Description
癌は、早期であればある程、診療にかかる精神的・身体的・経済的・社会的負担が軽く、根治の可能性も高い。進行すればする程、根治の可能性は低下し、切除不能の進行再発癌に至っては、多大の負担と損失のうえ、延命治療となる現実がある。癌は細胞の遺伝子の異常が原因となり発生するため、ほとんどの癌において人口10万対各年齢における発生頻度は4-6 乗に比例して増加する(Cancer Patterns in Canada, 1982)。このため、いずれの年齢や社会においても、癌を早期発見・早期治療できれば、様々な場面において負担と損失が小さくできることは明白である。
この事から、癌特有のニオイを検出すれば高精度の癌探知システムの構築が可能と考えられる。
すなわち、本発明は以下の通りである。
(2)線虫がシノラブディティス・エレガンス(Caenorhabditis elegans)である(1)に記載の方法。
(3)線虫が、野生型線虫、変異型線虫又はトランスジェニック線虫である(1)又は(2)に記載の方法。
(4)被検者由来の生体関連物質又はその処理物の匂いに対して、線虫が正の応答を示したときは、被検者は癌である、又は癌のリスクがあると判定する、(1)〜(3)のいずれか1項に記載の方法。
(6)生体関連物質又はその処理物が、体液、細胞、組織、又は細胞若しくは組織の培養物若しくは保存液である(1)〜(5)のいずれか1項に記載の方法。
(7)体液が尿である(6)に記載の方法。
(8)保存液が生理食塩水である(6)に記載の方法。
(9)
線虫を用いて匂いの受容体の同定を行うことを特徴とする、線虫における匂いの受容体の同定方法。
(10)前記受容体をコードする遺伝子の発現又は機能を阻害し、当該阻害された線虫における匂いに対する反応を検査するものである(9)に記載の方法。
(12)匂いが癌種の匂いである(9)〜(11)のいずれか1項に記載の方法。
(13)同定される受容体の種類が、癌種により、又は匂い物質の濃度に依存して異なるものである(9)〜(12)のいずれか1項に記載の方法。
(14)線虫がシノラブディティス・エレガンス(Caenorhabditis elegans)である(9)〜(13)のいずれか1項に記載の方法。
(15)被検者由来の生体関連物質又はその処理物の匂いに対する線虫の反応を指標として癌種を同定することを特徴とする癌種の同定方法。
(a)前記(1)〜(8)のいずれか1項に記載の方法により癌を検出し、
(b)前記工程(a)において癌であることが検出された試料について、前記(9)〜(14)のいずれか1項に記載の方法により同定された受容体を改変した改変体線虫を用いて、匂いに対する反応を検査し、
(c)前記改変体線虫と、前記工程(a)で使用した線虫との間で匂いに対する反応が異なるときは、同定された受容体に対応する癌種を、同定の対象癌種であると判定する。
(17)受容体の改変が、受容体の欠失、受容体の発現又は機能の阻害、及び受容体の高発現又は高機能化からなる群から選ばれる少なくとも1つである(16)に記載の方法。
(18)線虫を含む、癌の検出又は同定用キット。
(19)線虫がシノラブディティス・エレガンス(Caenorhabditis elegans)である(17)に記載のキット。

(1)癌の検出
本発明は、線虫を、被検者由来の生体関連物質又はその処理物の存在下で飼育し、例えば線虫の嗅覚による化学走性などを指標として癌を検出することを特徴とする癌の検出方法である。
本発明者は、被検者が癌であるか否かを検査するに際し、一態様として被検者由来のサンプルに対する線虫C. elegansの嗅覚による化学走性に注目した。
線虫であるセノラブディティス・エレガンス(Caenorhabditis elegans、以下「C. elegans」ともいう)は生物研究のモデル生物として、世界中の研究室で広く飼育、研究されているポピュラーな生物であり、飼育が容易で、嗅覚が優れている特徴がある。
健常者、及び癌患者の尿に対する線虫の反応を調べたところ、健常者の尿に対しては忌避行動を、癌患者の尿に対しては誘引行動を示し、30 検体を調べその精度は100%であった(図1)。
また、早期癌を含む、胃癌、結腸・直腸癌、膵臓癌の全てに反応したことから、がん探知犬の行動と同じく、様々な癌に共通した、癌特有の匂いに反応している事が示された。
この線虫を用いた癌診断システムは、以下の通り、従来の問題点の多くを解決できる。
ステージ0、1の早期癌についても、高精度で検出可能である。尿を採取した時点(2011 年)で既存の腫瘍マーカーで陰性と判断された検体について、このテストでは陽性を示した。この患者は、経過観察中の2 年間に癌を発症した。すなわち、既存の腫瘍マーカーでは検出できない癌を、本発明により検出することが可能である。
(ii) 多種類の癌の存在を単一の検査で診断することができる。すなわち、一度の検診で多くの種類の癌について診断することができる。これまでのところ、胃癌、結腸・直腸癌、食道癌、膵臓癌、前立腺癌、胆管癌、乳癌、悪性リンパ腫、消化管間葉性腫瘍、盲腸癌、肺癌について検出可能であることを確認している。
(iii)高感度である。
