本明細書に記載される宿主動物細胞は、特定のグリカン改変活性を有する1以上の酵素(例えば、グリコシダーゼまたはグリコールトランスフェラーゼ)を過剰発現するように遺伝子操作されている。遺伝子操作された宿主動物細胞は、本明細書に記載されるフコシダーゼおよびエンドグリコシダーゼの1以上をコードする外因性の(非天然の)遺伝物質、例えば外因性遺伝子などを保有する動物細胞である。酵素を過剰発現する宿主細胞は、該宿主細胞に対する野生型の対応するもの、すなわち、遺伝子操作された宿主細胞と同じ遺伝子改変を含有しない同じ種類の細胞における酵素のそれよりもより大きい(例えば、20%、50%、80%、100%、2倍、5倍、10倍、50倍、100倍、1,000倍、104倍または105倍高い)レベルで酵素を発現する遺伝子操作された宿主細胞を指す。いくつかの態様において、本明細書に記載される外因性酵素をコードする遺伝子は、好適な親動物細胞へと導入して、本明細書に記載される遺伝子操作された宿主動物細胞を作り出すことができる。外因性酵素は、操作された宿主動物細胞を作製するために用いられた親細胞中に存在しない酵素を指す。
該遺伝子操作された宿主動物細胞は、改変グリコシル化パターンを有する糖タンパク質(例えば、内因性または外因性)の製造に用いることができる。いくつかの態様において、上記の操作された宿主動物細胞を産生することに用いるための親宿主細胞は、外因性糖タンパク質をコードする1または複数の遺伝子を、既に保有している。他の態様において、対象となる糖タンパク質をコードする1遺伝子または複数の遺伝子は、当技術分野において知られた方法または本明細書に記載される方法によって、1以上のフコシダーゼおよび/またはエンドグリコシダーゼを発現する、遺伝子操作された宿主動物細胞へ導入されることができる。
i. フコシダーゼまたはエンドグリコシダーゼを産生するための発現ベクターの構築。
フコシダーゼまたはエンドグリコシダーゼのための発現ベクターを構築するために、フコシダーゼまたはエンドグリコシダーゼ遺伝子を常用の手技により単離し、ハムスター細胞のコドンの使用に基づいて、コドン最適化を施した。合成遺伝子を、GeneArt Corp.により作製し、pcDNA3.1 B(-) Myc-Hisベクター(Invitrogen, US)の制限部位Bgl II/EcoR Iにクローンした(図2、パネルA)。アルファ−フコシダーゼの発現カセットは、5’から3’に向かって、コザック配列、Igkリーダー配列、フコシダーゼのコード配列、および、His−タグをコードする配列を含む。図2、パネルB。エンドグリコシダーゼの発現カセットは、5’から3’に向かって、コザック配列、Igkリーダー配列、エンドグリコシダーゼのコード配列、および、Flagタグをコードする配列を含む。図2、パネルB。
抗体産生細胞株を、完全培地、8mMのL−グルタミンおよび抗クランピング剤(anti-Clumping Agent)の1:100希釈(Life Technologies, USA)を補充したCD FortiCHO(商標)培地中に0.3〜3.0×106生存細胞/mLで維持した。8%CO2インキュベーター内で、細胞を、130〜150rpmに設定した振盪プラットフォーム上に維持した。
本明細書に記載される方法に従って作製した組換え抗体を、還元し、アルキル化し、消化した(終夜、トリプシンで、25mMの重炭酸アンモニウム緩衝液の存在下(pH 〜8))。PNGase F溶液(3μL、Roche)を、200μLの消化した検体に添加して、および、混合物をさらに16時間37℃でインキュベートした。放出されたグリカンを、Sep-Pak(登録商標)C18カートリッジ(Waters)を用いてペプチドから分離した。Sep-Pak C18をアセトニトリルで、次いで水で洗浄した。PNGaseF消化した検体を、カートリッジ上に載せ、ペプチドがSep-Pak C18にバインドさせたまま、放出されたグリカンを1%エタノールで溶出させた。放出されたタンパク質オリゴ糖を、まず多孔質グラファイトカーボンカラム(PhyNexus)を用いて精製し、および次にパーメチル化した。全ての質量分析実験は、Orbitrap Fusion Tribrid質量分析計を用いて、ナノエレクトロスプレー源への直接的な注入により行った。
1. 単糖(GlcNAc)の糖型を有する抗体h4B12、リツキシマブおよびオマリズマブの産生
