あるいは、2つの相同ドメインは異なるLAMPタンパク質に由来する。2つの相同ドメインを含むこれらの構築物には、LAMPヒンジドメインも含まれ得る。抗原は全LAMP-1ルーメンドメインと全LAMP-1膜貫通/細胞質尾部との間に常に配置され、ルーメンドメインの断片は強い免疫応答の生成に効果的ではないと報告されているため、この段落で説明した改善されたLAMP構築物は先行技術の観点からは予想外である。
標的化の効率は、改善されたLAMP構築物を発現する抗原提示細胞がHAエピトープ特異的MHCクラスII拘束性T細胞を刺激する能力を測定することにより測定できる(例えば、米国特許第5,633,234号の実施例5を参照)。

一態様では、改善されたLAMP構築物を含むワクチン組成物が細胞に導入される。細胞は、核酸を複製するため、または改善されたLAMP構築物を発現するための宿主細胞であり得る。好ましくは、改善されたLAMP構築物を発現するための宿主細胞は抗原提示細胞である(以下でさらに説明する)。
例には、プラスミド、ファージ、自律複製配列(ARS)、動原体、およびインビトロまたは宿主細胞(細菌、酵母、昆虫細胞など)および/または標的細胞(例えば、哺乳動物細胞、好ましくは抗原提示細胞など)で複製できるかまたは複製され得る、および/または改善されたLAMP構築物をコードする配列を標的細胞内の所望の位置に運ぶための他の配列が含まれる。
実質的に類似のドメイン配列は、ストリンジェントなハイブリダイゼーション条件下で目的のドメイン配列に特異的にハイブリダイズする配列を選択することにより最初に特定することができる。相同、変異、または修飾されたドメイン配列の適合性を決定するためのアッセイの実行は、適切な活性を発現する配列のスクリーニングの問題に過ぎない。そのようなスクリーニングは、当技術分野では日常的である。
改善されたLAMP構築物の細胞内送達を促進するように設計された送達媒体は、(例えば、原形質膜、組織液、細胞内の区画など)非極性環境と極性環境の両方と相互作用する必要がある。したがって、好ましくは、送達媒体は、極性および非極性ドメインの両方、または改善されたLAMP構築物を細胞に移行させるための移行配列を含むように設計されている。
一態様では、改善されたLAMP構築物送達媒体は、ウイルスまたはウイルス粒子を含む。この態様において、好ましくは、改善されたLAMP構築物はウイルスベクターを含む。レトロウイルス、アデノウイルス、アデノ随伴ウイルス、ヘルペスウイルスなどのウイルスベクターは、多くの場合、2つの成分、すなわち修飾されたウイルスゲノムとそれを囲むコート構造で構成されているが(例えば、Smithら、1995、Ann.Rev.Microbiol.49:807-838を参照)、ウイルスベクターは裸の形で導入されたり、ウイルスタンパク質以外のタンパク質でコーティングされたりすることがある。現在のほとんどのベクターは、野生型ウイルスに似たコート構造を有する。この構造は、ウイルス核酸をパッケージングして保護し、標的細胞に結合して侵入する手段を提供する。
アデノウイルスゲノムは、ウイルス複製サイクルを完了するために必要な約30を超える遺伝子を運ぶ約36kbの線形二本鎖DNA分子で構成されている。初期の遺伝子は、抗ウイルス宿主免疫応答を調節すると考えられているE3領域を除いて、ウイルス複製に不可欠な4つの領域(E1~E4)に分けられる。E1領域(EIAおよびEIB)は、ウイルスゲノムの転写調節に関与するタンパク質をコードする。E2領域遺伝子(E2AおよびE2B)の発現は、ウイルス複製に必要なポリペプチドの合成をもたらす。E3領域によってコードされるタンパク質は、細胞傷害性T細胞および腫瘍壊死因子による細胞溶解を防ぐ(Wold and Gooding、1991、Virology 184:1-8)。E4領域によってコードされるタンパク質は、DNA複製、後期遺伝子発現、スプライシング、および宿主細胞の遮断に関与している(Halbertら、1985、J.Virol.56:250-257)。後期遺伝子は一般に、ウイルスキャプシドに寄与する構造タンパク質をコードする。さらに、アデノウイルスゲノムは、シス作用性5’および3’ITR(逆方向末端反復)およびDNA複製に不可欠なパッケージング配列を運ぶ。ITRにはDNA複製の起点があるが、感染粒子へのアデノウイルスDNAのパッケージングにはキャプシド形成領域が必要である。