30 検体のテストでは100%の感度・特異度で検出が可能であった。さらに、中規模テスト(242検体)を行っても、癌患者について100%の感度・95%の特異度で検出が可能であった。
尿サンプルの採取には、食事制限などの特別な条件を定めておらず、通常の定期検診で採取した尿サンプルを用いて解析できる。このため、被検者は苦痛を伴わず、他の尿検査と同時にサンプル採取が可能である。必要な尿の量は数μlで済む。
(v-1) 早い
短時間で解析可能。線虫の化学走性の解析は、1 時間半程度で行うことができる。トランスジェニック線虫を用いた嗅覚神経の応答の解析は、30分程度で行うことができる。
(v-2) 安価
例えば、1検体あたり、線虫飼育用シャーレ2 枚(1 枚約10 円)、走性解析用シャーレ3〜5枚(1 枚約10 円)。寒天はそれぞれのシャーレ1 枚あたり、2.5 円、10 円。その他試薬を合わせても、1 検体あたり100 円程度。人件費を合わせても、非常に安価に解析できる。
(v-3) 解析が容易であり、専門的な技術は必要としない。
線虫の走性解析は非常に容易であり、誰でも行うことができる。線虫の飼育も容易である。線虫に対する特別な訓練は必要とせず、通常飼育した線虫を用いて解析することができる。
(v-4) 多検体の解析が可能
実験者1 人当たり1 日に150 回程度の回数で走性解析を行うことができる。1 検体あたり3 回の走性を解析する場合、1 日に50検体を解析できる。この作業を自動化することも可能である。
(vi-1) システム全体が安価で導入しやすい
線虫の飼育に特別な部屋は必要ない。必要とする機器は、20℃インキュベーターと実体顕微鏡のみであり、安価かつ短時間でシステムの構築ができる。
(vi-2) 全世界に導入可能
高価な測定機器を必要としないことから、先進国だけでなく、全ての国に導入可能である。
全ての部位の癌を検出できることから、術後再発の可能性判断に応用できる。
匂いは嗅覚神経上の嗅覚受容体によって受け取られる。ヒトには約350種の嗅覚受容体が存在し、約1万種の匂いを識別できると言われている。匂いの種類に対して圧倒的に少ない受容体において、どのようにして膨大な種類の匂いを識別できるのかは不明であることから、それを明らかにするため、匂いと受容体との対応関係を明らかにする必要がある。しかし、そのような試みは部分的にしか行われておらず、特に生体内における対応関係はほとんどわかっていなかった。
(1)線虫
本発明の方法に用いる線虫は土壌自活線虫の一種であり、生物研究のモデル生物として広く研究に用いられている。本発明の方法に用いる線虫は、雌雄を問わないが、自家受精により増殖することができる点で雌雄同体のものが好ましい。また、本発明において使用する線虫は、シャーレの中で大腸菌を餌として飼育すればよく、飼育は容易である。親をシャーレに移しておけば、4 日後には子供が産まれて成虫にまで成長し、50〜100 倍に個体数が増える。その間、インキュベーターに入れて放置しておけばよく、特別な操作は必要ない。雌雄同体を使用する場合は、かけ合わせなどの操作も必要ない。飼育するのに必要な設備は20℃インキュベーター及び実体顕微鏡であり、システムの立ち上げは短時間に、安価に行える。
また、AWAに発現させるために使用するプロモーターとしては、例えばodr-10プロモーター(Sengupta, P., Chou, J. H. & Bargmann, C. I. odr-10 encodes a seven transmembrane domain olfactory receptor required for responses to the odorant diacetyl. Cell 84, 899-909 (1996).)が挙げられる。odr-10はAWAのみに発現誘導することが知られている。
カルシウムインディケーター遺伝子としては、Yellow Cameleon (YC) 遺伝子(Nagai, T., Yamada, S., Tominaga, T., Ichikawa, M. & Miyawaki, A. Expanded dynamic range of fluorescent indicators for Ca(2+) by circularly permuted yellow fluorescent proteins. Proc Natl Acad Sci U S A 101, 10554-10559 (2004).)、GCaMP遺伝子(Nakai, J., Ohkura, M. & Imoto, K. A high signal-to-noise Ca2+ probe composed of a single green fluorescent protein. Nat. Biotechnol. 19, 137-141 (2001). )などが挙げられ、これらの遺伝子は、例えばアクセッション番号AB178712、HM143847により塩基配列情報を得ることができる。また、これらの遺伝子は、addgeneにより入手することも可能である。
