Description
1.抗CD40抗体またはその結合フラグメントをコードする導入遺伝子カセットを含む腫瘍溶解性ウイルス(例えば、複製可能なウイルス)であって、導入遺伝子カセットが、配列番号12において付与されるアミノ酸配列またはそれに対して、特に、配列番号12のカセットに対して少なくとも95%同一(例えば、それに対して、特に、配列の全長に渡り96、97、98または99%同一)の配列を含む、腫瘍溶解性ウイルス。(代替の)項1:抗CD40抗体またはその結合フラグメントをコードする腫瘍溶解性アデノウイルス(例えば、複製可能な腫瘍溶解性アデノウイルス)であって、アデノウイルスが、配列番号1またはそれに対して少なくとも95%同一(例えば、それに対して96、97、98または99%同一)の配列を含む、腫瘍溶解性アデノウイルス。

4.配列番号1を含む、項1~3のいずれか1項に記載の腫瘍溶解性ウイルス。
5.配列番号1からなる、項4に記載の腫瘍溶解性ウイルス。
6.項1~5のいずれか1項に記載のウイルス、および医薬的に許容される賦形剤、希釈剤または担体を含む、医薬組成物。
7.処置における使用のための、請求項1~5のいずれか1項に記載の腫瘍溶解性ウイルスまたは項6に記載の医薬組成物。
8.癌、インスリン抵抗性、肥満および/または免疫不全の処置における使用のための、項1~5のいずれか1項に記載の腫瘍溶解性ウイルスまたは項6に記載の医薬組成物。
9.ウイルスまたは組成物が、癌の処置における使用のため、例えば、CD40を発現している癌(例えば、アップレギュレーションされた発現のCD40を伴う癌)の処置のためである、項6に記載の使用。
10.項1~5のいずれか1項に記載のウイルスまたは項6に記載の組成物およびさらなる抗癌療法を含む、併用療法(例えば、癌の処置における使用のため)。
11.さらなる抗癌療法が、化学療法である、項10に記載の併用療法。
12.さらなる抗癌療法が、チェックポイント阻害剤である、項10または11に記載の併用療法。
13.抗癌療法が、PD-1阻害剤、PD-L1阻害剤、CTLA-4阻害剤、TIM-3阻害剤、LAG-3阻害剤、TIGIT阻害剤、B7-H3(CD276)阻害剤、B7-H4(B7S1)阻害剤、B7H7(HHLA2)阻害剤、CD96阻害剤、VISTA阻害剤およびその2つ以上の組み合わせを含む群から選択される、項12に記載の併用療法。
14.阻害剤が、抗体またはその結合フラグメントである、項13に記載の併用療法。
15.さらなる抗癌療法が、同時刺激経路アゴニストである、項10~14のいずれか1項に記載の併用療法。
16.さらなる抗癌療法が、CD27アゴニスト、CD28アゴニスト、ICOSアゴニスト、TMIGD2(IGPR-1/CD28H)アゴニスト、CD226アゴニスト、OX40アゴニスト、4-1BBアゴニスト、およびその2つ以上の組み合わせを含む群から選択される、項15に記載の併用療法。
17.療法が、抗体またはその結合フラグメントである、項16に記載の併用療法。
18.さらなる抗癌療法が、免疫応答を活性化するか、または免疫応答の抑制、例えば、IL-10、TGFβ、IDO阻害剤、およびその2つ以上の組み合わせから選択される免疫応答の抑制を逆転させる、項10~17のいずれか1項に記載の併用療法。
19.さらなる癌療法が、腫瘍溶解性ウイルス(さらなる腫瘍溶解性ウイルス)、例えば、アデノウイルス、特に、群Bアデノウイルスのような、複製可能な腫瘍溶解性ウイルスである、項10~18のいずれか1項に記載の併用療法。
20.腫瘍溶解性ウイルス(さらなる腫瘍溶解性ウイルス)が、RNAi配列、抗体またはその結合フラグメント、ケモカイン、サイトカイン、免疫調節物質および酵素からなる群から選択される物質をコードする治療遺伝子をコードする、項19に記載の併用療法。
21.抗体またはその結合フラグメントが、OX40、OX40リガンド、CD27、CD28、CD30、CD40、CD40リガンド、CD70、CD137、GITR、4-1BB、ICOS、ICOSリガンド、CTLA-4、PD-1、PD-L1、PD-L2、VISTA、B7-H3、B7-H4、HVEM、ILT-2、ILT-3、ILT-4、TIM-3、LAG-3、BTLA、LIGHT、CD160、CTLA-4、PD-1、PD-L1、PD-L2、CD40、CD40リガンドおよびその2つ以上の組み合わせに特異的である、項20に記載の併用療法。
