本明細書の解釈を目的として、下記の定義が適用されることになると共に、適切であれば常に、単数形で使用される用語は複数形も含むことになり、逆もまた同様である。以下に示す任意の定義が、参照によって本明細書に組み込まれる任意の文書と矛盾する際は、示す定義が優先するものとする。
抗NSM抗体の使用を含む、非刺激性骨髄系細胞(NSM)の無能化及び/または検出を目的とする方法及び組成物が本明細書で提供される。NSMタンパク質を発現する非刺激性骨髄系細胞の標的化及び/または検出を目的とする方法及び組成物も本明細書で提供される。

本明細書では、刺激性骨髄系細胞(ある特定の態様では、SDCとも呼ばれる)は、免疫応答の刺激に有効な骨髄系細胞である(例えば、非刺激性骨髄系細胞と比較して、腫瘍微小環境における抗腫瘍応答の刺激に、より有効である)。いくつかの実施形態では、刺激性骨髄系細胞は、非刺激性骨髄系細胞と比較して、T細胞に対する抗原(例えば、腫瘍抗原)提示に有効であるか、または腫瘍特異的なT細胞応答の刺激に有用である。いくつかの実施形態では、刺激性骨髄系細胞は、非刺激性骨髄系細胞と比較して、T細胞に対する腫瘍関連抗原の取り込み、処理、及び/または提示の高い能力を示し得る。刺激性骨髄系細胞は、非刺激性骨髄系細胞と比較して、細胞傷害性Tリンパ球の高い再刺激能力を有し得るか、または場合によっては、有効な腫瘍細胞死滅刺激を行うことができる。刺激性骨髄系細胞は、非刺激性骨髄系細胞と比較して、抗原処理、抗原提示、及び/または抗原同時刺激に関与する遺伝子及び細胞表面マーカーの高い発現量を示し得り、当該遺伝子及び細胞表面マーカーには、限定はされないが、CD80、CD86、MHCI、及びMHCIIが含まれる。
本出願は、抗体及び組成物を提供し、当該組成物は、非刺激性骨髄系細胞を無能化する抗体を含む、NSMタンパク質に結合する抗体を含む。
(表B)CH2ドメイン及びエフェクター機能の操作
NSMタンパク質を発現する非刺激性ヒト骨髄系細胞の無能化及び/または検出を目的とする方法及び組成物が、本明細書で提供される。いくつかの実施形態では、本発明は、NSMタンパク質のホモログを発現する非ヒト哺乳類細胞に由来する非刺激性骨髄系細胞の無能化及び/または検出に関する。例えば、マウスにおけるNSMタンパク質の発現パターンは、そのヒトホモログと同等に限定されたパターンを示し得る。したがって、1つの実施形態では、NSMタンパク質を発現する非刺激性マウス骨髄系細胞の無能化及び/または検出を目的とする方法及び組成物が、本明細書で提供される。NSMタンパク質のホモログを類似の発現パターンで発現する任意の個体に由来する非刺激性細胞であって、その細胞が、NSMタンパク質のものと同等の発現パターンを示す非刺激性細胞の無能化及び/または検出を目的とする類似の方法及び組成物も、本明細書で提供される。
本出願は、抗体を含む組成物を提供し、当該組成物には、本明細書に記載の抗体のいずれか1つまたは複数を、1つまたは複数の医薬的に許容可能な添加剤と共に含む医薬組成物が含まれる。いくつかの実施形態では、組成物は、無菌である。医薬組成物は、一般に、有効量の抗体を含む。
使用方法
1つの態様では、本出願は、非刺激性骨髄系細胞と、ヒト抗体などの抗NSM抗体と、の接触方法を提供し、当該方法は、非刺激性骨髄系細胞の無能化をもたらす。
いくつかの実施形態では、本明細書で提供される方法は、個体における免疫関連の症状の治療に有用である。1つの実施形態では、個体はヒトであり、抗体はNSM抗体である。別の実施形態では、個体はマウスであり、抗体は、NSM抗体である。
