Description

本発明は、単離された抗PD-L1抗体、ならびにそれらの調製および使用に関する。
プログラム細胞死タンパク質1(PD-1)は、細胞の認識、結合、および接着プロセスに関与している細胞表面タンパク質および可溶性タンパク質を含む免疫グロブリンスーパーファミリーに属する細胞表面受容体である。このファミリーの初期のメンバーは、モノクローナル抗体の添加後のT細胞増殖の増大に対する機能的作用が原因で発見された(Hutloff et al. ある種の状況では、全長抗体ではなく抗体断片を使用することが有利である。断片のサイズが小さいほど、迅速なクリアランスが可能になり、より上手く固形腫瘍に接近するようになり得る。

本発明の局面には、限定されるわけではないが様々ながんおよび免疫疾患の治療を含む、PD-L1によって(例えばPD-1とPD-L1の相互作用によって)少なくとも部分的に媒介される疾患または病態の治療、予防、および/または診断において、本明細書において説明されるように、1種類または複数種類の抗PD-L1組成物(例えば、抗PD-L1抗体、抗PD-L1抗体の抗原結合断片、またはADC)を使用する方法が含まれる。非限定的な使用例を、下記にさらに詳細に説明する。
本発明の抗PD-L1組成物は、例えば、インビトロ、エクスビボ、およびインビボの治療方法で使用され得る。1つの局面において、本発明は、PD-L1タンパク質と1つまたは複数の受容体またはリガンド(例えばPD-1タンパク質)との相互作用を阻害するための方法を提供する。1つの局面において、本発明は、組成物中の抗PD-L1抗体またはその抗原結合断片がPD-L1に結合するのを許容する条件下で細胞を抗PD-L1組成物と接触させる工程を含む、インビボまたはインビトロのいずれかで細胞の成長または増殖を抑制するための方法を提供する。「細胞の成長または増殖を抑制すること」とは、細胞の成長または増殖を少なくとも約10%、20%、30%、40%、50%、60%、70%、80%、90%、95%、または最大で100%、減少させることを意味し、細胞死を誘導することを含む。特定の態様において、細胞は腫瘍細胞である。本発明の態様に従う抗PD-L1組成物は、(i)PD-L1タンパク質とPD-L1タンパク質が結合できる受容体もしくはリガンド(例えばPD-1タンパク質)との相互作用を阻害するか;(ii)インビボで腫瘍転移を抑制するか;(iii)インビボで腫瘍増殖を抑制するか;(iv)インビボで腫瘍サイズを小さくするか;(v)インビボでPD-L1発現腫瘍細胞に対して細胞障害活性を示すか;または(vi)インビボでPD-L1発現腫瘍細胞に対して細胞増殖抑制活性を示すことができる。いくつかの局面において、抗PD-L1組成物は、PD-L1タンパク質に結合し、それによってPD-1タンパク質の1つまたは複数の機能を阻害する(例えば、PD-1タンパク質の1つまたは複数の免疫抑制機能を阻害する)ことができる。したがって、いくつかの局面において、有用な対象治療方法は、PD-L1タンパク質を本明細書において説明される抗PD-L1抗体または抗原結合断片と接触させる工程であって、それによって、PD-1タンパク質の1つまたは複数の機能(例えば、1つまたは複数の免疫抑制機能)を阻害する、工程を含む。
本発明の局面には、試料をPD-L1ポリペプチドに結合する抗体と接触させる工程、および試料においてPD-L1ポリペプチドへのその抗体の結合を判定する工程であって、そのような結合の存在によって試料においてPD-L1ポリペプチドの存在が示される、工程を含む、PD-L1ポリペプチドを含むと推測される試料においてPD-L1ポリペプチドの存在を判定する方法が含まれる。任意で、試料は、PD-L1ポリペプチドを発現すると推測される細胞(例えばがん細胞)も含んでよい。この方法で使用される抗体は、任意で、検出可能に標識されるか、または固体支持体に結合されるなどしてよい。
本発明の態様に従う抗PD-L1組成物は、当技術分野において公知の様々なアッセイ法で利用することができる。1つの局面において、抗PD-L1組成物は、生物活性アッセイ法で使用することができる。生物活性アッセイ法は、例えば、抗PD-L1組成物が細胞の成長もしくは増殖を抑制する能力(例えば「細胞死滅」活性)またはプログラム細胞死(アポトーシス)を含む細胞死を誘導する能力を測定することができる。