22.サイトカインが、IL-1α、IL-1β、IL-6、IL-9、IL-12、IL-13、IL-17、IL-18、IL-22、IL-23、IL-24、IL-25、IL-26、IL-27、IL-33、IL-35、IL-2、IL-4、IL-5、IL-7、IL-10、IL-15、IL-21、IL-25、IL-1RA、IFNα、IFNβ、IFNγ、TNFα、TGFβ、リンホトキシンα(LTA)およびGM-CSF、例えば、IL-12、IL-18、IL-22、IL-7、IL-15、IL-21、IFNγ、TNFα、TGFβおよびリンホトキシンα(LTA)ならびにその2つ以上の組み合わせを含む群から独立して選択される、項20または21に記載の併用療法。

抗CD40モノクローナル抗体をコードする生成した第1のenadenotucirevウイルスゲノムを、NG-350(配列番号13)と命名した。
プラスミドpNG-350を、酵素AscIを用いた制限酵素切断により直線化して、ウイルスゲノムNG-350(配列番号13)を産生した。ウイルスNG-350を増幅させ、以下で与える方法に従い、精製した。
粒子収量に関するNG-350ウイルス活性および分泌された抗CD40抗体産生を、HEK293懸濁培養細胞株において評価した。HEK293細胞を、振盪フラスコにおいて1×106個のウイルス粒子/mLの密度で播種し、細胞1個当たり50個のNG-350ウイルス粒子(ppc)を感染させた。HEK293細胞をまた、NG-350親ウイルス、対照としてenadenotucirev(EnAd)を感染させた(50ppc)。感染の48および72時間後に、細胞上清を、5分間、1200rpm遠心分離することにより、収集した。試料の清透化した上清を、HPLCによりウイルス粒子濃度またはヒトIgG2ELISAにより抗CD40抗体濃度を評価するため使用した。
Resource Q(陰イオン交換)カラムを使用した高速液体クロマトグラフィー(HPLC)により、上清中のウイルス粒子濃度を定量した。ウイルス溶出を260nmにおいて検出し、260nmのシグナルピークを積分し、enadenotucirev標準曲線から濃度を計算することにより、ウイルス濃度を決定した。Ng-350およびenadenotucirev細胞からのウイルス産生の比較は、トータルの粒子産生に関するNG-350ウイルス活性および細胞上清におけるウイルス粒子産生が、enadenotucirevより有意に少ないことを示した(図2Aおよび2B)。
清透化した上清を、PBS10%BSAに2分の1希釈した。標準曲線および陰性対照試料を、製造元のプロトコール(IgG2ヒトSimpleStep ELISAキット、ab202402、Abcam)に従い調製した。試料および標準をELISAプレートに加え、製造元のプロトコールに従い、アッセイを行った。プレートのそれぞれのウェルにおける450nmでの吸光度を、プレートリーダー(BioTek)を使用して測定し、分泌されたIgG2抗CD40抗体の濃度を、標準曲線から書き入れることにより、決定した(図2C)。NG-350から、抗CD40を産生させ、分泌させたが、enadenotucirevが感染した細胞ではなかった。
制限酵素解析
NG-350ウイルスストック物質の同一性を、制限酵素解析を介してゲノム同一性を解析することにより、まず調べた。NG-350またはenadenotucirevウイルス粒子(3.5×1011個のウイルス粒子)を、PBSにおいて終容積200μLまで希釈した。製造元のプロトコールに従い、QIAgen Mineluteウイルススピンキットを使用して、ウイルスからDNAを抽出した。酵素AscIを用いて、2時間、37℃でpNG-350プラスミドを直線化し、次に、アガロースゲル電気泳動およびQiaQuick Gel抽出キット(Qiagen)を用いたゲル抽出により、精製することにより、アッセイのための対照DNAを調製した。制限酵素EcoRv/NheIの組み合わせ(図3A)または個々の制限酵素NcoIもしくはFspI(図3B)を使用して、精製した対照またはNG-350DNAを制限酵素切断した。切断したDNAを、アガロースゲル電気泳動により分離し、UVトランスイルミネーターを使用して、可視化した。FspIまたはEcoRV/NheIを用いた切断は、予測していなかった、NG-350ウイルスゲノム物質における追加のバンドを示した(図3、赤色の矢印)。
