Description

【発明の詳細な説明】 本発明は、人体中の沈着脂肪を処置するための組成物に
関する。
さらに詳細に述べると、本発明は、ａ）アデニレートシ
クシーゼアゴニスト性活性（ａｄｅｎｙｌａｔｅ　ｃｙ
ｃｌａｓｅａｇｏｎｋｓｔ＋ｃ　ａｃｔｉｖｉｔｙ）を
有する植物活性成分、あるいはｂ）抗ホスホジェステラ
ーゼ（ａｎｔｉｐｈｏｓｐｈｏｄｉｅｓｔｅ−ｒａｓｅ
）活性を有する植物活性成分、もしくは２種類の活性成
分ａ）とｂ）との混合物からなる配合を含む組成物に関
する。
肥満は、問題であり、それは西欧の人口の過半数に影響
を与え、且つ、その影響を受ける数が増加している。肥
満対象者の体重を減少させることは、薬理的手段による
か、あるいは、一定の食事療法で達成できる。しかしな
がら、両者の場合には、劇的減量が必ずしも、美的観点
からみて満足な結果を与えるものではない、それは、体
の一部の場所（特に腿、臀筋、ヒップ、膝）の脂肪沈積
が変化を受けないままであり、肉体的に不快な不釣合の
ため、結果として患者に心理的な悩みを残すからである
。
脂肪沈着と言う問題は、普通の体重を有する成年女子の
一部にも、また影響を与えている。
大抵の場合、沈着脂肪の処置用の、現在入手可能な薬剤
組成物あるいは化粧品組成物は、期待した効果をあげて
いない。
ブデニレートシクラーゼに対してアゴニスト作用を有す
る植物抽出物あるいはその特定の成分及び／もしくは抗
ホスホジェステラーゼ（ａｎｔｉｐｈｏｓｐｈｏｄｉｅ
ｓｔｅｒａｓｅ）活性を有する抽出物あるいは化合物を
含む組成物が、上述の体の部分に存在する沈積脂肪の減
少を早め、かつ、それを容易ならしめ、それにより、美
的観点からの生理的な欠点を除去し、かつ、強制的に体
重を減少せしめる結果になると言う、顕著な効果を得る
ことが、見出されたのは、驚くべきことである。
本発明によると、適切なアデニレートシクラーゼアゴニ
ストは、ヒルガオ科に属する植物、例えば、マルバアサ
ガオ（Ｉｐｏｍｅａ　ｈｅｄｅｒａｃｅａ）　、イポミ
アバラシチカ（Ｉｐｏｔａｅａ　ｐａｒａｓｓｉｔｉｃ
ａ）　、サツマイモ（Ｉｐ。
ｓｅａ　ｂａｔａｔａ）等、あるいは、クチビルバナ科
（Ｌａｂｆａｔａｅ）に属する植物、例えば、サルビヤ
（セージ）（ＳａＬｖｉａ　ｏｆｆｉｃｉｎａＬｉｓ）
、タンジンＣ３ａＬｖｉａ　ｍｅＬｔｉｏｒ−ｒｈｉｚ
ａ）及びマンネンロウ（Ｒｏｓｔａａｒｉｎｕｓ　ｏｆ
ｆｉｃｉｎａＬｉｓ）の抽出物である。抗ホスホジェス
テラーゼ（ａｎｔｉｐｈｏ−ｓｐｈｏｄｉｅｓｔｅｒａ
ｓｅ）に関する限り、有用な植物は、アピゲニン（ａｐ
ｉｇｅｎｉｎｅ）骨格から誘導される２！！１体フラボ
ンを含む植物であり、それるの中に、ギンナン（Ｇｉｎ
ｋｇｏ　ｂｔｌｏｂａ）、セコイアメスギ（Ｓｅｑｕｏ
ｉａ　ｓｅ峠ｅｒ−ｖｉｒｅｎｓ）、イチイ（Ｔａｘｕ
ｓ　ｂａｃｃａｔａ）、ヒモカスジ（ＳｅＬａ−ｇｉｎ
ｅＬＬａ　ｒｕｐｅｓｔｒｉｓ）等が、活性物質のソー
ス（ｓｏｕｒｃｅ）として用いることが出来る。これら
の植物、あるいはその特定のフラクシゴンの抽出物は、
単独であるいは、種々の割合でアデニレートシクラーゼ