本発明は、SDC及び/またはNSM細胞の定量を目的とする任意の方法論を包含する。さまざまな実施形態は、例えば、腫瘍試料における、SDC及び/またはNSM細胞の同定及び定量に関する。腫瘍試料は、当該技術分野において知られるように患者から得た腫瘍細胞を含む任意の組織であってよく、例えば、生検(例えば、針生検または例えば、4mmのパンチ生検といったパンチ生検)で取り出された腫瘍の一部であるか、または原発性もしくは転移性の細胞を含む、外科的に切除された実質的な全腫瘍である。SDCの存在量は、周縁領域と比較して、腫瘍の遠位領域で多いことに留意されることになる。典型的な試料では、この差異は、平均化されて取り除かれることになり、結果に影響を与ないことになる。しかしながら、過剰量の周縁材料が試料に含まれるのであれば、これによって結果に影響が及び、SDCの存在量が実際より低く数えられる可能性がある。
本発明のさまざまな実施形態は、腫瘍におけるSDCの存在量の決定に関する。そのような細胞の存在量は、さまざまな尺度によって評価してよい。そのような測定は、相対的な測定または絶対的な測定を含み得、直接的または間接的な測定を含み得る。例えば、相対的な存在量は、試料におけるNSM細胞に対するSDCの割合、試料におけるDC1細胞の数に対するSDCの割合、または試料内のすべての細胞型に対するSDCの割合、試料における全骨髄系細胞に対するSDCの割合、もしくは試料におけるHLA-DR+細胞に対するSDCの割合、を測定することによって決定してよい。いくつかの実施形態では、SDCの絶対的な存在量は、例えば、腫瘍組織のml当たりの全SDC数の尺度、または腫瘍組織のマイクログラム当たりの全SDC数の尺度として決定される。間接的な尺度には、SDCマーカー遺伝子の遺伝子発現レベルの単独評価、またはNSM細胞などの他の細胞型との、マーカー発現レベルの比較評価が含まれる。
本出願は、本明細書に記載の抗体組成物のいずれか1つまたは複数を含むキットを提供する。いくつかの実施形態では、二次抗体、免疫組織化学分析用の試薬、医薬的に許容可能な添加剤、及び説明書、ならびにそれらの任意の組み合わせのいずれかから選択される構成要素をさらに含む。1つの特定の実施形態では、キットは、1つまたは複数の医薬的に許容可能な添加剤と共に、本明細書に記載の抗体組成物のいずれか1つまたは複数を含む医薬組成物を含む。
マウスの腫瘍
腫瘍浸潤性骨髄系集団を精査するために、11色のフローサイトメトリーパネルを考案し、蛍光mCherryタンパク質及び卵白アルブミンを独立して共発現する癌遺伝子が開始するように操作された自発生乳腺腫瘍モデルであるPyMTChOVA(Engelhardt et al.,2012)使用して、段階的なゲート方針を用いた。出願人は、腫瘍性CD45+コンパートメントの特性を明らかにし、その多くが、貪食された腫瘍抗原を有しており、したがって、mCherryの蛍光を示している(図1A)。骨髄系特異的マーカーであるCD11b及び単球マーカーであるLy6Cによってすべての造血細胞を部分ゲートすることで、好中球及び単球を排除することが可能であった(データ未掲載)。MHCII+細胞内で、DCは、CD24高及びF4/80低である発現に基づいてマクロファージと区別され、そのいずれもあてはまらないか、単独であてはまれば、典型的には、この区別をする上で十分である。その後、健康な末梢組織で観測されたように、DCは、CD11b及びCD103の発現差異に基づいて2つの集団へと解析分類されることが明らかとなった(Hashimoto et al.,2011)。出願人は、2つのメラノーママウスモデル(B78ChOVA(mCherry及びOVAを発現するB16の変異体)、図1B及びBRAF V600E、データ未掲載)においてこうした集団を発見し、これは、マウス種(例えば、FVB PyMT、データ未掲載)、及び異所性腫瘍(ルイス肺癌、データ未掲載)にまたがるものであった。出願人は、識別及び議論を容易なものとするために、これ以後は、こうしたDC集団を「CD11b+DC1」及び「CD103+DC2」と称する。
