Description
本発明は一般に、TDP−43を特異的に認識する、抗体(その断片、誘導体及び変異体を含む)のような新規のTDP−43(43kDaのTAR DNA結合タンパク質)に特異的な結合分子に関する。さらに、本発明は、そのような抗体及び他のTDP−43に特異的な結合分子を含む医薬および診断用組成物、並びに血漿、脳脊髄液、脳及び他の試料でTDP−43を検出する及び特定するためのその使用、ならびに、たとえば、前頭側頭葉変性症及び/又は筋委縮性側索硬化症、アルツハイマー病、パーキンソン病及び他のTDP−43タンパク質症のようなTDP−43の発現における凝集又は他の異常に関連する疾病を治療するための受動ワクチン接種戦略を含む治療応用におけるその使用に関する。
lstein,Nature、256(1975),495−497によって最初に記載されたように、抗原刺激されたB細胞のマウス骨髄腫細胞株との効率的で選択的な融合とそれに続く安定な抗体産生ハイブリッドの選抜に基づく。しかしながら、マウスに基づく抗体のヒトにおける治療上の有用性は、非ヒト起源の結末としてのヒト抗マウス抗体(HAMA)の反応によって妨害される。ヒト又はヒト様のモノクローナル抗体を作製するためのアプローチは遺伝子操作を介して利用可能になった。しかしながら、これらの方法は通常、生理的なヒトの免疫応答の過程の間でヒト免疫系によって内因産生される抗体の特徴の多数を提示する抗体を作製するには好適ではないという欠点に悩まされる。さらに、これらの遺伝子操作された抗体は、生物学的に関連する天然の構造及び標的抗原の正常な生理的機能の背景において他のタンパク質及び/又は標的タンパク質との望ましくない交差反応を示し得る。外因性抗体の全身性投与の際の得られる副作用は、たとえば、望ましくない自己免疫疾患からアナフィラキシー反応に及び得る。これらの副作用は、たとえば、マウスのような非ヒト生物に元々起因するいわゆる「ヒト化抗体」、ならびに試験管内又はヒト抗体のレパートリーを発現するように遺伝子操作された異種マウスにおけるいわゆる「完全なヒト抗体」において報告されている。一方、病理上関連する抗原による能動免疫は、これらの抗原に対する制御不能の免疫応答及び結果的に危険な自己免疫応答を招き得る内因性抗原との交差反応を発生する患者の相当なリスクを負う。
従って、上述の限界を克服し、TDP−43の生物学的に関連する構造を特異的に認識する治療用及び診断用の抗体及び他の結合分子を提供するニーズがある。

本発明は、前述のTDP−43結合分子、たとえば、本発明の抗体、又は抗原結合性の断片、又はその変異体又は誘導体、又は本発明のポリヌクレオチド、ベクター又は細胞を含む組成物に関する。本発明の組成物はさらに薬学上許容可能な担体も含むことができる。その上、本発明の医薬組成物はさらに医薬組成物の用途に応じてインターロイキン又はインターフェロンのような剤を含むことができる。たとえば、TDP−43タンパク質症の治療での使用のために、追加の剤は、有機小分子、抗TDP−43抗体及びそれらの組み合わせから成る群から選択することができる。従って、特定の実施形態では、本発明は、TDP−43タンパク質症の予防的処置及び治療的処置のための医薬組成物若しくは診断用組成物を調製するための、対象におけるTDP−43タンパク質症の進行若しくはTDP−43タンパク質症の治療に対する応答をモニターするための、又はTDP−43タンパク質症を発症する対象のリスクを判定するための、TDP−43結合分子、たとえば、本発明の抗体若しくはその抗原結合断片の使用、又はその1つの実質的に同一の結合特異性を有する結合分子の使用、本発明のベクター又は細胞の使用に関する。
トープを認識するモノクローナル抗体WO−2のキメラ組換えFab(rFab)及び単鎖断片(scFv)の使用を記載している。操作された断片は、(i)アミロイドの線維化を防ぎ、(ii)事前に形成されたAβ1〜42小線維をバラバラにし、(iii)IgG全長分子と同様に効率的に試験管内でAβ1〜42が介在する神経毒性を阻害することができた。エフェクター機能を欠く小型Fab及びscFvの操作された抗体形式を使用することの認識される利点には、脳血管関門のさらに効率的な通過及び炎症性の副反応を誘発するリスクを最小にするることが挙げられる。その上、scFv及び単一ドメイン抗体は完全長抗体の結合特異性保持することに加えて、その標的の折り畳み、相互作用、修飾又は細胞内の局在の変化の可能性を伴って、単一遺伝子として及び哺乳類細胞にて細胞内で細胞内抗体として発現することができる;たとえば、概説については、Miller及びMesser,Molecular Therapy、12(2005),394−401を参照のこと。
(a)本発明の抗体、そのTDP−43結合断片又はそのいずれかの実質的に同一の結合特異性を有するTDP−43結合断片によって診断される対象からの試料にてTDP−43のレベルを評価することと;
(b)1以上の対照の対象におけるTDP−43のレベルを示す参照標準とTDP−43のレベルを比較することを含み、