変異型線虫は、例えば野生型線虫のゲノムに多型が生じた線虫、癌種それぞれの匂いに対する嗅覚受容体をを欠失した変異体などを意味する(後述の実施例において説明する)。
本発明において使用される試料は被検者(健常人、癌患者、癌が疑われる患者等、動物)由来の生体関連物質である。「生体関連物質」とは、被検者から採取された生体試料であり、例えば体液(尿、汗、唾液、便汁)、細胞(生検細胞等)、癌組織(生検組織、組織切片等)血液、呼気などが挙げられる。本発明においては、これらの生体試料そのものを使用することができるが、生体関連物質の処理物を使用することが好ましい。「処理物」とは、生体関連物質を物理的及び/又は化学的に処理したサンプルを意味する。
また、本発明においては、上記試料のほか癌細胞又は癌組織(生検組織、組織切片等)の保存液を使用することもできる。保存液としては、例えば生理食塩水、緩衝液、ホルマリン、DMSOなどが挙げられるが、これらに限定されるものではない。保存液には凍結保存に一般に使用される凍結保存用保存液が含まれ、凍結保存後は、融解して使用することができる。
まず、検出に必要な線虫を得るために増殖させる。
線虫(成虫)をシャーレ(大腸菌をまいたNGM 培地)に数匹置いて、3〜6日、好ましくは4日間、15〜25℃、好ましくは20℃で飼育する。これにより次の世代の線虫が300〜500 匹程度、成虫まで育つ。
次に、実際に検査を行うためのシャーレを作製する。シャーレに図2 のようなフォーマットを作成し、4 点にアジ化ナトリウム(NaN3)を置く(添加する)。アジ化ナトリウムは線虫を麻酔して動かなくするためのものであり、添加量は1M濃度では0.2〜3μl、好ましくは0.5μlである。
所定時間(1 時間程度)線虫を飼育する(泳がせる)。室温は23℃±1℃。
所定時間経過後、+側の個体数、−側の個体数を数え、chemotaxis index (下記式)を計算する。+側の個体数をN(+)、−側の個体数をN(-)とすると、
chemotaxis index= N(+) - N(-) / 全個体数
となる。
「正の応答」とは、線虫がサンプルに対して「好き」であること又は「興味ある」ことを意味し、「負の応答」とは、線虫がサンプルに対して「嫌い」であること又は「興味ない」ことを意味する。
化学走性を指標とする場合は、正の値(+)は正の応答(正の化学走性、好き)、負の値(−)は負の応答(負の化学走性、嫌い)を表す。
1 検体あたり1回の解析でもよいが、複数回の解析を行い、chemotaxis index の値の平均値を計算することにより値の精度を高めることができる。解析により得られた値(複数回行ったときはその平均値)が正の値の場合、癌である又は癌のリスク(可能性)があると予備的に又は確定的に判断できる。「癌である」との判断は、例えば癌の確定診断又は予備的診断の補助資料として使用することができ、「癌のリスクがある」との判断は、例えば健康診断や癌の初診等において癌を疑うための補助資料として使用することができる。
風見鶏行動において、「正の応答」とは、線虫がサンプルの方向に向きを変えることを意味する。また、ターン行動においては、サンプルが高い濃度から低い濃度に変わったときにターンすると、線虫は「負の応答」をしたといえる。体の屈曲度の場合は、頭部と尾部の間の距離が長いと、「正の応答」をとったといえる。
(i) 遺伝子組み換え線虫を利用した検出についても、上記の通り行うことができるが、線虫の嗅覚神経AWC、AWAに、神経内カルシウム濃度を測定できるインディケーター遺伝子を発現させたトランスジェニック線虫を用い、カルシウム濃度の変化(神経の応答)を指標として検出する。この方法は線虫数個体で解析することができるため、線虫の培養にかかるコストが低く、短時間で行うことができる利点がある。
測定に使用するインディケータータンパク質は、カルシウム結合タンパク質CaMと、CaMが結合するターゲットのM13とをつなぎ、その両端にCFP、YFPをつないだ融合タンパクである(遺伝子にコードされており、遺伝学的に生体内で発現させることができる)。カルシウム濃度が低いときは、CaMとM13が離れており、CFPとYFPも離れた位置にある(左図)。そのため、CFPを励起させる光を与えると、CFPから青い光が発せられる。一方、カルシウム濃度が高くなってCaMとM13が結合すると、CFPとYFPが近付き、両者の間で蛍光共鳴エネルギー移動(又はフェルスター共鳴エネルギー移動)(FRET)が生じる。すると、CFPを励起する光を与えても、YFPから黄色い光が発せられるようになる(右図)。そこで、青い光、黄色い光を同時に計測し、その比を計算すれば、カルシウム濃度変化がわかる。
インディケータータンパク質は、CaMとM13とをGFPにつないだ構造をしている融合タンパク質である。このタンパク質も、遺伝子によりコードされており、遺伝学的に生体内で発現させることができる。カルシウム濃度が高くなってCaMと結合すると、GFPの蛍光強度が上昇する。そこで、GFPの蛍光を測定すれば、カルシウム濃度の変化がわかる。
ところで、健常者の尿でも線虫の嗅覚神経は弱く反応するため、コントロールの尿(健常者の尿)を用意し、同じ線虫個体にコントロール尿、検体尿を順番に与えて、その反応の強さの違いにより、癌の検出を行うことが好ましい。