アゴニスト（ａｄｅｎｙｌａｔｅ　ｃｙｃｌａｓｅ　ａ
ｇｏｎｉｓｔｓ）あるいは、同一の目的のために、既に
用いられている他の活性成分、例えば、ヨヒンベン異性
体（ｙｏｈｉｍｂｉｎｅ　１ｓｏｓ＋ｅｒｓ）との混合
物として用いることができる。
アピゲニン（ａｐｉｇｅｎｉｎｅ）　２１体、あるいは
対応する誘導体、例えば、アメントフラボン（ａ霞ｅｎ
ｔｏｆｌａ−ｖｏｎｅ）　、ビロベチン（ｂｉｌｏｂｅ
ｔｉｎｅ）、シアドヒチシン（ｓｃｉａｄｏｐｉＬｉｓ
ｉｎｅ）、ギンクゴネチン輸ｉｎｋｇｏｎｅｔｊｎｅ）
等、あるいは類似の構造を有するフラボン２量体を用い
ることが、特に好ましい、アメントフランボン（ａｍｅ
ｎ　Ｌｏｔ　Ｉａｖｏｎｅ）は、ホスホジェステラーゼ
（ｐｈｏｓｐｈｏｄｉｅｓｔｅｒａｓｅ）の強力なイン
ヒビターであり、類似の構造を有する他の２量体も、ま
た同一の活性を有する。
さて、純粋の形態でのこれらのフラボンの混合物、ある
いは、アデニレートシクラーゼアゴニスト（ａｄｅ−ｎ
ｙｌａｔｅ　ｃｙｃｌａｓｅ　ａｇｏｎｉｓｔｓ）と共
に、それらを含む抽出物の混合物を、脂肪の沈積する場
所に局所的に通用すると、沈積脂肪を劇的に減少させる
ことが見い出された。
先に引用したイボミア（Ｉｐｏｓ＋ｅａ）科あるいはレ
イビエート　（Ｌａｂｊａｔｅ）科の抽出物が、アデニ
レートシクラーゼアゴニスト（ａｄｅｎｙｌａｔｅ　ｃ
ｙｃｌａｓｅ　ａｇｏｎｉｓｔｓ＞として、時に活性を
有する。
マルバアサガオ（Ｉｐｏｍｅａ　ｈｅｄｅｒａｃｅａ）
　、サツマイモ（Ｉｐｏａｅａ　ｂａＬａｔａ）、イボ
ミアパラシチ力（Ｉｐｏｍｅａｐａｒａｓｓｉｔｉｃａ
）、サルビヤ　（セージ）　（ＳａＬｖｉａ　ｏｆｆｉ
ｃｉ−ｎａｌｉＳ）、タンジン（Ｓａｔｖｉａ　ｍｅｔ
ｔｉｏｒｒｈｉｚａ）のアデニレートシクラーゼアゴニ
スト活性を、第１表に示す。

測定は、脳、心臓ホモゲネートあるいはＩ！１ｗｊ細胞
を用いてクロカワら（Ｋｕｒｏｋａｗａ　ｅｔ　ａＬ）
　　（バイオケム・バイオフィズ　アクタ（Ｂｉｏｃｈ
ｉｍ、　Ｂｉｏｐｈｙｓ、八ｃｔａ）　５８３，４６７
゜１９７９）の方法によりなされた。アデニレートシク
ラーゼ（ａｄｅｎｙｌａｔｅ　ｃｙｃｌａｓｅ）活性は
、（３Ｂ）　ＡＴＰからの（３■）環状ＡＭＰの生成に
基づいて評価された。活性割合は、基礎活性（ｂａｓａ
ｌ　ａｃｔｉｖｉｔｙ）に対するサンプルの活性をもっ
て、活性の百方率として、表された。
対照物質として、周知のアデニレートシクラーゼアゴニ
ストであるフォルスコリン（Ｆｏｒｓｋｏｌｉｎｅ）が
用いられた。
ホスホジェステラーゼの活性の測定は、ベレッツら（Ｂ
ｅｒｅｔｚ　ｅｔ　ａＬ、＞　（エクスペリエンチア（
Ｅｘｐｅｒｉ−ｅｎｔｉａ）、　３４，１０５４．１９
７７）の方法に従って行われた。
その方法は、ホスホジェステラーゼ製剤と過剰のヌクレ
オチドと共に（３１）−ｃ−ＡＭＰをインキュベーショ
ンし、その結果得られる（３Ｈ）生成物を測定すること
からなる。残留基質を除去するために、バッチプロセス
において、陰イオン交換樹脂が用いられた。フラボノイ
ド類によるＰＤ［！の抑制が、実験により研究された。