TDP−43のレベルと参照標準との間での差異又は類似性は対象がTDP−43タンパク質症に罹っていることを示す。
行時間型(SELDI−TOF)、高速液体クロマトグラフィ(HPLC)、高速タンパク質液体クロマトグラフィ(FPLC)、多次元液体クロマトグラフィ(LC)とその後の直列質量分光分析(MS/MS)及びレーザーデンシトメトリーから選択される1以上の技法によってTDP−43を解析することを含む当該技術で既知の好適な方法によって評価することができる。一実施形態では、TDP−43の前記生体内画像化は、ポジトロン放出断層撮影(PET)、単一光子放出型断層撮影(SPECT)、近赤外線(NIR)光学画像撮影又は磁気共鳴画像撮影(MRI)を含む。
本明細書で採用されるもののような従来の方法の詳細な記載は引用された文献にて見つけることができる。以下で特に指示されない限り、TDP−43に特異的なB細胞の特定及び当該特異性を示すTDP−43抗体の分子クローニング並びにそれらの組換え発現及び機能的な性状分析は、それぞれその全体が参照によって本明細書に組み入れられるWO2008/081008として公開された国際出願PCT/EP2008/000053の実施例及び補足の方法の項で記載されたように実施されているし、実施することができる。
ELISA
96穴ハーフエリアマイクロプレート(Corning)を炭酸ELISA緩衝液(pH9.6)中のそれぞれ5μg/ml及び3.3μg/mlの濃度での
(a)組換えの完全長Hisタグ付きのヒトTDP−43(米国、Biogen Idec)又は
(b)残基409と410でリン酸化修飾したTDP−43のC末端ドメインの残基390〜414から成る合成ペプチド(Shafer−N,DK)のいずれかによって4℃にて一晩コーティングした。
それぞれアミノ酸1〜259、260〜277及び350〜366(英国、Abcam
plc)に対応するTDP−43タンパク質の断片の混合物で標準の96穴10−スポットMULTI−SPOTプレート(米国、Meso Scale Discovery)をコーティングした。10μg/mlの各ペプチドを用い、PBSにて調製した。3%BSAを含有するPBS/Tによって室温にて1時間、非特異的な結合部位をブロックし、その後、B細胞調整培地と共に室温にて1時間インキュベートした。プレートをPBS/Tで洗浄し、次いでSULFO−タグと複合体化した抗ヒトポリクローナル抗体(米国、Mesoscale Discovery)と共にインキュベートした。PBS/Tによる洗浄の後、SECTORイメージャー6000、米国、Mesoscale Discovery)を用いた電気化学発光測定によって結合抗体を検出した。
健常なヒト対象から記憶B細胞を含有する試料を得た。選択した記憶B細胞の培養物の生きているB細胞を回収し、mRNAを単離し、逆転写酵素(米国、Clontech)によってcDNAを調製した。次いでネストPCR法を用いて免疫グロブリン重鎖及び軽鎖の配列を得た。
6×His−タグを付けたTDP−43(pCGB026)のための発現ベクターを、pRSET A発現ベクター(Invitrogen)にTDP−43配列をクローニングすることによって生成した。pCGB026によってBL21Star(DE3)pLysS大腸菌細胞(Invitrogen)を形質転換した。LBブロスを含有する1Lの振盪フラスコにておよそ1ODまで37℃で増殖させた後、IPTGを0.5mMまで加え、細胞を18℃で一晩増殖させた。低速遠心によって細胞をペレットにし、上清を捨て、細胞ペレットを−20℃での保存のために凍結した。
ヒトのTDP−43ドメインのコーディング配列を完全長cDNA配列(Q13148,TARDBP_HUMAN)からPCRによって増幅し、発現ベクターpRSET−A(米国、Invitrogen)にクローニングした。4つの発現ベクター(His−huTDP−43ドメインI、His−huTDP−43ドメインII、His−huTDP−43ドメインIII及びHis−huTDP−43ドメインIV)はそれぞれ4つのTDP−43ドメイン:N−末端ドメイン(配列番号94のアミノ酸残基2〜106)、RNA結合ドメイン1(配列番号94のアミノ酸残基99〜204)、RNA結合ドメイン2(配列番号94のアミノ酸残基183〜273)及びGlycine−リッチドメイン(配列番号94のアミノ酸残基258〜414)をコードしている。T7プロモータの制御下でTDP−43ドメインをコードするcDNAを含むDNA構築物を用いて、たとえば、BL21(DE3)(米国、New England Biolabs)のような適当な大腸菌株を形質転換し、0.5mMのイソプロピル−β−D−チオガラクトピラノシド(IPTG)の添加によって15mLの細胞培養物の発現を誘導した。37℃での4時間のインキュベート後、細胞を回収し、1mLの100mMのKCl、50mMのHEPES、2mMのEGTA、1mMのMgCl2、1mMのジチオスレイトール、0.1mMのPMSF、10%グリセロール及び0.1mg/mlのリゾチーム、pH7.5に再浮遊し、その後超音波処理した。4℃にて9000rpmで45分間の遠心の後、可溶性分画及び不溶性分画を回収した。同様に、対照の大腸菌から9000gの上清を回収した。必要であれば(TDP−43ドメインIV)、1mLの8M尿素、20mMのTris、200mMのKCl及び1mMのβ−メルカプトエタノールにて不溶性分画を可溶化した。可溶性分画及び可溶化分画をNi−NTA Superflowカラム(米国、Qiagen)に負荷し、His−TDP−43ドメインを製造元のプロトコールに従って精製した。組換えタンパク質の純度の等級をSDS−PAGE及びクマシー染色によって推定した。精製したタンパク質の濃度は280nmの吸収測定によって決定した。