出願人のゲーティング方針を検証するために、出願人は、ImmGenコンソーシアム(Gautier et al.,2012、Miller et al.,2012)によって定義される抗体パネルを適用した。出願人の「DC」の割り当てと一致して、B78chOVAモデル及びPyMTchOVAモデルにおいて、CD103+DC2は、CD135(Flt3)、CD117(cKit)、及びCD26を発現していた一方で、TAMの両集団はそれらを発現していなかった(図2A及びデータ未掲載)。驚くべきことに、CD11b+DC1は、検出可能なレベルのDCマーカーを発現しておらず、実際は、CD206、CD64、及びMerTKを含む「マクロファージ」マーカーのいくつかを発現しているという理由によって、TAM1及びTAM2とはさらに分離されるものであった(図2B及びデータ未掲載)。しかしながら、CD11b+DC1は、CD301b及びPDL2を僅かに発現しており、それらは両方共、皮膚において見出されるIRF4依存性の「DCTh2」集団を定義するために使用されてきたものである(図2C及びデータ未掲載)(Gao et al.,2013、Kumamoto et al.,2013)。
APCは、骨髄(BM)前駆細胞に由来すると共に、それがDC/マクロファージのサブセットへと分化することは、特定のサイトカインに依存している。こうした集団への分化を推進するサイトカインを決定するために、出願人は、qPCRによって、コロニー刺激因子(CSF)受容体の発現を、モデルを横断して調べた。TAM1、TAM2、及びCD11b+DC1においては、Csf1r(M-CSFR)が排他的に見出された一方で、DC1サブセット及びDC2サブセットにおいては、Csf2rb(GM-CSFR)が特異的に発現しており、Csf3r(G-CSF)は、すべてにおいて存在しなかった(図4A)。中和抗体での処理または異所性腫瘍を有するサイトカイン受容体欠損マウスのいずれかを使用して、出願人は、腫瘍におけるAPCのCSFサイトカインに対する依存性を機能的に試験した。
異なるAPCの系譜要件が確立されたため、次に出願人は、それらがT細胞応答を開始、関与、及び維持する能力について評価した。抗原の処理、提示、及び同時刺激に関して細胞を解析するために、出願人は、図2のRNA配列解析データを使用して、こうした経路に関与する遺伝子の転写物レベル及びタンパク質レベルを解析した。差異は、かなりのものであり、潜在的なAPC機能の広範囲にまたがるものであった(図5A)。注目すべきことに、CD80、CD86、及び2B4を含む、T細胞応答の制御に関与する分子の表面レベルは、集団間で同等であった一方で、CD103+DC2は、異なる転写特性を示し、当該特性は、T細胞相互作用を増進するであろうと予測される交差提示の増大、同時刺激の増進、及びケモカイン発現の増加と一致するものであった(図5A、図5B、及びデータ未掲載)。APC間においてMHCI及びMHCIIの発現に大きな違いはなく、例外として、CD103+DC2でMHCIが僅かに減少していた(図5C)。しかしながら、異所性腫瘍に由来し、エクスビボでのデキストランの取り込みによって外因的に測定された貪食能力では、TAM1/TAM2と比較して、CD103+DC2において有意な差が観測された(図5D)。
