上記キメラ抗原受容体をコードするヌクレオチド配列を合成して増幅し、上記ヌクレオチド配列をレンチウイルス発現ベクターへクローニングし、好ましくは、上記ScFv-CD8-4-1BB-CD3ζ融合タンパク質のコーディングヌクレオチド配列を合成し、ScFv-CD8-4-1BB-CD3ζ融合タンパク質のコーディングヌクレオチド配列を含むレンチウイルスベクターを得るステップaと、
レンチウイルスパッケージングに必要なプラスミド及びステップaで得られたレンチウイルス発現ベクターによりレンチウイルスパッケージング細胞株においてパッケージングを行い、レンチウイルスを製造するステップbと、
ステップbで得られたレンチウイルスをNK細胞に感染させ、ROBO1 CAR-NK細胞を得るステップcと、
を含む方法により製造される。
まず、ROBO1 CAR-NK細胞は、腫瘍系疾患の治療薬としてROBO1分子高発現腫瘍の治療に適用でき、且つサイトカインストームなどの有害現象がなく、従来の手術、化学療法、放射線療法に効果のない腫瘍に対して新しい治療法を提供する。また、NK92細胞をそこにCARを導入することで改変することにより標的性は大幅に向上し、CARにおける標的は抗腫瘍標的ROBO1であり、標的性を向上させるとともに、その抗腫瘍性能を大幅に向上させる。さらに、臨床前の実験により、現在使用されているROBO1CAR-NKの毒性や副作用はCAR-Tよりも小さく、殺傷効果はより高いことを発見した。つまり、本発明で構築されるROBO1 CAR-NK細胞は、まず、NK92細胞をそこにCARを導入することで改変することによりその標的性が向上し、そして、NK92細胞はリンパ腫患者の末梢血から分離して構築されたものであるため、MHC制限なしで標的細胞を識別でき、前感作なしで腫瘍細胞を殺傷できるとともに、一連のサイトカインを産生して生体の獲得免疫を調節できるので、ROBO1 CAR-NKは、二次免疫を調節できる作用も有する。しかし、人体に使用した後、腫瘍が形成される可能性がある。
NK-92細胞(CC(登録商標)CRL-2407);乳がんBT474、T47D、HCC1187、HCC1937、MCF7、MDAMB-231、MDAMB-453、MDAMB-468、ZR-75-1;肺がん細胞A549、H1299;肝がん細胞Huh7、SMMC7721及びHEPG2;膵臓がん細胞BxPC3、PANC1、Capan-2、及びLenti-X-293T細胞;結腸直腸がん細胞HT-29、LoVo及びHCT116は、いずれも中国科学院上海生科院細胞資源センターから購入した。
ScFv(Anti ROBO1-FN3)-CD8(CD8TM)-4-1BB-CD3ζ融合遺伝子配列(そのアミノ酸配列は配列番号8に示され、遺伝子配列は配列番号10に示される)及び変異型ScFv (Anti ROBO1-FN3)-CD8(CD8TM)-4-1BB-CD3ζ融合遺伝子配列(そのアミノ酸配列は配列番号9に示され、遺伝子配列は配列番号11に示され、その変異部位GCCはGCGであってもよい)をそれぞれ合成した。ScFV(Anti ROBO1-FN3)-CD8TM-4-1BB-CD3ζ融合遺伝子を例としてROBO1 CAR-NK細胞の製造過程を説明する。変異型ScFV(Anti ROBO1-FN3)-CD8TM-4-1BB-CD3ζ融合遺伝子を用いてROBO1M CAR-NK細胞を製造する過程はそれと同じである。
(1)トランスフェクションの24時間前、約8×106個の細胞/皿でLenti-X-293T細胞を15cmペトリ皿に接種した。トランスフェクション中に細胞が約80%のコンフルエントでペトリ皿に均一に分布することを確保するようにした。
溶液A:6.25ml 2×HEPES buffer緩衝液(5つの大きな皿を一緒にパッケージングした量;最も効果的である)。