ウイルス増幅中に生じる切断についての説明は、導入遺伝子カセットにおける予測できない不安定性であり、これが、VH領域とSV40ポリAの間の組換えをもたらし、それ故、抗体コード領域の大部分の喪失をもたらす。それ故、導入遺伝子カセットDNA配列は、この問題を克服するために修飾されることを必要とする。それ故、DNA配列を変更して、他のカセットおよびウイルス配列との相同性を低減し、少数の直接および逆方向反復を取り除いた(Oxford Genetics、UKが行った)。最適化したカセット配列を使用して、実施例4による新規プラスミドpNG-350Aを生み出した。
NG-350Aゲノムプラスミドを産生するために、プラスミドpEnAd2.4への、5’短いスプライスアクセプター配列(CAGG、配列番号2);重鎖リーダー配列(配列番号3)、抗CD40VH鎖(配列番号4)、抗体定常重鎖(配列番号5)、P2A高い効率の自己切断可能なペプチド(配列番号6)、軽鎖リーダー配列(配列番号7)、抗CD40VL鎖(配列番号8)、抗体定常軽鎖(配列番号9)およびSV40ポリ(A)テール(配列番号10)をコードするカセットの直接的挿入により、pNG-350を生成した。抗CD40抗体をコードするNG-350Aのアミノ酸配列は、NG-350ウイルスにおいてコードされるものと同一であり、カセット構造は同じであった(図1)。しかしながら、導入遺伝子カセットの核酸配列を修飾し、それは、NG-350のもの(配列番号11)と有意に異なった。
プラスミドpNG-350Aを、酵素AscIを用いた制限酵素切断により直線化して、ウイルスゲノムNG-350(配列番号1)を産生した。ウイルスNG-350Aを増幅させ、実施例1において記載した方法に従い、精製した。
導入遺伝子カセットにまたがる産物を生み出す、2種のプライマープローブセット(表2;DおよびK)を使用したPCR解析により、NG-350Aゲノム同一性を確認した。実施例3に詳述した方法に従い、NG-350A DNAおよび対照DNAを調製した。実施例3において詳述した方法に従い、プライマーセットDおよびKを使用して、PCR解析を行った。PCR産物の可視化により、予想したサイズの単一の産物を示した(図6)。いずれのプライマーセットを用いても、コンタミしている産物を検出しなかった。

ウイルス腫瘍溶解能
HT-29大腸癌腫細胞を、96ウェルプレートにおいて2.5e4細胞/ウェルの細胞密度において播種した。プレートを、4時間、37℃、5%CO2でインキュベーションし、その後、細胞に、1個の細胞当たり100~0.39個の粒子(ppc)の感染密度範囲で、EnAdまたはNG-350Aウイルス粒子のいずれかを感染させた。感染の72時間後、細胞タイター96MTS試薬(Promega:G3581)を使用して、HT-29細胞生存率を評価した。それぞれの感染密度における%細胞生存の定量により、EnAdに類似の、NG-350Aが、HT-29細胞に対する強力な腫瘍溶解性活性を示すことを示した(図7A)。
肺癌腫細胞(A549)または大腸癌腫細胞(HCT-116)に、100ppcのNG-350AもしくはNG-350A親ウイルス、enadenotucirevを24、48、72または96時間感染させるか、または感染させないままにした。大腸癌腫細胞(HT-29)または膀胱癌腫細胞(HTB-5、HT-1197およびHT-1376)に、100ppcのNG-350Aもしくはenadenotucirevを24、48、72、144および168時間感染させるか、または感染させないままにした。それぞれの時間ポイントにおいて、細胞上清を収集し、5分間、1200rpmで遠心分離することにより、清透化させた。製造元のプロトコールに従い、DNeasy血液および組織キット(Qiagen)を使用して、上清10μL(HT-29、A549もしくはHCT-116)または50μL(HTB-5、HT-1197およびHT-1376)からDNAを抽出した。EnAdウイルス粒子(2.5e10~2.5e5ウイルス粒子)を使用した標準曲線をまた、調製し、DNeasy血液および組織キット(Qiagen)を使用して抽出した。enadenotucirevE3遺伝子に特異的なプライマーとプローブセットを使用したqPCRにより、それぞれの抽出した試料または標準を解析した。