本発明の局面には、PD-L1タンパク質を本明細書において説明される抗PD-L1抗体と接触させることによってPD-1タンパク質とPD-L1タンパク質の相互作用を阻害する方法が含まれる。いくつかの態様において、この方法は、インビトロで実施される。
1.1 免疫化のプロトコールおよびスケジュール
(i)HISタグ付きのヒトPD-L1(Sino Bio、カタログ番号10084-H084)および(ii)HISタグ付きのカニクイザルPD-L1(Sino Bio、カタログ番号90251-C08H)の細胞外部分を含む組換え融合タンパクを、免疫化プロトコールにおいて抗原として利用した。PD-L1に対する完全なヒトモノクローナル抗体を作製するために、3つの異なる系統のマウスを免疫化しスクリーニングした。Balb/C(3)、C57/Black6(3)、およびスイスウェブスター(3)を、タンパク質を交互に用いて(ヒトPD-L1およびカニクイザルPD-L1)、0日目、7日目、14日目、21日目、28日目、および35日目に6回免疫化した。1回目の免疫化は、50ugのタンパク質を投与し、その後の免疫化は25ugを投与した。抗原の皮下投与は、MagicMouseアジュバントを用いて、左および右の鼠径リンパ節、左および右の上腕リンパ節、ならびに左および右の軸リンパ節の近くに50uL/部位の量で行った。9匹のマウスすべてから、1回目の免疫化の前の0日目(事前採血)および42日目にに血液を採取して、抗体応答に関する血清力価を評価した。血清を(後述の)ELISAによってスクリーニングし、十分な力価の抗ヒト-PD-L1免疫グロブリンを有しているマウスを融合用に使用した。屠殺および膵臓切除の3日前に、静脈内経由でマウスに組換えヒトPD-L1を追加免疫した。
スイスウェブスターマウスから単離したマウス脾細胞を、標準的なプロトコールに従って、マウス骨髄腫細胞株と融合させた。次いで、結果として生じるハイブリドーマを、抗原特異的抗体の産生についてスクリーニングした。
間接的な捕捉ELISA:
最初のスクリーニングにおいて、抗体を産生するマウス血清またはハイブリドーマの検出を、間接的ELISAプロトコールおよび血清またはハイブリドーマ上清の一点希釈物を用いて実施した。その後のスクリーニングは、マウス血清またはイブリドーマ上清の希釈物を用いて実施した。ELISAの形式は、模式的な図1に示したとおりである。
フローサイトメトリーに基づく結合反応性:
細胞表面でヒトPD-L1を発現するGranta細胞(DSMZ ACC番号342)またはPromega 187103細胞(Promega、カタログ番号187106)などの細胞株を用いて、図2に示すようにフローサイトメトリーによって、ハイブリドーマ上清中の抗ヒトPD-L1モノクローナル抗体の反応性および特異性を明らかにした。
FACS 2%FBS緩衝液(BD Pharmingen、カタログ番号554656)60uLに溶かした1.5×104個/ウェルの細胞を「v」底96ウェルプレートに添加した。12ウェルマルチチャンネルピペットを用いて、ハイブリドーマプレートの各ウェルから得られる上清10uLを、それらのウェルに対応する細胞に添加した。3つのハイブリドーマ上清プレートを用いてこれを繰り返して、3つのハイブリドーマウェルから得られる上清を、細胞を含む各ウェル中にまとめ合わせた。陽性対照29E.2A3(BioLegend、カタログ番号329702)およびIgG2b kアイソタイプ対照(BioLegend、カタログ番号400302)をこれらの細胞に1ug/mLの濃度で添加し、対照ウェルと呼んだ。これらのプレートを4℃で30分間インキュベートした。細胞をFACS 2%FBS緩衝液150μLで洗浄した後、プレートを1200rpmで5分間、遠心分離し(Sorvall Legend XIR遠心分離機)、細胞ペレットを乱さずに上清を穏やかに吸引した。これらの細胞をAF647ヤギ抗マウスIgG(Jackson ImmunoResearch、カタログ番号115-605-164)と共に4℃で30分間インキュベートすることによって、抗体結合を検出した。上記のように洗浄工程を繰り返し、1:100の7_AAD(BD Pharm、カタログ番号68981E)を含む60μLのFACS 2%FBS中に細胞を再懸濁した。これらの試料の1000個の事象を、FACSCalibur(Becton Dickinson)を用いて各試料について獲得し、FLOJO(登録商標)フローサイトメトリープラットフォームで解析を行った。上清において検出された結合を、陽性対照およびアイソタイプ対照と比較し、陽性ウェルに印を付けた。
