Description

すべての有核細胞は、クラスI主要組織適合性複合体(MHC-I)経路介して表面に抗原ペプチド(AP)を提示する。APは8~10個のアミノ酸であり、固有の細胞活性を反映している(Caron et al.)。その提示は、免疫細胞(主に細胞傷害性CD8+ T細胞(CTL)とCD4+ Tヘルパー細胞)による潜在的に危険な要素の監視を誘導しているため、APは現在開発されている治療用抗癌ワクチンの標的である。有望であるにもかかわらず、腫瘍関連抗原(TAA)を標的とする治療用ワクチンによる臨床試験の結果は期待を満たしていない。主な失敗は、免疫抑制機構と最適ではない抗原の選択に関連している(Mellman et al.; Burg et al.)。腫瘍の免疫選択を促進し、予後不良と相関する重要な事象の1つは、腫瘍細胞によるMHCクラスI抗原提示の喪失またはダウンレギュレーションである(Watson et al.; Liu et al.)。これらは、MHCクラスI経路のコンポーネントに欠陥があるために、CTLおよびナチュラルキラー細胞の認識を逃れる可能性ががある(Leone et al.)。MHCクラスI抗原提示の全体的な減少とともに、MHCIクラスI免疫ペプチド(MIP)と呼ばれる細胞表面に提示される抗原の性質は、免疫認識にとって非常に重要である。乳癌のHer/neuや結腸癌のCEAなど、特定のTAAが特定され免疫療法で標的化されている癌では、腫瘍細胞表面でのこのTAA発現の喪失が免疫回避につながる(Lee et al.; Kmieciak et al.)。それに対抗するため、現在の戦略は、標的となる癌ペプチドの範囲を拡大し、MHC抗原提示を復活させることを目指している。

n=3~10であるCnH2n+1基は、飽和の直鎖または分岐鎖の脂肪族基である「アルキル」基を指す。例えば、プロピル、イソプロピル、ブチル、イソブチル、tert-ブチル、ペンチル、ヘキシル、ヘプチル、オクチル、ノニルまたはデシル基が挙げられる。好ましくは、nは3~8または3~6である。
発明者らは、ヒトおよびマウスの癌細胞株で発現したPTP由来抗原モデルの抗原提示を調べ、イソギンゲチン、より好ましくはIP2、によるin vitro処理によりこの抗原の提示が増加することを観察した。さらに、発明者は、肉腫を有するマウスのDMSOに溶解したイソギンゲチンによるin vivo治療が、免疫依存的に腫瘍成長を遅らせることを示した。その効果を改善するため、水に溶解するイソギンゲチン誘導体IP2を試験したところ、驚くべきことに、それがイソギンゲチン自体よりもはるかに強力な癌増殖阻害剤であることを観察した。免疫不全のNu/Nuマウスでは、天然物や誘導体は腫瘍成長に影響を及ぼさないため、それらの効果は免疫応答に依存していると結論付けた。これらの結果は、PTP由来抗原の提示を調節できることを示しており、発明者は、イソギンゲチンの他の誘導体に反して、抗癌反応を促進し、癌を治療するために使用することができる、IP2スプライシング阻害剤または式(I)で示される化合物という、市場開発に新しい有望な分子を提供する。
哺乳動物の例としては、ヒトおよび非ヒトの動物、たとえば家畜(ウシ、ヒツジ、ネコ、イヌ、およびウマなど)、非ヒト霊長類(サルなど)、ウサギ、およびげっ歯類(たとえば、マウスやラット)が挙げられる。治療は、好ましくは、年齢や性別を問わず、それを必要とするヒトを患者として意図する。
対象は、例えば、癌を患っており、癌治療、典型的には化学療法に耐性のあるヒトである。
したがって、本明細書はまた、典型的には、好ましくは前記癌の治療において、少なくとも1つの別の抗癌剤と同時、個別または連続的に使用するための組み合わせ製剤として、式(I)で示される化合物、好ましくは「IP2」化合物(IP2-6NaまたはIP2-4Na)と、薬学的に許容される担体を含む組成物を記載する。