112.5μg pRRLSIN-ScFv (anti ROBO1-FN3)(target plasmid);39.5μg pMD2.G(VSV-G envelop);73μg pCMVR8.74(gag,pol,tat,rev);625μl 2Mカルシウムイオン溶液。溶液Bの総体積は6.25mlであった。
NK92細胞密度を(2~3)×105個/mlに調整し、ウイルス:細胞培地=1:5の体積比(V/V)で実施例2で製造されたウイルスを添加し、同時にポリブレン8μg/mlを添加した。4h後に、等量の新鮮な完全培地を補充して細胞密度を1×105個/mlに調整し、引き続き培養した。翌日、すべての細胞を遠心分離し、新鮮な培地を加え、引き続き培養した。細胞密度が(2~3)×105個/mlに維持されるように1~2ごとに培養液を補充した。72h後にCAR抗体染色を行い、フローサイトメトリーによりROBO1 CAR NK-92陽性細胞を選別して拡大培養した。培地の色の変化、細胞密度、細胞形態を毎日観察し、記録した。
実施例4:ROBO1-iCaspase9 CAR-NK細胞の製造

実施例4で製造されたROBO1-iCaspase9CAR-NK細胞の全ゲノムをシークエンシングしてNK-92細胞ゲノムでの挿入部位を検出した。シークエンシング結果を図7に示す。PCR方法によりシークエンシング結果を検証したところ、iCaspase9遺伝子が2番染色体に挿入され、ROBO1遺伝子が10番染色体に挿入された。
実施例4で構築されたROBO1-iCaspase9CAR-NK細胞を培養し、異なるクローンのROBO1-iCaspase9CAR-NK細胞をそれぞれATCG427B-1D11、ATCG427B-1E1、ATCG427B-1F5、ATCG427B-1F7、ATCG427B-1F9、ATCG427B-2C3、ATCG427B-2D5と表記し、そしてフローサイトメトリーによりこれらの細胞におけるタグ抗体Flag及びCD19の陽性率を検出し(具体的には、それぞれフローサイトメトリー用抗体Flag及びCD19により細胞を暗所で15min染色した後、PBSで2回洗浄し、検出する)、細胞安定性の指標とした。結果を図8に示す。
異なる量のAP1903医薬品を実施例6でスクリーニングされた自殺遺伝子を持つROBO1 CAR-NK細胞(ATCG427B-1F7及び2D5)に加え、自殺遺伝子を持つROBO1 CAR-NK細胞(ATCG427Bと表記する)アポトーシスに対する影響を検出し、AP1903医薬品をROBO1 CAR-NK細胞に加えて対照とし、3つの実験群に分け、異なる濃度のAP1903医薬品(0nM、10nM、50nM)で細胞を2h、4h及び24h処理し、次いで細胞アポトーシスの状況を確認した。結果を図11に示す。それぞれ4h及び24h処理した後の実験群に対して新鮮な培地を交換して引き続き培養し、3日後に細胞の生存率をカウントした。結果を図12に示す。さらに、4h処理した後の0nM、10nM実験群に対して新鮮な培地を交換して24h培養した後、写真を撮った。結果を図13に示す。
異なる乳がん細胞BT474、T47D、HCC1187、HH1937、HCC38、MCF7、MDAMB-231、MDAMB-453、MDAMB-468及びZR-75-1を培養した後、Western Blotにより異なる乳がん細胞におけるROBO1の発現状況を検出した。結果を図14aに示す。ImageJソフトウェアによりWestern Blotで測定したROBO1の光学密度と対照GAPDHの光学密度との比を分析した。結果を図14bに示す。
1.乳がん細胞に対する自殺遺伝子を持つROBO1 CAR-NK細胞の殺傷