本発明においては、線虫を用いて匂いの受容体の同定を行うことを特徴とする、線虫における匂いの受容体の同定方法を提供する。
本発明の同定方法は、受容体をコードする遺伝子の発現又は機能を阻害し、当該阻害された線虫における匂いに対する反応を検査する工程を含む。
嗅覚受容体遺伝子の発現又は機能阻害は、前記の通りRNAiを利用した阻害、アンチセンス核酸を利用した阻害、ドミナントネガティブ型変異遺伝子の発現による阻害などが挙げられるが、RNAiを利用した阻害が好ましい。
また、同定される受容体の種類は、又は匂い物質の濃度に依存して異なる。従って、どの濃度の試料に対して反応するかを検査し、高濃度の試料に対する受容体、及び低濃度の試料に対する受容体を同定することができる。
本発明においては、線虫走性テストにより癌種を特定することが可能となる。従って、本発明においては、被検者由来の生体関連物質又はその処理物の匂いに対する線虫の反応を指標として癌種を同定することを特徴とする癌種の同定方法を提供する。
(a)まずSTEP 1として、前記本発明の検出方法により癌を検出する。例えば、N2株を用いて、癌種の有無を検査する。
(b)次に、STEP 2として、各癌種の受容体の変異体や受容体遺伝子の発現、機能を変化させた株を用いて、癌種を特定する。
前記工程(a)において癌であることが検出された試料について、前記同定された嗅覚受容体を欠失させた変異体線虫や受容体遺伝子の発現、機能を変化させた株を用いて、匂いに対する反応を検査する。
例えば、前記変異体線虫や受容体遺伝子の発現または機能を阻害した線虫のうち匂いに対する反応を示さなかった線虫において同定された受容体に対応する癌種を、同定の対象癌種であると判定する。あるいは、受容体遺伝子を高発現または高機能化した線虫のうち匂いに対する反応が亢進した線虫において同定された受容体に対応する癌種を、同定の対象癌種であると判定する。
例えば、大腸癌の匂いの受容体変異体が誘引行動を示さない場合、大腸癌であると判断(診断)できる(図37)。
本発明は、線虫を含む癌の検出用キットを提供する。本発明のキットには線虫が含まれるが、本発明の検出方法を実施するために必要な1種以上の成分を含めることができる。このような成分としては、例えば緩衝液、培養液、アジ化ナトリウム、大腸菌、シャーレ、寒天などが挙げられる。また本発明のキットは、必要な成分のうちの一部のみを含む部分的キットであってもよく、その場合には、ユーザーが他の成分を用意することができる。また、本発明のキットには、検出法又は同定法を説明した使用説明書を含めることができる。
また、本発明は、線虫と、生体関連物質又はその処理物及び前記線虫を収容する収容部と、前記収容部の線虫の行動を探知する探知部とを備える癌の検出システムを提供する。
本発明のシステムは、線虫の少なくとも1匹の動画像をリアルタイムで撮影することができる少なくとも1つの探知部10を含む。探知部10は線虫の画像、個体数、動きの軌跡等のデータを取得するデバイスであり、顕微鏡又はカメラ、例えば蛍光顕微鏡、デジタル顕微鏡、デジタルビデオカメラなどを備える。顕微鏡及びカメラには、線虫の動きを追随できる自動追尾(追跡)システムを備えることができ、線虫1匹ごとに又は複数匹を同時に追跡する。そして、その軌跡から線虫の移動距離を測定する。あるいは、所定のエリアに集合している線虫を撮影し、個体数を数える。また、顕微鏡及びカメラには、蛍光強度を検出できるセンサーを備えることもできる。
検査条件設定手段111は、GUI(Graphical User Interface)により、計算に必要な条件を、マウスやキーボードから入力するための手段であり、入力された情報は、グラフにより確認することができる。
線虫の特徴は、欠失変異体であること、遺伝子発現等を阻害した株であること、測定のために蛍光タンパク質をコードする遺伝子(例えばGFP遺伝子、RFP遺伝子等)が導入されたことなどが含まれる。但し、条件はこれらに限定されるものではなく、検査目的に応じて適宜設定することができる。
線虫の反応検査手段112は、検査条件設定手段111又はデータベース120から癌を検出又は同定するための計算式(例えばchemotaxis index)を選択するとともに、それぞれの計算式に基づいて、線虫の反応を計算する手段である。この手段では、所定エリア内の線虫の個体数の測定、1匹の線虫が動いた総距離の測定、1匹の線虫が発する蛍光強度の検出などを行い、被検サンプルに反応した線虫の行動を記録する。例えば、1匹の線虫に着目してサンプルに誘引されて移動したときの距離を測定し、各線虫の移動距離を合算して総和を求める。あるいは、線虫に蛍光タンパク質遺伝子を導入したときは、サンプルに反応した線虫の蛍光強度を測定する。これも1匹の線虫あたりの蛍光強度を測定して線虫の総和を求めてもよく、一定のエリアに集まった線虫全体から発する蛍光強度を、エリア単位で測定することもできる。
癌の判定手段113は、線虫の匂いに対する反応をもとに癌の有無を検出し、又は癌の種類を同定する手段である。