およびITRのないAAVコード領域(AAV repおよびcap遺伝子)を含むAAVヘルパープラスミドACG2と同時にトランスフェクトすることによってパッケージングされる。その後、細胞をアデノウイルスAd5に感染させる。ベクターは、当技術分野で知られている方法(例えば、塩化セシウム密度勾配超遠心分離など)を使用して細胞ライセートから精製し、検出可能な複製競合AAVまたはアデノウイルスがないことを確認するために検証することができる(例えば、細胞変性効果バイオアッセイによる)。AAV力価は、プロテイナーゼKによる消化後に調製されたウイルスDNA試料を用いた定量的PCRによって決定され得る。好ましくは、そのような方法によって生成されるベクター力価は、mlあたり約5x1012~1x1013DNase耐性粒子である。
本明細書に記載の改善されたLAMP構築物を使用して自己複製RNAウイルスベクター構築することもできる。例えば、アルファウイルス、フラビウイルス、麻疹ウイルスおよびラブドウイルスを使用して、自己複製RNAウイルスワクチンを生成することができる。自己複製RNAウイルスの好ましい株には、狂犬病ウイルス(RABV)、水疱性口内炎ウイルス(VSV)、西ナイルウイルス、クンジンウイルス、セムリキ森林ウイルス(SFV)、シンドビスウイルス(SIN)および/またはベネズエラ馬脳炎ウイルス(VEE)が含まれるが、これらに限定されない。
本発明による改善されたLAMP構築物は、構築物を含む他の細胞(「送達細胞」)によって標的細胞に送達することができる。構築物を細胞に導入する方法は当技術分野で知られており、細胞の核へのDNAのマイクロインジェクション(Capechiら、1980、Cell 22:479-488)、CaP04によるトランスフェクション(Chen and Okayama、1987、Mol.Cell Biol.7:2745 2752)、エレクトロポレーション(Chuら、1987、Nucleic Acid Res.15:1311-1326)、リポフェクション/リポソーム融合(Feignerら、1987、Proc.Natl.Acad.Sci.USA 84:7413-7417)および粒子衝撃(Yangら、1990、Proc.Natl.Acad.Sci.USA 87:9568-9572)が含まれる。適切な細胞には、自己細胞および非自己細胞が含まれ、異種細胞が含まれてもよい。送達細胞は、細胞の死を誘導することにより(例えば、細胞に誘導性自殺遺伝子を提供することにより)、標的細胞にその内容物を送達するように誘導され得る。
本発明による改善されたLAMP構築物を含む組成物は、細胞への改善されたLAMP構築物の導入を促進し、および/または特定の治療効果を高め、および/または抗体産生を高めるための1つ以上のアクセサリー分子を含み得る。
本発明による改善されたLAMP構築物は、限定ではないが原核細胞(例えば、大腸菌、ブドウ球菌種、バチルス種)、酵母細胞(例えば、サッカロマイセス属)、昆虫細胞、線虫細胞、植物細胞、両生類細胞(例えば、アフリカツメガエル)、鳥類細胞および哺乳動物細胞(例えば、ヒト細胞、マウス細胞、哺乳動物細胞株、解剖組織などからの初代培養哺乳動物細胞)を含む様々な宿主細胞で発現され得る。
本発明の好ましい態様では、本明細書に記載の改善されたLAMP構築物は、天然または操作された抗原提示細胞に導入される。