別の特定の態様において、前記方法はさらに、癌または癌治療の副作用を予防または治療するための、有効量の別の治療活性化合物を投与することを含む。
細胞培養
MCA205マウス肉腫細胞株は、標準条件下で、1%グルタミン、1%ピルビン酸ナトリウム、1%非必須アミノ酸、1%ペニシリン/ストレプトマイシンおよび10%FBS(Life Technologies)の存在下、RPMI 1640培地(Life Technologies)中、5%CO2下、37℃で培養する。B16F10マウス黒色腫細胞株、MRC5ヒト線維芽細胞株およびA375ヒト黒色腫細胞株を、標準条件下で、1%グルタミン、1%ペニシリン/ストレプトマイシンおよび10%FCSを含むDMEM培地(Life Technologies)中、5%CO2下、37℃で培養する。A549ヒト肺癌細胞株は、標準条件下で、1%Hepes、1%ピルビン酸ナトリウムおよび10%FBSの存在下、DMEM/F12 + Glutamax I中、5%CO2下、37℃で培養する。安定なMCA205-グロビン-SL8-イントロン細胞株は、選択のため、2mg/ml G418(Life Technologiesより入手したジェネティシン)を添加して、MCA205細胞株と同じ条件下で培養する。安定なB16F10-グロビン-SL8-イントロン細胞株は、選択のため、2mg/ml G418(Life Technologiesより入手したジェネティシン)を添加して、B16F10細胞株と同じ条件下で培養する。SL8/Kb特異的(B3Z)T細胞レポーターハイブリドーマを、標準条件下で、1%グルタミン、0.1%β-ガラクトシダーゼ、1%ペニシリン/ストレプトマイシンおよび10%FCSの存在下、RPMI 1640培地(Life Technologies)中、5%CO2下、37℃で培養する。
MCA205およびB16F10マウス細胞株は、各製造元のプロトコルに従って、トランスフェクション試薬jetPRIME(Ozyme)またはGeneJuice(Millipore)を用い、プラスミドYFP-グロビン-SL8-イントロンまたはPCDNA3 emptyプラスミド(ネガティブコントロール)でトランスフェクトする。A375、A549およびMRC5ヒト細胞株は、製造元のプロトコルに従って、トランスフェクション試薬jetPRIME(Ozyme)を用い、マウスH2-Kb分子をコードするプラスミドを12時間トランスフェクトした後、プラスミドYFP-グロビン-SL8-イントロンでトランスフェクションする。トランスフェクションの24時間後、細胞を異なる用量のイソギンゲチン(Merk Millipore)、IP2またはIM2P2(本明細書では「M2P2」とも呼ばれる)で一晩処理する。次に、細胞をPBS 1Xで3回洗浄し、5×104細胞を1×105 B3Z細胞と共培養する。陽性対照ウェルには、4μg/mlの合成ペプチドSL8を添加する。次に、細胞を5%CO2で37℃にて一晩インキュベートする。細胞を1200rpmで5分間遠心分離し、PBS 1Xで2回洗浄し、次の溶解バッファーで振とうしながら4℃で5分間溶解する:0.2%TritonX-100、0.2%DTT、0.5M K2HPO4、0.5M K2HPO4。溶解物を3000rpmで10分間遠心分離し、上清を96ウェルオプティプレート(Packard Bioscience、Randburg、SA)に移す。33mMのメチルウンベリフェリβ-D-ガラクトピラノシド(MUG)を含む暴露緩衝液を添加し、プレートを室温で3時間インキュベートする。最後に、FLUOstar OPTIMA(BMG LABTECH Gmbh、Offenburg、Germany)を使用して、β-ガラクトシダーゼ活性を測定する。結果は平均±SEMとして表す。* P <0.05、** P <0.01、*** P <0.001(対応のないスチューデントt検定)。