検査条件として個体数を採用した場合、対照のエリアに移動した線虫の個体数に対し、検査対象エリアに移動した線虫の個体の比又は差などを求める。検査条件として移動距離を採用した場合、対照の線虫の移動距離と比較して何%多ければ反応したか、その差又は比を求めることができる。予めこの差又は比の一定値を境界値として設定しておけば、この境界値は癌の判定の判断基準として使用される。また、検査条件として蛍光強度を採用した場合は、対照の線虫の蛍光強度と比較して何%多ければ反応したといえるか等、上記移動距離と同様に境界値を設定することができる。
測定されたデータと、境界値と、サンプル情報(癌種の情報等)とを対比して、所定の癌であるかどうかを判定する。
判定結果出力手段114は、検出又は同定された癌種又は癌の有無に基づいてその情報を出力する手段であり、癌種及びその確率(リスク)を表示する。表示はグラフでも表でもよい。上記(ii)により計算された線虫の行動のアニメーションを表示することもできる。
入力された検査条件と検査結果は、関連付けられてデータ蓄積手段としてデータベース120に保存される。
保存された検査条件と計算結果は、再度データベース120から、あるいは検査条件設定手段111と判定結果表示手段114から読み込むことができる。
以下、実施例により本発明をさらに具体的に説明する。但し、本発明はこれら実施例に限定されるものではない。
(i) 線虫の飼育
野生型線虫N2 の成虫を6cm シャーレ(大腸菌をまいたNGM 培地)に5〜6 匹置き、4日間、20℃で培養した。次の世代の線虫を300〜500 匹程度、成虫まで育てた。
9cm シャーレに図2 に示すフォーマットを作成し、4 点にアジ化ナトリウム(NaN3)を0.5μl ずつ置いた。
線虫飼育プレートにwash buffer 1ml をかけ、浮いてきた線虫をbuffer ごとチューブに回収した。しばらく置いておくと線虫が下に沈むので、沈んだところで上清を廃棄した。さらにチューブにwash buffer 1ml を入れ、線虫が下に沈んだところで上清を廃棄した。この洗浄を3 回繰り返し、大腸菌を除去した。
次に、シャーレの中心に線虫を100 匹程度置き、 1 時間線虫を飼育した(泳がせた)。室温は23℃±1℃で行った。
1 時間後、+側の個体数、−側の個体数を数え、chemotaxis index を計算した。
1 検体あたり、5 回解析を行い、5 回分のchemotaxis index の値の平均値を計算した。
結果を図1に示す。図1より、健常者由来の対照の尿(c1〜c10)はすべて負(−)のchemotaxis index(忌避反応)を示したのに対し、癌患者由来の尿(p1〜p20)はすべて正(+)のchemotaxis index(誘引)を示し、100%の精度で癌を検出することができた。
なお、図1においてエラーバーはSEMを表す。
マイクロ流路を用いたイメージング実験においては、尿サンプルは薄いチューブ中に流す必要があることから、尿中の沈殿物及び固形物を遠心及びろ過により除去した (pore size 0.22 μm, MillexGP, Merck Millipore)。 AWC及びAWAニューロンをモニターするため、それぞれodr-1及びodr-10プロモーターによりYellow Cameleon遺伝子(YC3.60)を神経細胞に発現させた。カルシウムイメージングは、公知方法で行った(Uozumi, T. et al. Temporally-regulated quick activation and inactivation of Ras is important for olfactory behaviour. Sci Rep 2, 500 (2012); Shinkai, Y. et al. Behavioral choice between conflicting alternatives is regulated by a receptor guanylyl cyclase, GCY-28, and a receptor tyrosine kinase, SCD-2, in AIA interneurons of Caenorhabditis elegans. J Neurosci 31, 3007-3015 (2011))。
YC3.60の蛍光画像は、Leica DMI3000B顕微鏡(40倍対物レンズ)及びORCA-D2 デジタルカメラ(Hamamatsu) を用いて取得した。すべての画像は、露光時間200msで取得した。AWC又はAWA神経のCFP及びYFPの蛍光強度を取得し、CFP の蛍光強度に対するYFPの蛍光強度の比について、Metamorphソフトウエア(Molecular devices)により解析した。この蛍光強度比は、YFP強度/CFP強度 (= R)として計算し、10-sウインドウの比の平均 (-10-0 s)をR0としてセットした。
図7において、左2つのパネルは対照尿、右2つのパネルは胃癌患者由来の尿を用いて試験した結果である。図7はAWC嗅覚神経の尿刺激(尿あり→なし)に対するカルシウム濃度変化(Yellow CameleonのYFP/CFP ratio変化)を示す図である。健常者の尿(対照)と比較して、癌患者の尿に対して有意に強く反応した。図8は平均の蛍光強度比(YFP/CFP ratio)の変化量を表す。***はp<0.001で有意であることを示す。