圧縮プレート/ウェルから得られた陽性ウェルのそれぞれを個々のプレート/ウェルに復元し、3つの圧縮プレートのそれぞれの個々のウェルから得られるハイブリドーマ上清を、上記のプロトコールに従ってPromega 187103細胞に対する陽性染色について個別に試験した。結合をアイソタイプ対照と比較して、抗PD-L1抗体を産生するハイブリドーマを同定し、PD-L1発現細胞に対して高レベルの結合を示しているELISA陽性ウェルを次のスクリーニングに進めた。ハイブリドーマ上清のFACSスクリーニングについてのデータ例を図3のパネルBに示している。
ハイブリドーマの1回目のスクリーニングから得られる陽性上清の滴定を、前述の間接的ELISAプロトコールを用いて実施し、その際、上清はウェルに添加する前に1%ミルク中で段階的に3倍希釈した。選択されたハイブリドーマ上清中のマウス抗体の濃度を、濃度が公知である市販の29E.2A3抗体を用いる間接的ELISAによって推定した。次いで、PD1/PD-L1遮断アッセイ法において陽性上清の機能性について試験した。
前述のスクリーニングアッセイ法は、プレート上にコーティングされた抗原または細胞表面の抗原としてのPD-L1に結合できるクローンを同定することができるが、本発明者らが開発したい抗体は、PD-1とPD-L1の相互作用を効果的に阻止することが要求される。上記で同定されたFACSおよびELISA陽性抗体がこの相互作用を阻止する能力を試験するために、本発明者らは、発光に基づく読取りを用いる市販のPD-1/PD-L1遮断アッセイ法(Promega)を使用した(図4)。
典型的なハイブリドーマでは、複数の重鎖および軽鎖が発現され、これらのうちの1つまたは複数は、機能アッセイ法で認められる活性に関与している配列であり得る。次世代シーケンシング(NGS)法を適用して、ELISA、FACS、および遮断アッセイ法で陽性であると確認されたハイブリドーマに由来する配列を同定した。簡単に説明すると、RNAをハイブリドーマ(1×106個の細胞)から抽出し、RACE-PCRプロトコールを用いて一本鎖DNAを生じさせた。重鎖および軽鎖の両方を増幅するために、マウスIgG領域およびマウスIgK領域に対する縮重プライマーを使用した。これらのプライマーは、FR1からFR4までの可変領域を包含するように設計した。試料にバーコードを付け、MiSeq NGSシークエンサー(Illumina)を用いてペアエンド法で配列決定した。得られたFastqファイルは、Panoply Bio(Carlsbad、CA)で特別仕様のソフトウェアを用いて解析された。これらの配列を、共通の部分があるペアエンドについて位置合わせし、マウスフレームワーク領域を用いてCDRを同定した。各配列の頻度に基づいてデータを集めて整理した。いくつかの切断が、この配列アセンブリ過程で発生する場合があり、したがって、クローニングおよびさらなる試験のために選択される配列は、以下の基準に基づいた。
(1)3つのCDRすべてが存在すること。
(2)配列のファミリーに複数回出現すること。
(3)CDRの頻度が最も高いこと。
(4)N末端に明らかな切断がないこと。
1 設計された変異を有しているDNA構築物の作製
DNA構築物の合成。各々の発現ベクターにサブクローニングするために適合する制限部位を両端に有している、設計された変異を有しているDNA構築物はすべて、商業的販売業者(例えばGenescript、Lake Pharma)によって合成された。
a トランスフェクション