C57BL/6J雌マウスはHarlanから入手する。NU/NUヌードマウスは、Charles Riverから入手する。7週齢で、マウスの右脇腹に5×104 MCA205-グロビン-SL8-イントロン細胞または4×104 B16F10-グロビン-SL8-イントロン細胞を、マトリゲル(VWR)とともに皮下注射する。投与の5日後に、マウスを、PBS、イソギンゲチン(Merk Milllipore)、IP2またはIM2P2で腹腔内治療する。投与の15日後、マウスを再び同じ薬物で腹腔内治療する。腫瘍の面積を、27日目まで3~4日ごとに記録する。動物実験はすべて、フランスおよびヨーロッパの法律および規制に従って実施した。結果は平均±SEMとして表す。* p <0.05、** P <0.01、*** P <0.001(すべてのグループを比較するTukeyの多重比較検定によるANOVA)。
イソギンゲチン処置は、癌細胞におけるイントロン由来抗原の抗原提示を増加させる。
最近の研究において、発明者は、パイオニア翻訳産物(「PTP」)が、in vitroでの内因性MHCクラスI経路のペプチドの主要な供給源であることを示した。癌細胞表面での提示PTP由来抗原を調節するために、発明者は、黒色腫A375、肺癌A549、および正常線維芽細胞肺MRC5細胞株に対するイソギンゲチン処置の影響を試験した。その目的のために、すべての細胞は、MHCクラスI Kb分子およびβ-グロビン遺伝子構築物内のイントロンからのSL8エピトープをそれぞれ一時的に発現させる。図1A、1Bおよび1Cに示すように、天然のイソギンゲチン化合物は、試験した癌細胞株でイントロンPTP依存の抗原提示の増加を引き起こし、用量依存的な効果を示す。同様に、同じ実験を、マウスの腫瘍細胞株である、1つの黒色腫(B16F10)および1つの肉腫(MCA205)細胞株で行った。いずれのマウス細胞株も、β-グロビン遺伝子構築物内のイントロンに由来するPTP-SL8エピトープを一時的に発現させた。ヒト細胞株での以前の結果と一致して、イソギンゲチンは、マウス細胞株において、用量依存的効果を示し、PTP依存的抗原提示の増加を誘発する。これらの結果は、PTP抗原またはPTP由来抗原の産生と提示を、イソギンゲチン処理によって癌細胞株において積極的に調節できることを示している。発明者は、この分子をPTPの産生と提示に依存する特定の抗腫瘍免疫応答を強化するための能動的な免疫調節剤として使用できるという仮説を支持する。
イソギンゲチンがイントロンコード領域の細胞表面でのSL8エピトープの提示を増加させるという事実は、pre-mRNAがスプライシングされた機構が対になっていないときの抗原提示の源であるという考えを支持する。そして、発明者は、イソギンゲチンが、エクソンコード領域からの抗原性エピトープの増加を誘発するが、スプライシングする必要のないcDNA構築物からは誘発しないと推測する。その目的のため、上記の両方のマウス細胞株は、β-グロビン遺伝子構築物内のエクソンからPTPs-SL8エピトープを一時的に発現するか、SL8エピトープがその右側にあるOva cDNAを一時的に発現させた。予想どおり、天然のイソギンゲチン化合物は、試験した癌細胞株においてエクソンおよびイントロン-PTP依存的な抗原提示の増加を引き起こし、用量依存的な効果を示し(図2A、2B、2D、2E)、一方、Ova cDNA構築物によってコードされたSL8エピトープの産生には効果を示さなかった(図2Cおよび2F)。これらの結果は、イソギンゲチンが癌細胞におけるPTP抗原またはPTP由来抗原の提示のブースターとして作用するためには、スプライシングが必要であることを示している。