CFSE染色方法(A Mathematical Model of Natural Killer Cell Activity, Anna Scherbakova,1,2 Helen Lust,1,2 Hele Everaus,1,2,3 Alar Aints1,2,4*を参照)により、実施例5でスクリーニングされた自殺遺伝子を持つROBO1 CAR-NK細胞(ATCG427B-1F7、ATCG427B-2D5)の殺傷効果を検出し、ROBO1 CAR-NK及びNK-92細胞を対照とした。異なる乳がん細胞には、BT474、T47D、HCC1187、HCC1937、MCF7、MDAMB-231、MDAMB-453、MDAMB-468及びZR-75-1が含まれる。
(1)標的細胞を事前に処理し、1×106個の細胞/1μlでCFSEを加え、暗所でインキュベーターで15minインキュベートし、10%のBSAで停止させ、PBSで2回洗浄し、24ウェルプレートに調製された500μl腫瘍細胞懸濁液(8×104個の細胞/ウェル)を加えた。培養プレートをインキュベーターで12h前培養した。
(2)エフェクター細胞と標的細胞との比が0.5:1及び1:1であるように500μlのエフェクター細胞を加え、各実験につき3重測定し、エフェクター細胞と標的細胞を2時間インキュベートした。
(3)2時間後、上清を1.5のEP管に移し、各ウェルに200μlのパンクレアチンを加え、1min消化した後、移された上清を用いて細胞をピペッティングし、遠心分離し、PBSで再懸濁させ、1ulの7-AADを加え、暗所で15minインキュベートした後、検出した。
(4)殺傷率=(標的細胞の殺傷率-自発的死亡率)/(1-自発的死亡率)100%;
上記1と同じ方法により他の種類の腫瘍細胞を2h殺傷し、実施例5でスクリーニングされた自殺遺伝子を持つROBO1 CAR-NK細胞(ATCG427B-1F7、ATCG427B-2D5)の異なる腫瘍細胞に対する殺傷効果を検出した。異なる腫瘍細胞には、肺がん細胞A549、H1299;肝がん細胞Huh7、SMMC7721;膵臓がん細胞BxPC3、PANC1、Capan-2が含まれる。培養した後、殺傷を行った。結果を図16a-cに示す。図から分かるように、ROBO1-iCaspase9細胞の肺がん及び肝臓がんに対する殺傷効果は比較的良好である。
健常者を3名ランダムに選択し、それぞれ50ml新鮮な血液を採取し、PBSを用いて1:1で希釈し、新しい遠沈管を取り、Ficoll(リンパ球分離液)を入れ、管を45度傾斜し、希釈した血液をFicoll:血液=2:1でFicoll液面の上方にゆっくりと加え、2000rpmで30min遠心分離した後、ピペットを雲層に直接挿入し、雲層(即ち、単一の核細胞層)を吸い出し、新しい遠沈管に入れ、洗浄して上清を捨て、1mlのRPMI-1640培地を加え、ピペッティングして均一に混合することにより、PBMC細胞懸濁液を製造し、それをそれぞれPBMC-1、PBMC-2、PBMC-3とし、正常乳腺細胞及びPBMCにおけるROBO1の発現状況を検出した。結果を図17aに示す。上記1と同じ方法により、ROBO1-iCaspase9細胞の正常乳腺細胞PBMC-1、PBMC-2、PBMC-3及びMDA-kb2に対する殺傷効率を検出し、殺傷過程においてPBMC-1、PBMC-2、PBMC-3及びMDA-kb2細胞に対するNK92とROBO1 CAR-NKの殺傷率を比較した。結果を図17bに示す。図から分かるように、ROBO1-iCaspase9CAR-NK細胞は、インビトロで正常乳腺及びPBMCに対する殺傷が顕著ではないことで、自殺遺伝子を持つROBO1 CAR-NK細胞医薬品の安全性がさらに証明されている。