以前に出願人は、腫瘍から直接的に取得したとき、抗原を取り込んでいる骨髄系コンパートメントの総体がナイーブCD8+T細胞を刺激することは可能であるが、予め活性化されたCD8+T細胞を刺激することは不可能であることを発見した(Engelhardt et al.,2012)。しかしながら、CD103+DC2に特有の交差提示表現型に基づき、出願人は、腫瘍から新たに単離したそれぞれの集団のT細胞刺激能力を試験することを試みた。卵白アルブミンに特異的なOT-I CD8+T細胞と12時間共培養した後に、TCRシグナル伝達を強固に誘導する能力を有していたのはCD103+DC2集団のみであり、このことは、ナイーブOT-I CD8+T細胞及び予め活性化されたOT-I CD8+T細胞の両方において、早期T細胞活性化マーカーであるNur77及びCD69が増加したことによって証明された。重要なことに、このことは、異所性マウスモデル及び自発性マウスモデルの両方において一貫性を有していた(図6A及びデータ未掲載)。色素標識したOT-I CD8+T細胞との共培養を延長することで、CD11b+DC1集団及びCD103+DC2集団が、ナイーブCD8+T細胞を増殖させる最も強固な刺激因子であることが明らかとなり、貪食性腫瘍骨髄系細胞において以前に同定された刺激能力のほとんど全体が、こうしたDC内に特異的に存在することが示された(図6B~6C、及びデータ未掲載)。興味深いことに、CD103+DC2は、確立されたCTLの強力な増殖を特異的に誘導する能力を有しており、当該CTLは、他の集団によって刺激されなかったことから、CD103+DC2は、腫瘍におけるCTLの優れた交差提示刺激因子であることが示唆された(それぞれ図6D~6E、及びデータ未掲載)。
希少CD103+DC2のT細胞刺激能力が特有であることを考慮し、出願人は、こうした細胞の腫瘍内での空間的な組織化、及びそのT細胞との相互作用動力学に対する理解をインビボ及びインビトロの両方で試みた。生存している自発性腫瘍において、インビボでこうした集団を区別するために、PyMTchOVA対立遺伝子をCx3cr1-eGFP対立遺伝子及びCd11c-mCherry対立遺伝子へと交差させ、骨髄系コンパートメントにおいて3つの特異的な蛍光集団を生成させた(データ未掲載)。両DCサブセット(DC1/DC2)は、赤色で特異的に標識された(CD11c-mCherryのみ)一方で、TAM1集団及びTAM2集団は、それぞれ緑色(Cx3cr1-eGFP)ならびに黄色(CD11c-mCherry及びCx3cr1-eGFP)に標識された。この蛍光手法を、2光子生体内イメージングと共に使用して、出願人は、TAM1集団及びTAM2集団が腫瘍性病巣に密接して優先的に縁へと移動していることを観測した。この帯域は、出願人が以前に、T細胞が優先的に補足されることを発見した帯域である(Engelhardt et al.,2012)。対照的に、赤色のDCサブセットは、典型的には、腫瘍病巣から遠位にあるコラーゲンに富む別の帯域において見出され、遠位に局在化しているすべてのAPCの70%近くを構成するものであった(図7A及びデータ未掲載)。
出願人は、完全に侵襲性かつ増殖性の株であるEG7.1と比較して、自発的に退縮するEG7腫瘍細胞株では、CD11b+DC1とCD103+DC2との割合がほとんど反転していることを発見し、このことは驚きであった。自発的に退縮するEG7腫瘍細胞株は、これ以後はEG7.2と称す。この侵襲性の増殖株は、出願人が他のすべての侵襲性の腫瘍において観測したDCの相対的な割合を維持していた(データ未掲載)一方で、この自発的に退縮するモデルは、異常に多い数のCD103+DC2を含んでいた(データ未掲載)。出願人は、CD103+DC2を欠いているIrf8KO腫瘍モデルにおいては、腫瘍増殖が増加するが、Irf4条件的KOモデルでは増加しないことも観測した(データ未掲載)。まとめると、DC2の腫瘍性の存在量は、腫瘍制御において重要な役割を担い得ることが示唆されるが、増殖の差異は、その骨髄系細胞集団及びそのCTL刺激能力を超えて、こうした株における変動の大きさに相当し得るものである。