Claims (38)
- 核酸シークエンシングのための方法であって、
フローセルに材料を通すことであって、前記材料が、核酸シークエンシングのための試薬を含む、前記通すことと、
前記フローセルを光で照らすことと、
カメラセンサを使用して、前記フローセルからの光を、対物レンズを通して撮像することであって、前記対物レンズが、
単純面を有する複数のレンズと、
開口絞りであって、前記開口絞りが物理的な開口絞りである、前記開口絞りと、
前記開口絞りから25ミリメートル以内に配置された非球面レンズと
を備える、前記撮像することと
を含む、前記方法。 - 前記フローセルは、0.3ミリメートル以上2.0ミリメートル以下の厚さを有する窓を備え、前記カメラセンサによって撮像された光は、前記対物レンズを通過する前に前記窓を通過する、請求項1に記載の方法。
- 前記対物レンズは、前記フローセルに近接する第1のレンズを備え、前記第1のレンズは、10ミリメートル以下3ミリメートル以上の距離だけ前記フローセルから離されている、請求項1または請求項2に記載の方法。
- 前記複数のレンズは光学縦列を形成し、前記光学縦列は第1のレンズから最終レンズまで延在し、前記複数のレンズは4つのダブレットレンズを備える、請求項1~3のいずれか1項に記載の方法。
- 前記光学縦列にはトリプレットレンズが無い、請求項4に記載の方法。
- 前記対物レンズは、0.6~0.8の間の開口数を有する、請求項1~5のいずれか1項に記載の方法。
- 前記対物レンズは少なくとも4mm2の視野を有する、請求項1~6のいずれか1項に記載の方法。
- 前記フローセルは、1.5ミクロン以下の被写界深度で撮像される、請求項1~7のいずれか1項に記載の方法。
- 前記対物レンズは少なくとも4mm2の視野を有する、請求項1~8のいずれか1項に記載の方法。
- 前記フローセルは、1.5ミクロン以下の被写界深度で撮像される、請求項1~9のいずれか1項に記載の方法。
- 核酸のアレイが前記フローセルの表面に付着される、請求項1~10のいずれか1項に記載の方法。
- 前記カメラセンサを使用して、前記対物レンズを通して撮像された前記フローセルからの光に基づいて、前記核酸のアレイを検出することをさらに含む、請求項11に記載の方法。
- 前記核酸のアレイを検出することは、前記核酸のアレイの特徴を解像することを含み、前記特徴のそれぞれが、約500μm2より小さい平均面積を有する、請求項12に記載の方法。
- 核酸シークエンシングのための前記試薬は、前記カメラセンサを使用して、前記対物レンズを通して撮像された前記フローセルからの光に基づいて検出される標識を有するヌクレオチドを含む、請求項1~13のいずれか1項に記載の方法。
- 前記標識は発光団を含む、請求項14に記載の方法。
- 前記フローセルを光で照らすことは、前記対物レンズを通して前記フローセルに光を伝送させることにより、前記フローセルを照らすことを含む、請求項1~15のいずれか1項に記載の方法。
- 核酸のアレイを撮像するための顕微鏡の対物レンズのための装置であって、
単純面を有する複数のレンズであって、
前記複数のレンズが光学縦列を形成し、前記光学縦列が、第1のレンズから最終レンズまで延在しており、
前記複数のレンズは、4つのダブレットレンズを備え、トリプレットレンズを備えない、前記複数のレンズと、
開口絞りであって、
前記開口絞りは、物理的な開口絞りであり、
前記開口絞りは、前記第1のレンズと前記最終レンズとの間に配置される、前記開口絞りと、
前記第1のレンズと前記最終レンズとの間の非球面レンズであって、前記非球面レンズは、前記開口絞りから25ミリメートル以内に配置される、前記非球面レンズと
を備える、前記装置。 - 前記非球面レンズは、前記開口絞りから10ミリメートル以内に配置される、請求項17に記載の装置。
- 前記第1のレンズは、前記最終レンズよりも試料に近いように構成され、前記開口絞りは、前記第1のレンズから測定して、前記光学縦列内の70%~90%の間の距離に配置される、請求項17または請求項18に記載の装置。
- 前記非球面レンズは、40ミリメートル以上60ミリメートル以下の直径を有する、請求項17~19のいずれか1項に記載の装置。