なお、本実施例において尿中の沈殿物及び固形物は遠心分離及びろ過により除去したが、この処理によって線虫の化学走性には影響しなかった(図9)。
AWA嗅覚神経についても、尿の添加により応答が観察された(図10、11)。
(1)癌細胞株を用いた癌の検出
ヒト癌細胞の培養上清を用いた癌の検出を行うため、大腸癌(結腸直腸癌)細胞としてSW480、COLO201及びCOLO205、乳癌細胞としてMCF7、胃癌細胞としてNUGC4、MKN1及びMKN7を用いた。
SW480, COLO201及びCOLO205は独立行政法人医薬基盤研究所JCRBセルバンク(Japanese Collection of Research Bioresources Cell Bank (Tokyo, http://cellbank.nibio.go.jp))から入手し、それ以外の細胞は東北大学加齢医学研究所 医用細胞資源センター(Cell Resource Center for Biomedical Research, Institute of Development, Aging and Cancer (Tohoku University, Sendai, Japan))から入手した。すべての細胞株は、10% FBS添加RPMI 1640 培地を用い、37 °C、5% CO2エアレーション下でコンフルエントではない状態で維持した。培地上部の清澄な層の培養液を試験に用いた。細胞を培養した後の培地は、アッセイプレート(図12)の「+」の位置にスポットした。培地自体の匂いによる影響を無くすために、細胞を培養した培地のスポット位置とは反対側の位置に、同じ濃度で希釈した対照の培養液をスポットした(図12)。
図13中、各細胞における左側のバーは1/106希釈、右側のバーは1/107希釈の癌細胞培養培地を用いた結果である。また、*はp<0.05、**はp<0.01、***はp<0.001で有意であることを表す(Dunnett's test)。また、図13中、エラーバーはSEMを表す。
MEM、EMEM、RPMIは培地のみ。KMST-6、CCD-112CoNは線維芽細胞(入手先はそれぞれRBC、ATCC)。
また、様々な濃度の線維芽細胞培養培地及び癌細胞培養培地に対する走性、並びに人間の癌組織に対する線虫の走性を検討した。手法は以下の通り。
線維芽細胞培養培地及び癌細胞培養培地を水で各種濃度(原液〜10-9)に薄め、それに対する野生型線虫の化学走性を観察した。人間の癌組織、健常組織についてはインフォームドコンセプトを得た上で癌患者から切除し、それを直径0.1~0.8mmに細分して使用した。
その結果、繊維芽細胞についてはいずれの濃度でも誘引行動は示さないのに対し、癌細胞については10-6、10-7の濃度で有意な誘引行動が観察された(図17、18)。
人間の癌組織片を生理食塩水に入れて-20℃で保存した(保存期間:3か月)。その生理食塩水の希釈液に対する線虫の走性を検討した。
癌患者からインフォームドコンセプトを得た上で癌組織を直径0.5cm切除し、20mlの生理食塩水に入れた。その生理食塩水を水で10-2〜10-4の濃度に薄め、それに対する野生型線虫の化学走性を観察した。
その結果、癌組織を入れた生理食塩水に対しては誘引行動を示すのに対し、健常組織を入れた生理食塩水に対しては忌避行動を示した(図20)。
odr-3変異体は癌組織の食塩水に誘引行動を示さないことから、線虫は匂いを感じていると言える。
本発明の方法が高精度であることを確認するため、242個の尿サンプル(218の対照サンプル、24の癌患者由来のサンプル)を用いて試験を行った(表1)。表1は被検者の背景を示す。
解析対象となる腫瘍マーカーとして、血清CEA、血清抗p53抗体 (Anti-p53 Ab) 及び尿N1,N12-ジアセチルスペルミン(DiAcSpm)を用いた。これらの腫瘍マーカーと比較して、本発明の方法(NSDT)による感度は極めて高かった(図15、表2)。なお、感度(%)は、癌患者由来のサンプルに対する陽性の割合である。
方法:
健常者の尿サンプル3検体(c1、c2、c3)、癌患者の尿サンプル5検体(p2、p5、p8、p17、p18)について水で各種濃度(原液〜10-5)に薄め、それらに対する野生型線虫の化学走性を調べた。
図21より、10倍希釈が好ましいことが示された。図21において、各濃度に記載の棒グラフは、左側3本のグラフが健常者由来の尿、右側5本のグラフが癌患者由来の尿を用いたときの結果を示す。
(1)材料及び方法
線虫培養及び線虫株
線虫(C. elegans)は、大腸菌(E. coli)OP50とともに培養したeri-1変異体を除いて、食物源として大腸菌(Escherichia coli)NA22を含む線虫増殖培地(NGM)プレート(36)上で、標準的な条件下で20℃にて培養した。使用した野生型線虫は、Bristol N2株であった。他の線虫株として、GR1373:eri-1(mg366)、VC2123:sri-14(ok2865)、CX3410:odr-10(ky225)、及びMT7929:unc-13(e51)を用いた。