96穴マイクロプレート(Corning)を、炭酸ELISAコーティング緩衝液(pH9.6)にてそれぞれ6.6μg/ml又は3.3μg/mlの濃度に希釈されたヒト完全長TDP−43、TDP−43ドメインI(配列番号94のアミノ酸残基2〜106)、TDP−43ドメインII(配列番号94のアミノ酸残基99〜204)、TDP−43ドメインIII(配列番号94のアミノ酸残基183〜273)、TDP−43ドメインIV(配列番号94のアミノ酸残基258〜414)又はTDP−43のドメインのC末端の残基390〜414(配列番号94を参照)を占め、残基409/410にてリン酸化の修飾を持つ合成ペプチド(Schafer−N、DK)によって4℃にて一晩コーティングした。2%BSA(スイス、ブックスのSigma)を含有するPBS/Tによって室温にて1時間、非特異的な結合部位をブロックした。抗体を室温にて1時間インキュベートした。HRと複合体化されたロバ抗ヒトIgGγ特異抗体(米国、Jackson ImmunoResearch)又はHRPと複合体化されたヤギ抗マウスIgG(H+L)特異的二次抗体(米国、Jackson ImmunoResearch)のいずれかを用いて結合を測定し、その後、標準の比色アッセイにてHRPの活性を測定した。
組換えのヒト完全長TDP−43、TDP−43ドメインI(配列番号94のアミノ酸残基2〜106)、TDP−43ドメインII(配列番号94のアミノ酸残基99〜204)、TDP−43ドメインIII(配列番号94のアミノ酸残基183〜273)、TDP−43ドメインIV(配列番号94のアミノ酸残基258〜414)のそれぞれ300ngをSDS-PAGE(NuPAGE、12%Bis−Tris Gel;スイスバ
ーゼルのInvitrogen)によって分離し、その後、ニトロセルロース膜に電気ブロットした。2%BSA(スイス、ブックスのSigma)を含有するPBS/Tによって室温にて1時間、非特異的な結合部位をブロックした。ブロットを一次抗体(10nM)と共に一晩インキュベートし、その後、HRPと複合体化されたロバ抗ヒトIgGγ特異的二次抗体(米国、Jackson ImmunoResearch)又はHRPと複合体化されたヤギ抗マウスIgG(H+L)特異的二次抗体(米国、Jackson ImmunoResearch)のいずれかと共にインキュベートした。ECL及びImageQuant350検出(スイス、オテルフィンゲンのGE Healthcare)によってブロットを発色させた。
抗体で処理したTDP−43タンパク質症のマウスモデルにおける作業記憶の改善は、2試行Y迷路試験を用いて調べることができる(たとえば、その全体が参照によって本明細書に組み入れられるPennanen, Genes Brain Behav. 5 (2006), 369-79)。迷路
の3つのアームは長さ22cm、幅5cm及び深さ15cmである。迷路を取り囲む黒色カーテン上に白黒の抽象的なヒントがある。実験は暗相の間、6ルクスの周囲の光レベルで実施される。各実験は訓練期間と観察期間を含む。訓練期間の間、マウスは3つのアームのうち2つ(出発アーム及び第2のアーム)に割り振られ、4分間自由に探索できるが、第3のアーム(新規のアーム)にアクセスすることはできない。次いでマウスを迷路から取り出し、1.5〜5分間、保持ケージに閉じ込める一方で、70%アルコールで迷路を十分に清掃し、嗅覚のヒントを除去する。次いで観察のためにマウスを迷路に戻し、4分間3つのアームすべてにアクセス可能にする。各アームに入った順、入った回数、各アームで費やした時間を記録する。それから、他の2つのアーム(出発アーム及び第2のアーム)で費やした時間の平均に対する新規の第3のアームで費やした時間の比を算出し、タウオパシーのマウスモデル及び相当する野生型マウスにおける異なる処理群の間で比較する。齧歯類は通常、以前訪れたものに戻るよりも迷路の新しいアームを探索することを好む。抗体の効果は、その疾病に関連する作業記憶の損傷のために無処理のマウスの非弁別行動と比べた処理したTDP−43タンパク質症モデルマウスによるこの嗜好を取り戻すことに関してモニターすることができる。従って、1に近い比は損傷された作業記憶を示す。高い比ほど、さらに良好な作業記憶を示す。TDP−43タンパク質症モデルマウスにおける損傷された作業記憶は、ヒトTDP−43の過剰発現から生じるTDP−43の病理によると考えられる。従って、抗TDP−43抗体で処置したマウスで見られる対照マウスよりも有意に高い比は、抗TDP−43抗体がTDP−43の病理に対して治療効果を有することを示すであろう。