改善されたLAMP構築物によってコードされるペプチドワクチンも本発明の範囲内である。好ましくは、抗原は区画/オルガネラ(またはそれが送達される後続の区画/オルガネラ)内で処理され、免疫応答を調節することができるMHCクラスII分子に結合したエピトープを生成する。
ポリヌクレオチドとしての改善されたLAMP構築物、改善されたLAMP構築物のコード化タンパク質、および/または細胞(本明細書に記載の改善されたLAMP構築物を発現する抗原提示細胞など)は、例えば当技術分野で説明されている方法など、当業者に周知の方法により抗体を生成するために使用できる。例えば、前出のSutcliffeら;前出のWilsonら;Chowら、Proc.Natl.Acad.Sci.USA 82:910-914(1985);およびBittleら、J.Gen.Virol.66:2347-2354(1985)を参照。インビボ免疫化を使用する場合、改善されたLAMP構築物によってコードされたタンパク質および/または本明細書に記載したような抗原を含む改善されたLAMP構築物を含むポリヌクレオチドで動物を免疫化してもよい。本発明の好ましい一実施形態は、ポリヌクレオチドとしての改善されたLAMP構築物でのプライミング、改善されたLAMP構築物のコード化タンパク質、および/または細胞(本明細書に記載の改善されたLAMP構築物を発現する抗原提示細胞など)に続く抗原でのブースティングである。さらなる好ましい実施形態では、特に抗体レパートリーの多様性および力価を考慮すると、本明細書に記載の改善されたLAMP構築物でのプライミングとそれに続く抗原でのブースティングが特に考えられ、さらに強い免疫応答を生み出すために使用できる。
例えばELISAアッセイで、直接または間接的に(例えば、ビオチン化AviTagを介して)固体表面に吸着した遊離抗原を使用して検出することができる抗抗原抗体の有用な力価を得るために、例えば約2週間の間隔で、何度かのブースター注射(組換え抗原タンパク質など)が必要になる場合がある。免疫動物の血清中の抗抗原抗体の力価は、抗抗体の選択によって、例えば、固体支持体上の抗原への吸着および当技術分野で周知の方法による選択された抗体の溶出によって増加し得る。
したがって、本発明は、異種プロモーターに機能的に連結された抗抗原抗体(例えば、抗体全体、その重鎖または軽鎖、またはその一部、または本発明の一本鎖抗体、またはその断片もしくは変異体)をコードするポリヌクレオチドを含む宿主細胞を含む。好ましい実施形態では、抗体分子全体の発現のために、以下に詳述するように、免疫グロブリン分子全体の発現のために、重鎖と軽鎖の両方をコードするベクターを宿主細胞で共発現させることができる。

IgG部分のジスルフィド結合によるジスルフィド結合二量体構造を有するIgG融合タンパク質は、単量体ポリペプチドまたはその断片単独よりも他の分子の結合および中和においてより効率的であることがわかっている。例えば、Fountoulakisら、J.Biochem、270:3958-3964(1995)を参照。本明細書に記載の抗抗原抗体または改善されたLAMP構築物のコード化ポリペプチドをコードする核酸を、エピトープタグ(例えば、血球凝集素(「HA」)タグまたはフラグタグ)としての目的の遺伝子と組換えて、発現したポリペプチドの検出および精製を補助することもできる。例えば、Janknechtらによって説明されたシステムによりヒト細胞株で発現した非変性融合タンパク質の容易な精製が可能になる(Janknechtら、1991、Proc.Natl.Acad.Sci.USA 88:8972-897)。このシステムでは、遺伝子のオープンリーディングフレームが6つのヒスチジン残基からなるアミノ末端タグに翻訳融合されるように、目的の遺伝子をワクシニア組換えプラスミドにサブクローニングする。タグは、融合タンパク質のマトリックス結合ドメインとして機能する。組換えワクシニアウイルスに感染した細胞からの抽出物をNi2+ニトリロ酢酸-アガロースカラムにロードし、ヒスチジンタグ付きタンパク質をイミダゾール含有緩衝液で選択的に溶出することができる。