ROBO1 CAR-NK細胞が異体細胞として人体に注入された後、体内のPBMC免疫細胞は、異体由来細胞に対して殺傷効果を発揮する可能性がある。ROBO1 CAR-NK細胞がPBMCによって除去されるか否かを検証するために、インビトロで、健常者を3名ランダムに選択し、採血し、分離してPBMC細胞を製造し(製造方法は上記方法と同じ)、それぞれPBMC-4、PBMC-5、PBMC-6と表記し、上記1と同じ方法によりROBO1-iCaspase9CAR-NK細胞に対するPBMC-4、PBMC-5、PBMC-6の殺傷を検出し、各群の実験においてNK92及びROBO1 CAR-NK細胞に対するPBMCの殺傷を比較した。結果を図18に示す。図18から分かるように、PBMCはインビトロでROBO1-iCaspase9細胞に対する殺傷が顕著ではなく、これによって、自殺遺伝子を持つROBO1 CAR-NK細胞医薬品は、インビボで特異的殺傷作用をより良好に発揮できることがさらに証明されている。
を合成した(配列番号12;最初と最後の斜体で下線付きの部分は酵素切断部位である)(その簡略化構造を図20に示す)。さらにそれをレンチウイルスベクターへクローニングし、図19に示されるレンチウイルスベクター(1942-3-hEGFRtと呼ばれる)(そのヌクレオチド配列は配列番号13に示される)を得た。さらに3プラスミドシステム(補助プラスミドpMD2.G及びpCMVR8.74)によりウイルスをパッケージングし、そしてウイルス感染により実施例3で製造されたROBO1CAR-NK細胞(又はROBO1M CAR-NK細胞;以下の実施例に用いるのはROBO1M CAR-NK細胞)に感染させ、次いでフローサイトメトリーによりROBO1とEGFRtが共に陽性である細胞、即ち自殺遺伝子を持つROBO1 CAR-NK細胞(ROBO1-EGFRt CAR-NK細胞;427Eとも呼ばれる)を選別し、スクリーニングされた細胞をモノクローナルスクリーニングして培養し、その後、培養した細胞をフローサイトメトリーにより自殺遺伝子を持つROBO1 CAR-NK細胞の陽性率を検出した。結果を図21a-bに示す。図から分かるように、APC及びPE蛍光標識の信号値はいずれも顕著に高くなり、2つの自殺遺伝子を持つROBO1 CAR-NKモノクローナル細胞(ROBO1-EGFRt CAR-NK細胞、即ち427E)が正常にスクリーニングにされ、それぞれROBO1-EGFRt-3C10-2D10及びROBO1-EGFRt-3C10-1F11(それぞれ427E-3C10-2D10及び427E-3C10-1F11とも呼ばれる)と表記した。この2つのモノクローナル細胞のマザークローンはROBO1-EGFRt-3C10(427E-3C10)である。
実施例10で製造されたモノクローナル細胞株及び上記2つのモノクローナル細胞株のマザークローンに対して腫瘍特異的殺傷の実験を行った。実験過程は以下の通りである。まず、Westernにより乳がん細胞以外の他の異なる種類のがん細胞におけるROBO1タンパク質の発現レベルを検出し、具体的には、異なるがん細胞株を収集し、総タンパク質を抽出して分解し、BCAキットを用いて総タンパク質を定量し、次いでSDS-PAGEゲルによりROBO1タンパク質に対して電気泳動、転写膜を行い、最後に現象してROBO1タンパク質を検出した。一部の乳がん細胞におけるROBO1の発現状況を図14aに示す。結腸直腸がん(例えば、HT-29、LoVo及びHCT116)、肝臓がん(例えば、SMMC7721、Huh7及びHEPG2)の検出結果を図22に示す。結果により、異なる種類のがん細胞におけるROBO1タンパク質の発現には確かに明らかな違いがあることが示されている。