その実験では、ｃ−ＡＭＰの濃度が、１ｕＰｌで、一定
に保持された。　ｌ−５０（ｃ−ＡＭＰ分解を５０％抑
制するフラボノイド類の濃度）は、付加されたフラボノ
イドの用量の対数に対して、３５−７５％の範囲にある
抑制の２値を挿入することにより計算された。
この試験における最も重要な２量体フラボノイド類の！
−５０値は、第２表に報告されている。
第２表 評価は、１μＨ基質をもっておこなわれ、■−５０は、
上述のごとく計算された。
美的観点から望ましくない体の部分の処置に当たり、２
量体フラボンに富むギンナン（Ｇｉｎｋｇｏ　ｂｉｌｏ
ｂａ）の抽出物とマルバアサガオ（Ｉｐｏｍｅａ　ｈｅ
ｄｅｒａｃｅａ）のしトロアルコール抽出物との配合物
、あるいは、ギンナン（Ｇｉｎｋｇｏ　ｂｉＬｏｂａ）
かまたはヒモカズラ（ＳｅＬａｇｉ−ｎｅＬＬａ　ｒｕ
ｐｅｓｔｒｉｓ）の抽出物とタンジン（Ｓａｔｖｉａ　
ｍｅＬ−ｔ　１ｏｒｒｈｉｚａ）の抽出物の配合物は、
特に興味がある。
沈着脂肪を減少させるために、上述の抽出物による処置
は、望ましくなさの程度により、２〜３日、あるいは数
週間、継続しなくてはならない。
アデニレートシクラーゼ（ａｄｅｎｙｌａｔｅ　ｃｙｃ
ｌａｓｅ）のアゴニスト作用あるいは抗ホスホジェステ
ラーゼ（ａｎｔｉｐｈｏｓｐｈｏｄｉｅｓｔｅｒａｓｅ
）作用、あるいはアルファ２−アドレナリンリセブター
ブロッキング作用を有する植物の抽出物、あるいは、そ
の純粋な活性成分、例えば、α−ヨヒンベンの用量は、
０．１■から０．２■の間で変化させることができる。
抽出物の形態か、あるいは純粋な成分の形態でのこれら
の製品は、通常の賦形剤を含有する配合物中に混ぜるこ
とができる。賦形剤の例としては、例えば、レミングト
ンのファマスーテカル、サイエンス、ハンドブック、マ
ック出版社、米国、ニューヨーク（Ｒｅｍｉｎｇｔｏｎ
’ｓ　Ｐｈａｒｓａｃｅｕｔｉｃａｌ　５ｃｉｅｎｃｅ
　１ｌａｎｄｂｏｏｋ。
Ｍａｃｋ　Ｐｕｂ、　Ｃｏ、、　Ｎ、Ｙ、、　Ｕ、Ｓ、
＾、）に報告されている。
特に好ましい賦形剤は、天然あるいは合成の燐脂質で、
それは皮膚からの活性成分の吸収を高めるものである。
配合剤中の活性成分の一日の用量は、患者の状況及び症
状の如何により、また、−日に２回あるいはそれ以上に
分けて投与することができる０通常は、完全な治療のサ
イクルには、適切な食餌と併用して１０〜１８０日の処
置を要する。
本発明の組成物の活性成分は、アルコールあるいは脂肪
族ケトン単独で、あるいは水と混合して、あるいは、活
性成分が脂溶性の場合には、芳香族及び／もしくは脂肪
族炭化水素あるいは酢酸エチルをもって、植物材料を抽
出することによって調製される。
得られた抽出物は、水性相と選択した溶媒との間で活性
成分を分配することにより、不活性物質あるいは望まし
くない物質を除去して精製することができる。該抽出物
の精製のための望ましい方法は、抽出物が水性抽出によ
るか、あるいは親水性溶媒で得られる場合には、樹脂に
吸収させることによる。
吸収された活性成分は、異なった極性あるいは高い誘電
率を有する溶媒を用いて、樹脂から選択的に解離するこ
