一実施形態では、BYは導入遺伝子カセットを含み、前記カセットは、B7タンパク質またはそのフラグメントをコードする導入遺伝子と、調節エレメントの組合せなどの調節エレメントとを含む。
一実施形態では、本開示のウイルスは、感染癌細胞の表面に発現するタンパク質を複数コードし、ここでは、少なくとも1つがB7タンパク質またはその活性フラグメントであり、例えば、癌細胞表面に発現するか、細胞外空間内に分泌される2つ、3つ、4つまたはそれ以上の異なるタンパク質、特に2つまたは3つのタンパク質がウイルスによってコードされている。この場合のタンパク質には融合タンパク質が含まれる。一実施形態では、本開示のウイルスは、例えばともに癌細胞表面に発現する2つの異なるB7タンパク質、その活性フラグメントまたはこれらの組合せをコードする。
本明細書で使用され癌細胞に対する選択性を有する腫瘍溶解性ウイルスは、例えば、癌細胞に優先的に感染することおよび/またはウイルス生活環が調節不全を起こしたp53、例えば癌細胞に過剰発現するp53などの遺伝子に依存することを理由に、癌細胞を優先的に死滅させるウイルスを指す。一実施形態では、腫瘍溶解性ウイルスは、例えばEnAdのように、癌細胞に優先的に感染し、次いでそのゲノムを複製し、キャプシドタンパク質を産生して新たなウイルス粒子を生じる。
本明細書の文脈における複製可能なウイルスは、in vitroおよびin vivoの細胞内で、すなわちパッケージング細胞系による補助がなくても複製するのに必要な機構をすべて備えているウイルスを指す。ウイルスベクター、例えば少なくともE1A領域が欠損し、補完的なパッケージング細胞系で複製可能なウイルスベクターは、本明細書の文脈では複製可能なウイルスではない。
結合は、あるDNA配列と別のDNA配列とを連結している共有結合、例えばウイルスゲノムのある区画と別の区画とを連結している共有結合を指す。したがって、本明細書の式(I)および(Ia)の可変物が結合を表す場合、その結合によって表される特徴または要素は存在しない、すなわち欠失している。
本明細書で使用される「遺伝子発現調節因子」(または調節因子/調節エレメント)は、通常は転写または翻訳を開始または増大させることによって、遺伝子発現に何らかの役割を果たす、プロモーター配列、エンハンサー配列またはスプライスアクセプター配列などの遺伝子要素を指す。
本開示はこのほか、本明細書に記載されるウイルスの医薬製剤に及ぶ。
さらなる態様では、本開示は治療、特に癌治療に使用するための本明細書に記載されるウイルスまたはその製剤に及ぶ。
実施例1:T細胞活性化抗原CD80を発現するEnAdウイルスの作製
プラスミドpEnAd2.4を用いて、ヒトT細胞活性化抗原CD80(配列番号11)をコードするカセットを直接挿入することによりプラスミドpNG−330を作製した。pNG−330カセットは、5’の短いスプライスアクセプター配列CAGG、ヒトCD80のcDNA配列および3’ポリアデニル化配列(配列番号5)を含むものである。挿入した導入遺伝子カセットの模式図を図3Aに示す。DNAシーケンシングによりプラスミドの構築を確認した。
本明細書でNG−330−00などのウイルスに言及する場合、それは単に特定の1回分にまとめた「00」のウイルスNG−330に言及したことになる。ほかのウイルスにも同様の命名法が使用され得る。プラスミドpNG−330を酵素AscIで制限消化することによって直線化し、ウイルスゲノムNG−330(配列番号16)を作製した。消化したDNAをフェノール/クロロホルム抽出によって精製し、分子生物学グレードの95%超エタノール300μlおよび3M酢酸ナトリウム10μl中、−20℃で16時間沈殿させた。沈殿したDNAを14000rpmで5分間遠心分離することによってペレット化し、70%エタノール500μlで洗浄した後、再び14000rpmで5分間遠心分離した。清浄なDNAペレットを風乾させ、lipofectamineトランスフェクション試薬15μlを含有するOptiMEM 500μlに再懸濁させ、室温で30分間インキュベートした。次いで、集密度70%まで増殖させた293細胞の入ったT−25フラスコにトランスフェクション混合物を滴加した。細胞とトランスフェクション混合物を5%CO2、37℃で2時間インキュベートした後、細胞に細胞培地(グルタミンを含み2%FBSを添加した高グルコースDMEM)4mlを加え、フラスコを5%CO2、37℃でインキュベートした。