IgG2 ELISA
100ppcのEnAd、2つの異なるバッチのNG-350A(NG-350A B1もしくはNG-350A B2)を24、48または72時間感染させた、または感染させないままの、A549細胞を、IgG2 ELISA(IgG2ヒトSimpleStep ELISAキット、ab202402、Abcam)による抗CD40抗体発現の解析のため使用した。培養上清をそれぞれのウェルから取り出し、5分間、1200rpmで遠心分離して、細胞デブリを取り除いた。清透化した上清を、PBS10%BSAに2分の1希釈した。標準曲線および陰性対照試料を、製造元のプロトコールに従い調製した。試料および標準をELISAプレートに加え、製造元のプロトコールに従い、アッセイを行った。プレートのそれぞれのウェルにおける450nmでの吸光度を、プレートリーダー(BioTek)を使用して測定し、分泌されたIgG2抗CD40抗体の濃度を、標準曲線の記入形態により、決定した(図9Aおよび9B)。
100ppcのEnAd、2つの異なるバッチのNG-350A(NG-350A B1もしくはNG-350A B2)を24、48または72時間感染させた、または感染させないままの、A549細胞を、CD40結合ELISAによる抗CD40抗体発現の解析のため使用した。
10ppcのEnAd、NG-350Aを用いて24もしくは48時間またはNG-165(対照抗体[抗VEGF]を発現するウイルス)を用いて48時間のいずれかで感染させた、あるいは感染させていないままのA549細胞を、それらの膜で発現したCD40分子を介した活性化に対して応答してアルカリホスファターゼ(Invivogen)を分泌するCD40+HEK-Blueレポーター細胞を使用した、抗CD40抗体機能活性の解析のため使用した。感染後、A549細胞由来の培養上清をそれぞれのウェルから取り出し、5分間、1500rpmで遠心分離して、細胞デブリを取り除いた。
EnAdウイルス活性に半寛容である、肺癌腫細胞(A549)、大腸癌腫細胞(HCT-116)または肺線維芽細胞細胞(MRC-5)を使用して、癌細胞についてのNG-350Aウイルスの選択性を示した。細胞株に72時間、100ppcのNG-350AもしくはNG-350A親ウイルス、enadenotucirevを感染させるか、または感染させないままにした。
細胞上清を収集し、5分間、1200rpmで遠心分離することにより、清透化させた。製造元のプロトコールに従い、Sigma Genelute DNA抽出キットを使用して、上清10μL(A549、HCT-116)または100μL(MRC-5)からDNAを抽出した。EnAdウイルス粒子(2.5e10~2.5e5ウイルス粒子)を使用した標準曲線をまた、調製し、Sigma Geneluteキットを使用して抽出した。enadenotucirevE3遺伝子に特異的なプライマーとプローブセットを使用したqPCRにより、それぞれの抽出した試料または標準を解析した。
IgG2 ELISA(IgG2ヒトSimpleStep ELISAキット、ab202402、Abcam)により、細胞上清にあった抗体発現を評価した。培養上清をそれぞれのウェルから取り出し、5分間、1200rpmで遠心分離して、細胞デブリを取り除いた。清透化した上清を、PBS10%BSAに2分の1希釈した。標準曲線および陰性対照試料を、製造元のプロトコールに従い調製した。試料および標準をELISAプレートに加え、製造元のプロトコールに従い、アッセイを行った。プレートのそれぞれのウェルにおける450nmでの吸光度を、プレートリーダー(BioTek)を使用して測定し、分泌されたIgG2抗CD40抗体の濃度を、標準曲線の記入形態により、決定した(図11B)。