それ故に、CD103+DC2が、効率的なCTL媒介性の腫瘍退縮に必要であるかどうかを正式に試験するために、出願人は、増殖性EG7.1腫瘍モデルを対象とし、活性化した腫瘍特異的T細胞による養子T細胞療法を実施して退縮を解析した(Helmich and Dutton,2001)。出願人は、こうした実験をzDC-DTRマウスにおいて実施した。当該マウスでは、腫瘍においてCD103+DC2を特異的に除去することが可能である(図3D)。腫瘍部位に対するCD103+DC2の影響を分離すると共に、LNでのプライミングのどのような影響も排除するために、(1)LNでのプライミングを必要とせず、典型的には、LNを通過しない活性化したOT-I CD8+CTL芽細胞の使用と、(2)LNへと流入する移入した希少CTL T細胞がLNから流出することを防ぐSIP1RアンタゴニストであるFTY-720での動物の処理と、である2つの方針を含めた実験を出願人は特別に設計した。FTY-720単独の影響が、移入したCTLが腫瘍の退縮を媒介することに対して与えた影響は最小限であった(データ未掲載)。しかしながら、出願人は、FTY-720と関連させてCD103+DC2を除去すると、CTLが効率的に腫瘍退縮を媒介する能力に対して重大な影響を与え、これによって、T細胞が媒介する腫瘍退縮が大幅に遅延することを発見した(図8A)。
ヒト腫瘍に対して、CD103+DC2の存在量が重要な役割を果たすかどうかを決定するために、出願人は、適合した予後データと共に、多数のヒト癌型に由来する相対的な遺伝子発現量を定量するTCGAアレイデータ(Cancer Genome Atlas Research et al.,2013、Hoadley et al.,2014)を利用した。出願人は、CD103+DC2を特徴付ける高レベル転写物を選択するために、出願人のRNA配列解析データを使用すると共に、TAM1/TAM2/CD11b+DC1細胞を特徴付けるが、CD103+DC2では低下している遺伝子のサブセットも選択した(それぞれ図8Bの上部及び下部の遺伝子)。出願人は、こうしたマウス遺伝子のヒトホモログを同定し、すべての癌型に由来する患者のTCGAデータにおけるこうした「特性」の発現をアッセイし、予後との関連性を評価した。比例ハザード生存率解析において、共変数として癌型をモデルに適合させることで、出願人は、こうした集団に由来する個々の遺伝子が予後に対してもたらす恩恵(ハザード比(HR)として示される)は僅かなものにすぎないことを観測した。しかしながら、出願人がCD103+とCD103-との遺伝子発現データの比を定義し、これをCox解析内の連続変数として使用すると、生存率の増加との高度に有意な関連性(BH p=0.00019)が存在することが観測された(図8B)。
メラノーマバイオインフォマティクス解析
腫瘍内APCの表現型は極めて多様であり、骨髄系は、マクロファージ及び樹状細胞の両方に類似した集団を複数形成することに寄与している。しかしながら、「マクロファージ」は、腫瘍の進行に対して抑制性である(DeNardo et al.,及びHanada et al.)一方で、おそらく、腫瘍内樹状細胞は刺激性である(Sandel et al.)のではないかと、長期にわたって考えられてきた。このことを明確かつ詳細に理解するための努力がなされてきたが、こうした細胞型同士を明瞭に区別するすべが存在しないことで大いに阻まれてきた。最近、出願人は、腫瘍内骨髄系細胞を区別するために、RNA配列解析と共に高次元のフローサイトメトリーを試みた。出願人は、実際に、交差提示を行う樹状細胞の小集団は、CTLに対して高度に刺激性であるが、他の真の「樹状細胞」サブセット、ならびに非常に多く存在するマクロファージ集団は、腫瘍抗原反応性のCD8T細胞の刺激に失敗することを発見した。希少な刺激性樹状細胞(SDC)は、マウスでは、インテグリンCD103の発現によって定義され、ヒトでは、CD141/BDCA3の発現によって定義される(Broz et al.)。