結腸癌細胞系HT−29および肺癌細胞系A549を用いてNG−330ウイルスまたはEnAdウイルスの複製(qPCRにより評価した)およびCD80の膜発現(フローサイトメトリーにより評価した)(図4および5)を比較した。NG−330は、EnAdに由来し、EnAd後期遺伝子L5(ファイバー)の後にヒトT細胞活性化抗原CD80をコードする導入遺伝子カセットを含むウイルスである。挿入されているカセットの模式図を図3Aに示す。NG−330ウイルスの作製については実施例1に詳述されている。A549癌細胞系またはHT−29癌細胞系を6ウェルプレートに7.5e5細胞/ウェル(A549細胞)または2.e6細胞/ウェル(HT−29細胞)の細胞密度で播いた。プレートを37℃、5%CO2で18時間インキュベートした後、細胞にEnAdウイルス粒子またはNG−330ウイルス粒子を1細胞当たり100個(ppc)感染させるか、感染させずにおいた。感染から24時間後、48時間後または72時間後にアッセイを実施した。
100ppcのEnAdまたはNG−330を24時間、48時間または72時間、感染させるか感染させずにおいたHT−29細胞系およびA549細胞系をqPCRによるウイルスDNAの定量化に用いた。細胞上清を収集し、1200rpmで5分間遠心分離することによって清澄化した。Sigma Genelute DNA extraction Kitを製造業者のプロトコル通りに用いて上清10μlからDNAを抽出した。EnAdウイルス粒子(2.5e10〜2.5e5vp)を用いた標準曲線も作成し、Sigma Genelute Kitを用いて抽出した。EnAd E3遺伝子特異的プライマー−プローブセットを用いたqPCRにより、各抽出試料または標準物質を分析した。
100ppcのEnAdまたはNG−330を24時間、48時間または72時間、感染させるか感染させずにおいたHT−29細胞系およびA549細胞系を細胞表面でのCD80導入遺伝子発現の解析に用いた。トリプシン処理によって腫瘍細胞をプレート表面から剥がし、遠心分離した後、1%BSA/PBSに再懸濁させた。次いで、試料を緩衝液、Cy5とコンジュゲートしたマウスアイソタイプ対照抗体またはCy5とコンジュゲートした抗ヒトCD80抗体(クローン2D10)と5℃で1時間インキュベートした。生存細胞を識別するため、全試料にZombie Aqua live/deadによる共染色も実施した。試料を1%BSA/PBSで3回洗浄した後、フローサイトメトリー(FACS、Attune)による細胞生存率および細胞表面のCD80タンパク質発現の解析に供した。解析では、A549細胞(図5A)、HT−29(図5B)細胞ともに、NG−330で処理した細胞の表面にはCD80を検出することができるが、EnAdで処理した細胞、未処理の細胞のいずれもCD80を検出することができないことがわかった。
HT−29結腸癌細胞を2.5e4細胞/ウェルの細胞密度で96ウェルプレートに播いた。プレートを37℃、5%CO2で4時間インキュベートした後、感染密度が1細胞当たり100〜0.39粒子(ppc)の範囲のEnAdウイルス粒子またはNG−330ウイルス粒子を細胞に感染させた。感染から72時間後、Cell Titre 96MTS試薬(Promega社:G3581)を用いてHT−29細胞の生存率を評価した。各感染密度での細胞の生存%を定量化することにより、HT29細胞でのNG−330の腫瘍溶解能がEnAdと同程度であることが明らかになった(図6)。

プラスミドpEnAd2.4を用いて、可溶性変異型のFMS様チロシンキナーゼ−3リガンド(Flt3L、配列番号13)、MIP1α(アイソフォームLD78β、配列番号14)およびIFNα(配列番号12)をコードするカセットを直接挿入することによってプラスミドpNG−345を作製する。pNG−345カセットには、5’の短いスプライスアクセプター配列CAGG;ヒトFlt3LのcDNA;高効率自己切断型P2Aペプチド配列(配列番号7);ヒトMIP1αのcDNA;高効率自己切断型T2Aペプチド配列(配列番号10);ヒトIFNαのcDNAおよび3’ポリアデニル化配列(配列番号5)が含まれている。挿入した導入遺伝子カセットの模式図を図3Dに示す。CDNAシーケンシングによりプラスミドの構築を確認する。

プラスミドpNG−348を酵素AscIで制限消化することによって直線化し、ウイルスゲノムNG−348(配列番号96)を作製する。実施例1で詳述した方法に従ってウイルスNG−348を増幅し精製する。
癌細胞系を用いてEnAdと比較したNG−343ウイルス活性の特徴付け