マウスが最も活発である暗相の始まりでマウスを調べる。ポールは登るのを円滑にするように布で覆われた長さ50cm及び幅1cmの木製の棒で構成される。ポールの基はマウスのホームケージの中に置かれる。マウスをポールの上端に載せ、下方を向く時間及びホームケージに降りてくる時間を30分間隔で5試行にわたって記録する。最良の成績の試行を解析する。
マウスが最も活発である暗相の始まりでマウスを調べる。試験は薄明かり(40ルクス)で行う。高架式十字迷路は2つの開放アームと2つの閉鎖アーム(アームの長さ:30cm、幅:5cm)から成る。開放アームは小さな1cmの縁を有し、閉鎖アームには15cmの壁が接する。試験の開始時、マウスを開放アームに面して高架式十字迷路の中央に置く。マウスが5分間迷路を探索する間、ビデオで追跡する。開放アーム及び閉鎖アームで費やした時間及びカバーされる距離を測定し、解析する。

組換えの完全長TDP−43、TDP−43ドメインI(配列番号94のアミノ酸残基2〜106)、TDP−43ドメインII(配列番号94のアミノ酸残基99〜204)、TDP−43ドメインIII(配列番号94のアミノ酸残基183〜273)、TDP−43ドメインIV(配列番号94のアミノ酸残基258〜414)をSDS−PAGEによって分離した。市販の TDP−43特異抗体TARDBP(M01)、クローン2E2−D3(台湾のAbnova)をヒトTDP−43検出の陽性対照として使用した一方で、抗ヒトIgGのFcγ特異抗体を陰性対照として使用した。ヒト由来の抗体の特異的なTDP−43ドメインへの結合はウエスタンブロット解析によって測定した。
50%効果濃度(EC50)の決定
ヒトTDP−43に対するヒト由来のTDP−43特異抗体の50%効果濃度(EC50)を決定し、標的特異性を評価するために、炭酸ELISAコーティング緩衝液(pH9.6)にて5μg/mlの濃度に希釈した組換え完全長のTDP−43(米国、Biogen Idec、大腸菌抽出物)及びBSA(スイス、ブックスのSigma)によって96穴マイクロプレートを4℃にて一晩コーティングした。或いは、炭酸ELISAコーティング緩衝液(pH9.6)にて3.3μg/mlの濃度に希釈した残基409/410にてリン酸化の修飾を持つTDP−43のC末端ドメインの残基390〜414に相当する合成ペプチド(Schafer−N,DK)及びBSA(スイス、ブックスのSigma)によって96穴ハーフエリアマイクロプレートを4℃にて一晩コーティングした。2%BSA(スイス、ブックスのSigma)を含有するPBSによって室温にて1時間、非特異的な結合部位をブロックした。TDP−43特異抗体を指示された濃度に希釈し、室温にて1時間インキュベートした。HRPと複合体化されたロバ抗ヒトIgGγ特異的二次抗体(米国、Jackson ImmunoResearch)を用い、その後、標準の比色アッセイにてHRPの活性を測定することによって結合を測定した。EC50値はGraphPad Prism(米国、サンディエゴ)を用いた非線形回帰によって推定した。
の結合は認められなかった。抗体NI−205.3F10、NI−205.8A2、NI−205.15F12、NI−205.113C4、NI−205.25F3、及びNI−205.87E7はヒトTDP−43に結合したが、ホスホ-TDP−43のC末端ペ
プチドには結合しなかった。これらの抗体のヒトTDP−43タンパク質に対するEC50値は1〜18nMの範囲だった。NI−205.21G1は4.1nMのEC50で完全長TDP−43に結合し、49.5MのEC50での、より低い親和性でホスホ-TD
P−43のC末端ペプチドを認識した。抗体NI−205.68G5及びNI−205.20A1はそれぞれ、16.9及び15.8nMのEC50でホスホ-TDP−43のC
末端ペプチドへの優先的な又は専らの結合を示し、TDP−43の409位でのセリン及び/又は410位でのセリンのリン酸化がNI−205.68G5及びNI−205.20A1による結合に必要とされることを示唆している。
重なり合うペプチドの走査を用いて、ヒト由来のTDP−43特異抗体によって認識されるヒトTDP−43タンパク質の中でのエピトープをマッピングした。まとめてヒトTDP−43(Q13148,TARDBP_HUMAN)の配列全体を表す11アミノ酸が重なり合う配列の101の直鎖15量体を伴ったPepscan膜(PepSpots,ドイツ、ベルリンのJPT Peptide Technologies)。ニトロセルロース膜上でペプチドのスポットを作り、次いでメタノール中で5分間活性化し、その後、TBSにて10分間室温で洗浄した。Roti(登録商標)−Block(ドイツ、カールスルーエのCarl Roth GmbH Co.KG)によって室温にて2時間、非特異的な結合部位をブロックした。ヒト由来のTDP−43特異抗体(1μg/ml)をRoti(登録商標)−Blockにて室温で3時間インキュベートした。HRP抱合のロバ抗ヒトIgGγ特異的な二次抗体(米国、Jackson ImmunoResearch)を用いて一次抗体の結合を測定した。ECL及びImageQuant350検出(スイス、オテルフィンゲンのGE Healthcare)によってブロットを発色させた。
