Description
本実施形態の細胞検出方法について、図面を参照して説明する。なお、本実施形態では、試料液に血球細胞が含まれる場合で、目的の細胞が有核赤血球である場合を例示して説明する。
分取工程では、フローサイトメトリー法を実現するフローサイトメータ10が用いられ、試料液の細胞由来の第1情報が取得され、細胞が容器に分取される。
Violet 570、及びBrilliant Violet 421等を挙げることができる。
撮像工程では、容器に分取された細胞を撮像する。細胞を撮像するとは、細胞を対象として撮像する意味であって、実際には細胞ではない異物(ゴミ等)を撮像する場合も含んでいる。撮像工程では、容器20に分取された細胞を遠心分離装置によりウェルの底面に細胞を移動させることが好ましく、また、撮像するため画像撮像装置30を用いることが好ましい。画像撮像装置30として、撮像装置を備える蛍光顕微鏡を挙げることができる。
決定工程にいて、撮像工程により撮像された細胞の画像に基づいて、細胞由来の第2情報を取得し、容器20に分取された細胞の中から、解析する細胞を決定する。上述したように、フローサイトメトリーでは、得られた光学的な情報に基づいて細胞を分取するため、ウェルにゴミ、又は目的とは異なる細胞が回収される場合がある。細胞由来の第2情報を取得し、解析する細胞を決定することにより、ゴミ、又は目的とは異なる細胞を取り除くことができる。結果として、目的の細胞のみを効率的に、かつ精度良く遺伝子解析を行うことができる。予め、目的となる細胞と目的でない細胞について蛍光の強度、細胞の丸さ、細胞を透過した光によって生ずる色、細胞の大きさ、の情報を取得しておき、これらの取得情報から、目的の細胞である確からしさを表す範囲を決定し、その範囲を表す閾値を決定し、その閾値を、目的の解析する細胞として決定することができる。そのように取得し各種情報を細胞由来の第2情報として用いることができる。例えば、目的細胞が有核赤血球等であれば国際公開WO2016021309号公報や国際公開WO2016021311号公報に記載の選別方法における各種情報を用いることができる。
決定工程の後に、容器20から解析する細胞のみを取り出す取出工程を有することが好ましい。解析を行う細胞のみ取り出しすることにより、遺伝子解析または、その前処理を行うことにより、遺伝子解析の時間を短縮することができる。ここで、「細胞のみ取り出しする」とは、細胞だけを取り出して培養液等の他の成分を取り出さないか、又は、細胞の周囲に存在する培養液と共に目的の細胞を取り出しする場合を意味する。
Chain Reaction)に基づく試薬PicoPLEXWGA kit(New
EnglandBiolabs社)、GenomePlex Single Cell Whole Genome Amplification kit(Sigma−Aldrich社)、国際公開WO2012/166425A2号に開示されている、MALBAC法(MultipleAnnealing and Looping−BasedAmplification Cycles)に係る試薬を用いることができる。また、鎖置換型DNA合成反応に基づく試薬として、例えば、GenomiPhi(GEヘルスケア社、GenomiPhiは登録商標)、REPLI−g(Qiagen社、REPLI−gは登録商標)も同様に用いることができる。本実施形態では、PicoPLEXWGA kit(New England Biolabs社)を用いることが好ましい。

Claims (15)
- フローサイトメトリー法により試料液の細胞由来の第1情報を取得し、前記第1情報に基づいて、目的の細胞を、開口を有するウェルが配列された容器に分取する分取工程と、
前記容器に分取された細胞を撮像する撮像工程と、
前記撮像工程により撮像された細胞の画像に基づいて、細胞由来の第2情報を取得し、前記分取された細胞の中から、解析する細胞を決定する決定工程と、
を有する細胞検出方法であって、
前記分取工程は、前記ウェルの位置と前記第1情報とを関連付け、かつ前記決定工程において、関連付けられた前記ウェルの位置と前記第1情報とに、前記第2情報を関連付けることを含み、
前記決定工程は、予め、目的となる細胞と目的でない細胞について情報を取得し、前記取得された情報から閾値を決定し、前記第2情報と前記閾値とに基づいて解析する細胞として決定することを含む、細胞検出方法。 - 前記分取工程の前に、前記細胞を染色する工程をさらに有する請求項1に記載の細胞検出方法。
- 前記容器には、開口を有する複数のウェルが配列されている請求項1又は2に記載の細胞検出方法。
- 前記撮像工程において、遠心分離により前記ウェルの底面に前記細胞を移動させることを含む請求項1から3の何れか一項に記載の細胞検出方法。
- 前記撮像工程において、前記容器に分取された細胞を前記容器のウェルの開口と反対の側から前記細胞を撮像することを含む請求項1から4の何れか一項に記載の細胞検出方法。
- 前記第1情報は、前方散乱光、側方散乱光、及び蛍光の少なくとも一つを含み、前記第2情報は蛍光、細胞の形状、透過色、及び大きさの少なくとも一つを含む請求項1から5の何れか一項に記載の細胞検出方法。
- 前記試料液が血球細胞を含む請求項1から6の何れか一項に記載の細胞検出方法。
- 前記容器は、前記ウェルが行と列とに配置される請求項1から7の何れか一項に記載の細胞検出方法。
- 前記決定工程の後に、前記容器から前記解析する細胞のみを取り出す取出工程を有する請求項1から8の何れか一項に記載の細胞検出方法。
- 前記取出工程において、キャピラリー又はピペットを用いて前記解析する細胞のみを取り出す請求項9に記載の細胞検出方法。
- 前記取出工程で取り出した前記細胞を、PCR用のチューブ又はPCR用のプレートに移動する請求項9又は10に記載の細胞検出方法。
- 前記第1情報及び前記第2情報と、前記PCR用のチューブ又は前記PCR用のプレートの位置情報とを関連づける請求項11に記載の細胞検出方法。
- 前記容器は、容器内部に培養液を含む請求項1から12の何れか一項に記載の細胞検出方法。
- 前記培養液は少なくとも1つの染色液を含む請求項13に記載の細胞検出方法。
- 前記撮像工程の後に、前記培養液を、染色液を含まない培養液に置換する工程を更に含む請求項14に記載の細胞検出方法。