結腸癌細胞系HT−29または肺癌細胞系A549を用いてNG−343ウイルスまたはEnAdウイルスの複製(qPCRにより評価した)、CD80導入遺伝子発現(フローサイトメトリーにより評価した)またはIFNα導入遺伝子発現(ELISAにより評価した)を比較した。NG−343は、EnAdに由来し、EnAd後期遺伝子L5(ファイバー)の後に位置するヒトT細胞活性化抗原CD80およびヒトサイトカインのインターフェロンα2bをコードする導入遺伝子カセットを含むウイルスである。挿入したカセットの模式図を図3Bに示す。NG−343ウイルスの作製については実施例3に詳述されている。A549癌細胞系またはHT−29癌細胞系を12ウェルプレートに7.5×105細胞/ウェル(A549細胞)または1.4×106細胞/ウェル(HT−29細胞)の細胞密度で播いた。プレートを37℃、5%CO2で18時間インキュベートした後、細胞にEnAdまたはNG−343を1細胞当たり100ウイルス粒子(ppc)で感染させるか、感染させずにおいた。感染から24時間後、48時間後または72時間後にアッセイを実施した。

10ppcのEnAdまたはNG−343を24時間、48時間または72時間、感染させるか感染させずにおいたHT−29細胞系またはA549細胞系の上清について、分泌IFNαの発現をELISAにより分析した。各ウェルから培養上清を取り出し、1200rpmで5分間遠心して細胞残屑を除去した。上清を5%BSA/PBSアッセイ緩衝液で希釈(1:2、1:50または1:100)し、Verikine Human IFN alpha Kit(Pbl assay science社)を製造業者のプロトコル通りに用いてELISAを実施した。
10ppcのEnAdまたはNG−343を48時間または72時間、感染させるか感染させずにおいたA549細胞系を細胞表面でのCD80導入遺伝子発現の解析に用いた。感染後のしかるべき時点で、トリプシン処理によってA549細胞をプレート表面から剥がし、遠心分離した後、1%BSA/PBSに再懸濁させた。次いで、試料を緩衝液、Cy5とコンジュゲートしたマウスアイソタイプ対照抗体またはCy5とコンジュゲートした抗ヒトCD80抗体(クローン2D10)と5℃で1時間インキュベートした。生存細胞を識別するため、全試料にZombie Aqua live/deadによる共染色も実施した。試料を1%BSA/PBSで3回洗浄した後、フローサイトメトリー(FACS、Attune)による細胞生存率および細胞表面でのCD80タンパク質発現の解析に供した。CD80発現対Live/dead染色の解析から、感染から48時間、72時間後ともに、NG−343で処理した細胞には細胞表面にCD80を検出することができるが、EnAd未感染対照(UIC)細胞には検出することができないことがわかった(図8)。ウイルス処理から72時間後の細胞生存率は包括的なCD80発現解析を実施するのに十分なものではなかったが、NG−343で処理した細胞では生存細胞、死滅細胞ともに、この時点で高レベルのCD80を検出することができた(図8D、下パネル)。
NG−343ウイルスによってコードされるIFNαおよびCD80の導入遺伝子が、NG−343感染またはEnAd感染を許容する細胞にのみ選択的に発現することを示すため、EnAd感染を許容することが知られている癌細胞(HT−29)、許容性でないことが既に特徴付けられている線維芽細胞系(WI−38およびMRC−5)およびEnAd感染にごくわずかな許容を示す気管支上皮細胞系(BE)でのウイルス複製(qPCRにより評価した)、IFNα発現(ELISAにより評価した)およびCD80発現(フローサイトメトリーにより評価した)を測定した。簡潔に述べれば、12ウェルプレートに細胞を播き、播種から18時間後、WI38細胞、MRC5細胞またはBE細胞には100ppcのNG−343ウイルスまたはEnAdウイルスを感染させ、HT−29細胞には10ppcのNG−343ウイルスまたはEnAdウイルスを感染させた。細胞をウイルス粒子と4時間インキュベートした後、細胞から感染培地を除去し、新鮮な培地と交換した。4時間にわたる感染から1時間後または72時間後、細胞上清を回収してqPCR分析またはELISA分析に供し、細胞をトリプシンで処理してプレートから剥がし、フローサイトメトリーによる解析に供した。

IFNα発現の検出には、細胞上清を収集し、1200rpmで5分間遠心分離することによって清澄化した。上清を5%BSA/PBSアッセイ緩衝液で希釈(1:2、1:50または1:100)し、Verikine Human IFN alpha Kit(Pbl assay science社)を製造業者のプロトコル通りに用いてELISAを実施した。