上記の結果は、エクソンまたはイントロン配列によってコードされるPTP依存性抗原の供給源とは無関係に、イソギンゲチンの天然物が、処置された腫瘍細胞株の細胞表面でのin vitroでの産生および提示を増加できることを示している。次の明白な疑問は、DMSOに溶解しなければならないイソギンゲチンが、in vivoで腫瘍の成長とCD8+ T細胞の増殖に同じ効果をもたらすかどうかを確認することであった。その目的のため、β-グロビン遺伝子のイントロンからのSIINFEKL(SL8)エピトープ(グロビン-イントロン-SL8)を安定して発現するMCA205肉腫細胞をマウスに皮下接種した。この接種の5日後、マウスに規定用量のイソギンゲチンを腹腔内にワクチン接種した。次いで、10日後に同じ用量を再度注射した。その間、腫瘍の成長を2~3日ごとにモニターした(図3A)。発明者は、6および18mg/kgのイソギンゲチンで処置したマウスで、接種後27日目に腫瘍成長の有意な50%の減少を観察した(図3B)。この効果に対する免疫応答の要件を評価するために、発明者は、以前に記載したのと同じ設定で免疫不全nu/nuマウスで18mg/kgのイソギンゲチン処置の影響を試験し、腫瘍成長に効果がないことを観察した(図4D)。これらの結果は、イソギンゲチン処置後の腫瘍サイズの縮小に、生体内での能動的な免疫応答の存在が必要であることを示している。発明者らは次に、抗腫瘍応答を増加させるために、イソギンゲチンの誘導体を生成することにした。事実、イソギンゲチンは水に不溶であり、DMSO溶媒にしか溶けないため、腹腔内の薬物動態の効率は低い。
毒性のある担体または共溶媒(DMSO)を使用せずに、イソギンクゲチンをより広範なin vivoでの検証に利用できるようにするには、その溶解度を高める戦略を見つける必要があると考えられた。本発明の化合物は、イチョウ(Ginko biloba L.)の葉から抽出される、より一般的にはビフラボノイドと呼ばれる、小さなポリフェノール分子である市販のイソギンゲチンから調製した。DMSOに溶解した天然物がマウスに十分に吸収されない可能性があることを考慮して、発明者は、天然化合物を共溶媒に溶解させるよりも優れた薬物動態を有する、スプライソソームの阻害剤および癌細胞株に対する能動的な免疫調節剤を意味する、その機能を保持する誘導体化合物を生成することにした。
スキーム2(図6参照)
腫瘍細胞株に対する能動的な免疫調節剤としての新しい化合物を試験するために、発明者は、最初にそれらをインビトロアッセイで試験することを決めた。本明細書でともに「IP2」とも呼ばれる誘導体2および/または2’、および「IM2P2」と呼ばれる誘導体4、はいずれも水に溶解することができた。β-グロビン遺伝子のイントロンに由来するPTP-SL8エピトープを一時的に発現するMCA205細胞(グロビン-イントロン-SL8)を、15μM、25μMまたは35μMのIP2で処理した後、発明者は、PTP依存の抗原提示の増加を観察した(図4A)。対照的に、15μM、25μM、35μMのIM2P2(図4B)または化合物10の製品(図4C)で発明者のPTPエンコード構築物を発現するMCA205細胞を処理しても、発明者のPTP由来抗原の提示は増加しない。次に、発明者は、抗腫瘍成長および特定の抗腫瘍免疫応答の誘導因子に関して、これらの誘導体の効果をin vivoで調べることにした。その目的のために、β-グロビン遺伝子のイントロン設定からSIINFEKL(SL8)エピトープを安定的に発現するMCA205肉腫細胞(グロビン-イントロン-SL8)をマウスに皮下接種した。次に、この接種の5日後に、マウスの各グループにそれぞれ18mg/kgのイソギンゲチン、IP2またはIM2P2を腹腔内にワクチン接種した。10日後、同じ用量の各化合物を再び注射した。その間、腫瘍の成長を2~3日ごとにモニターした(図3A)。18mg/kgのIP2で処理したマウスのグループでは、18mg/kgのイソギンゲチンで処理したマウスのグループと比較して、腫瘍成長の劇的な減少が観察された(図4D)。並行して、反対に、18mg/kgのIM2P2で処置したマウスは、腫瘍成長の減少を示さなかった(図4D)。