本発明によるワクチン材料は、本明細書に記載の免疫刺激性が改善されたLAMP構築物を含んでもよく、または組換え微生物、または免疫刺激性が改善されたLAMP構築物を発現する抗原提示細胞であってもよい。本発明によるワクチン材料を含む改善されたLAMP構築物の調製および個体の免疫化のためのそのような改善されたLAMP構築物の投与は、当業者に周知の免疫化の原理に従って達成される。
表1に記載の少なくとも1つの癌抗原を含む本明細書に記載の改善されたLAMP構築物を使用して、癌および/または過剰増殖性障害を有する患者を治療することができる。
例には、エプスタイン-バーウイルス関連リンパ腫が記録されている患者、HPV関連子宮頸癌患者、慢性HCV患者、または癌遺伝子もしくは腫瘍抑制遺伝子に明確な再配列もしくは変異がある患者が含まれる。さらに好ましい実施形態は、CMV感染に起因する癌を含む過剰増殖性障害を有する患者を含む。
一実施形態では、改善されたLAMP構築物は、悪性腫瘍の経過中の任意の適切な時間に患者に注入され得る。例えば、表1に記載の少なくとも1つの癌抗原を含む改善されたLAMP構築物は、腫瘍負荷が低い段階で注入されるであろう。改善されたLAMP構築物が個体の抗原提示細胞(例えば、樹状細胞など)に導入される代替実施形態では、抗原提示細胞または成熟抗原提示細胞の前駆体は、個体の骨髄または末梢血から静脈穿刺によって採取される。これらの細胞は培養で確立され、続いて改善されたLAMP構築物で形質導入される。形質導入が行われた後、これらの抗原提示細胞は患者に注入される。
本発明はさらに、本明細書に記載された方法の実施を促進するキットを含む。一態様では、キットは、本明細書に記載の改善されたLAMP構築物と、改善されたLAMP構築物を受け取るための細胞とを含む。キットは、細胞をプロフェッショナルAPC内に設計するための1または複数の核酸をさらに含み得る。ただし、一態様では細胞はプロフェッショナルAPCである。細胞は共刺激分子を発現してもしなくてもよい。好ましい態様では、細胞が共刺激分子を発現しない場合、改善されたLAMP構築物によりコードされる抗原は自己抗原である。別の態様では、異なるMHC分子(例えば、ヒトで発現されることが知られている)を発現する細胞のパネルが提供される。さらなる態様では、キットは、細胞(例えば、脂質ベースの製剤、ウイルスパッケージング材料、細胞など)への改善されたLAMP構築物の進入を促進する試薬を含む。なおさらなる態様では、改善されたLAMP構築物によってコードされる抗原に特異的な1つ以上のT細胞株が提供され、改善されたLAMP構築物が免疫応答を誘発、調節または増強する能力を検証する。
ここで、以下の実施例を参照して本発明をさらに説明する。以下の実施例は単なる例に過ぎず、本発明の範囲内にある限り詳細を変更することができることを理解されたい。
図1に示す改善されたLAMP構築物は、当業者に周知の標準的な分子生物学技術を使用して構築することができる。例えば、ポリヌクレオチドを含むプラスミドは、図1に示すILC-1~ILC-6の異なる構造を生じるように設計できる。図1に示すLAMPドメインは、図3~図10に示すアミノ酸配列に由来することができる。好ましくは、LAMPドメインは、図3~図10に示すヒトLAMPタンパク質に由来する。各ドメインの境界は、図2Aおよび図2Bから導出できる。ヒト配列と比較した場合に同等のドメインを識別することにより、対応するドメインはオーソロガス配列からもクローニングできることが想定される。癌抗原は、表1に記載されているように、個別に、または組み合わせて、記載されているLAMP構築物にクローニングできる。
