腺細胞、小腸細胞、肝臓細胞、及び腎臓細胞からなるグルー プから選ばれた細胞集団からなる請求の範囲25の方法。
- 31.Invitroで細胞集団が接触を受け、そして少くともその1部がトラ ンスダクション後に患者に再導入される請求の範囲25の方法。
- 32.組換え体レトロウイルスベクターがPLJを含む請求の範囲27の方法。

- 34.細胞がPLJ−CFTRクローン1、及び3から20までからなるグルー プから選ばれたPLJ−CFTRクローンに由来する請求の範囲23の細胞。
- 35.推定上の欠陥性CFTRを含む核酸配列のための相補検定法であって、 (a)CFTR欠陥細胞集団; (b)CFTR欠陥細胞集団に感染し、CFTR遺伝子をトランスファーするこ とができ、検定される核酸配列を担っている組換え体ベクター; (c)組換え体ベクターによる感染後、細胞におけるCFTRの存在、および/ またはCFTR機能レベルについてのスクリーニングのための手段、からなる方 法。
- 36.核酸配列中の推定上のCF突然変異について検定する方法であって、 (a)その核酸配列を組込んだベクターを提供する;(b)CFTR欠陥細胞集 団を提供する;(c)CFTR欠陥細胞集団をトランスダクションに都合のよい 条件で、トランスダクションが起るに足るだけの時間、組換えたベクターで感染 させる;そして(d)感染後、トランスデュースされた細胞集団においてCFT R機能の相補についてスクリーニングする、工程からなる方法。