腫瘍細胞をNG−343ウイルスで処理することによって自然免疫細胞応答が未処理またはEnAd処理と比較して増強されるかどうかを明らかにするため、新たに単離した末梢血単核球(PBMC)と、NG−343もしくはEnAdを感染させた腫瘍細胞または未感染の腫瘍細胞とを共培養した。自然免疫細胞集団のフローサイトメトリー解析または共培養上清のELISA分析により免疫細胞活性化を評価した。簡潔に述べれば、A549肺癌細胞を12ウェルプレートに4×105細胞/ウェルの密度で播いた。20時間後、細胞に10ppcのEnAdウイルスまたはNG−343ウイルスを感染させるか感染させずにおいた後、37℃、5%CO2で24時間インキュベートした。次いで、健常ヒトドナーから密度勾配遠心分離により単離したPBMCを、PBMCとA549細胞の比が5:1になるようA549培養ウェルに加えた。PBMCの添加から48時間後、プレートから共培養上清を回収した。この時点での樹状細胞活性化を分析するため、細胞を緩衝液、Alexa Fluor 488、PE、PerCP/Cy5.5、BV605もしくはBV412とコンジュゲートしたマウスアイソタイプ対照抗体、Alexa Fluor 488とコンジュゲートした抗CD14抗体、PEとコンジュゲートした抗CD80抗体、PerCP.Cy5.5とコンジュゲートした抗HLA−DR、BV605とコンジュゲートした抗CD3またはBV421とコンジュゲートした抗PD−L1抗体と5℃で1時間インキュベートした。生存細胞を識別するため、全試料にZombie Aqua live/deadによる共染色も実施した。試料を1%BSA/PBSで3回洗浄した後、フローサイトメトリー(FACS、Attune)による解析に供した。染色がCD14、CD3ともに陰性であるが、HLA−DRが陽性である生存細胞を樹状細胞集団と定義した。この集団についてDC活性化マーカーのCD80およびPD−L1の発現を比較した(図10)。以上の解析から、NG−343を感染させた腫瘍細胞は、DC表面でのCD80およびPD−L1の両方の表面レベルをEnAd感染腫瘍細胞培養物または未感染腫瘍細胞培養物よりも上昇させ得ることが明らかになった。
癌細胞系を用いてEnAdと比較したNG−347ウイルス活性の特徴付け
CD80導入遺伝子発現(フローサイトメトリーにより評価した)およびIFNαまたはMIP1α(CCL3)の導入遺伝子発現(ELISAにより評価した)を、結腸癌細胞系HT−29または肺癌細胞系A549をNG−347で処理したものとEnAdで処理したものとで比較した。NG−347は、EnAdに由来し、ヒトT細胞活性化抗原CD80、ヒトサイトカインのインターフェロンα2bおよびヒトケモカインMIP1α(LD78βアイソフォーム)をコードする導入遺伝子カセットを含むウイルスである。導入遺伝子の発現はウイルス内在性主要後期プロモーターの制御下にある。挿入したカセットの模式図を図3Fに示す。NG−347ウイルスの作製については実施例6に詳述されている。A549癌細胞系またはHT−29癌細胞系を12ウェルプレートに7.5×105細胞/ウェル(A549細胞)または1.4×106細胞/ウェル(HT−29細胞)の細胞密度で播いた。プレートを37℃、5%CO2で18時間インキュベートした後、細胞に1細胞当たり100個(ppc)のEnAdウイルス粒子またはNG−347ウイルス粒子を感染させるか、感染させずにおいた。感染から24時間後、48時間後または72時間後にアッセイを実施した。
100ppcのEnAdまたはNG−347を24時間、48時間または72時間感染させるか感染させずにおいたHT−29細胞系またはA549細胞系の上清について、分泌IFNαまたは分泌MIP1αの発現をELISAにより分析した。
感染から48時間後のHT−29細胞およびA549細胞の表面でのCD80タンパク質発現を比較した。細胞を回収し、実施例9で詳述した方法に従って染色した。細胞を、細胞生存率および細胞表面でのCD80タンパク質発現についてフローサイトメトリーによって解析した。生存細胞のこの時点におけるCD80発現の解析から、NG−347で処理したHT−29細胞の約96%およびNG−347で処理したA549細胞の約99%の表面にCD80を検出することができるが、EnAd処理対照には染色が検出されないことがわかった(図11C)。
癌細胞系を用いてEnAdと比較したNG−345ウイルス活性の特徴付け
Flt3リガンド導入遺伝子、IFNα導入遺伝子およびMIP1α導入遺伝子の発現(ELISAにより評価した)を、結腸癌細胞系HT−29または肺癌細胞系A549をNG−345で処理したものとEnAdで処理したものとで比較した。NG−345は、EnAdに由来し、可溶性変異型のヒトFlt−3リガンド、ヒトサイトカインのインターフェロンα2bおよびヒトケモカインMIP1α(LD78βアイソフォーム)をコードする導入遺伝子カセットを含むウイルスである。導入遺伝子の発現はウイルス内在性主要後期プロモーターの制御下にある。挿入したカセットの模式図を図3Dに示す。NG−345ウイルスの作製については実施例4で詳述されている。A549癌細胞系またはHT−29癌細胞系を12ウェルプレートに7.5×105細胞/ウェル(A549細胞)または1.4×106細胞/ウェル(HT−29細胞)の細胞密度で播いた。プレートを37℃、5%CO2で18時間インキュベートした後、細胞に1細胞当たり100個(ppc)のEnAdウイルス粒子またはNG−345ウイルス粒子を感染させるか、感染させずにおいた。感染から24時間後、48時間後または72時間後にアッセイを実施した。

