Description

コンピュータ可読形式の配列表は、本願と共に電子出願され、その全体が参照により本願に援用される。配列表は、MONS389WO_ST25.txtと名付けられたファイルの中に入っており、それは、(オペレーティングシステムMS Windowsの測定で)大きさが122キロバイトであり、2016年12月19日に作成されたものである。
新規遺伝子導入植物の生産は、有益な形質、例えば強化された雑草抑制方策を可能にするための向上した除草剤耐性を呈する、大幅に改善された作物植物の潜在性を提案する。しかしながら、作物において有益な形質を生じさせるのに有用なタンパク質が知られているものの、遺伝子導入植物細胞におけるこれらの組換えタンパク質の効果的な細胞内局在化(標的指向化として知られる)及びプロセシングは大きな障壁であり続けている。それゆえ、組換えタンパク質を効果的に植物細胞内で局在させることができる新規な輸送ペプチドが必要とされている。
配列番号1は、Arabidopsis thalianaアルビノ薄緑(APG6)CTPのアミノ酸配列である。
除草剤耐性タンパク質を細胞内で局在させるための葉緑体輸送ペプチド(CTP)は当該技術分野において既知であるが、CTPと除草剤耐性タンパク質とのいかなる組み合わせについても、細胞内局在化及びプロセシングの効果的な度合いを予測することは困難である。局在化及びプロセシングは、除草剤耐性タンパク質の発現レベル及び機能を決定付け、したがって当該タンパク質を含む遺伝子導入細胞、植物または種子の除草剤耐性表現型に影響を与える。遺伝子導入植物において、ジカンバモノオキシゲナーゼ(DMO)及びプロトポルフィリノーゲンオキシダーゼ(PPO)を含めた有用な除草剤耐性タンパク質を使用して様々なCTPが試験されてきた。しかしながら、タンパク質の不十分または不完全なプロセシング及び局在化がみられることが多い。
本明細書中で使用する場合、「組換え」という用語は、遺伝子操作の成果である、したがって自然界では通常見つからないものである、人間の介入によって作り出された非天然のDNA、ポリペプチド、タンパク質、細胞、種子または植物を指す。「組換えDNA分子」は、天然に生じない、人間の介入の結果であるDNA配列を含むDNA分子、例えば、互いに異種である少なくとも2つのDNA分子の組み合わせを含むDNA分子である。組換えDNA分子の一例は、配列番号18〜27からなる群から選択される配列を含むDMOタンパク質をコードするDNA配列に機能可能に繋げられた、配列番号1〜3からなる群から選択される配列を含むCTPをコードするDNA分子である。DMOタンパク質の例を以下の表1に示す。

本発明の一態様は、本発明によって提供される組換えDNA分子を含んでいる遺伝子導入植物細胞、遺伝子導入植物組織、遺伝子導入植物及び遺伝子導入種子を含む。組換えDNA分子を含むこれらの細胞、組織、植物及び種子は、除草剤に対する耐性を呈する。
ダイズプロトプラストアッセイを用いて、DMO配列(配列番号27)に機能可能に繋げられた5つのうち1つのCTPを含んでいる組換えタンパク質の相対的な葉緑体指向効率を評価した。組換えタンパク質のサイトゾル及び葉緑体での分布を監視するために、組換えのCTPとDMOとの組み合わせ(本明細書においてCTP−DMOと呼ぶ)をコードするカセットに緑色蛍光タンパク質コード配列を、緑色蛍光タンパク質がDMOのカルボキシ末端と融合するように追加した。

DMOに機能可能に繋げられた別個の2つのCTPのうちの1つをコードする組換えDNA分子を含んでいるDNA構築物の発現を、遺伝子導入トウモロコシ細胞及び植物において分析した。

表9.F1雑種のジカンバ損傷及び収穫高についての圃場試験
タンパク質翻訳効率(タンパク質合成)を増強しかつタンパク質蓄積を増大させるべくAPG6 CTPを最適化した。最適化されたAPG6 CTP(配列番号2)は、APG6 CTP(配列番号1)の位置3及び4においてスレオニン(T)からセリン(S)へのアミノ酸変更を有する。ダイズにおいて各々DMOに機能可能に繋げられた2つのCTPを比較するために、DNA構築物を作った。

除草剤細菌スクリーニング系を使用して、PPO除草剤に対して耐性である新規PPOを同定した。このスクリーニング系は、PPO除草剤に対して感受性でないPPOを同定するために、PPO除草剤を含むLB液体培地でのノックアウトE.coli株の増殖アッセイを用いるものであった。