- 前記装置は1.5ミクロン以下の被写界深度を有する、請求項17~20のいずれか1項に記載の装置。
- 前記装置は、0.6~0.8の間の開口数を有する、請求項17~21のいずれか1項に記載の装置。
- 前記装置は少なくとも1mm2の視野を有する、請求項17~22のいずれか1項に記載の装置。
- 前記4つのダブレットレンズは、第1のレンズおよび第2のレンズを有する第1のダブレットレンズを備えており、前記第1のダブレットレンズの前記第1のレンズは凹面および凸面を有し、前記第1のダブレットレンズの前記第2のレンズは凹面および凸面を有する、請求項17~23のいずれか1項に記載の装置。
- 前記4つのダブレットレンズは、第1のレンズおよび第2のレンズを有する第2のダブレットレンズをさらに備えており、前記第2のダブレットレンズの前記第1のレンズは凹面および凸面を有し、前記第2のダブレットレンズの前記第2のレンズは凹面および凸面を有する、請求項24に記載の装置。
- 前記4つのダブレットレンズは、第1のレンズおよび第2のレンズを有する第3のダブレットレンズをさらに備えており、前記第3のダブレットレンズの前記第1のレンズは2つの凸面を有し、前記第3のダブレットレンズの前記第2のレンズは2つの凹面を有し、前記第2のダブレットレンズは前記第1のダブレットレンズと前記第3のダブレットレンズとの間にある、請求項25に記載の装置。
- 前記4つのダブレットレンズは、第1のレンズおよび第2のレンズを有する第4のダブレットレンズをさらに備えており、前記第4のダブレットレンズの前記第1のレンズは2つの凹面を有し、前記第4のダブレットレンズの前記第2のレンズは2つの凸面を有し、前記第3のダブレットレンズは前記第2のダブレットレンズと前記第4のダブレットレンズとの間にある、請求項26に記載の装置。
- 前記第2のダブレットレンズと前記第3のダブレットレンズとの間の第1のシングレットレンズであって、前記第1のシングレットレンズは2つの凸面を有する、前記第1のシングレットレンズをさらに備える、請求項27に記載の装置。
- 前記第2のダブレットレンズと前記第3のダブレットレンズとの間の第2のシングレットレンズであって、前記第2のシングレットレンズは2つの凸面を有する、前記第2のシングレットレンズをさらに備える、請求項28に記載の装置。
- 前記非球面レンズは、前記第3のダブレットレンズと前記第4のダブレットレンズとの間にある、請求項29に記載の装置。
- 前記開口絞りは、前記第3のダブレットレンズと前記第4のダブレットレンズとの間にある、請求項30に記載の装置。
- 前記第1のダブレットレンズから前記第4のダブレットレンズを通過した後に光線が無限遠補正されない、請求項31に記載の装置。
- 前記対物レンズは、前記対物レンズの屈折力に影響を与える22を超えないレンズ面、4つのダブレットレンズからの16面、2つのシングレットレンズからの4面、および前記非球面レンズからの2面を有する、請求項17~32のいずれか1項に記載の装置。
- DNAシークエンシング用のシステムであって、
請求項17~33のいずれか1項に記載の装置と、
核酸のアレイの付着のために構成された固体支持体と、
前記対物レンズを使用して前記アレイを撮像するように構成されたカメラと
を備える、前記システム。 - 前記対物レンズと前記カメラとの間のチューブレンズであって、前記チューブレンズは、前記対物レンズからの光を前記カメラのセンサに集束させるように構成されている、前記チューブレンズをさらに備える、請求項34に記載のシステム。
- 前記固体支持体はフローセルの一部であり、前記フローセルは、0.3ミリメートル以上2.0ミリメートル以下の厚さを有するカバースリップを有する、請求項34または請求項35に記載のシステム。
- 前記対物レンズの第1のレンズは、前記固体支持体からある距離だけ離されており、前記距離は、10ミリメートル以下3ミリメートル以上である、請求項34~36のいずれか1項に記載のシステム。
- 前記固体支持体に付着させた前記核酸のアレイをさらに備える、請求項34~37のいずれか1項に記載のシステム。