免疫応答を調節する能力について、実施例1に記載の改善されたLAMP構築物の能力を試験することができる。例えば、雌のBALB/cマウスを0、14、28日目にナノパスを使用して、PBS100ul中の改善されたLAMP構築物50ugおよびGMCSF5ugの投与量で免疫化することができる。実験は、最後の投与の4週間後に終了する。
サバイビンは、タンパク質のアポトーシス阻害剤(IAP)ファミリーの最小メンバーであり、アポトーシスの阻害と細胞周期の調節に関与する。これらの機能的属性により、サバイビンは、多様な機能、すなわち細胞増殖と細胞死の調節を示す独自のタンパク質になる。腫瘍におけるサバイビンの発現は、アポトーシスの阻害および細胞死の減少率と相関するだけでなく、化学療法への抵抗性および腫瘍の攻撃性とも相関する[1-6]。したがって、サバイビンは癌ワクチンおよび治療薬の重要な標的である[7-9]。サバイビンは、ヒトおよび胚性幹細胞と多くの体性幹細胞タイプの両方で顕著に発現されることもわかっており、幹細胞の発生と維持における未だ未解明の役割を示している。
SureBlue TMBマイクロウェルペルオキシダーゼ基質およびTMB停止液は、KPL(メリーランド州ゲーサーズバーグ)から購入した。
プレートを200μl/ウェルのPBSで2回、200μl/ウェルのPBS-T(0.05%Tween/PBS)で2回洗浄した。希釈した検出抗体(PBS-T/0.5%BSA中50μl/ウェル)を添加し、室温で振とうしながらプレートを2時間インキュベートした。プレートをPBSで4回洗浄した。PBSで希釈したストレプトアビジン-アルカリホスファターゼ(50μl/ウェル)を添加し、2時間インキュベートした。プレートをPBSで4回洗浄し、50μl/ウェルの3-アミノ-9-エチルカルバゾール(AEC、BD Bioscience)基質で10分間発色させた。水道水の流水で洗浄することにより発色を停止させた。暗所で室温で72時間乾燥させた後、AID ELISPOT高解像度リーダーシステムおよびAID ELISPOTソフトウェアバージョン3.5(Autoimmun Diagnostika GmbH)を使用して、着色スポットをカウントした。

0日目に、雌のBALB/cマウスに同系の7000 4T1乳癌細胞を皮下接種することができる。腫瘍が触知可能になったら、ナノパスを使用して、PBS100ul中のワクチン50ugおよびGMCSF5ugを皮内投与する。原発腫瘍をノギスで測定し、腫瘍体積を式p/6(長さx幅)3/2を使用して計算する。腫瘍接種後の日数の関数としての平均腫瘍体積を測定できる。Kaplan-Meierプロットを使用して、終了時点での全生存率を表示できる。
腫瘍壊死因子受容体スーパーファミリーメンバー14(TNFRSF14)またはCD270としても知られるヘルペスウイルス侵入メディエーター(HVEM)は、TNF受容体スーパーファミリーのヒト細胞表面受容体である。近年、HVEMは、造血細胞、様々な実質細胞(乳癌、黒色腫、結腸直腸、卵巣癌細胞など)、および腸上皮に高度に発現していることが見出されている。HVEMは、BTLAまたはLIGHT(TNFSF14)への結合を介して、T細胞を阻害または刺激する双方向タンパク質である。
抗抗原抗体は、動物への注射を用いて抗体を産生する様々な標準的な方法で調製できる。(Current Protocols,Chapter 2を参照。)例えば、本明細書に記載の癌抗原を含む改善されたLAMP構築物を発現する細胞を非ヒト脊椎動物に投与して、ポリクローナル抗体を含む血清の産生を誘導する。好ましい方法では、LAMP/抗原タンパク質の調製物を調製および精製して、天然汚染物質を実質的に含まないようにする。次に、そのような調製物を非ヒト脊椎動物に導入して、より高い比活性のポリクローナル抗血清を産生する。