本発明は、IP2として本明細書で特定された生成物が抗腫瘍免疫応答、したがって腫瘍増殖に対して非常に好ましい効果を有することを初めて実証する。発明者らは実際に、天然物イソギンゲチンの特定の誘導体が、in vitroおよびin vivoで抗腫瘍免疫応答の強力な刺激物質であることを実証した。この種の作用機序を示すこのような小分子は前例のないものである。発明者のデータは、標的分子療法の枠組み内で新しい抗癌治療の適用に道を開くものである。
スプライシング阻害は、癌細胞のイントロンとエクソンに由来する抗原の提示を増加させる
癌細胞は、細胞表面でMHCクラスI(MHC-I)分子に提示される、抗原性の低下とT細胞認識の回避につながるペプチドのプールと量を形成するさまざまな細胞内メカニズムを示す。発明者は、PTPがin vitroの内因性MHC-I経路のペプチドの主要な供給源であることを示した。さらに、彼らは、スプライシング阻害剤イソギンゲチンでHEK細胞を処理することにより、PTP依存抗原提示に対するスプライシング阻害の好ましい影響の最初の証拠を提供した。後者は、そのアセンブリの初期段階でスプライソソームを阻害することが報告されている。癌細胞株の抗原性と免疫認識を改善するために、発明者は、イソギンゲチンが癌細胞表面での腫瘍関連PTP由来抗原(TA-PTP)の発現と提示を積極的に調節できるかどうかを調べた。その目的のために、ヒト黒色腫細胞株A375、ヒト肺癌細胞株A549および正常ヒト線維芽細胞肺細胞株MRC5を、β-グロビン遺伝子構築物内のイントロンからMHC-I H2-Kb分子およびPTP-SL8エピトープを一時的に発現させ(グロビン-SL8-イントロン)、異なる濃度のイソギンゲチンで18時間処理した。すべての結果は、処置で細胞表面でのH2-Kb分子の全体的な発現の変調によって偏らないよう、SL8ペプチドの細胞外添加有り無しでのB3Z活性化の比率に基づいて表した。3つの細胞型で、イソギンゲチンでの処置は用量依存的にイントロン由来SL8抗原提示を増加させる(図7A)。使用したイソギンゲチンの濃度は、処置で生存率が80%を超えることが示されているため、ヒト細胞に対して毒性はなかった(table 1)。
細胞表面での抗原の豊富さは、CD8+ T細胞応答の規模の大きさを決定し、免疫優性を定義する際の重要なパラメーターであることが実証されている(Doherty et al.、2006)。SL8ペプチドは、in vivoで免疫原性が高いことが広く示されている。インビトロでのSL8特異的T細胞活性化に注目して、発明者は、スプライシング阻害後の癌細胞表面でのSL8発現の豊富さの変化を観察した。この仮説をin vivoで試験するために、発明者は最初に、イントロン由来SL8ペプチドを発現する腫瘍の成長に対するイソギンゲチン処置の影響に注目した。その目的のために、グロビン-イントロン-SL8構築物を安定して発現するMCA205肉腫細胞およびB16F10黒色腫細胞の両方をマウスに皮下接種した。腫瘍接種後5、10、および15日目に、マウスに規定用量のイソギンゲチンを腹腔内注射し、腫瘍成長をモニターした(図9A)。MCA205グロビン-SL8-イントロン(MCA205 GI)腫瘍を有するマウスにおいて、発明者は、12および18mg/kgのイソギンゲチンで処理した場合、接種後27日目に腫瘍サイズの50%以上の有意な減少を観察した(図9B)。B16F10グロビン-SL8-イントロン(B16F10 GI)腫瘍の成長に対するイソギンゲチン処置の影響は、MCA205 GIよりも低いた、この薬物は依然として腫瘍の成長を大幅に遅らせる(図9C)。イソギンゲチン処置後のin vivoでのSL8の過剰発現と腫瘍成長の減少との関連を評価するために、発明者はMCA205またはB16F10野生型(WT)細胞を接種したマウスで同じ実験を行った。12および18 mg/kgのイソギンゲチンでの処置後も、MCA205 WT(図9D)またはB16F10 WT(図9E)腫瘍成長の有意な減少は観察されなかった。これらの結果は、免疫優性エピトープ(本明細書におけるSL8ペプチド)の発現は、イソギンゲチンがin vivoで腫瘍成長に影響を与えるために必要であることを示唆している。
