Description
蛍光を発する分子(蛍光体)からの光放出のプロセスはヤブロンスキー図で説明することができる(図1Aを参照)。図1Aは、分子の励起状態からの様々な減衰経路を示すヤブロンスキー図を示す。図の下部に、エタノール中のヘマトポルフィリンからの蛍光スペクトルが示される。省略形は、Sn:一重項状態、Tn:三重項状態、Abs:吸収、Sc:散乱、IC:内部変換、F:蛍光、IX:項間交差、P:燐光、A:他の分子への移動である。さらにいくつかのプロセスの近似の時間スケールが寿命(LT)として図1Aに右下に示され、さらにτと表記される。
ポイントモニタ装置とは対照的に、蛍光イメージングシステムは多数のポイントの蛍光信号を検出することができる。したがって、注目する区域の2次元画像が生成される。典型的なシステムはチューナブルフィルタと共にカメラを含む(図5aを参照)。透照での同様のセットアップが図5bに概略的に示される。チューナブルフィルタにより所望の検出波長を容易に選択することができ、約20nm幅のスペクトル解像度を達成することができる。
蛍光イメージングのうちの特に興味あるサブセクションは本実施形態の非線形蛍光体を使用するものである。本用途の関連において、「非線形マーカー」はルミネセンスマーカーであり、マーカーのルミネセンス(L)は励起光(E)の光束に線形には依存しない。したがって、非線形マーカーは、L=k×E^xに従うルミネセンスを有し、ここで、x>1であり、kは正の定数である。非線形マーカーは、さらにL=k×E^x+b、L=k(E)×E^x+b、L=k(E)×E^x+b(E)、またはL=k×E^x+b(E)の関係に従うルミネセンスを有することがあり、ここで、kおよびbは一定であるか、または励起光(E)の局所場に依存する(すなわちk(E)およびb(E))材料定数であるしたがって、従来のルミネセンスイメージングと比較して、非線形マーカー(または蛍光体)は励起に1つよりも多くの光子を必要とすることがある。これは励起体積を強烈に減少させ、より局在化した励起ポイントを与える。このようにして、以下で実証されるように、ルミネセンスイメージングのコントラストおよび解像度が改善される。さらに詳細には、吸収および散乱媒体のルミネセンスイメージングにおける散乱光のコントラストおよび解像度が改善される。本発明の実施形態はこの効果を利用する。
蛍光断層撮影
物体の表面から放出された蛍光の平面画像はいくつかの態様に関する情報を含む。分光の特徴は蛍光体のタイプをもたらし、強度は蛍光体の濃度に関連する。
・励起光の吸収および散乱。蛍光体は光を放出するために励起されなければならず、したがって、励起光は蛍光体の場所に達しなければならない。
・励起光源の位置。同じ励起光とすると、蛍光体の近くに位置づけられた光源は、その光源から遠くに位置づけられた光源と比較してより多くの蛍光体を励起することになる。
・蛍光体の位置およびサイズ。ここで、蛍光体は、内部構造をもつ、すなわち輪郭のはっきりした、蛍光マーカーの均一な分布を含む領域として扱われる。サイズおよび位置に応じて、放出された蛍光は境界上において異なる外観を有することになる。
・放出光の吸収および散乱。放出は、組織を通って伝搬するとき減衰させられる。通常、放出の光学的性質は励起光のものと同じではない。
・放出光の収集位置。収集された強度は、検出された場所(境界上の)に依存する。これは、放出部位(蛍光体の位置)からの伝搬経路、および収集部位(境界)の不均等に起因する。
小動物のイメージング
今日、医学的診断用にヒトの患者に使用するのにインドシアニン(ICG)のみがFDA承認されているが、小動物のイメージングでは可能性のある蛍光体は多数ある。これは、細胞の生物医学的現象のイメージングに蛍光を利用する様々な顕微鏡的技法の使用によって開始された過去何年にもわたるプローブ開発における研究の加速の結果である。
これまでの主な用途はICGまたはICGの誘導体を使用する乳癌診断である。蛍光タンパク質は、人体に用いる上での選択肢とならないのは明らかであり、したがって、蛍光体イメージングは非特異的分子プローブを機能化にすることによって達成されることになる。
例えばアップコンバートナノ結晶における放出蛍光の二次依存により、蛍光断層撮影は改善される。
組織光学の分野では、組織との光相互作用が研究される。光学的に、生体組織は、水よりわずかに高い屈折率をもつ不均質で吸収性の媒体である。光が組織と相互作用すると、多数の散乱および吸収事象が生じると予想され、これらの事象の可能性は高度に波長依存である。組織は高濃度の水を有するので、水からの吸収が低い波長領域からの光を使用するのが有利であり、これは使用可能波長に究極的制限を強いることになる。しかし、いくつかの実施形態におけるように経皮的非侵襲性用途では、光は肌を透過する必要があり、それは使用可能波長にさらなる制約を課することになる。
外因性蛍光体のいくつかの例は、蛍光タンパク質(FP)、NIR色素(ND)、量子ドット(QD)、または光感作物質(PS)がある。
非線形蛍光体の例はナノ粒子(NP)であり、以下でより詳細に説明される。
アップコンバージョンは、2つ以上の光子が吸収され、入射光子よりも高いエネルギーの光子が解放されるときに生じる非線形プロセスである。
アップコンバートナノ結晶は、近赤外線(NIR)励起時に反ストークスシフト光を効率的に放出する独特の性質のために生物医学的イメージング用途における蛍光体として本明細書で開示される。これは、自己蛍光が存在しない領域の蛍光信号の検出を可能にする。