RNAiアッセイは、Ahringerライブラリー(37)を用いて、eri-1(mg366)(19)に対して、餌によるRNAi法(feeding RNAi法)によって行った。
9個体のeri-1変異体の成虫を、イソプロピルβ−D−1−チオガラクトピラノシド(0.19g/L)、アンピシリン(60mg/L)、及び大腸菌(E. coli)を含むNGMプレート上に置き、4日間培養した。次に、成虫を化学走性アッセイに使用した。化学走性アッセイは、以前に報告されているように行った(6, 17)。化学走性アッセイについて、本発明者は、それぞれ1μlの10-3若しくは10-4希釈された匂い物質(低濃度)、又はそれぞれ1及び5μlの未希釈の匂い物質(高濃度)を使用したそれぞれの実験において、30〜50個体を用いた。
RNAiスクリーニング結果の統計分析は以下のように行った。
本発明者は、浸透圧刺激のために4M NaClを用い、浸透圧忌避行動について以前に報告されているようにアッセイした(38)。
特異的ニューロンにおけるsri-14の機能をノックダウンするための導入遺伝子の構築は、以前に報告されているように行った(24)。sri-14の標的領域(1.6kbのゲノム配列)を以下の2つのプライマーを用いて増幅した。

Tr、5'-ctgctgcgtttttcgtatca-3'(配列番号2)
遺伝子発現は、ASHについてはsra-6(20)プロモーターによって、及びAWCについてはceh-36(39)とsrd-17プロモーターによって行った。
本発明者は、AWA、AWB、AWC、及びASHニューロンの除去のためにマウスのカスパーゼ−1(mCasp1)を用いた。これらは、それぞれ、odr-10(11)、str-1(15)、ceh-36(39)、及びsra-6(20)プロモーターによって発現させた。また、介在ニューロンの阻害のためにunc-103(gf)を用いた。unc-103(gf)のAIA−、AIB−、AIY−、又はAIZ−特異的発現は、それぞれ、gcy-28.d(29)、npr-9(40)、ttx-3(41, 42)、又はlin-11(43, 44)プロモーターによって行った。
PureLink RNAミニキット(Ambion)を用いて単離された全量RNAは、製造業者の指示書に従って、gDNA Remover(Toyobo)を用いたReverTra Ace qPCRマスターミックスによってcDNAに変換した。sri-14 cDNAを下記の2つのプライマーを用いて増幅し、Nhe IとKpn Iで消化し、pPD-DESTベクター(Invitrogen)に挿入した。

AWA、AWB、AWC、及びASHニューロンの応答をモニターするために、本発明者は、それぞれ、odr-10、str-1、odr-3、及びsra-6プロモーター(11, 15, 20, 45)を用いて、YC3.60を発現する株を作製した。カルシウムイメージングは、以前に報告されているように(33, 46, 47)、マイクロ流体デバイスを用いて行われた。マイクロ流体デバイスを用いた実験について、線虫の鼻が、ジアセチル[10-5希釈(低濃度)と10-3希釈(高濃度)]又は匂いのない溶液を含む流水に曝露されるように、マイクロチャネルに線虫を捕捉することによって、それぞれの線虫を固定した。室温は20℃から23℃に設定した。YC3.60の蛍光画像は、40×対物レンズと3CCDデジタルカメラ(C7780、Hamamatsu Photonics)を備えたZeiss Axioplan 2を用いて得た。すべての画像は、露光時間を200msにして回収した。AWA、AWB、AWC、及びASH細胞体のタイムスタックを捕捉し、AquaCosmosソフトウェア(ver. 2.6、Hamamatsu Photonics)を用いて、黄色蛍光タンパク質(YFP)とシアン蛍光タンパク質(CFP)の発光比について分析した。この比は、YEP強度/CFP強度(R)として計算され、10秒ウィンドウ(-10〜0秒)における平均比は、R0として設定した。
匂い−受容体対のRNA干渉スクリーニング
特定の匂い物質に対する応答に必要とされる嗅覚受容体を網羅的に同定するために、本発明者は、システマティックRNA干渉(RNAi)スクリーニングを行った。線虫(C. elegans)におけるニューロンのRNAiは、RNAiに対する線虫(C. elegans)ニューロンの低感受性のため(18)、野生型線虫においては有効でないことから、RNAi増強型線虫株eri-1(mg366)(19)を用いた。
感覚ニューロンに発現する遺伝子は嗅覚受容体として働く可能性があることから、これらの遺伝子の発現パターンを調べた。
ODR-10はジアセチル受容体であり、odr-10変異体は、低いジアセチル濃度に対する化学走性に欠陥がある(11)。従来報告されているように(11)、本発明者は、odr-10変異体が高濃度ジアセチルに対して、正常な化学走性を示すことを確認した(図24A)。これは、ジアセチルに対して他の受容体が存在し、ODR-10が低濃度に特異的である可能性がある(21)ことを示唆している。RNAiスクリーニングによって、5つの嗅覚受容体候補遺伝子(srh-25、srh-79、srh-216、srh-281、及びsri-14)が高濃度ジアセチルに関与するものとして得られた(表3及び図25)。