表4は、PepSpotを用いて特定された様々なヒト由来のTDP−43特異抗体について特定された結合エピトープを要約する。
TDP−43抗体結合の能力の検証のために、ヒトALS患者又はヒトFTLD患者の脊髄及び脳の切片を使用した。TDP−43病態に対する抗体の結合を免疫組織化学染色によって評価した。ヒト組換えTDP−43抗体の結合はヒトFTLD−TDP−43症例の組織(10人の患者症例、7人の対照症例)にて特徴付けた。免疫組織化学は、5μmのパラフィン包埋切片にて実施し、EDTAに基づいたエピトープ検索の使用を含み、その後、、それ以外は標準の、DAB(Pierce)を伴ったEliteABCキット(Vector Laboratories)による免疫ペルオキシダーゼ手順を実施した。以下の一次抗体を使用した:陽性対照としてヒトTDP−43に対して産生させたマウスモノクローナル抗体2E2−D3(Abnova);ここ/上記で記載される組換えヒトTDP−43抗体NI205.3F10、NI205.51C1、NI205.21G2、NI205.8A2、NI205.15F12、NI205.25F3、NI205.87E7、NI205.21G1、NI205.68G5、NI205.20A1。ヘマトキシリンで切片を対比染色して細胞核を示した。
TDP−43抗体の前臨床の検証のための実験をTDP−43タンパク質症のマウスモデルにて実施する。末梢注射又は浸透圧ミニポンプを介した脳室内注入によってヒトTDP−43抗体を投与する。血液及びCSF試料の分析、及び体重、一般的な臨床的印象、及び、たとえば、オープンフィールド、Y−迷路、高架式十字迷路、新規の目的認識、握力、足握力持久力、ポール試験、挑戦的なビームウォーク、ロータロッド又はさもなければ当該技術で既知のものを介して見られるような運動及び認知の損傷の兆候の分析によって治療効果をモニターする。
43の25kDaのC末端断片を選択的に発現するトランスジェニックマウス(Caccamo
et al., Am J Pathol. 180(1):293-302 (2012));前脳にて野生型のヒトTDP−43(hTDP−43)を条件付きで発現するトランスジェニックマウス(Cannon et
al., Acta Neuropathol. 123(6):807-23 (2012));及びヒトVCP/p97の野
生型及び疾患を生じる型の普遍的な発現を伴うトランスジェニックマウス(Custer et al., Hum Mol Genet. 19(9):1741-55 (2010))にて実施される。別の実施形態では
、TDP−43抗体を検証する実験は、野生型TDP−43、核局在シグナルを欠損するTDP−43、又は配列番号94の残基220〜414を含むTDP−43の切り詰められたC末端断片をコードするAAVベクターで形質移入されたマウスで実施される。Tatomら、Mol.Ther.17(2009),607−613。
ヒトTDP−43に対するヒト由来のTDP−43特異抗体の50%効果濃度(EC50)及びその標的特異性を実質的に上記実施例2に記載されたように測定した。手短には、炭酸ELISAコーティング緩衝液(pH9.6)にて5μg/mlの濃度に希釈した組換え完全長のTDP−43(米国、Biogen Idec、大腸菌抽出物)及びBSA(スイス、ブックスのSigma)によって96穴マイクロプレートを4℃にて一晩コーティングした。或いは、炭酸ELISAコーティング緩衝液(pH9.6)にて3.3μg/mlの濃度に希釈した残基409/410にてリン酸化の修飾を持つTDP−43のC末端ドメインの残基390〜414に相当する合成ペプチド(Schafer−N,DK)及びBSA(スイス、ブックスのSigma)によって96穴ハーフエリアマイクロプレートを4℃にて一晩コーティングした。2%BSA(スイス、ブックスのSigma)を含有するPBSによって室温にて1時間、非特異的な結合部位をブロックした。ヒトTDP−43特異抗体を指示された濃度に希釈し、室温にて1時間インキュベートした。HRPと複合体化されたロバ抗ヒトIgGγ特異的二次抗体(米国、Jackson ImmunoResearch)を用い、その後、標準の比色アッセイにてHRPの活性を測定することによって結合を測定した。EC50値はGraphPad Prismソフトウエア(米国、サンディエゴ)を用いた非線形回帰によって推定した。41D1、21G1、31D2及び8F8の抗体で得られた例となる滴定曲線を図4に示す。