天然のイソギンゲチンの誘導体を合成し、スプライシングを阻害する能力、in vitroでPTP由来抗原を増加させる能力、およびin vivoで腫瘍成長を減少させる能力について試験した。誘導体IP2(図10BのIP2-6NaおよびIP2-4Naを参照)およびM2P2(図10C)は、イチョウ(Ginko biloba L.)の葉から抽出される、一般的にビフラボノイドと呼ばれる市販のイソギンゲゲチン(図10A)から合成した。図5に合成スキームを示す。簡単に言うと、IP2、即ち化合物2および2’(スキーム中の名称)の合成は、ジエチルクロロホスファイトのin situ形成を用いたリン酸化により、1を得た。さらに、ヨードトリメチルシランによるエチルエステル保護基の切断によりリン酸中間体を得、これをすぐに水酸化ナトリウムで処理し、リン酸二ナトリウムプロドラッグ2および2’への実用的な経路を完結させた。M2P2分子の合成では、化合物1の残りの2つのフェノール基をヨウ化メチルを用いてアルキル化し、化合物3を得た。化合物1から化合物2および2’を調製する同様の条件下で、化合物3を処理してリン酸二ナトリウムプロドラッグ4を得る一方、塩基性条件下でも反応させて化合物5を得た。
イソギンゲチンと比較してIP2およびM2P2化合物の潜在的な免疫調節効果を試験するために、2つの分子をまずin vitroでPTP由来抗原のMHC-I提示を増加させる能力について試験した。その目的のために、MCA205およびB16F10細胞を、グロビン-イントロン-SL8構築物を一時的に発現させ、15μMまたは35μMのIP2またはM2P2で処理した。IP2で処置した場合、発明者がイソギンゲチン処理後に観察したのと同様、MCA205およびB16F10細胞でのイントロンSL8由来抗原提示が増加する(図11AおよびB、左パネル)一方、M2P2は。MCA205での提示が減少し、B16F10細胞では影響は見られない(図11AおよびBの右パネル)。これらの結果は、グロビン-SL8-イントロン遺伝子スプライシングを阻害するIP2およびM2P2のそれぞれの能力と興味深い相関関係がある。実際、M2P2は、B16F10において、スプライシングにもSL8抗原提示にも影響を与えない。逆に、M2P2はMCA205のスプライシングを強力に阻害し、SL8の提示に悪影響を及ぼす。これらの結果とともに、発明者らは、細胞表面でのH2-Kb分子の発現がIP2およびM2P2で処理された細胞株では影響を受けないことを示した(図12Aおよび12B)。したがって、イントロン由来のエピトープの提示にプラスの影響を与える処置には、スプライシングの厳密な調節が必要と思われる。これらの結果は、イソギンゲチン誘導体IP2が、天然物と同様にin vitroでPTP由来抗原の提示のブースターとして作用することを示している。
全体として、これらの結果は、in vitroで観察されたPTP由来抗原の提示の増加と処置後のin vivoでの腫瘍成長の減少との相関関係を示している。興味深いことに、イソギンゲチンとは対照的に、IP2処置は、PTPに由来する高度に免疫優性のSL8エピトープを有しない腫瘍の成長を遅らる。発明者は、スプライシング阻害剤IP2が、抗腫瘍応答を促進する細胞表面での免疫優性エピトープの出現を増大すると考える。
免疫系の要件、特に腫瘍に対するIP2の有効性に対するT細胞応答の要件を調べるために、発明者は、T細胞を欠損し、B細胞とNK細胞を欠損していない、Nu/Nu無胸腺ヌードマウスにおける効果に注目した。前にイソギンゲチンで試験したように、グロビン-イントロン-SL8構築物またはWTを安定して発現するMCA205またはB16F10細胞をマウスに皮下接種した。腫瘍接種後5、10、15日目に、マウスの各グループを、免疫適格マウスで観察された腫瘍増殖に対して最も効率的なIP2の用量で腹腔内処置した。したがって、MCA205GI、MCA205 WTおよびB16F10 GIまたはWTを有するマウスを、それぞれ18mg/kg、24mg/kgおよび36mg/kgで処置した。各条件で、腫瘍成長に対するIP2処置の影響は認められなかった(図13AおよびB)。