HT−29結腸癌細胞を96ウェルプレートに2.5e4細胞/ウェルの細胞密度で播いた。プレートを37℃、5%CO2で4時間インキュベートした後、感染密度が1細胞当たり100〜0.39粒子(ppc)の範囲のEnAdウイルス粒子、NG−347ウイルス粒子またはNG−348ウイルス粒子を細胞に感染させた。感染から72時間後、Cell Titre 96 MTS試薬(Promega社:G3581)を用いてHT−29細胞の生存率を評価した。各感染密度での細胞の生存%を定量化することにより、NG−347およびNG−348の腫瘍溶解能がEnAdと同程度であることが明らかになった(図13Aおよび13B)。
HT−29結腸癌細胞を12ウェルプレートに4e5細胞/ウェルの細胞密度で播いた。プレートを37℃、5%CO2で24時間インキュベートした後、感染密度が1.6e7〜2e6vp/mLの範囲のEnAdウイルス粒子、NG−347ウイルス粒子またはNG−348ウイルス粒子を細胞に感染させた。ウイルスタンパク質であるヘキソンの抗体染色によりHT−29細胞の感染を検出した。各被検希釈物について6つ組のウェルの1ウェル当たり6視野を手作業でカウントすることにより染色細胞を定量化した。ウイルス力価から各ウイルスの粒子/感染力比(P:I)を算出したところ、NG−347およびNG−348の両方は感染力比がEnAd参照対照と同程度であることが明らかになった(図13C)。
CD80導入遺伝子発現(フローサイトメトリーにより評価した)を、結腸癌細胞系DLD−1または肺癌細胞系A549をNG−347で処理したものとNG−348で処理したものとEnAdで処理したもので比較した。A549癌細胞系またはDLD−1癌細胞系を12ウェルプレートに7.5e5細胞/ウェルの細胞密度で播いた。プレートを37℃、5%CO2で18時間インキュベートした後、細胞に1細胞当たり10個(ppc)のEnAdウイルス粒子、NG−348ウイルス粒子またはNG−347ウイルス粒子を感染させるか、感染させずにおいた。感染から24時間後、48時間後、72時間後または96時間後のA549細胞またはDLD−1細胞の表面でのCD80タンパク質発現を比較した。各時点で細胞を回収し、実施例9で詳述した方法に従って染色した。細胞を、細胞生存率および細胞表面でのCD80タンパク質発現についてフローサイトメトリーにより解析した。感染から72時間後のA549細胞におけるCD80発現の解析から、NG−347処理細胞またはNG−348処理細胞の95%超の表面にCD80を検出することができることがわかった(図14Aおよび14B)。感染から96時間後、A549細胞の大部分がウイルス処理によって溶解していたため、FACS解析は実施しなかった。DLD−1細胞については、96時間後までに、NG−348による処理では50%超の細胞に、NG−347処理では70%超の細胞に発現を検出することができた(図15Aおよび15B)。EnAdにも未処理対照にも染色は検出されなかった。
NG−348ウイルス粒子またはEnAdウイルス粒子を感染させるか感染させずにおいたA549肺癌細胞と、ヒトPBMCドナーから単離したT細胞とを共培養した。A549細胞およびT細胞の両方の表面について、NG−348ウイルスがコードするCD80での発現の選択性をフローサイトメトリーにより評価した。細胞表面活性化マーカー(フローサイトメトリーによる)、脱顆粒のマーカーとしてのCD107a細胞表面発現(フローサイトメトリーによる)ならびに刺激性サイトカインであるIL−2およびIFNγの分泌(ELISAによる)を解析することによってT細胞活性化を評価した。
実施例14で示した結果と同じように、NG−348を感染させたEpCam+A549細胞の80%超の表面にCD80発現を検出することができたが、EnAdを感染させた細胞にも未感染対照細胞にもCD80発現を検出することができなかった(図16)。これに対し、CD3+T細胞では細胞表面に検出可能なCD80の発現はみられず、少なくとも上記の実験条件下では、導入遺伝子発現が共培養した腫瘍細胞に選択性を示すことが示された。