重鎖、軽鎖、およびJ鎖のDNAをCHO細胞またはHEK293細胞にトランスフェクトした。典型的には、発現ベクター用のDNAを1:1:1の比でPEIと混合し、次いでCHO-S細胞に添加した。確立されている技術に従って、CHO-S細胞を用いるPEIトランスフェクションを実施した(Biotechnology and Bioengineering, Vol 87, 553-545を参照されたい)。
i Capture Select IgM(BAC, Thermo Fisher): トランスフェクトされたCHO細胞上清から得られたIgMタンパク質を、製造業者のプロトコールに従って(GE Life Sciences)、Capture Select IgMアフィニティーマトリックスを用いる免疫沈降によって、ある程度精製した。室温で2時間インキュベーションした後、遠心分離によってアフィニティーマトリックスを上清から分離させた。PBSで3回、マトリックスをさらに洗浄した後、PBSを注意深く除去した。NuPage LDSタンパク質緩衝液(Life Technology)と共に5分間インキュベートすることによって、捕捉されたタンパク質をマトリックスから溶出させた。
i 非還元SDS PAGE: 非還元SDS PAGEは、大きさに基づいてネイティブIgM およびその変異型を分離する。ホモ二量体の重鎖および軽鎖から構成される五量体IgMは、分子量が約1,000,000のタンパク質バンドを生じる。ゲルに載せる前に、25℃で30分間、NuPage LDS試料緩衝液(Life Technologies)をIgMタンパク質試料に添加した。NativePage Novex 3〜12%ビス-トリスゲル(Life Technologies)をNovexトリス-酢酸SDS泳動用緩衝液(Life Technologies)と共に使用した。色素の前部がゲルの底面に到達するまで、ゲルを泳動させた。
前と同じように、PD-L1を表面で発現する凍結したCHO細胞(Promega CS187103)を37℃で解凍し、10%ウシ胎児血清(Invitrogen)を添加したDMEM中に再懸濁した。細胞7,500個を含む培地を37℃および5%CO2で16時間、384ウェルプレートのウェル中に添加した。培地を、3C5-1G12抗体または対照抗体を含む10uLのRPMI+2%FBSに交換した。操作されたジャーカット細胞(Promega CS187105)10,000個を含むさらに10uLの培地をウェルに添加し、37℃および5%CO2で5時間インキュベートした。細胞を、ルシフェリンを含む20μLの溶解緩衝液(Promega、Cell Titer Glo)と混合して、ルシフェラーゼレポーター活性を測定した。EnVisionプレートリーダーによって光出力を測定した。Prismソフトウェアを用いて4パラメーター曲線当てはめによって、EC50を明らかにした。
組換えヒトPD-L1 Fcキメラ(Sino Biological、カタログ番号10084-H02H)、組換えカニクイザルPD-L1 Fcキメラ(Sino Biological、カタログ番号90251-C02H)、組換えマウスPD-L1 Fcキメラ(Sino Biologicals、カタログ番号50010-M02H)、および組換えヒトPD-L2 Fcキメラタンパク質(Sino Biological、カタログ番号10292-H02H)を、4℃で一晩インキュベーションすることによって、1ug/mLの濃度でELISAプレートに直接コーティングした。これらのプレートをPBS 0.05%Tweenで3回洗浄し、ペーパータオルに数回軽くたたきつけて乾かした。室温で1時間、200uLの1%ミルクで、すべてのプレートをブロッキングした。上記のようにプレートを洗浄し乾かした後に、3C5.2G12 IgG抗体、3C5.2G12 IgM抗体、および3C5.2G12 IgM+wtJ抗体をそれぞれ、3ug/mLから始めて0.1ng/mLまで、1%ミルク中で1:3希釈した。希釈した抗体50μLを全プレートの各ウェルに添加した。