腫瘍の消化
次に、NSM及びSDCが、複数の異なるヒト腫瘍型にわたって存在するかどうかを決定した。転移性メラノーマ、頭頸部扁平上皮癌(HNSC)、及び結腸癌に由来するヒト腫瘍組織の生検を、SDC集団及びNSM集団の存在を対象として、フローサイトメトリーによって分析した。代表的なフローサイトメトリーは、段階的なゲーティングによって、記載のヒト腫瘍型(転移性メラノーマ、頭頸部扁平上皮癌(HNSC)、及び結腸癌)のすべてにおいてNSM集団及びSDC集団の両方が同定されたことを示している。図13を参照のこと。
次に、ある特定のNSMタンパク質が、NSMの細胞表面に発現しているかどうか、及びそうしたNSMタンパク質に抗NSM抗体が結合し得るかどうかを決定した。ある特定のNSMタンパク質が、SDCに発現しているかどうかも決定した。
消化した腫瘍組織におけるSDC集団及びNSM集団それぞれでのCCR7の特異的な発現をフローサイトメトリーによって分析した。データはすべてヒトの転移性メラノーマ細胞に由来する。図15は、NSM及び他の免疫細胞と比較して、CCR7がヒトSDCに特異的に発現していることを示す。
次に、ある特定のSDCタンパク質が、SDCの細胞表面に発現しているかどうか、及びそうしたSDCタンパク質に抗SDC抗体が結合し得るかどうかを決定した。ある特定のSDCタンパク質が、NSMに発現しているかどうかも決定した。
TREM2(クローン237920、RnD)、及びMS4A7(市販ポリクローナル、Human Protein Atlas)の発現を対象として、健康な野生型B6マウスの骨髄(BM)及び脾臓をフローサイトメトリーによって分析した。
次に、抗NSM抗体が、NSMを有する細胞をインビボで特異的に枯渇させることができるかどうかを決定した。
次に、抗NSM抗体がインビボで腫瘍の増殖を減少させることができるかどうかを決定した。
それぞれTdTomato及びGFPを発現する対照、及びNSMタンパク質を発現するEL4形質移入細胞を、例えば、それぞれの細胞型を5x10^5個として、1:1の比で混合し、野生型(WT)B6雄性マウスの腹腔へと注射する。2時間後、動物に対して、抗NSM抗体(例えば、100ug)、Fc対照、またはPBS対照を注射する。24時間後にマウスを屠殺し、腹膜洗浄によって収集し、腹膜から回収した細胞をフローサイトメトリーによって数える。抗NSM抗体は、NSMタンパク質を有する細胞をインビボで特異的に枯渇させる。抗NSM抗体は、TREM2、MS4A7、C5AR1、LYVE1、ABCC3、LILRB4、MRC1/CD206、SIGLEC1、STAB1、TMEM37、MERTK、及び/またはTMEM119に結合する。NSMタンパク質は、TREM2、MS4A7、C5AR1、LYVE1、ABCC3、LILRB4、MRC1/CD206、SIGLEC1、STAB1、TMEM37、MERTK、及びTMEM119から選択される。
6週齢の雄性B6マウスに対して癌細胞(例えば、MC38結腸癌)をT0で注射する。処理群へと無作為化したマウスに対して、腫瘍移植後の複数日(例えば、5日目、7日目、11日目、15日目)に、Fc対照または抗NSM抗体を腹腔内注射して処理する。投薬を決定する。例えば、Fc対照は、200ug/日で投与し、抗NSM抗体は、それぞれの日(例えば、5日目、7日目、11日目、及び15日目)に、40ug、20ug、20ug、及び40ug投与する。ノギスで腫瘍を測定し、腫瘍体積を決定する。抗NSM抗体は、対照と比較して、腫瘍増殖を減少させる。抗NSM抗体は、対照と比較して、実験マウスにおいて腫瘍に対する免疫応答を増進する。抗NSM抗体は、TREM2、MS4A7、C5AR1、LYVE1、ABCC3、LILRB4、MRC1/CD206、SIGLEC1、STAB1、TMEM37、MERTK、及び/またはTMEM119に結合する。