とができる。この分離のために用いられる樹脂は、通常
、アンバーライト（１及びデオライト（１）　のような
ポリスチレン母体を有するものである。
デオライトＳ−７３０”’およびアンバーライトＸＡＤ
２　”’を用いるのが好ましい。
後者の方法により、タンジン（Ｓａｌｖｉａ　ｔｎｅｌ
ｔｉｏｒｒ−ｈｉｚａ）の活性成分及びクチビルバナ（
Ｌａｂ　ｉａ　ｔａｅ）科に属する他の植物の活性成分
を、好ましくは８：２の割合のトルエン／酢酸エチルの
混合物を用いて、選択的に解離することができる。イボ
メア属（Ｉｐｏｍｅａｓｐｅｃｉｅｓ）の活性成分は、
ｎ−ブタノールあるいはｎ−ブタノールとトルエンとの
混合物で、ｐ）Ｉを約１，５に酸性化した後、水溶液か
ら抽出するのが好ましい。上述の方法で得られた抽出物
は、化粧品組成物か薬剤組成物に直接配合し、人体の治
療に用いることができる。
抗ホスホジェステラーゼ（ａｎｔｉｐｈｏｓｐｈｏｄｉ
ｅｓｔｅｒａｓｅ）成分に関する限り、その調製に用い
られた方法は、２量体フラボンに通常用いられる方法で
ある。
次の実施例は、本発明の説明のため示される。
実施例１ マルバアサガオ　ｒ　ｏａｅａ　ｈｅｄｅｒａｃｅａ　
　　’のマルバアサガオ（Ｉｐｏｍｅａ　ｈｅｄｅｒａ
ｃｅａ）の種、１００ｇが砕かれ、４００戚のエタノー
ル／水６：４（ν／ｖ）の混合物で、数回、抽出される
。
薬剤不純物とフロキュレーションされた物質（ｆｌ−ｏ
ｃｃｕｌａｔｅｄ　ｍａｔｅｒｉａｌｓ）を除去するた
めに、抽出物を合せ、これを濾過し、その上で、真空中
で乾燥するまで濃縮される。褐色の固体、１４ｇが得ら
れ、それはすぐに生物的施用ができるものである。
実施例２ ンジン５ａｔｖｉａ　ｍｅＬｔＬｏｒｒｈｉｚａ　　　
　　　　の災 タンジン（Ｓａｌｖｉａ　ｍｅＬｌｉｏｒｒｈｉｚａ）
の根、５００ｇが砕かれ、２１のエタノール／水（１：
１）の混合物で、５回、抽出される。薬剤不純物を除去
するために、抽出物を合せ、これを濾過し、その上で、
真空中で５００−になるまで濃縮された。（抽出物／薬
剤の割合ｌ：１ｖ／ｗ）。濁った懸濁液が、５００ｄの
酢酸エチルで、４回、抽出される。Ｎａ１Ｓｏ、で濾過
の上、有機相が、真空中で乾燥するまで濃縮される。タ
ンジノン（ｔａｎｓｈｉｎｏｎｅ）の様な物質に富む赤
色の抽出物２ｇが得られた。この精製された抽出物は、
水に難溶であり、肥満による見苦しさの治療用に、油性
乳剤に配合することによってのみ用いることができる。
実施例３ サルビヤ（セージ）　（Ｓａｌｖｉａ　ｏｆｆｉｃｉｎ
ａＬｉｓ）の葉及び茎５００ｇが精油を除去するために
、蒸気蒸留される。
蒸留の後、薬剤は、アルコールで枯渇するまで、継続し
て抽出される。
溶媒は、真空中で５００ｄになるまで濃縮され、水性残
留物が３００ｊｄの酢酸エチルで、２回抽出され、有機
相は、乾燥するまで濃縮される。
葉緑素その他の不活性物質を含む残留物は、ｎ−へキサ
ンで抽出されたアルコールの中に、溶解される。
このアルコール相を、乾燥するまで濃縮するとアルコー
ル相は、サルビヤ（セージ）　（ＳａＬｖｉａ　ｏｆｆ
ｉｃｉｎａＬｉｓ）活性フラクシコンとなる。