安全性研究は、自殺スイッチインデューサー(セツキシマブ)に対する殺傷/エフェクター細胞の存在でのROBO1-EGFRt CAR-NK細胞の感度を研究するものである。インデューサーであるセツキシマブによりROBO1-EGFRt CAR-NK細胞(実施例10で製造されたモノクローナル細胞株及びこの2つのモノクローナル細胞株のマザークローン)のアポトーシスを誘導し、インビトロで構築されたNK92-CD16(高親和力)エフェクター細胞を添加して自殺スイッチの有効性を評価した。それぞれエフェクター細胞の作用濃度、エフェクター/標的比、作用時間、作用システムなどについて考察した結果、インデューサーであるセツキシマブの最適化作用条件は、インビトロ、作用時間4時間、作用濃度1μg/ml、NK92-CD16(高親和力)、ROBO1-EGFRt CAR-NK細胞のエフェクター/標的比25:1(E:T)であった。検出方法は、CFSE(A Mathematical Model of Natural Killer Cell Activity, Anna Scherbakova,1,2 Helen Lust,1,2 Hele Everaus,1,2,3 Alar Aints1,2,4*を参照)を採用し、7-AAD/CFSE二重染色により細胞アポトーシス効率を検出した。具体的には、構築されたNK92-CD16エフェクター細胞を事前にCFSEで標識し、そしてエフェクター/標的比でROBO1-EGFR CAR-NK及びNK92-CD16細胞をプレートに敷いた後、それぞれ作用濃度が0、1及び10μg/mlのセツキシマブを加え、4時間作用した後、7-AADにより10min染色し、そしてフローサイトメーターにより検出した(実験を並行して3回繰り返した)。検出結果を図24に示す。結果から分かるように、スクリーニングされたサブクローン細胞株において、セツキシマブによって誘導されたアポトーシス効率は40%~50%であった。

前記EGFRtは、EGFRの細胞外ドメインIII、細胞外ドメインIV及び膜貫通領域を含み、
前記EGFRtのヌクレオチド配列は、配列番号12に示されることを特徴とする、請求項1に記載の核酸。 - 前記抗原結合ドメインは、ROBO1のFN3ドメインに特異的に結合する抗体又はその抗原結合断片であり、
前記抗原結合断片は、ScFvであることを特徴とする、請求項1に記載の核酸。 - 前記ScFv-CD8-4-1BB-CD3ζ融合タンパク質のアミノ酸配列は、配列番号9に示されることを特徴とする、請求項1に記載の核酸。
- 請求項1から9のいずれか1項に記載の核酸を含むことを特徴とする、自殺遺伝子を持つ構築物。
- 前記自殺遺伝子がiCaspase9である場合、前記構築物のヌクレオチド配列は、配列番号7に示され、或いは
前記自殺遺伝子がEGFRtである場合、前記構築物のヌクレオチド配列は、配列番号13に示されることを特徴とする、請求項10に記載の構築物。 - 請求項1から9のいずれか1項に記載の核酸によってコードされることを特徴とする、自殺遺伝子を持つキメラ抗原受容体。
- 請求項12に記載のキメラ抗原受容体を発現することを特徴とする、自殺遺伝子を持つROBO1 CAR-NK細胞。
- 肺がん細胞、膵臓がん細胞、肝がん細胞、乳がん細胞、結腸がん細胞、前立腺がん細胞又は胃がん細胞を効果的に殺傷させるか又は殺すことができることを特徴とする、請求項13に記載の自殺遺伝子を持つROBO1 CAR-NK細胞。
- 請求項13又は14に記載の自殺遺伝子を持つROBO1 CAR-NK細胞の製造方法であって、
請求項1から9のいずれか1項に記載の核酸を合成して増幅し、前記核酸をレンチウイルス発現ベクターへクローニングし、自殺遺伝子を持つレンチウイルスベクターを得るステップ(1)と、
