Claims (18)
- 1.HIV蛋白についてのDNAコーディング配列と、ドロソフィラ細胞内での 該コーディング配列の転写および翻訳に必要な調節要素とからなるHIV蛋白遺 伝子発現ユニット。
- 2.調節要素がアクチン5Cプロモーターまたはドロンフイラ・メタロチオネイ ン・ブロモ−ターからなる請求項1記載の遺伝子発現ユニット。
- 3.gp120またはgp160DNAコーディング配列からなる請求項2記載 の遺伝子発現ユニット。
- 4.pgp160Δ32、pgp120FΔ32、pgp120Δ32またはp gp120Δ274中に存在するHIV DNAコーディング配列からなる請求 項3記載の遺伝子発現ユニット。
- 5.請求項1記載の遺伝子発現ユニットからなるDNAベクター。
- 6.請求項5記載のベクターでトランスフェクションしたドロソフィラ細胞。
- 7.請求項6記載の昆虫細胞の培養にて産生されるHIVgp120またはgp 160蛋白。
- 8.HIV感染に対する保護を刺激するワクチンであって、免疫保護量および非 毒性量の請求項7記載の蛋白からなるワクチン。
- 9.請求項7記載の蛋白からなるヒト患者におけるHIV感染検出用診断薬。
- 10.HIV蛋白遺伝子発現ユニットでトランスフェクションした、該蛋白の発 現能を有するドロンフィラ細胞を適当な培地にて培養することを特徴とするドロ ソフィラ細胞におけるHIV蛋白の産生方法。
- 11.細胞を、ヒグロマイシンBホスホトランス7フェーゼについてのコーディ ング配列を有する第1のベクター、およびHIV蛋白遺伝子発現ユニットについ てのコーディング配列を有する第2のベクターでトランスフェクションする請求 項10記載の方法。
- 12.第1のベクターがpCOHYGROである請求項11記載の方法。
- 13.第2のベクターが、pgp160Δ32、pgp120FΔ32、pgp 120Δ32またはpgp120Δ274中に存在するHIV遺伝子発現ユニッ トからなる請求項11記載の方法。
- 14.ドロソフィラ細胞がディー・メラノガスターS2細胞である請求項10記 載の方法。
- 15.S2細胞をベクターpCOHYGROと、pgp160Δ32、pgp1 20FΔ32、pgp120Δ32またはpgp120Δ274中に存在する遺 伝子発現ユニットからなるベクターで共トランスフェクションする請求項14記 載の方法。
- 16.HIV結合物質を請求項7記載の蛋白と接触させ、該物質と該蛋白との間 の結合の存在を検定し、かかる結合がHIVの結合を示すことを特徴とするHI V結合物質の同定方法。
- 17.非変性gp160蛋白産生物およびマチュアgp120蛋白に存在するエ ピトープと反応能を有するモノクローナル抗体を含有するアフィニティー樹脂を 用いることを特徴とする請求項7記載のHIV蛋白の精製方法。
- 18.非変性gp160蛋白産生物およびマチュアgp120蛋白に存在するエ ピトープと反応能を有するモノクローナル抗体を含有するアフィニティー樹脂を 用いることを特徴とする請求項10記載の方法によって産生されるHIV蛋白の 精製方法。