動物にポリヌクレオチドを直接注入する方法は、当技術分野で十分に説明されている。
例えば、米国特許第5,676,954号;第6,875,748号;第5,661,133号を参照。例えば、抗原を含む改善されたLAMP構築物をコードするポリヌクレオチドを、拘束された覚醒マウス(雌の6-12週齢BALB/cまたはヌード、nu/nu、Harlan Sprague Dawley(インディアナ州インディアナポリス)から入手)の四頭筋に注射することができる。一実施形態では、50μl溶液中の50μgのポリヌクレオチドを、使い捨ての滅菌プラスチックインスリン注射器およびマイクロピペット先端部から切り取ったプラスチックカラーを備えた28G 1/2針(ニュージャージー州フランクリンレイクスのBecton-Dickinson、Cat.No.329430)を使用して、Hartikka、Jら、Hum.Gene Ther.7:1205-1217(1996)に記載されているようにマウスに注射することができる。
次に、各マウスに、リポソーム150μg(Lipofection TM)と複合体を形成した抗原を含む改善されたLAMP構築物をコードするポリヌクレオチド30μgを総量30μlで25ゲージ針を用いて尾静脈穿刺により静脈内注射する。動物の世話は研究を通して維持されるべきであり、「実験動物の使用と管理に関する指針(Guide for the Use and Care of Laboratory Animals)」、Institute of Laboratory Animal Resources,Commission on Life Sciences,National Research Council、National Academy Pressに従って実施すべきである。
哺乳動物で抗体を産生する方法は、当技術分野で周知である。一例では、抗原を含むLAMP構築物に対するポリクローナル抗血清は、抗原を含む合計500μgの改善されたLAMP構築物で病原体を含まないウサギを免疫化することによって2か月間にわたって産生される。例えば、抗原を含む改善されたLAMP構築物をPBSに溶解し、等量のフロイントアジュバントで乳化することができる。最後のブースターの後、ウサギの血清を分離して、ポリクローナル抗血清の力価を決定することができる。
癌ワクチンの開発は40年以上にわたって追求されてきた。しかし、様々な癌ワクチン製剤の多数の臨床研究にもかかわらず、治療用癌ワクチン接種の全体的な臨床的利益は約20%である1、2、3、4、5。有効性を改善するための障害には、a)癌関連抗原である自己タンパク質の免疫原性の低さ、b)制御性T細胞の誘導6、およびc)ワクチン接種により得られる低い免疫応答が含まれる。最近の報告は、多くの腫瘍における臨床反応の改善と腫瘍微小環境におけるワクチン誘発性Th1免疫浸潤との相関を示唆している7、8、9、10、11。
マウス 6~8週齢の雌のC57BL/6アルビノマウスをEnvigoから購入し、Immunomic Therapeutics、Inc.(マサチューセッツ州ロックビル)の動物施設で飼育した。

多形性膠芽腫(GBM)脳癌は、5年生存率が10%未満の攻撃的なヒト脳癌である(Schuessler Aら、2014)。現在の治療には腫瘍の外科的切除と放射線療法が含まれるが、治療の成功で延長される生存期間中央値は約15か月に過ぎない。現在、GBMについて免疫療法を含むいくつかの臨床試験が評価されている。興味深いことに、GBM腫瘍は抗原pp65を含むCMV抗原を発現することが最近発見された。CMVは発癌性ウイルスではないため、GBMでのCMVの役割は不明であるが、治療の観点から、CMV抗原の存在は、免疫ベースのGBM治療に既存の抗ウイルス免疫を活用する独自の機会を提供する。