アップコンバートナノ粒子の機能化は、全体がすべての目的のために本明細書に組み込まれるX. Gao等、In vivo cancer targeting and imaging with semiconductor quantum dots、Nature Biotechnology、22、8:969〜976、2004に記載されているような量子ドットの機能化と同様の方法で行うことができる。Gao等では、アップコンバート希土類元素ドープナノ粒子に適用可能である方法が記載されている。
ルミネセンスイメージングにおける非線形マーカーの適用可能性を確認するために、アップコンバートナノ結晶が実験セットアップで使用された。生体内用途で蛍光体として使用するための妥当性を実証するために、2つの実験が行われた。
エピ蛍光セットアップが本実験で使用された。ファントムの光学的性質は、小動物で見いだされるものの範囲内にある660nmでμ’s=6.5cm−1およびμa=0.44cm−1となるように選ばれた。
非線形蛍光体および従来の蛍光体の使用によるFMTのシミュレーションが、図5bに示されるような透過蛍光セットアップで行われた。シミュレートされた組織ファントムは半径43mmの半無限円柱(508)としてモデル化された。光学的性質はλ=660nmでμ’s=10cm−1およびμa=0.4cm−1であり、16個の均一間隔の光源−検出器ポイント(509)が形状の1つの平面のまわりにあった。蛍光体は、図5bに示されるようにファントムの全体を通って延びる棒のように密に一緒にして配置された。
図9は、線形蛍光体(902)と非線形蛍光体(903)との間の断層撮影再構成の比較を示す。図9の図は二次蛍光体の一例として提示されている。(この場合二次であるように選ばれた)
さらに、本開示は、アップコンバートナノ粒子の独特な非線形パワー依存を利用して、同時に2つのビームで励起することを含むことによってラスター走査セットアップの情報量をさらに増加させるための蛍光拡散光学断層撮影の方法を実証する。情報の増加がより正確な再構成をもたらすことが見いだされた。

Claims (15)
- 拡散ルミネセンス分子イメージングによって散乱媒体中の領域を画像化する方法であって、前記領域が前記散乱媒体中でマーカー位置に配置された少なくとも1つのルミネセンスマーカーを含み、前記ルミネセンスマーカーがアップコンバートナノ粒子である非線形ルミネセンスマーカーであり、
単一ビーム励起及び二重ビーム励起の両方を得るために、少なくとも1つの光源位置から励起体積中に複数の光源によって同時に放出された励起光によって前記非線形ルミネセンスマーカーを励起する段階と、
前記励起光による前記ルミネセンスマーカーからのルミネセンスを検出器によってルミネセンス光検出位置で検出する段階と、
前記検出されたルミネセンスの前記励起光強度への非線形依存に基づいて前記ルミネセンスマーカーを画像化する段階と、を含み、
前記ルミネセンスマーカーを画像化する段階が、
各励起位置における励起光の強度を拡散方程式を用いて計算し、行列である励起場を得る段階と、
x次の前記非線形依存に従って前記励起場のx次積を計算する段階と、
前記励起場のx次積に基づいて、行列である前記ルミネセンスマーカーからの放出場を計算する段階と、
励起及び検出の対のそれぞれに関して前記励起場と放出場との間の積を計算して、感受性プロファイルを得る段階と、
前記感受性プロファイルとマーカーの分布との積と測定データとの差を最小にすることにより二乗問題を解くことによって前記アップコンバートナノ粒子の内部分布を得る段階と、を含み、

それぞれの光源位置に対して、前記マーカーから放出された全方向における全ルミネセンス強度を決定するために前記ルミネセンスを合計する段階であって、それによって前記ルミネセンスが前記複数の光源位置の各々に対して前記ルミネセンスマーカーの全ルミネセンス強度を有する、段階と、
前記複数の光源位置の各々に対して前記全ルミネセンス強度の画像を製作することによって前記ルミネセンスマーカーを画像化する段階と
を含む、請求項4に記載の方法。 - 前記全ルミネセンス強度が前記ルミネセンスマーカーの前記ルミネセンスの合計によって与えられる、請求項5に記載の方法。
- 第1の波長を有する第1の光源によって第1の光源位置から前記ルミネセンスマーカーを励起する段階と、
第2の波長を有する第2の光源によって第2の光源位置から前記ルミネセンスマーカーを励起する段階と
を含む、請求項1から6のいずれか一項に記載の方法。 - 前記第1の波長が前記第2の波長と実質的に同一である、請求項7に記載の方法。
- 前記第1および第2の光源位置の少なくとも一方が前記マーカー位置に関連して移動される、請求項7に記載の方法。
- 前記拡散ルミネセンスイメージングが拡散ルミネセンス断層撮影を含む、請求項1から9のいずれか一項に記載の方法。
- 前記光源位置が前記マーカー位置に関連して移動されるように、複数の異なる光源位置間で前記複数の光源を走査する段階と、
前記複数の異なる光源位置の各々に対する前記ルミネセンスマーカーのルミネセンス画像を供給するために前記ルミネセンスを検出する段階と、
を含む、請求項10に記載の方法。 - 散乱媒体中の注目する領域の拡散ルミネセンス分子イメージングのためのシステムであって、前記散乱媒体の前記ルミネセンス分子イメージングで使用するためのルミネセンスマーカーを含み、前記ルミネセンスマーカーが前記散乱媒体中に配置されたアップコンバートナノ粒子である非線形ルミネセンスマーカーであり、
単一ビーム励起及び二重ビーム励起の両方を得るために、複数の光源によって励起体積中に同時に放出された励起光によって前記ルミネセンスマーカーを励起するために、少なくとも1つの光源位置によって位置づけられた複数の光源と、
前記励起光による前記ルミネセンスマーカーからのルミネセンスを検出するルミネセンス光検出位置における検出器と、を備え、