6週齢の雄性B6マウスに対して癌細胞(例えば、MC38結腸癌)をT0で注射する。処理群へと無作為化したマウスに対して、腫瘍移植後の複数日(例えば、5日目、7日目、11日目、15日目)に、Fc対照または抗NSM抗体を腹腔内注射して処理する。マウスは、免疫療法を以前に受けたことがあるか、現在受けているか、またはこれから受けることになる。免疫療法には、チェックポイント阻害物質を抑制する免疫療法、T細胞のチェックポイント阻害物質を抑制する免疫療法、抗PD1、抗PDL1、抗CTLA4、養子T細胞療法、CAR-T細胞療法、樹状細胞ワクチン、単球ワクチン、T細胞及び抗原提示細胞の両方に結合する抗原結合タンパク質、BiTE抗原結合タンパク質、トール様受容体リガンド、ならびに/またはサイトカインが含まれ得る。当該技術分野において利用可能な情報及び通常技術を使用して投薬を決定し、例えば、Fc対照は、200ug/日で投与し、抗NSM抗体は、それぞれの日(例えば、5日目、7日目、11日目、及び15日目)に、40ug、20ug、20ug、及び40ug投与する。ノギスで腫瘍を測定し、腫瘍体積を決定する。免疫療法と併せて抗NSM抗体を同時投与することで、非同時投与対照(例えば、単独療法)と比較して、腫瘍増殖の減少が増進する。免疫療法と併せて抗NSM抗体を同時投与することで、非同時投与対照(例えば、単独療法)と比較して、腫瘍に対する免疫応答が増進する。抗NSM抗体は、TREM2、MS4A7、C5AR1、LYVE1、ABCC3、LILRB4、MRC1/CD206、SIGLEC1、STAB1、TMEM37、MERTK、及び/またはTMEM119に結合する。

図21は、原発性ヒトHNSC腫瘍組織において、TREM2タンパク質の発現がNSM集団(CD14+TAM)に限定されており、SDC(BDCA3+DC)を含むCD14陰性CD11c陽性細胞ではほとんど発現していないか、または全く発現していないことを示す。TREM2に特異的な市販抗体(RnD、クローン237920)による染色を、消化したヒトHNSC腫瘍組織に対して実施し、二次対照染色(抗ラットIgG、Jackson Immunoresearch)と比較して、フローサイトメトリーによって分析した。この図は、ヒト腫瘍組織において、NSM細胞でNSM遺伝子産物が特異的に発現しており、SDC細胞では発現していないことを示す。集団は、生存、CD45+、系譜陰性、HLA-DR+、CD11c+でゲートし、CD14の発現によって分割した。

Claims (21)
- 骨髄細胞で発現するトリガー受容体2(TREM2)の細胞外ドメインに結合する抗体またはその抗原結合断片による治療を必要とするまたは該治療に応答する可能性のある対象を同定するために、癌を有する対象におけるTREM2陽性(TREM2+)骨髄細胞を検出する方法であって、
方法が、該対象から得られた試料におけるTREM2+骨髄細胞の存在を決定する段階を含み、
TREM2+骨髄細胞の存在が、対象が、該治療を必要とするまたは該治療に応答する可能性のあることを示し、
該抗体または抗原結合断片が、ヒトFcドメインを含み、かつ
抗体依存性細胞媒介性細胞傷害活性、抗体依存性細胞貪食活性、補体依存性細胞傷害活性、および抗体媒介性貪食(antibody-mediated phagocytosis)活性のうちの少なくとも1つを有する、
方法。 - 前記試料が、針生検、パンチ生検、または外科的切除によって得られる、請求項1に記載の方法。
- 前記試料が、癌組織試料または血液試料である、請求項1または2に記載の方法。