単糖の糖変異体は、上述した二糖変異体からフコシダーゼ開裂反応によって作製することができた。多数の入手可能な酵素からの検索およびLC/MS/MSによるグリコール−ペプチド解析により、最適化された開裂反応条件では、α−1,6−フコシダーゼを用いて効率的な脱フコシル化を達成することができたということ、および、より高い開裂効率は、より低いNF/F比と関連していることが示された。あるいは、単糖の糖変異体は、エンドグリコシダーゼ(Endo S)およびα−1,6−フコシダーゼを含む、連続して組み合わせられた2つの反応酵素で得ることができた。結果物のモノ−GlcNAc糖変異体を、図3、パネルAに示した。
酵素発現をウェスタンブロットで検出したところ、図4、パネルBに示すとおりであった。
かかる効率性は、均等なグリカン形態を有する抗体の作製におけるトランスグリコシル化のための重要なステップを表す。
本明細書に開示される特徴の全ては、任意の組み合わせで組み合わせてもよい。本明細書において開示される各々の特徴は、同じ、均等な、または類似の目的の役割をする代替的な特徴により置き換えてもよい。ゆえに、他に明示的に述べられない限り、開示される各々の特徴は、一般的な一連の均等物または類似の特徴の一例に過ぎない。
Claims (18)
- フコシダーゼおよびエンドグリコシダーゼを発現する、遺伝子操作された宿主哺乳動物細胞であって、操作された宿主哺乳動物細胞が、改変グリコシル化を有する糖タンパク質を産生し、エンドグリコシダーゼが、Endo S酵素、Endo F2酵素またはEndo F3酵素である、前記遺伝子操作された宿主哺乳動物細胞。
- フコシダーゼが、哺乳動物フコシダーゼまたは細菌フコシダーゼである、請求項1に記載の遺伝子操作された宿主哺乳動物細胞。
- エンドグリコシダーゼが、Endo S酵素である、請求項1または2に記載の遺伝子操作された宿主哺乳動物細胞。
- 外因性糖タンパク質をさらに発現する、請求項1〜3のいずれか一項に記載の遺伝子操作された宿主哺乳動物細胞。
- 糖タンパク質が、抗体、Fc融合タンパク質、サイトカイン、ホルモン、成長因子または酵素である、請求項4に記載の遺伝子操作された宿主哺乳動物細胞。
- 糖タンパク質が、抗体である、請求項5に記載の遺伝子操作された宿主哺乳動物細胞。
- 哺乳動物細胞が、チャイニーズハムスター卵巣(CHO)細胞であり、エンドグリコシダーゼが、Endo S酵素である、請求項1〜6のいずれか一項に記載の遺伝子操作された宿主哺乳動物細胞。
- (i)ヒトFUCA1、ヒトFUCA2、Cricetulus griseusフコシダーゼ アルファ-L-1、Chryseobacterium meningosepticum α1,6−フコシダーゼまたは細菌フコシダーゼBF3242、および(ii)Endo Sを発現する、請求項1〜7のいずれか一項に記載の遺伝子操作された宿主哺乳動物細胞。
- 脱フコシル化糖タンパク質の製造方法であって、
(a)外因性糖タンパク質、(b)フコシダーゼおよび(c)エンドグリコシダーゼを発現する、遺伝子操作された宿主哺乳動物細胞を提供すること;
糖タンパク質、フコシダーゼおよびエンドグリコシダーゼの産生を可能にする条件下で宿主哺乳動物細胞を培養すること;および
糖タンパク質を単離するために宿主哺乳動物細胞または培養上清を回収すること、を含み、
エンドグリコシダーゼが、Endo S酵素、Endo F2酵素またはEndo F3酵素である、前記方法。 - フコシダーゼが、哺乳動物フコシダーゼまたは細菌フコシダーゼである、請求項9に記載の方法。
- エンドグリコシダーゼが、Endo S酵素である、請求項9または10に記載の方法。
- 糖タンパク質が、抗体、Fc融合タンパク質、サイトカイン、ホルモン、成長因子または酵素である、請求項9〜11のいずれか一項に記載の方法。
- 糖タンパク質が、抗体である、請求項12に記載の方法。
- 哺乳動物細胞が、チャイニーズハムスター卵巣(CHO)細胞であり、エンドグリコシダーゼが、Endo S酵素である、請求項9〜13のいずれか一項に記載の方法。

- 請求項1に記載の遺伝子操作された宿主哺乳動物細胞の、作製方法であって、フコシダーゼおよびエンドグリコシダーゼをコードする1以上の発現ベクターを、哺乳動物細胞へ導入することを含み、エンドグリコシダーゼが、Endo S酵素、Endo F2酵素またはEndo F3酵素である、前記方法。
- 外因性糖タンパク質をコードする発現ベクターを哺乳動物細胞へ導入することをさらに含む、請求項17に記載の方法。