実施例４ 緑色のギンナン（Ｇｉｎｋｇｏ　ｂｉＬｏｂａ）の葉５
００ｇが枯渇するまでメタノールで抽出される。溶媒が
、小量になるまで濃縮され、残留物が、１００！！１）
のメタノール／水ｌ：１の混合物で希釈され、この相は
、１００ｍのクロロホルムで、２回抽出される。クロロ
ホルム溶液は、真空中で乾燥するまで蒸発され、残留物
は、１００ｄの６０％メタノール／水で、溶解され、ｎ
−ヘキサンで抽出される。ヘキサン抽出液は、水性メタ
ノールが乾燥されるまで濃縮されている間に、除去され
る。この残留物は、アメントフラボン（ａｍｅｎｔｏ−
ｆｌａｖｏｎｅ）骨格を有する約４０％の２Ｎ体フラボ
ノイドを含み、沈着脂肪の減少に有用な配合物中の活性
成分として用いることができる。
実施例５ クリーム１００ｇ毎に次の成分を含む。

Claims (7)

    【特許請求の範囲】
  1. （１）ａ）アテニレートシクラーゼアゴニスト活性（ａ
    ｄｅｎｙｌａｔｅｃｙｃｌａｓｅａｇｏｎｉｓｔｉｃａ
    ｃｔｉｖｉｔｙ）を有する植物活性成分、あるいはｂ）
    抗ホスホジエステラーゼ（ａｎｔｉｐｈｏｓｐｈｏｄｉ
    ｅｓｔｅｒａｓｅ）活性を有する植物活性成分、もしく
    は２種類の活性成分ａ）とｂ）との混合物を含む、余分
    な沈積脂肪を処置するための薬剤・化粧品組成物。
  2. （２）ａ）型の活性成分がマルバアサガオ（Ｉｐｏｍｅ
    ａｈｅｄｅ−ｒａｃｅａ）、イポミアバラシチカ（Ｉｐ
    ｏｍｅａｐａｒａｓｓｉｔｉｃａ）、サツマイモ（Ｉｐ
    ｏｍｅａｂａｔａｔａ）、サルビヤ（セージ）（Ｓａｌ
    ｖｉａｏｆｆｉｅｉｎａｌｉｓ）、タンジン（Ｓａｌｖ
    ｉａｍｅｌｔｉｏｒｒ−ｈｉｚａ）あるいは、マンネン
    ロウ（Ｒｏｓｍａｒｉｎｕｓｏｆｆｉｃｉ−ｎａｌｉｓ
    ）抽出物からなる請求項（１）による組成物。
  3. （３）ｂ）型の活性成分がギンナン（Ｇｉｎｋｇｏｂｉ
    ｌｏｂａ）、セコイアメスギ（Ｓｅｇｏｏｉａｓｅｍｐ
    ｅｒｖｌｒｅｎｓ）、イチイ（Ｔａｘｕｓｂａｌｃａｔ
    ｅ）またはヒモカズラ（Ｓｅｌａｇｉｎｅｌｌａｒｕｐ
    ｅｓｔｒｉｓ）抽出物からなる請求項（１）による組成
    物。
  4. （４）ｂ）型の活性成分が、アピゲニン（ａｐｉｇｅｎ
    ｉｎｅ）、アメントフラボン（ａｍｅｎｔｏｆｌａｖｏ
    ｎｅ）、ビロベチン（ｂｉｌｏｂｅｔｉｎｅ）、シアド
    ビチシン（ｓｃｉａｄｏｐｉｔｉｓｉｎｅ）、ギンクゴ
    ネチン（ｇｉｎｇｋｏｎｅｔｉｎｅ）または類似のフラ
    ボン２量体からなる請求項（１）による組成物。
  5. （５）アルファ−ヨヒンベン（ａｌｐｈａ−ｙｏｈｉｍ
    ｂｉｎｅ）を含む請求項（１）から（４）のいずれかに
    よる組成物。
  6. （６）タンジン（Ｓａｌｖｉａｍｅｌｔｉｏｒｒｈｉｚ
    ａ）抽出物を含む請求項（１）から（５）のいずれかに
    よる組成物。
  7. （７）賦形剤として天然あるいは合成燐脂質混合物を含
    む請求項（１）から（６）のいずれかによる組成物。