さらに、IP2の有効性に対するCD8+ T細胞の特定の要件を評価するために、発明者は、マウスにおけるin vivo CD8+ T細胞枯渇の影響を試験した。マウスにMCA205 GI細胞を皮下接種した後、抗CD8+ T細胞抗体またはアイソタイプ2A3による計画的な処置を行った。5日目、10日目および15日目にIP2処置を前と同様に投与した。興味深いことに、抗CD8+ T細胞抗体処置は、IP2処置の抗腫瘍効果を完全に無効にした(図13C)。したがって、この結果は、腫瘍成長に対するIP2処置の効果がCD8+ T細胞応答に依存していることを裏付けており、CD8+ T細胞応答は腫瘍細胞に対する抗原駆動型細胞傷害活性を裏付けている。
最後に、MCA205 GIを接種し、続いて上記のようにIP2で処置したマウスの約50%は、処置後に腫瘍がなくなった。最初の腫瘍接種の100日後、これらのマウスの右脇腹にMCA205 GI腫瘍細胞を、左脇腹にB16F10細胞を再接種した。B16F10腫瘍は時間とともに成長したが、MCA205 GIはマウスでは成長しなかった(図13D)。これらの結果は、IP2処置後、マウスがMCA205 GI腫瘍に特異的な長期抗腫瘍反応を発現したことを示している。
特定の抗腫瘍CD8+ T細胞の増殖を促進することにより腫瘍成長の調節に対する特定のIP2の有効性を評価するために、発明者は、in vivoでの確立された腫瘍に対するIP2の影響を試験した。グロビン-イントロン-SL8構築物を安定して発現するMCA205肉腫腫瘍細胞(15.105)を皮下注射し、10日間進行させた後、IP2処置の開始前にランク付けして同等の腫瘍量のグループに割り当て、腫瘍サイズ40 mm2、50 mm2、100 mm2の3つのグループにした。その後、3~4日ごとに、各グループのマウスをそれぞれ24mg/kgのIP2で腹腔内処置した。この用量では、試験したグループに関係なく、非処置のマウスと比較して、IP2処置後に、MCA205 GI腫瘍成長の顕著な減少が観察された(図14)。腫瘍サイズが100mm2(丸印)に達した場合でも、24 mg/kgのIP2を5回投与すると、未処置のマウス(黒丸印)と比較して、生存期間が約30日間長くなり有意な腫瘍退縮を誘発することができた(グレーの丸印)。
結論として、免疫調節剤としてIP2を使用するこれらすべての実験は、この分子が長期抗腫瘍応答を誘導することにより、マウスが非常に早期に処置される場合(腫瘍が触知できる場合)に完全な腫瘍拒絶を誘導するのに効率的であることを示し、また、IP2処置が確立された腫瘍の腫瘍成長を減少させることができることを示している。

- 対象における、癌の治療における使用、癌の転移予防における使用、および/または癌の再発予防における使用のための請求項1に記載の化合物。
- 少なくとも1つの別の抗癌剤および/または放射線療法と組み合わせた、請求項2に記載の使用のための化合物。
- 癌が癌腫、肉腫、リンパ腫、胚細胞腫瘍、芽細胞腫、白血病および多発性骨髄腫から選択される、請求項2または3に記載の使用のための化合物。
- 癌腫が黒色腫、肺癌または乳癌である、請求項4に記載の使用のための化合物。
- 少なくとも1つの別の抗癌剤が、化学療法剤、免疫チェックポイントブロッカーおよび抗癌ワクチンから選択される、請求項3~5のいずれかに記載の使用のための化合物。
- それを必要とする対象における抗癌免疫応答を刺激するための使用のための、請求項1に記載の化合物。
- 対象における、癌の治療、癌の転移予防、および/または癌の再発予防における使用のための、請求項8に記載の組成物。