T細胞活性化マーカーであるCD25およびHLA−DRの生存CD3+単一細胞での発現によって、T細胞活性化のフローサイトメトリー解析を評価した。これらのデータから、CD25を発現するT細胞の数(図17Aおよび17B)もT細胞表面でのCD25発現レベルの平均値(図17C)も、NG−348を感染させたA549細胞と培養したT細胞の方がEnAdまたは未感染対照よりも有意に高いことがわかった。具体的には、未処理対照とEnAd(CD25発現T細胞はそれぞれ26.9%±3.4%および25.3±3.5%)との比較ではT細胞の活性化状態に差がみられなかったのに対し、NG−348と共培養した細胞の大部分にCD25のアップレギュレーションがみられた(83.2±1.5%)。CD4 T細胞およびCD8 T細胞のサブセットについても、そのCD4の発現に基づきCD3+T細胞にゲートをかけることによりCD25発現を解析した。これらの解析から、NG−348で処理した共培養物中のCD4+T細胞サブセット、CD4_T細胞サブセットともに、EnAdおよび未感染対照と比較してCD25発現が有意にアップレギュレートされることがわかった(図18)。
生存CD3+T細胞表面でのCD107a発現の解析から、A549細胞にNG−348を感染させた場合(細胞の8.3%±1.7%)、脱顆粒したためにCD107aで染色されたT細胞の数がEnAd(細胞の0.6%±0.2%)または未処理対照(細胞の0.1%±0.02%)と比較して有意に増大することがわかった(図20)。CD25のアップレギュレーションと同じく、CD4+T細胞サブセット、CD4_T細胞サブセットともに、EnAdまたはA549対照と比較してCD107a発現の有意な増大がみられた(図21)。
IL−2またはIFNγの発現の検出には、共培養上清を5%BSA/PBSアッセイ緩衝液で希釈し(1:100〜1:1000の範囲)、Human IL−2 Ready Set go Kit(Affymetrix社)またはHuman IFN gamma Ready set go kit(Affymetrix社)を製造業者のプロトコル通りに用いてELISAを実施した。
NG−348ウイルス粒子またはEnAdウイルス粒子を感染させるか感染させずにおいたA549肺癌細胞と、ヒトPBMCドナーから単離したCD4+T細胞またはCD8+T細胞とを共培養した。培養上清中への刺激性サイトカインIFNγの分泌によりT細胞活性化を評価した。
NG−347ウイルス粒子またはEnAdウイルス粒子を感染させるか感染させずにおいたA549肺癌細胞と、ヒトPBMCドナーから単離したT細胞とを共培養した。細胞表面活性化マーカー(フローサイトメトリーによる)および刺激性サイトカインIFNγの分泌(細胞上清のELISA分析による)を分析することによりT細胞活性化を評価した。
T細胞活性化マーカーCD69の生存CD3+単一細胞での発現によって、T細胞活性化のフローサイトメトリー解析を評価した。これらのデータから、CD69を発現するT細胞の数は、NG−347を感染させたA549細胞と培養したT細胞の方がEnAdまたは未感染対照よりも有意に高いことがわかった(図24)。
IL−2またはIFNγの発現の検出には、共培養上清を5%BSA/PBSアッセイ緩衝液で希釈し(1:100〜1:1000の範囲)、Human IFN gamma Ready set go kit(Affymetrix社)を製造業者のプロトコル通りに用いてELISAを実施した。
プラスミドpEnAd2.4を用いて、ヒトT細胞活性化抗原CD80(配列番号11)およびC末端V5タグを有する膜係留キメラ型の一本鎖Fv抗ヒトCD3e(配列番号99)をコードするカセットを直接挿入することによってプラスミドpNG−348Aを作製した。pNG−348カセットには、5’の短いスプライスアクセプター配列(配列番号2);膜係留型抗ヒトCD3e ScFvのcDNA;C末端V5タグ(配列番号100);高効率自己切断型P2Aペプチド配列(配列番号7);ヒトCD80のcDNA配列および3’ポリアデニル化配列(配列番号5)が含まれている。NG−348A導入遺伝子カセットの模式図を図26Aに示す。DNAシーケンシングによりプラスミドの構築を確認する。
プラスミドpNG−348Aを酵素AscIで制限消化することによって直線化し、ウイルスゲノムNG−348A(配列番号101)を作製する。実施例1で詳述した方法に従ってウイルスNG−348Aを増幅し精製する。
