Description
アロニア抽出物(ABF)[(株)jbklab]20mgをリン酸緩衝液(Phosphate buffer)pH3(PB3)水溶液20mlに沈殿物が見えないように溶解させ、フコイダン[(株)haewon biotech]200mgを蒸留水20mlに沈殿物が見えないように溶解させた後、フコイダン溶液にアロニア抽出物溶液を添加し、室温で72時間撹拌させて、アロニア抽出物−フコイダン複合体(Cyaplex−F8)を製造した。
1.アロニア抽出物−フコイダン複合体の形成確認
初日から6日目までアロニア抽出物(ABF)溶液とアロニア抽出物−フコイダン複合体サンプルとを得て、前記サンプルを1/10に希釈した後、分光光度計(UV−1601、Shimadzu,JAPAN)で光学密度を測定して結果を得た。また、アロニア抽出物(ABF)溶液とアロニア抽出物−フコイダン複合体原液とで光学写真を得た。
完成された複合体2mlをポリスチレンキュベット(DTS0012)に入れた後、ゼータ電位&ナノ粒度分析器(Zetasizer nano ZS,Malvern Instruments Ltd.,England)を用いて複合体のサイズを確認した。また、完成された複合体10μlをカバーガラスに載せて60℃オーブンで一晩中乾燥させ、走査電子顕微鏡(S−4800、HITACHI,Ltd.,U.S.A)でイメージを得た。
前記アロニア抽出物−フコイダン複合体の製造方法で作られた複合体40mlを13000rpm、30分間遠心分離した後、上澄み液は除去し、残っている沈殿物は、PB3(pH3)とphosphate buffer saline(pH7)とに再分散させた後、経時的に分光光度計でアントシアニン溶液と共に測定して、構造的安定性の増進効果を確認した。
1.アロニア抽出物−フコイダンナノ粒子の試験管内細胞毒性の確認
アロニア抽出物とフコイダン複合体の癌細胞に対する抗ガン効果と正常組織細胞に対する毒性とを確認した。
アロニア抽出物(ABF)50μg/mlが含まれたアロニア抽出物(ABF)溶液またはフコイダン50μg/mlの濃度が含まれたフコイダン溶液と50μg/mlの濃度のアロニア抽出物(ABF)及び500μg/mlの濃度のフコイダン濃度が含まれたアロニア抽出物−フコイダン複合体試料を準備した。
発癌物質と腫瘍促進剤とが処理された実験動物でアロニア抽出物−フコイダン複合体の発癌抑制効果を確認した。
アロニア抽出物−フコイダン複合体と抗ガン剤との併用投与による抗ガン効果を確認した。
以前実験で確認されたように、アロニア抽出物−フコイダン複合体が白血球数値を増加させると確認されることによって、アロニア抽出物−フコイダン複合体が免疫細胞に及ぼす影響を確認した。
ドキソルビシン処理前、あらゆる実験群で血液200μlを採取し、RBC lysis buffer 500μLを処理した後、4℃で5分間反応させ、DPBSを用いて10倍希釈して、1,500rpm、3分間遠心分離した後、抗CD3及び抗CD8を用いて血中細胞毒性T細胞のレベルを確認し、抗CD49b(DX5)を用いて血中NK細胞の流細胞分析(Flow cytometry)を行った。
Claims (6)
- アロニア抽出物及びフコイダンからなり、前記アロニア抽出物のうち、シアニジンの陽イオンと前記フコイダンの陰イオンとのイオン結合が形成され、シアニジン分子間のπ−π結合が形成されたことを特徴とするアロニア抽出物−フコイダン複合体を有効成分として含有する免疫増強剤。
- 前記アロニア抽出物−フコイダン複合体は、免疫因子の発現を誘導して免疫細胞の活性を増加させることを特徴とする請求項1に記載の免疫増強剤。
- アロニア抽出物及びフコイダンからなり、前記アロニア抽出物のうち、シアニジンの陽イオンと前記フコイダンの陰イオンとのイオン結合が形成され、シアニジン分子間のπ−π結合が形成されたことを特徴とするアロニア抽出物−フコイダン複合体を有効成分として含有する免疫抗ガン剤。
- 前記アロニア抽出物−フコイダン複合体は、免疫因子の発現を誘導して免疫細胞の活性を増加させて、癌細胞を死滅させることを特徴とする請求項3に記載の免疫抗ガン剤。
- 前記癌細胞は、固形癌の癌細胞であることを特徴とする請求項4に記載の免疫抗ガン剤。
- アロニア抽出物及びフコイダンからなり、前記アロニア抽出物のうち、シアニジンの陽イオンと前記フコイダンの陰イオンとのイオン結合が形成され、シアニジン分子間のπ−π結合が形成され、抗ガン剤投与による体重または免疫因子の減少及び悪液質からなる群から選択される副作用を緩和させることを特徴とするアロニア抽出物−フコイダン複合体を有効成分として含有する抗ガン補助剤。