種々のCTPに機能可能に繋げられたPPO酵素を遺伝子導入ダイズ植物において発現させ、遺伝子導入植物をPPO除草剤耐性について分析した。

PPO H_N10(配列番号43)に機能可能に繋げられたAPG6(配列番号1)を遺伝子導入ワタ植物において発現させるために、植物形質転換用ベクターを作出し、遺伝子導入植物をPPO除草剤耐性について分析した。植物形質転換用ベクター及びAgrobacterium tumefaciensを当該技術分野において知られている標準的方法と共に用いて、ワタDP393を形質転換した。再生されたR0遺伝子導入小植物体を温室内で栽培し、結果として生じるR0植物から採取した葉の試料をPCRで分析して、事象の単一のコピーを含有する植物を同定した。温室内で、各々が固有の事象に相当している遺伝子導入単一コピーR0植物に対して、V2段階で施す20g活性成分/ヘクタールのS−3100の除草剤処理によって噴霧を行った。さらに、温室内で遺伝子導入多コピー(2コピー/植物以上)に対して、V2段階で施す20g活性成分/ヘクタールのS−3100の除草剤処理によって噴霧を行った。処理から3日後に損傷評価を行った。
Claims (27)
- ジカンバモノオキシゲナーゼ(DMO)またはプロトポルフィリノーゲンオキシダーゼ(PPO)をコードするDNA配列に機能可能に繋げられた葉緑体輸送ペプチド(CTP)をコードするDNA配列を含む、組換えDNA分子であって、前記CTPが、配列番号1および2からなる群から選択される配列を含む、前記組換えDNA分子。
- 前記CTPをコードする前記DNA配列が、配列番号7〜12からなる群から選択される配列を含む、請求項1に記載の組換えDNA分子。
- 前記DMOまたはPPOが、配列番号18〜27及び40〜59からなる群から選択されるポリペプチド配列を含む、請求項1に記載の組換えDNA分子。
- DMOまたはPPOをコードする前記DNA配列が、配列番号28〜37及び61〜102からなる群から選択される配列を含む、請求項3に記載の組換えDNA分子。
- 前記CTPが、DMOタンパク質に機能可能に繋げられており、前記CTPが、配列番号1および2からなる群から選択される配列を含む、請求項1に記載の組換えDNA分子。
- 前記CTPが、PPOタンパク質に機能可能に繋げられており、前記CTPが、配列番号1及び2からなる群から選択される配列を含む、請求項1に記載の組換えDNA分子。
- 植物細胞内で機能する異種プロモーターに機能可能に繋げられた請求項1に記載のDNA分子を含むDNA構築物。
- 請求項1に記載のDNA分子を含んでいる、遺伝子導入植物、植物細胞、植物部分または種子。
- 前記植物が単子葉植物である、請求項8に記載の遺伝子導入植物、植物細胞、植物部分または種子。
- 前記植物がトウモロコシ植物またはコムギ植物である、請求項9に記載の遺伝子導入植物、植物細胞、植物部分または種子。
- 前記植物が双子葉植物である、請求項8に記載の遺伝子導入植物、植物細胞、植物部分または種子。
- 前記植物がダイズ植物、ワタ植物またはBrassica植物である、請求項11に記載の遺伝子導入植物、植物細胞、植物部分または種子。
- 除草剤耐性植物を生産する方法であって、
a)植物細胞を請求項7に記載のDNA構築物で形質転換するステップと、
b)前記DNA構築物を含んでいる形質転換植物細胞から植物を再生するステップと
を含む、前記方法。 - 再生された植物が、ジカンバ及びPPO阻害剤からなる群から選択される除草剤に対して耐性である、請求項13に記載の方法。
- 除草剤耐性植物を生産する方法であって、
a)請求項1に記載のDNA分子を含んでいる親植物をそれ自体または第2植物と交配して1つ以上の子孫植物を生産するステップと、
b)前記DNA分子を含んでいる子孫植物を選抜するステップと
を含む、前記方法。 - 前記子孫植物が、ジカンバ及びPPO阻害剤からなる群から選択される除草剤に対して耐性である、請求項15に記載の方法。
- 前記子孫植物が、S−3100、ホメサフェン、アシフルオルフェン、ラクトフェン、フルミオキサジン、スルフェントラゾン及びサフルフェナシルからなる群から選択されるPPO阻害剤除草剤に対して耐性である、請求項16に記載の方法。
- 請求項1に記載のDNA分子を植物細胞内に導入することを含む、ジカンバモノオキシゲナーゼ(DMO)またはプロトポルフィリノーゲンオキシダーゼ(PPO)を発現させる方法。
- 導入することが、前記植物細胞を形質転換することを含む、請求項18に記載の方法。
- 作物栽培環境における雑草生長を抑制する方法であって、
a)請求項8に記載の植物または種子を作物栽培環境に植付けるステップと、
b)雑草生育を抑制するのに有効な量のジカンバまたはPPO阻害剤除草剤を前記作物栽培環境に散布するステップと
を含む、前記方法。 - 前記除草剤が前記植物または種子を害さない、請求項20に記載の方法。
- 前記植物または種子が単子葉の植物または種子である、請求項20に記載の方法。
- 前記植物がトウモロコシ植物またはコムギ植物である、請求項22に記載の方法。
- 前記植物または種子が双子葉の植物または種子である、請求項20に記載の方法。
- 前記植物がダイズ植物、ワタ植物またはBrassica植物である、請求項24に記載の方法。
- 前記除草剤がジカンバである、請求項20に記載の方法。