DP−43C末端ペプチドへの結合は認められなかった。抗体NI−205.1A9、NI−205.3F10、NI−205.14W3、NI−205.98H6、NI−205.44B2、NI−205.9E12A、NI−205.8A2、NI−205.15F12、NI−205.10D3、NI−205.38H2、NI−205.29E11、NI−205.9E12D、NI−205.31C11、NI−205.113C4、NI−205.25.25F3、NI−205.10H7、NI−205.8C10、及びNI−205.87E7は、ナノモルのEC50でヒトTDP−43に結合したが、ホスホ-TDP−43C末端ペプチドには結合しなかった。これらの抗体についてのEC5
0値は1〜18nMの範囲であった(表6を参照)。抗体NI−205.21G1(図4B及びF)は4.1nMのEC50で完全長のTDP−43に結合し、49.5nMのEC50での低親和性でホスホ-TDP−43C末端ペプチドを認識した。抗体NI−20
5.31D2(図4C及びG)、NI−205.14H5、NI−205.36D5、NI−205.19G5及びNI−205.68G5は、0.7〜17nMの範囲であるEC50値でホスホ-TDP−43C末端ペプチドに優先的に結合することを示した(表6
を参照)。対照的に、抗体NI−205.8F8、NI−205.8F8(図4D及びH)及びNI−205.20A1は、それぞれ5nM、7nM及び16nMのナノモルEC50での高親和性でヒトのホスホ-TDP−43C末端ペプチドに専ら結合したというこ
とは、セリン409及び/又はセリン410のリン酸化が結合に必要とされるという考えに矛盾しなかった。
直接ELISA
実施例1に記載したように直接ELISAを実質的に実施した。手短には、アミノ酸2〜106(ドメインI)、99〜204(ドメインII)、183〜273(ドメインIII)、258〜414(ドメインIV)及び完全長TDP−43(2−414)を含むTDP−43(配列番号94)の断片を6.6μg/mlの等しいコーティング濃度でELISAプレートにコーティングした。ヒト由来の抗体の結合を直接ELISAによって測定した。得られた結果の例を図5に示す。抗体NI−205.41D1(図5A)、NI−205.14W3(図5C)、NI−205.44B2(図5E)、NI−205.10D3(図5G)、及びNI−205.10H7(図5L)は、TDP−43ドメインIV(aa258〜414)に特異的に結合した。抗体NI−NI−205.98H6(図5D)は、TDP−43ドメインIII(aa183〜273)に特異的に結合した。抗体NI−205.1A9(図5B)、NI−205.38H2(図5H)、及びNI−205.31C11(図5K)は、TDP−43ドメインII(aa99〜204)を特異的に認識したが、抗体NI−205.9E12A(図5F)、NI−205.29E11(図5I)、NI−205.9E12D(図5J)、及びNI−205.8C10(図5M)は、TDP−43ドメインI(aa2〜106)に結合した。ヒト由来のTDP−43特異抗体はすべて完全長のヒトTDP−43を認識した。異なる組換えTDP−43ドメインのコーティング効率性を制御するために、完全長TDP−43及び特定のTDP−43ドメインに結合する市販の抗体用いた:(Fig.5W):i)Ab50930(英国、Abcam)、TDP−43ドメインI;ii)TARDBPモノクローナル抗体(M01)、クローン2E2−D3(台湾のAbnova,Abnova23435)、TDP−43ドメインIII及びiii)Ab82695(英国、Abcam),TDP−43ドメインIV。対照抗体は完全長TDP−43及びその特定のTDP−43ドメインに結合した。
組換えの完全長TDP−43、TDP−43ドメインI(配列番号94のアミノ酸残基2〜106)、TDP−43ドメインII(配列番号94のアミノ酸残基99〜204)、TDP−43ドメインIII(配列番号94のアミノ酸残基183〜273)、TDP−43ドメインIV(配列番号94のアミノ酸残基258〜414)をSDS−PAGEによって分離した。ヒト由来の抗体の特定のTDP−43ドメインへの結合をウエスタンブロット解析によって測定した。
排他的な結合を示す2つの抗体であるNI−205.68G5(図6N)及びNI−205.20A1(図6O)は完全長TDP−43又はその断片を認識しなかった。市販のTDP−43に特異的な抗体2E2−D3(台湾、AbnovaのAbnova23435)(図6P)をヒトTDP−43の検出の陽性対照として用いたのに対して、抗ヒトIgGγ特異的な抗体(図6Q)を陰性対照として用いた。
ヒトTDP−43のタンパク質配列を網羅する個々のペプチド間で11のアミノ酸が重なり合う101の直鎖15量体のペプチドを伴ったPepscan膜(PepSpots,ドイツ、ベルリンのJPT Peptide Technologies)を用いてヒトTDP−43タンパク質の範囲内でヒトに由来するTDP−43に特異的な抗体によって認識されるエピトープをマッピングした。Pepscanマッピングは実質的に実施例3に記載されたように実施した。手短には、ニトロセルロース膜上でペプチドのスポットを作り、次いでメタノール中で5分間活性化し、その後、TBSにて10分間室温で洗浄した。表7はPepSpotを用いて特定された異なるヒト由来のTDP−43に特異的な抗体についての結合エピトープを要約する。