懸濁液HEK293細胞を、1×106個の細胞/mLにてエルレンマイヤーフラスコにおいて播種し、100ppcのNG-350Aを感染させた。72時間後、5%FBSおよびタンパク質分解酵素阻害剤カクテル(1:2000)を細胞に加え、懸濁液を15分間、4600rpmで遠心分離した。上清を注意深く取り出し、500kDaの分子量カットオフ中空ファイバー膜を通して濾過して、NG-350Aウイルス粒子を抗CD40抗体から分けた。抗CD40抗体を含有する、濾過工程由来の素通り画分を、30kDaの分子量カットオフを有する第2の中空ファイバー膜を通過させた。AKTAを使用したタンパク質Aカラム上での第2の濾過工程由来の残余分から、抗CD40抗体を精製した。精製した抗体を濾過滅菌し、-80℃において保存した。製造元のプロトコールに従い、IgG2ヒトSimpleStep ELISAキット、ab202402、Abcamを使用したIgG2 ELISAにより、精製した抗体の濃度を決定した(図12)。
英国、オクスフォードにあるNHSの血液および移植ユニットから供給源されたNC24白血球コーンから、Ficoll-Paque勾配遠心分離により、PBMCを単離した。CD14マイクロビーズキット(MiltenyiBiotec)を使用して、CD14+単球を単離した。次に、単球をカウントし、遠心分離し(300×g)、GM-CSF(800U/mL)およびIL-4(500U/mL)を添加した10%RPMI培地中5×105個の細胞/mLにて再懸濁した。単球懸濁液40mLを、1つのT175フラスコに移した。
英国、オクスフォードにあるNHSの血液および移植ユニットから供給源されたNC24白血球から、Ficoll-Paque勾配遠心分離により、PBMCを単離した。Pan B細胞単離キット(MiltenyiBiotec)を使用して、CD19+B細胞を単離した。次に、B細胞を、10%RPMI培地中24ウェルプレートにおけるウェル毎に1×106個の細胞密度で播種した。それらを、増大する濃度の精製した抗CD40導入遺伝子抗体、ヒトCD40Lで処理するか、または処理しないままにした。次に、プレートを、48時間インキュベーションし、その後、上清および細胞を回収した。
A549腫瘍細胞を、T175フラスコまたはHyperflaskにおいて、それぞれ、1個のフラスコ当たり10×106または50×106個の細胞の密度にて播種した。4時間後、細胞に、1個の細胞当たり10個のEnAdまたはNG-350Aウイルス粒子を感染させた。72時間後、上清を回収し、300kDaのカットオフ除外カラムを使用して、ウイルスを枯渇させた。50kDaのカットオフサイズ除外カラムを使用して、ウイルスを枯渇させた上清を、抗体について続いて濃縮した。これらのウイルスを枯渇させた抗体濃縮分画を、-80℃において保存した。IgG2 ELISAを使用して、抗CD40抗体タイターを決定し、実施例8において記載したHEK Blueレポーター細胞アッセイを使用して、機能性を確認した。
実施例13において記載したものと類似の研究において、A549細胞を、T25フラスコにおいて密度4×106個の細胞において播種した。4時間後、細胞に、1個の細胞当たり10個のEnAdまたはNG-350Aウイルス粒子を感染させた。72時間後、上清を回収し、1600rpmにおいて5分間遠心分離した。この研究において、これらの清透化した上清を、ウイルスを除去するよりむしろ、使用(1時間以内)まで37℃において維持した。従って、上清は、ウイルスと抗CD40抗体導入遺伝子産物の両方を含む、NG-350A(またはEnAd対照)感染腫瘍細胞の産物を含有する。
英国、オクスフォードにあるNHS血液および移植ユニットから供給源されたNC24白血球コーンから、Ficoll-Paque勾配遠心分離により、PBMCを単離した。Pan B細胞単離キット(MiltenyiBiotec)を使用して、B細胞を単離した。次に、細胞を、10%RPMI培地中48ウェルプレートにおける1個のウェル当たり1.25×105個の細胞密度で播種した。それらを、実施例13において記載したNG-350A感染細胞由来の異なる濃度のウイルス枯渇抗CD40抗体濃縮上清を用いて処理するか、または未処理(培地)のままにした。B細胞をまた、対照としてウイルス枯渇EnAdウイルス上清を用いて処理した。次に、プレートを、24時間および48時間インキュベーションし、その後、細胞を回収した。細胞を、CD23、CD54、CD86およびHLA-DRに対する抗体を用いて染色し、その後、フローサイトメトリー解析を行った。細胞に、単一(FSC-H versus FSC-A)生細胞(LIVE/DEAD(登録商標)Fixable Aqua negative)上にゲートを掛けた。図23は、ウイルス枯渇NG-350A抗CD40抗体含有上清による、3人の異なるドナー(177、178、179)由来のB細胞上での全4種の表面マーカー(CD23、CD54、CD86およびHLA-DR)の選択的アップレギュレーションを示す。