陽性対照抗体(マウス抗ヒトPD-L1、29E.2A3(BioLegend、カタログ番号329702)およびそのアイソタイプ対照マウスIgG2b k(BioLegend、カタログ番号400302)を3ug/mLから始めて0.1ng/mLまで、1%ミルク中で段階的に3倍希釈し、用量設定されたこれらの抗体50uLを各プレート/ウェルに添加した。同様に、ラット抗マウスPD-L1(BioLegend、カタログ番号124302)およびラットIgG2bアイソタイプ(BioLegend、カタログ番号400622)、抗ヒトPD-L2(BioLegend、カタログ番号345502)およびマウスIgG1アイソタイプ(BioLegend、カタログ番号4011402)を前述したように用量設定し、各プレート/ウェルに添加した。これらのプレートを室温で1時間インキュベートした。これらを上記のように洗浄し乾かし、次いで、すべての3C5.2G12を、西洋ワサビペルオキシダーゼ(HRP)とコンジュゲートされた1:5000ヤギ抗ヒトκ軽鎖抗体(Jackson Immuno Research、カタログ番号115-035-071)50uLと共に室温で1時間インキュベートした。対照ウェルのために、適切な二次抗体、すなわちヤギ抗マウスHRP(Southern Biotech、カタログ番号1033)およびマウス抗ラットHRP(Southern Biotech、カタログ番号3070-05)を希釈し、上記のようにインキュベートした。洗浄および乾燥段階を繰り返した後、TMB基質(BD OptiIEIA、カタログ番号555214)50uLを用いて20〜30分間、プレートを発色させた。50uLの2N H2SO4を用いて反応を停止し、Spectramax Gemini分光光度計を用いて450nmでの吸光度を読み取った。段階希釈した3C5.2G12についてのデータ例を図9のパネルA、パネルB、およびパネルCに示している。
3C5-2G12抗体のヒト化を両方の型(IgGおよびIgM)で実施した。試験のために作製した配列および組合せを下記に示す。

PEPSCAN(商標)システムを用いる固体支持体上への直接的合成によってPD-L1(Q9NZQ7、SEQ ID NO: 48)の細胞外ドメインのアミノ酸19〜132をカバーする重複15アミノ酸直線状ペプチドすべてのライブラリーを作製することにより、ペプチドエピトープマッピングを実施した。
以下の方法によって、抗体の交差阻止解析を行った。ヒトPD-L1を発現するCHO細胞(反応1回当たり20,000〜30,000個)を、3C5(IgG)抗体、S70抗体、または無関係なアイソタイプ対照IgG抗体のいずれかの用量設定された非標識希釈物と共に4℃で30分間インキュベートした。細胞を洗浄し、次いで、Alexa Fluor(登録商標)647で標識した一定濃度の3C5またはS70(1μg/mL)と共に4℃で30分間インキュベートした。FACSCalibur(商標)システムを用いて蛍光染色強度を測定した。同一の抗体による競合は陽性対照であり、無関係な抗体による競合の欠如は陰性対照であった。交差阻止の結果を図11のパネルAおよびパネルBに示す。これらの結果から、3C5およびS70によるヒトPD-L1への結合の相互の交差阻止が実証される。前述の本発明は、理解を明確にするために、例示および例としていくらか詳しく説明してきたが、添付の特許請求の範囲の精神および範囲から逸脱することなく、いくつかの変更および修正をそれに加え得ることが、本発明の教示に照らして、当業者には容易に明らかになる。
Claims (17)
- 重鎖可変領域および軽鎖可変領域を含む、単離された抗PD-L1抗体またはその抗原結合断片であって、重鎖可変領域が、
(i)配列GFSLTSYDIS(SEQ ID NO: 4)を含むHVR-H1;
(ii)配列VIWTGVGTN(SEQ ID NO: 5)を含むHVR-H2;および
(iii)配列DPYYYGMDY(SEQ ID NO: 6)を含むHVR-H3
を含み、
かつ軽鎖可変領域が、
(iv)配列RASQDISIWLS(SEQ ID NO: 1)を含むHVR-L1;
(v)配列KASNLHT(SEQ ID NO: 2)を含むHVR-L2;および
(vi)配列LQSQSFPRT(SEQ ID NO: 3)を含むHVR-L3
を含む、