- 前記細胞選別方法が、蛍光活性化細胞選別、フローサイトメトリー、磁気活性化細胞選別、マイクロラフト選別(microraft sorting)、または親和性に基づく細胞分離の少なくとも1つを含む、請求項5に記載の方法。
- 前記対象が、免疫療法を以前に受けたことがある、請求項1~6のいずれか一項に記載の方法。
- 前記免疫療法が、チェックポイント阻害物質を抑制する免疫療法、T細胞のチェックポイント阻害物質を抑制する免疫療法、抗PD1、抗PDL1、抗CTLA4、養子T細胞療法、CAR-T細胞療法、樹状細胞ワクチン、単球ワクチン、T細胞及び抗原提示細胞の両方に結合する抗原結合タンパク質、BiTE抗原結合タンパク質、トール様受容体リガンド、またはサイトカインの少なくとも1つである、請求項7に記載の方法。
- 前記免疫療法が、ペンブロリズマブ、ニボルマブ、またはイピリムマブの少なくとも1つを含む、請求項7または8に記載の方法。
- TREM2の細胞外ドメインに結合する、抗体またはその抗原結合断片を含む、対象において癌を治療するためのまたはTREM2+骨髄細胞を死滅、無能化、もしくは枯渇するための医薬組成物であって、
該対象が、請求項1~9のいずれか一項に記載の方法によって、前記治療を必要とするまたは前記治療に応答する可能性がある対象と同定されており、
該抗体またはその抗原結合断片が、ヒトFcドメインを含み、かつ
抗体依存性細胞媒介性細胞傷害活性、抗体依存性貪食活性、補体依存性細胞傷害活性、または抗体媒介性貪食活性によって、TREM2+骨髄細胞を死滅、無能化、または枯渇するのに有効な量で医薬組成物中に存在する、
医薬組成物。 - 前記抗体が、モノクローナル抗体、IgG1抗体、IgG4抗体、アフコシル化(afucosylated)抗体、ヒト抗体、ヒト化抗体、キメラ抗体、または全長抗体の少なくとも1つである、請求項10に記載の医薬組成物。
- ヒトFcが、ヒトIgG1 Fcである、請求項11に記載の医薬組成物。
- 前記抗体が、アフコシル化抗体である、請求項11に記載の医薬組成物。
- 前記抗体が、ヒト化抗体である、請求項11に記載の医薬組成物。
- 医薬組成物の投与により、TREM2+骨髄細胞の死、TREM2+骨髄細胞のアポトーシス、TREM2+骨髄細胞の溶解、TREM2+骨髄細胞の貪食、およびTREM2+骨髄細胞における増殖抑止の少なくとも1つが誘導される、請求項10~14のいずれか一項に記載の医薬組成物。
- 前記癌が、固形癌または液性癌である、請求項10~15のいずれか一項に記載の医薬組成物。
- 前記癌が、メラノーマ、腎臓癌、肝胆道癌、頭頸部扁平上皮癌、膵臓癌、結腸癌、膀胱癌、神経膠芽腫、前立腺癌、肺癌、および乳癌からなる群より選択される、請求項16に記載の医薬組成物。
- 前記医薬組成物が免疫療法と併用され、かつ
該免疫療法が、前記抗体または抗原結合断片と同時にまたは前記抗体または抗原結合断片の後に前記対象に投与される、
請求項10~17のいずれか一項に記載の医薬組成物。 - 前記免疫療法が、チェックポイント阻害物質を抑制する免疫療法、T細胞のチェックポイント阻害物質を抑制する免疫療法、抗PD1、抗PDL1、抗CTLA4、養子T細胞療法、CAR-T細胞療法、樹状細胞ワクチン、単球ワクチン、T細胞及び抗原提示細胞の両方に結合する抗原結合タンパク質、BiTE抗原結合タンパク質、トール様受容体リガンド、またはサイトカインの少なくとも1つである、請求項18に記載の医薬組成物。
- 前記医薬組成物が免疫療法と併用され、かつ
該免疫療法が、前記抗体または抗原結合断片と同時に前記対象に投与され、
該免疫療法が、ペンブロリズマブ、ニボルマブ、またはイピリムマブの少なくとも1つを含む、
請求項10~19のいずれか一項に記載の医薬組成物。 - 前記医薬組成物が免疫療法と併用され、かつ
該免疫療法が、前記抗体または抗原結合断片の後に前記対象に投与され、