プラスミドpEnAd2.4を用いて、C末端V5タグを有する膜係留キメラ型の一本鎖Fv抗ヒトCD3e(配列番号99)またはV5タグを有さない同Fv抗ヒトCD3e(配列番号15)をコードするカセットを直接挿入することによってプラスミドpNG−420およびpNG−420Aを作製した。pNG−420カセットには、5’の短いスプライスアクセプター配列CAGG;膜係留型抗ヒトCD3e scFvのcDNAおよび3’ポリアデニル化配列(配列番号5)が含まれている。pNG−420Aカセットには、5’の短いスプライスアクセプター配列cagg;膜係留型抗ヒトCD3e ScFvのcDNA;C末端V5タグ(配列番号100)および3’ポリアデニル化配列(配列番号5)が含まれている。NG−420遺伝子導入カセットおよびNG−420A導入遺伝子カセットの模式図を図26Bおよび26Cに示す。DNAシーケンシングにより各プラスミドの構築を確認する。
プラスミドpNG−420およびpNG−420Aを酵素AscIで制限消化することによって直線化し、ウイルスゲノムNG−420(配列番号102)およびNG−420A(配列番号103)を作製する。実施例1で詳述した方法に従ってウイルスNG−420およびNG−420Aを増幅し精製する。
A549ヒト肺癌細胞およびMRC5ヒト線維芽細胞をEnAdウイルス、NG−347ウイルスまたはNG−348ウイルス(10ppc)と培養して、これらの細胞型によるウイルスゲノム複製、ウイルスヘキソン発現および導入遺伝子発現を比較した。72時間培養した後、細胞を表面マーカーまたは上清のFACS解析用に染色し、ヘキソン発現または導入遺伝子発現のウイルスゲノム複製(qPCR)解析またはmRNA(RT−qPCR)解析用に細胞溶解物を調製した。
単離CD3+T細胞を500ppcまたは5000ppcのEnAdウイルス、NG−347ウイルスおよびNG−348ウイルスと3日間培養することにより、各ウイルスのヒトT細胞に対する選択性/活性を評価した。選択性/活性は、a)CD69、CD4、CD80、CD25およびCD3を標的とする抗体で染色したT細胞のフローサイトメトリー解析、b)ヒトMIP1αタンパク質、IFNαタンパク質およびIFNγタンパク質の分泌に関するELISA分析、c)ウイルス複製のqPCR解析ならびにd)遺伝子発現のRT−qPCR解析によって評価した。
精製T細胞ではなく未分離ヒトPBMCを用いて同じ活性評価の実施を含むことにより、実施例21に記載したものと同様のウイルス選択性実験を実施した。実施例21のヒトT細胞の場合と同じく、この実験のデータ全体から、ヒトPBMCによるウイルス複製および導入遺伝子発現はみられなかったことがわかる。図35〜37には5000ppcのEnAd、NG−347またはNG−348を用いたデータが示されているが、500ppcを用いても同様のデータが得られた(不掲載)。図35にはウイルスゲノム複製およびヘキソンmRNA発現が示されており、図36および37には導入遺伝子のmRNA発現が示されている。アッセイのバックグラウンドは、培地に各ウイルスを添加した後、細胞溶解物試料の場合と同じように処理したアッセイで得られたシグナルに従って設定した。ウイルスを加えた上記のPBMC培養物はいずれも、CD3+T細胞、CD40+細胞(主としてB細胞)ともにCD80導入遺伝子の検出可能な発現はみられなかった(不掲載)。
この実施例は、実施例15〜17、21および22の実験とほぼ同じデザインであるが、今回の実験では、ヒトPBMCまたは精製T細胞と、ウイルスで前処理した(48時間)A549腫瘍細胞またはMRC5線維芽細胞とを共培養した。A549細胞またはMRC5細胞を10ppcのEnAd、NG−347、NG−348で処理するか、処理せずにおき(UTC)、48時間培養して、ウイルス複製および任意の導入遺伝子発現が起こるのに十分な時間をとった。次いで、この培養物にPBMCまたはT細胞を加えて24時間または48時間放置し、ウイルス処理細胞がT細胞を活性化することができるかどうかを評価した。
CD14+単球細胞を抗体コート磁気ビーズによる分離によってPBMCから単離し、ヒトIL−4およびGM−CSFと培養して樹状細胞に分化させた。3日間培養した後、細胞を5000ppcのEnAd、NG−347またはNG−348で処理するか、処理せずにおいた。陽性活性化対照として、一部の細胞をLPSで刺激した。2日後、上清を採取してサイトカインELISAに供し、細胞に表面活性化マーカー発現をみる染色を実施してフローサイトメトリーにより解析した。表8に示されるように、いずれのウイルスも共刺激分子CD80およびCD86のアップレギュレーションを誘導し、上で確認された粒子媒介性自然免疫細胞活性化効果が、NG−347およびNG−348のゲノムに導入遺伝子を組み込むことによって変化することはないことがわかった。このほか、いずれのウイルスもMIP1αおよびIFNαの同レベルの分泌を刺激した(図42)。
