Description
化学及び治療薬の分野において、改善された生理化学的性質を有する式(I)の新しい塩及び結晶形態の同定と、そのような塩及び結晶形態を再現可能に作製するための方法が必要とされている。
本発明は、系統名4-アミノ-N-(1-((3-クロロ-2-フルオロフェニル)アミノ)-6-メチルイソキノリン-5-イル)チエノ[3,2-d]ピリミジン-7-カルボキサミドを有し、以下の式によって表されうる、汎RAF阻害剤の式(I)の塩及び結晶形態に関する。
[0031]工程1:式(I)の化合物の二(p-トルエンスルホン酸塩)の非晶質形態の調製
[0040]式(I)の化合物の遊離塩基3gを反応器に入れ、ついで、50mLのACNを加えた。懸濁混合物に、ACN(10mL)中のp-トルエンスルホン酸一水和物(2.2当量)を加えた。得られた固形物を20~25℃で24時間撹拌し、濾過し、ついで50mLのACNで洗浄した。濾過した固形物をオーブンで50℃で18時間乾燥させた。収量:4.92g(95%)。
[0046]工程1:式(I)の化合物の粗ジエタンスルホン酸塩の調製
[0055]工程1:式(I)の化合物の粗ジメタンスルホン酸塩の調製
[0064]200.0gの式(I)の化合物の二塩酸塩(2HCl)の結晶形態を10Lの反応器に入れ、ついで1.0LのDMSOと4.0LのMeOHを加えた。得られた懸濁液を55~60℃に加熱して、メカニカルスターラーで撹拌しながら式(I)の化合物を溶解させた。反応器の温度を55~60℃に維持し、200rpmで撹拌しながら、347mLのDIPEAを反応混合物に2時間かけて添加した。反応器の温度を55~60℃に維持し、50~60rpmで弱く撹拌しながら、得られた混合物を加熱し、36時間撹拌して、沈殿物を形成した。反応混合物を20~25℃に冷却し、ついで6時間撹拌した。生成された沈殿物を濾過し、ついで10.0LのMeOHで洗浄した。濾過した固形物を真空オーブンで40℃において48時間乾燥させた。収量:146g(94.6%)。
Claims (5)
- ビス塩酸塩の多形形態Iが、Cu-Kα光源で照射した場合、5.89°±0.2°、7.77°±0.2°、8.31°±0.2°、16.68°±0.2°、及び26.89°±0.2°の回折角2θ値のピークから選択される3以上のピークを有する粉末X線回折パターンによって特徴付けられる、請求項1に記載の化合物。
- ビス塩酸塩が三水和物である、請求項1又は2に記載の化合物。
- 請求項1から3の何れか一項に記載の化合物と、少なくとも一種の薬学的に許容される賦形剤とを含む薬学的組成物。
- 異常細胞増殖疾患がプロテインキナーゼの異常活性化によって引き起こされる、哺乳動物の異常細胞増殖疾患を治療するための請求項4に記載の薬学的組成物。