別の研究において、NG-350Aの活性を、6種の癌指標に渡る広範な異なる腫瘍細胞株(結腸直腸、前立腺、膵臓、乳房、卵巣および膀胱)に対して評価した。細胞に、1個の細胞当たり1または100個の粒子(ppc)のenadenotucirev(EnAd)またはNG-350Aを感染させ、3~11日間培養し、これにより、NG-350A感染腫瘍細胞におけるウイルスゲノム複製、ウイルス介在性腫瘍退縮、ウイルス導入遺伝子発現(mRNAおよびタンパク質レベルにおける)ならびに機能的ウイルス導入遺伝子発現を評価した。全ての実験において、NG-350Aの活性を、EnAdのものと比較した。A549(非小細胞肺癌腫)細胞を陽性対照として、CT26(マウス大腸腫瘍)細胞を陰性対照として使用した。
腫瘍細胞株を、175cm2のフラスコにおいて、L-グルタミン、非必須アミノ酸、ピルビン酸ナトリウムおよび10%胎児ウシ血清(FBS)を添加した高グルコース(成長培地)において培養した。使用前、細胞を顕微鏡を使用して検査して、60~80%の集密度を確実にした。次に、PBSおよび0.25%トリプシン-EDTAを加えて、細胞を洗浄して、細胞をフラスコの底からはがした。細胞がはがれるまで、細胞を2~10分間、37℃、5%CO2においてインキュベーションし、その後、トリプシンを成長培地を使用して脱活性化した。細胞懸濁液を混合し、300gにて5分間スピンした。上清を捨て、細胞ペレットを、アッセイ培地;2%(10%の代わりに)FBSを添加した上記のDMEM培地10mLに再懸濁した。細胞をカウントして、細胞懸濁液を、アッセイ培地を使用して希釈して、濃度5×105個の細胞/mLにし、その100μLを、平底96ウェルプレートに播種した(それぞれの細胞株を、1種のウイルス当たり2つのウェルに播種し、時間ポイント毎に1枚のプレートを用いて、1枚のプレート当たり未感染の対照(UIC)2つのウェルを含めた)。EnAdおよびUICについての試料を、一方のセットのプレートに播種し(感染させ)、NG-350AおよびUICについての試料を、別のセットのプレートに播種した(感染させた)。プレートを、37℃、5%CO2においてインキュベーションし、その後、細胞播種の4~6時間後に接種した。

さらなる一連の実験において、NG-350Aウイルスのインビボウイルス粒子介在性効果をモニターし、マウスにおける1回または複数回静脈内投与後のEnAdのものと比較した。メスCD-1マウスに、ビークル対照またはEnAdもしくはNG-350Aいずれかの2.2×1010個の粒子をIV注射した。注射の6時間、24時間、48時間、または7日後に、マウスの心臓から採血し、血液を抗凝固剤チューブに収集し、処理して血漿を回収した。血漿試料をELISAアッセイにより試験して、ウイルス粒子に対する実際のサイトカイン応答を検出し、図29において示すデータは、1群当たり3~6匹のマウスを表す。NG-350A投与による刺激されたMCP-1(図29A)、IL-6(図29B)およびTNFα(図29C)レベルは、EnAdについてのものと類似した。黒色の実線は、平均を表す。ベースラインは、ビークル対照群の平均+3×SDを表す。
免疫適格CD-1マウスにおける1、3および5日目での2.2×1010個のウイルス粒子の3回の静脈内(IV)投与のそれぞれの投与後、NG-350Aの血液薬物動態を、enadenotucirev(EnAd)と比較した。
