Claims (20)
- 核酸シークエンシングのための方法であって、
フローセルに材料を通すことであって、前記材料が、核酸シークエンシングのための試薬を含む、前記通すことと、
前記フローセルを光で照らすことと、
カメラセンサを使用して、前記フローセルからの光を、対物レンズを通して撮像することであって、前記対物レンズが、
単純面を有する複数のレンズと、
開口絞りであって、前記開口絞りが物理的な開口絞りである、前記開口絞りと、
前記開口絞りから25ミリメートル以内に配置された非球面レンズと
を備える、前記撮像することと
を含む、前記方法。 - 前記フローセルは、0.3ミリメートル以上2.0ミリメートル以下の厚さを有する窓を備え、前記カメラセンサによって撮像された光は、前記対物レンズを通過する前に前記窓を通過する、請求項1に記載の方法。
- 前記複数のレンズの1つは、前記フローセルに近接する第1のレンズであり、前記第1のレンズは、10ミリメートル以下3ミリメートル以上の距離だけ前記フローセルから離されている、請求項1に記載の方法。
- 前記複数のレンズは光学縦列を形成し、前記光学縦列は、前記複数のレンズの第1のレンズから前記複数のレンズの最終レンズまで延在し、前記複数のレンズは4つのダブレットレンズを備える、請求項1に記載の方法。
- 前記光学縦列にはトリプレットレンズが無い、請求項4に記載の方法。
- 前記対物レンズは、0.6~0.8の間の開口数を有する、請求項1に記載の方法。
- 前記対物レンズは少なくとも4mm2の視野を有する、請求項6に記載の方法。
- 前記対物レンズは少なくとも4mm2の視野を有する、請求項1に記載の方法。
- 前記フローセルは、1.5マイクロメートル以下の被写界深度で撮像される、請求項1に記載の方法。
- 核酸のアレイが前記フローセルの表面に付着される、請求項1に記載の方法。
- 前記カメラセンサを使用して、前記対物レンズを通して撮像された前記フローセルからの光に基づいて、前記核酸のアレイを検出することをさらに含む、請求項10に記載の方法。
- 前記核酸のアレイを検出することは、前記核酸のアレイの核酸部位を解像することを含み、前記核酸部位のそれぞれが、500μm2より小さい平均面積を有する、請求項11に記載の方法。
- 核酸シークエンシングのための前記試薬は、前記カメラセンサを使用して、前記対物レンズを通して撮像された前記フローセルからの光に基づいて検出される標識を有するヌクレオチドを含む、請求項1に記載の方法。
- 前記標識は発光団を含む、請求項13に記載の方法。
- 前記フローセルを光で照らすことは、前記対物レンズを通して前記フローセルに光を伝送させることにより、前記フローセルを照らすことを含む、請求項1に記載の方法。