さらに、レンチウイルスパッケージング細胞株、及びステップ(1)で得られた自殺遺伝子を持つレンチウイルスベクターを含む3プラスミドシステムによりレンチウイルスをパッケージングして自殺遺伝子を持つレンチウイルスを得た後、前記自殺遺伝子を持つレンチウイルスをROBO1 CAR-NK細胞に感染させることで自殺遺伝子をそのゲノムに導入し、自殺遺伝子を持つROBO1 CAR-NK細胞を得るステップ(2)と、
を含むことを特徴とする、製造方法。 - 前記ROBO1 CAR-NK細胞は、
請求項1から9のいずれか1項に記載の核酸を合成し、ScFv-CD8-4-1BB-CD3ζ融合タンパク質のコーディングヌクレオチド配列を含むレンチウイルスベクターを得るステップaと、
レンチウイルスパッケージングプラスミド及びステップaで得られたレンチウイルス発現ベクターによりパッケージング細胞株においてパッケージングを行い、レンチウイルスを製造するステップbと、
ステップbで得られたレンチウイルスをNK細胞に感染させ、ROBO1 CAR-NK細胞を得るステップcと、
を含む方法により製造されることを特徴とする、請求項15に記載の製造方法。 - 請求項13若しくは14に記載の自殺遺伝子を持つROBO1 CAR-NK細胞又は請求項15若しくは16に記載の製造方法により製造される自殺遺伝子を持つROBO1 CAR-NK細胞を含む、ことを特徴とする医薬組成物。
- 前記医薬組成物はインデューサーをさらに含み、
前記自殺遺伝子がiCaspase9である場合、前記インデューサーはAP1903又はAP20187であり、前記インデューサーの濃度は0~50nMであり、
前記自殺遺伝子がEGFRtである場合、前記インデューサーはセツキシマブであり、その作用濃度が1μg/mlであることを特徴とする請求項17に記載の医薬組成物。 - 前記自殺遺伝子を持つROBO1 CAR-NK細胞と腫瘍細胞とのエフェクター/標的比は(0.5~5):1であることを特徴とする請求項17に記載の医薬組成物。
- 前記エフェクター/標的比は(0.5~1):1であることを特徴とする、請求項19に記載の医薬組成物。
- 薬学的に許容される塩、担体、賦形剤及び補助材料をさらに含むことを特徴とする、請求項17に記載の医薬組成物。
- がんを治療及び/又は予防するための医薬品の製造における請求項13若しくは14に記載の自殺遺伝子を持つROBO1 CAR-NK細胞又は請求項15若しくは16に記載の製造方法により製造される自殺遺伝子を持つROBO1 CAR-NK細胞の使用であることを特徴とする、使用。
- 前記がんは、肝臓がん、乳がん、結腸がん、膵臓がん、前立腺がん、胃がん又は肺がんであることを特徴とする、請求項22に記載の使用。
- 前記医薬品は、腫瘍内投与剤形の医薬品、腫瘍内注射剤形の医薬品又は静脈内注入剤型の医薬品を含むことを特徴とする、請求項22に記載の使用。
- がんを治療及び/又は予防する注射剤であって、前記注射剤は請求項13若しくは14に記載の自殺遺伝子を持つROBO1 CAR-NK細胞、請求項15若しくは16に記載の製造方法により製造される自殺遺伝子を持つROBO1 CAR-NK細胞、又は請求項17から21のいずれか1項に記載の医薬組成物を含む、
ことを特徴とするがんを治療及び/又は予防する注射剤。 - 前記がんは、肝臓がん、乳がん、結腸直腸がん、膵臓がん、前立腺がん、胃がん又は肺がんであることを特徴とする請求項25に記載の注射剤。
- 前記がんは、乳がん、結腸直腸がん又は肝臓がんであることを特徴とする、請求項25に記載の注射剤。
- 前記自殺遺伝子を持つROBO1 CAR-NK細胞の注射量は(0.5~5)×109細胞/回であることを特徴とする、請求項25に記載の注射剤。
- 前記注射剤の投与方法は、腫瘍内注射、静脈注射、胸腔内注射又は局所的介入であることを特徴とする、請求項25に記載の注射剤。
- 前記注射剤の投与方法は静脈注射であることを特徴とする、請求項29に記載の注射剤。