1回の静脈内投与後に組織から生きたウイルスの回復を測定することにより、NG-350Aを用いたマウスの投与後のウイルス体内分布を評価した。メスCD-1マウスに、ビークル対照またはNG-350Aの2.2×1010個の粒子をIV注射した。マウスの群を、投与の6時間、24時間、8日または28日後のいずれかにおいて安楽死させ、それらの肝臓、肺および脾臓を切除し、直ちにドライアイス上で凍結した。試料を後に融解し、タンパク質保存溶解バッファーにおいてホモジナイズし、溶解物を希釈し、陽性対照および陰性対照としてそれぞれの器官についてのNG-350A添加組織ホモジネートと共に、コンフルエントなA549単層に加えた。単層を96時間培養し、その後、固定し、抗アデノウイルスヘキソン抗体を使用した免疫染色アッセイの対象にした。図33におけるそれぞれのウェルの試料写真により、データを表す。ヘキソン陽性細胞は、茶色に染まった(図において濃い灰色として示す)。24時間より後では、マウスの肝臓、肺または脾臓(マウスにおけるウイルス体内分布の主要部位)において、生きたウイルスを検出することはできなかった。
この一連の研究において、NG-350Aウイルス活性を、ヒト腫瘍異種移植片を生じる免疫不全マウスにおいてインビボで評価した。

Claims (20)
- 抗CD40抗体またはその抗原結合フラグメントをコードする導入遺伝子カセットを含む腫瘍溶解性群Bアデノウイルスであって、導入遺伝子カセットが、配列番号12に示される塩基配列を含む、腫瘍溶解性群Bアデノウイルス。
- Enadenotucirevである請求項1に記載の腫瘍溶解性群Bアデノウイルス。
- 配列番号1またはそれに対して少なくとも95%同一の塩基配列を含む、請求項1又は2に記載の腫瘍溶解性群Bアデノウイルス。
- 配列番号1からなる塩基配列を含む、請求項3に記載の腫瘍溶解性群Bアデノウイルス。
- 請求項1~4のいずれか1項に記載の腫瘍溶解性群Bアデノウイルス、および医薬的に許容される賦形剤、希釈剤または担体を含む、医薬組成物。
- 処置における使用のための、請求項1~4のいずれか1項に記載の腫瘍溶解性群Bアデノウイルスまたは請求項5に記載の医薬組成物。
- 癌、インスリン抵抗性、肥満および/または免疫不全の処置における使用のための、請求項1~4のいずれか1項に記載の腫瘍溶解性ウイルスまたは請求項5に記載の医薬組成物。
- 前記腫瘍溶解性群Bアデノウイルスまたは組成物が、癌の処置のためである、請求項1~4のいずれか1項に記載の腫瘍溶解性ウイルスまたは請求項5に記載の医薬組成物。
- 請求項1~4のいずれか1項に記載の腫瘍溶解性群Bアデノウイルスまたは請求項5に記載の組成物およびさらなる抗癌剤を含む、併用剤。
- 前記さらなる抗癌剤が、化学療法剤である、請求項9に記載の併用剤。
- 前記さらなる抗癌剤が、チェックポイント阻害剤である、請求項9に記載の併用剤。
- 前記抗癌剤が、PD-1阻害剤、PD-L1阻害剤、CTLA-4阻害剤、TIM-3阻害剤、LAG-3阻害剤、TIGIT阻害剤、B7-H3(CD276)阻害剤、B7-H4(B7S1)阻害剤、B7H7(HHLA2)阻害剤、CD96阻害剤、VISTA阻害剤およびその2つ以上の組み合わせを含む群から選択される、請求項11に記載の併用剤。
- 前記阻害剤が、抗体またはその抗原結合フラグメントである、請求項12に記載の併用剤。
- 前記さらなる抗癌剤が、同時刺激経路アゴニストである、請求項9~13のいずれか1項に記載の併用剤。
- 前記さらなる抗癌剤が、CD27アゴニスト、CD28アゴニスト、ICOSアゴニスト、TMIGD2(IGPR-1/CD28H)アゴニスト、CD226アゴニスト、OX40アゴニスト、4-1BBアゴニスト、およびその2つ以上の組み合わせを含む群から選択される、請求項14に記載の併用剤。
- 前記抗癌剤が、抗体またはその抗原結合フラグメントである、請求項15に記載の併用剤。
- 前記さらなる抗癌剤が、免疫応答を活性化するか、または免疫応答の抑制を逆転させる、請求項9~16のいずれか1項に記載の併用剤。
- 前記さらなる抗癌剤が、腫瘍溶解性ウイルスである、請求項9に記載の併用剤。
- 前記腫瘍溶解性ウイルスが、群Bアデノウイルスである、請求項18記載の併用剤。
- 前記腫瘍溶解性ウイルスが、RNAi配列、抗体またはその抗原結合フラグメント、ケモカイン、サイトカイン、免疫調節物質および酵素からなる群から選択される物質をコードする治療遺伝子をコードする、請求項18または19に記載の併用剤。
