Claims (1)
- 【特許請求の範囲】 (1)外来蛋白質またはペプチドが酵素グルタチオン−S−トランスフェラーゼ と融合されている融合蛋白質の発現をコードするヌクレオチド配列を含む組換え DNA分子。 (2)ヌクレオチド配列が、外来蛋白質またはペプチドが酵素のCOOH−末端 と融合されている融合蛋白質の発現をコードする、請求項1記載の組換えDNA 分子。 (3)酵素が、シストソーマ・ジャポニクム(日本住血吸虫)の26kDaグル タチオン−S−トランスフェラーゼである、請求項1または請求項2記載の組換 えDNA分子。 (4)ヌクレオチド配列が、外来蛋白質またはペプチドが開裂可能な結合を介し て酵素と融合されている融合蛋白質の発現をコードする、請求項1、2または3 記載の組換えDNA分子。 (5)開裂可能な結合が、部位特異性プロテアーゼにより開裂され得る結合であ る、請求項4記載の組換えDNA分子。 (6)開裂可能な結合が、トロンビン、血液凝固因子Xaまたはレニンにより開 裂可能な結合である、請求項5記載の組換えDNA分子。 (7)請求項1〜6のいずれか1項記載のヌクレオチド配列が挿入されている発 現ベクター。 (8)融合蛋白質の発現を目的として前記ヌクレオチド配列に機能し得るように 結合された発現制御配列を含む、請求項7記載の発現ベクター。 (9)細菌性プラスミドである、請求項7または請求項8記載の発現ベクター。 (10)請求項7〜9のいずれか1項記載の発現ベクターにより形質転換された 宿主細胞。 (11)エシェリヒア・コリである、請求項10記載の宿主細胞。 (12)融合蛋白質が回収可能な量で発現される条件下での請求項10または請 求項11記載の宿主細胞の培養工程を含む、融合蛋白質の製造方法。 (13)細胞培養からの融合蛋白質の回収工程を含む、請求項12記載の方法。 (14)宿主細胞を溶解させ、リゼイトを固定グルタチオンと接触させることに よりリゼイトから融合蛋白質を分離する工程により、融合蛋白質が回収される、 請求項13記載の方法。 (15)請求項12〜14のいずれか1項記載の方法により融合蛋白質を製造し 、次いで融合蛋白質を開製して外来蛋白質またはペプチドを酵素から分離させる 工程を含む、蛋白質またはペプチドの製造方法。 (16)請求項12〜14のいずれか1項記載の方法により製造される融合蛋白 質または請求項15記載の方法により製造される蛋白質。 (17)酵素グルタチオン−S−トランスフェラーゼに対応する第1配列および 異種蛋白質またはペプチドに対応する第2配列を含む融合蛋白質。 (18)酵素グルタチオン−S−トランスフェラーゼとして発現され得るヌクレ オチド配列、次いでグルタチオン−S−トランスフェラーゼと融合した外来蛋白 質またはペプチドとして発現され得るヌクレオチド配列の挿入を目的とする少な くとも1つの制限エンドヌクレアーゼ認識部位が挿入されている、発現ベクター 。 (19)酵素が、シストソーマ・ジャポニクムの26kDaグルタチオン−S− トランスフェラーゼである、請求項18記載の発現ベクタ(20)ヌクレオチド 配列が、酵素および外来蛋白質またはポリペプチド間の開裂可能な結合として発 現され得る配列を含む、請求項18または請求項19記載の発現ベクター。 (21)開裂可能な結合が部位特異性プロテアーゼにより開裂され得る結合であ る、請求項20記載の発現ベクター。 (22)融合蛋白質の発現を目的としてヌクレオチド配列に機能し得るように結 合された発現制御配列を含む、請求項18〜21のいずれか1項記載の発現ベク ター。 (23)細菌性プラスミドである、請求項18〜22のいずれか1項記載の発現 ベクター。 (24)pSjlO、pSjlO DBamlおよびpSjlO DBam7S top7から成る群から選ばれるプラスミドを含む、請求項18記載の発現ベク ター。 (25)pGEX−1、pGEX−2TおよびpGEX−3Xから成る群から選 ばれるプラスミドを含む、請求項18記載の発現ベクター。