1組の実験では、EnAdを陰性対照とし、NG−347ウイルス、NG−348ウイルスおよびNG−420ウイルスによる導入遺伝子発現の機能を評価するT細胞活性化レポーターアッセイとして、JurkatDual細胞を腫瘍細胞との共培養に用いた。JurkatDual細胞は、2つの異なるレポーター遺伝子、すなわち、T細胞受容体複合体を介するシグナル伝達に応答する分泌型ルシフェラーゼを産生するNFκBレポーター遺伝子およびIFNα応答性分泌型アルカリホスファターゼ(SEAP)レポーター遺伝子を安定に発現する。A549細胞を10ppcのウイルスと2日間前培養した後、JurkatDual細胞を加えて一晩(18〜24時間)共培養し、次いで、上清を収集してルシフェラーゼ活性およびSEAP活性のアッセイに供した。図43に示されるように、NG−347を感染させたA549細胞は、そのIFNαの産生と一致するSEAP産生を選択的に誘導した(図11を参照されたい)が、ルシフェラーゼ活性は誘導しなかった。これに対し、膜抗CD3−ScFvを発現してT細胞受容体複合体を活性化するNG−348は、ルシフェラーゼを誘導したが、SEAPは誘導しなかった。
EnAdおよびNG−348を免疫適格CD1マウスにIV投与した後のin vivo薬物動態、生体内分布および粒子媒介性全身サイトカイン誘導活性を比較した。5×109個のEnAd粒子またはNG−348粒子をマウスの静脈内に投与し、投与から2分後、10分後、30分後、60分後および120分後に採血した。全血のDNAを抽出し、ウイルスゲノムのレベルをみるqPCRによって解析した(図46)。両ウイルスの血液からのクリアランスはほぼ同じ動態を示した。同様に、MCP−1サイトカイン応答(肝クッパー細胞などの自然免疫の粒子媒介性活性化の尺度)の誘導も両ウイルスで同程度であり、組織生体内分布パターンもほぼ同じであった(図46)。
CB17 SCIDマウスの皮下にHCT116細胞を移植し、腫瘍が70mm3を超えてからEnAdウイルス、NG−347ウイルスもしくはNG−348ウイルス(ウイルス粒子数5×109個)または対照を腫瘍内(IT)または静脈内(IV)に注射した。IV投与マウスでは、IV投与から3分後、15分後および30分後に各グループ3匹のマウスから血液試料を採取し、DNAを抽出し、qPCRにより血中のウイルスゲノムレベルを評価した(薬物動態[PK]解析)。データ(図47)から、NG−347とNG−348のPKはEnAd(および互い)に類似していることがわかる。6時間後、各グループ3匹のマウスから腫瘍、肝臓、肺および脾臓を摘出した。ホモジナイズした組織からDNAを抽出し、qPCRによりウイルスゲノムレベルを解析した(生体内分布解析)。データ(図48A)から、ウイルスの組織生体内分布が3種類とも類似していることがわかる。7日または14〜21日後、各グループ3匹のマウスから腫瘍を摘出し、ホモジナイズして腫瘍溶解物とし、これを用いてDNAおよびRNAの両方を調製した。抽出したDNAのqPCR解析によって、2つの時点での腫瘍中のウイルスゲノムレベルを測定した。データ(図48B)から、IV投与マウス、IT投与マウスともに腫瘍のウイルスゲノムレベルが投与したウイルス量よりも高く、ウイルスが組織内で複製されたことがわかり、第7日のゲノムレベルはIV投与よりもIT投与の方が高いが、第14日〜21日の時間枠ではともに同程度に高い。ウイルスは3種類とも同等のレベルまで複製された。
in vivoのNG−348ウイルスの活性および腫瘍細胞依存性を評価するため、ヒトPBMC(5×107個)、A549ヒト腫瘍細胞(5×106個)およびEnAdまたはNG−348(5×109ppc)を様々に組み合わせて免疫不全SCIDベージュマウスの腹膜内に注射し、細胞を注射してから15分以内にウイルスまたは対照(生理食塩水)を投与した。3日後、腹膜腔を生理食塩水5mLで洗浄し、回収した細胞をT細胞活性化マーカーのパネル(CD25、CD69およびHLA−DR)を用いたフローサイトメトリー解析によって解析し、CD3+T細胞集団にゲートをかけた後のT細胞活性化のレベルを評価した。2つの独立した実験で得られたデータ(表9)から、NG−348がin vivoでヒトT細胞活性化を選択的に腫瘍細胞依存性に引き起こすことがわかる。

- 癌細胞に対する選択性を有する複製可能な腫瘍溶解性B群アデノウイルスであって、前記アデノウイルスに内在するプロモーターの制御下に導入遺伝子を含み、且つスプライスアクセプター配列を含み、前記導入遺伝子が、B7タンパク質をコードするDNA配列を含み、
前記導入遺伝子とスプライスアクセプター配列が、アデノウイルスL5遺伝子の停止コドン−ポリA認識部位とアデノウイルスE4遺伝子の停止コドン−ポリA認識部位との間に位置し、
前記導入遺伝子が、内在性の主要後期プロモーターの制御下にある、