最近の報告では、独自のプロトコルでLAMP-pp65 mRNAをトランスフェクトしたDCを使用した自己免疫療法により、GBM患者の生存率が大幅に改善されることが示された。具体的には、フロリダ大学のDr.Duane A.Mitchellは、エクスビボで自己DCを生成し、LAMP-pp65 mRNAでパルスし、GBM患者に注射して、GBM患者の生存率を大幅に改善したことを示した。pp65をトランスフェクトしたDCのプロトコルでは、DC注射の24時間前に破傷風/ジフテリアワクチンを注射して注射部位を事前調整する必要もあった(Mitchell DAら、2015)。
前立腺癌は、男性で最も頻繁に診断される癌であり、米国では男性の癌による死亡の2番目に多い原因である1。局在性疾患の場合、手術と放射線療法は治癒的な治療法であり続けるが、尿路症状や性機能障害などの悪影響があり、生活の質に悪影響を及ぼす可能性がある。転移性疾患の場合、初期治療としての化学療法は現在、アンドロゲン除去療法単独(ADT)2と比較して生存期間を延長するように見える。ADTは、a)骨ミネラル密度の減少と骨粗鬆症による脆弱性骨折のリスク増加、b)去勢抵抗性前立腺癌(CRPC)の発生に関連する。免疫系を利用して前立腺癌細胞を認識し殺す方法が開発されている。そのような試みの1つは、無症候性または最小症候性の転移性CRPC(mCRPC)3の治療のために前立腺酸性ホスファターゼ(PAP)を標的とするFDA承認の自己細胞免疫療法であるSipuleucel-Tにつながっている。前立腺癌に対するさらなる免疫療法は、多くのワクチン候補、および標的モノクローナル抗体を使用するアプローチを含めて開発中である4,5。
プレートをPBSで4回洗浄した。PBSで希釈したストレプトアビジン-HRP(50μl/ウェル)を添加し、1時間インキュベートした。プレートをPBSで洗浄し、50μl/ウェルのAEC発色液で最大30分間発色させた。水道水の流水で洗浄することにより発色を停止させた。暗所の室温で24~72時間乾燥させた後、AID ELISPOT高解像度リーダーシステムおよびAID ELISPOTソフトウェアバージョン3.5(Autoimmun Diagnostika GmbH)を使用して、着色スポットをカウントした。
(CD3クローン17A2はT細胞を活性化できるため、4oCの温度が重要である)。
1時間(または一晩)インキュベーション後、細胞をPerm緩衝液で2回洗浄し、フローサイトメーターで細胞を取得した。取得中のイベント/秒は8000未満であった。試料あたり106個を超える細胞を取得した。

- 請求項1~18のいずれか一項に記載の樹状細胞を含む医薬組成物。
- 前記組成物を、疾患または障害を軽減または治療するのに十分な量でそれを必要とする対象に投与される、疾患または障害を軽減または治療するための請求項19に記載の医薬組成物。
- 前記組成物を、多形性膠芽腫を治療するのに十分な量でそれを必要とする対象に投与される、多形性膠芽腫を治療するための請求項19に記載の医薬組成物。
- 前記組成物が、プライミングステップおよび/または少なくとも1回のブースティングステップで使用される、請求項20または21に記載の医薬組成物。
- 前記組成物が、プライミングステップで使用される、請求項22に記載の医薬組成物。
- 前記組成物がブースティングステップで使用される、請求項22に記載の医薬組成物。
- プライミングに使用される抗原が、ブースティングに使用される抗原と同じである、請求項22~24のいずれか一項に記載の医薬組成物。
- プライミングに使用される抗原が、ブースティングに使用される第2の抗原と同じタンパク質に由来する、請求項22~25のいずれか一項に記載の医薬組成物。
- プライミングおよび/またはブースティングに2つ以上の抗原が使用される、請求項22~26のいずれか一項に記載の医薬組成物。