10μg/mlの等しいコーティング濃度で完全長TDP−43(2〜414)及びアミノ酸258〜414(ドメインIV)、258〜384、258〜375、258〜362、258〜353、258〜319、317〜414及び340〜414を含むTDP−43C末端断片をELISAプレートにコーティングした。特定のTDP−43へのNI−205.41D1抗体の結合は直接ELISAによって測定した。得られたデータの例を図7Aに示す。NI−205.41D1は、断片258〜319及び340〜414を除いてすべての組換え断片に結合し、NI−205.41D1の結合エピトープはTDP−43のC末端領域317〜353にあることを示した。NI−205.41D1は完全長TDP−43に結合した。
純粋な完全長のTDP−43タンパク質は凝集する天然の傾向を有する。従って、標準の精製条件下ではほんのわずかな量の完全長のTDP−43しか回収されなかった。従って、我々は、天然のタンパク質構造を保存しながらタンパク質の凝集を防ぐことが知られるKSCN及びアルギニン、穏やかなカオトロピック剤を用いて大量の機能的な6×His−SUMOタグを付けた完全長のヒトTDP−43を大腸菌から単離するための組換え発現及び精製の戦略を開発した。
Technologies)を5nMの濃度で、示したようなインキュベート緩衝液にてTDP−43(0〜20nMの最終濃度)と共に25℃で30分間インキュベートした。645nmの励起及び665nmの放射にて96穴プレート形式での各濃度のTDP−43について、プレートリーダー蛍光計Envision(PerkinElmer)によって蛍光偏光を測定した。Sigmaplot(Systat Software社)を用いた密接な結合のための二次方程式(Morrison方程式)を用いてKdsを算出した。RNA/TDP−43結合の定比性を決定するために、合計100nMのRNA濃度(以前決定したKd値よりも>7×高い)について同じ配列の95nMの標識していないRNAの添加と共に、同じ蛍光偏光の実験設定を用いた。定比性は、一方を部分的に結合した状態のデータ点を用いて適合させ、他方を完全に結合した状態のデータ点を用いて適合させた2本の直線の切片を測定することによって算出した。
ヒトの皮質、海馬及び脊髄のFTLD−U及び対照の組織をIDIBAPS Biobank(バルセロナのBanc de Teixits Neurologics)から入手した。免疫組織化学を、EDTAに基づいたエピトープ検索を用いて5μmのパラフィン包埋切片にて実施し、その後、それ以外では標準のDAB(Pierce)を伴ったEliteABCキット(Vector Laboratories)による免疫組織化学手順を実施した。50nM濃度での本発明のヒトTDP−43抗体を用いて免疫組織化学を実施した。ヒトTDP−43に対するマウスモノクローナル抗体2E2(Abnova)、TDP−43 p409/p410に対して作ったウサギポリクローナル抗体p409/p410(CosmoBio)、及びTDP−43 p409/p410に対して作った対して作ったウサギポリクローナル抗体p409/p410(CosmoBio)を用いて対照染色を行った。
ける細胞質のTDP−43(図11D)、顆粒細胞における核及び細胞質の蓄積(図11E)、並びにジストロフィ性神経突起におけるTDP−43(図11F)を認識した。第2のリン酸化特異抗体p409/p410(CMDSKS(p)S(p)GWGM(配列番号325)に対して作った; Hasegawa et al., Ann Neurol, 64(1):60-70 (2008))は、錐体細胞(図11G)、顆粒細胞(図11H)並びにジストロフィ性神経突起(図11I)における核及び細胞質でのTDP−43の蓄積に結合した。
1日目と4日目に30mg/kgの抗ヒトTDP−43抗体又は等量のPBSをTDP−43_G348Cトランスジェニックマウス(Swarup et al., Brain 134 (2011),
2610-2626)に腹腔内注射する。5日目に麻酔下にて1単位/mlのヘパリンを含有するPBSでマウスを潅流する。血液、脳及び脊髄を分析のために採取する。脳の右半球を−80℃で凍結し、脳の左半球及び脊髄を10%中性ホルマリンにて4℃で2日間後固定し、その後、パラフィンブロックに包埋し、切片にする。血漿はアリコートで−80℃にて保存する。
Roth GmbH + Co. T865.1)によって対比染色する。脱水ステップの後、スライドをカバーグラスで覆い、その後、Olympus dotSlide 2.1視覚顕微鏡システムで走査する。抗体処置した動物で見られるが、対照動物では見られないニューロンの抗ヒトTDP−43染色は、ヒト抗TDP−43抗体がニューロンに侵入することを示す。