低濃度のジアセチル(10-4希釈溶液)はAWAニューロンによって感知され(6)、中間濃度(10-1希釈溶液)はAWCニューロンによって感知される(22, 23)。これらはともに誘引を媒介する。しかしながら、どの感覚ニューロンが、高濃度のジアセチル(未希釈)を感知し、忌避応答に関与するかは知られていない。以前の研究において、ASH及びAWB感覚ニューロンは、高濃度イソアミルアルコール(17)を検出し、忌避を媒介することが示されている。さらに、AWC及びAWBニューロンは、誘引と忌避の両方に関与する(17, 27)。
本発明者による線虫の行動実験の結果と従来の研究結果(11)は、ジアセチルに対する濃度依存的な嗜好性変化が、2つのタイプの受容体、AWAニューロンにおけるODR-10とASHニューロンにおけるSRI-14によって媒介されることを示唆している。そこで本発明者は、野生型株、odr-10変異体、及びsri-14変異体において、遺伝的にコードしたカルシウム指示薬であるYellow Cameleon(YC)3.60(30)を用いたカルシウムイメージングによって、様々な濃度のジアセチルに対するAWA及びASHニューロンの応答をモニターした。
本発明者は、特定の匂い物質について嗅覚受容体を網羅的に同定するためにRNAiスクリーニングを用いた。線虫(C. elegans)は、嗅覚受容体及び嗅覚シグナリングが哺乳動物のものと類似しているため(23, 34)、嗅覚解析のためのモデル生物と考えられている。さらに、嗅覚シグナルが神経回路において伝達される経路を辿ることができる全神経ネットワークが記述されている(35)。しかしながら、大部分の匂い物質は、特定の受容体又は受容体オリゴマーとの対応関係がわかっておらず、匂い物質と嗅覚受容体の間の相互作用の機構は知られていない。結論として、どのようにして匂いシグナルがインプットされるか、どのようにして嗅覚シグナルが神経回路上を伝達されるか、どのようにして線虫(C. elegans)における少数のORNが非常に多くの匂い物質を識別し得るのかについて理解されていない。RNAiスクリーニングによって得た受容体候補のさらなる解析は、特定の匂い物質に関する嗅覚受容体を同定し、これらの機構の理解を与えるために役立つ。
そこで本実施例においては、野生株とともに上記遺伝子変異株を用いて、癌検出の精度を検討した。
乳癌、食道癌、胆管癌、直腸癌、盲腸癌、前立腺癌、膵臓癌、肺癌、消化管間葉性腫瘍の癌患者の尿サンプル、健常者の尿サンプル、中規模実験で偽陽性を示した健常者の尿サンプルについて、野生株、遺伝子変異株の化学走性を調べた。
遺伝子変異株は、ほとんどすべての癌種の尿に誘引行動を示さなかった(図34)。
遺伝子変異株は、他の匂いに対しては正常な走性を示すことから、基本的な嗅覚は働いていると言える。従って、遺伝子変異株には癌種の匂いを受容する受容体に欠損があると予想される(図35)。
次世代シークエンサー(イルミナ社)を用いて、実施例7で得られた遺伝子変異株の全ゲノムを解読し、N2のゲノム配列と比較し、変異のある受容体遺伝子を探索した。
その結果、癌種により、反応する受容体が異なることが分かった。
これにより、線虫走性テストにより癌種を特定することが可能となる。
受容体ノックダウン株における乳癌患者の尿に対する走性を調べた結果を図36に示す。
次に、STEP 2として、各癌種の受容体の変異体を用いて、癌種を特定する。例えば、大腸癌の匂いの受容体変異体が誘引行動を示さない場合、大腸癌と診断できる(図37)。

- 嗅覚受容体の欠失、嗅覚受容体の発現又は機能の阻害、及び嗅覚受容体の高発現又は高機能化からなる群から選ばれる少なくとも1つで改変された変異体線虫と、癌患者の体液、細胞、組織、又は細胞若しくは組織の培養物若しくは保存液とを用い、
前記変異体線虫の、前記体液、細胞、組織、又は細胞若しくは組織の培養物若しくは保存液に対する反応を検査することにより、嗅覚受容体を同定することを特徴とする、線虫における嗅覚受容体の同定方法。 - 前記変異体線虫が、前記嗅覚受容体をコードする遺伝子の発現が阻害された変異体線虫である請求項1に記載の方法。
- 嗅覚受容体遺伝子の発現阻害がRNAiによるものである請求項2に記載の方法。
- 同定される嗅覚受容体の種類が、癌種により異なるものである請求項1〜3のいずれか1項に記載の方法。
- 線虫がシノラブディティス・エレガンス(Caenorhabditis elegans)である請求項1〜4のいずれか1項に記載の方法。
- 請求項1〜5のいずれかに記載の方法により線虫における嗅覚受容体を同定し、当該嗅覚受容体の欠失、当該嗅覚受容体の発現又は機能の阻害、及び当該嗅覚受容体の高発現又は高機能化からなる群から選ばれる少なくとも1つにより線虫を改変し、変異体線虫を製造することを特徴とする、変異体線虫の製造方法。