10mg/kg、3mg/kgの抗体溶液又は等量のPBS対照をTDP−43_G348Cトランスジェニックマウス(Swarup et al., Brain 134 (2011), 2610-2626)に腹腔内注射する。各処置群は20〜25匹のマウスを有する。処置は週1回で26週間実施する。或いは、処理は週2回で13週間実施する。2週間ごとに体重をモニターする。処置の終了時、麻酔下でマウスを潅流する。脳、脊髄及び血液を採取する。脳半分及び脊髄を10%ホルマリンにて3日間後固定し、その後パラフィンブロックに包埋する。これらの組織ブロックから切り出した4〜6μm厚さの切片を免疫組織化学的検討に使用する。脳の他の半分を秤量し、生化学分析のために−80℃で深く凍結する。
Claims (26)
- 重鎖の可変ドメイン(VH)及び軽鎖の可変ドメイン(VL)を含む、抗TAR−DNA結合タンパク質43kDa(TDP−43)抗体又はそのTDP−43結合断片であって、前記VHが、相補性決定領域(CDR)であるVH−CDR1、VH−CDR2及びVH−CDR3を含み、前記VLが、CDRであるVL−CDR1、VL−CDR2及びVL−CDR3を含み、ここで、前記VH−CDR1、VH−CDR2及びVH−CDR3が、それぞれ、配列番号260、261及び262で示されるアミノ酸配列を含み、前記VL−CDR1、VL−CDR2及びVL−CDR3が、それぞれ、配列番号264、265及び266で示されるアミノ酸配列を含む、抗TDP−43抗体又はそのTDP−43結合断片。
- 重鎖の可変ドメイン(VH)及び軽鎖の可変ドメイン(VL)を含む抗TAR−DNA結合タンパク質43kDa(TDP−43)抗体又はそのTDP−43結合断片であって、ここで、前記VHが、配列番号259で示されるアミノ酸配列からなり、前記VLが、配列番号263で示されるアミノ酸配列からなる、抗TDP−43抗体又はそのTDP−43結合断片。
- ヒト抗体である、請求項1又は2に記載の抗TDP−43抗体又はそのTDP−43結合断片。
- キメラ抗体である、請求項1又は2に記載の抗TDP−43抗体又はそのTDP−43結合断片。
- TDP−43結合断片である、請求項1又は2に記載の抗TDP−43抗体又はそのTDP−43結合断片。
- 抗体TDP−43結合断片が、単鎖Fv断片(scFv)、F(ab’)断片、F(ab)断片またはF(ab’)2断片である、請求項5に記載の抗TDP−43抗体又はそのTDP−43結合断片。
- 請求項1または請求項2に記載の抗TDP−43抗体又はそのTDP−43結合断片をコードするヌクレオチド配列を含む単離されたポリヌクレオチド。
- 重鎖の可変ドメイン(VH)をコードするヌクレオチド配列及び軽鎖の可変ドメイン(VL)をコードするヌクレオチド配列を含む、抗TAR−DNA結合タンパク質43kDa(TDP−43)抗体又はそのTDP−43結合断片をコードする単離されたポリヌクレオチドであって、前記VHをコードするヌクレオチド配列が配列番号308で示される核酸配列からなり、前記VLをコードするヌクレオチド配列が配列番号309で示される核酸配列からなる、単離されたポリヌクレオチド。
- 請求項7または請求項8に記載のポリヌクレオチドを含む単離されたベクター。

- 抗TDP−43抗体又はそのTDP−43結合断片を調製するための方法であって、
(a)請求項10に記載の宿主細胞を培養する工程、および

- ヒトの体においてTDP−43に対する治療剤および/または診断剤を生体内検出または標的化するための組成物であって、請求項1〜6又は12〜14のいずれか1項に記載の抗TDP−43抗体又はそのTDP−43結合断片を含む、組成物。
- 前記生体検出が、ポジトロン放出断層撮影(PET)、単一光子放出型断層撮影(SPECT)、近赤外線(NIR)光学画像撮影又は磁気共鳴画像撮影(MRI)を含む、請求項21に記載の組成物。
- TDP−43タンパク質症の進行を診断またはモニターするのに有用なキットであって、請求項1〜6又は12〜14のいずれか1項に記載の少なくとも1つの抗TDP−43抗体又はそのTDP−43結合断片を含むことを特徴とする、キット。
- 前記TDP−43タンパク質症が、アルツハイマー病、ALS、FTLD、およびパーキンソン病から成る群から選択される、請求項23に記載のキット。
- 前記TDP−43タンパク質症が、嗜銀性顆粒痴呆、アルツハイマー病、筋委縮性側索硬化症(ALS)、グアムのALS/パーキンソン型認知症複合体、大脳皮質基底核変性症、レビー小体認知症、ハンチントン病、レビー小体病、運動ニューロン病、前頭側頭葉変性症(FTLD)、前頭側頭認知症、ユビキチン陽性の封入体を伴った前頭側頭葉変性症、海馬硬化症、封入体ミオパシー、封入体筋炎、パーキンソン病、パーキンソン病認知症、Kii半島におけるパーキンソン/認知症複合体、及びピック病から成る群から選択される、請求項18に記載の方法。
- 前記TDP−43タンパク質症が、アルツハイマー病、ALS、FTLD、およびパーキンソン病から成る群から選択される、請求項18に記載の方法。
