Description
別途定義しない限り、本明細書で用いられる全ての技術用語および科学用語は、当業者によって一般的に理解されるものと同じ意味を有する。
ってもよいが、最も免疫原性であるこれらのアミノ酸を、免疫原性が低いものと置きかえる。それにも拘らず、上記で考察されたCDR移植は、ヒト化抗体を得るための唯一の方法ではない。例えば、CDR領域だけの改変はCDRループの三次元構造および抗体のそのリガンドに対する全体の親和性を決定する際に役割を果たすフレームワーク残基にとって一般的ではないため、CDR領域だけの改変では不十分であり得る。したがって、非ヒト親抗体分子が、ヒトに対する免疫原性が低いものとなるように改変され、ヒト抗体との全体的配列同一性が常に必要ではないような任意の手段を実施することができる。したがって、例えば、ごくわずかの残基、特に、抗体分子上に露出され、分子内に埋まらず、したがって、宿主免疫系にとって容易に接近可能ではない残基のわずかな置換により、ヒト化を達成することもできる。例えば、Studnickaら、Prot Eng 7(6)805〜814頁、1994;Mol Imm 44:1986〜1988頁、2007;Simら、J Immunol 151:2296頁(1993);Chothiaら、J Mol Biol 196:901頁(1987);Carterら、Proc Natl Acad Sci USA 89:4285頁(1992);Prestaら、J Immunol 151:2623頁(1993)、WO2006/042333および米国特許第5,869,619号を参照されたい。あるいは、抗体は、他の技術、例えばCDR移植(EPO0239400;WO91/09967;および米国特許第5,530,101号および第5,585,089号)、ベニヤリングまたはリサーフェシング(EPO0592106;EPO0519596;Padlan、1991、Molec Imm 28(4/5):489〜498頁;Studnickaら、1994、Prot Eng 7(6):805〜814頁;およびRoguskaら、1994、PNAS 91:969〜973頁)および鎖シャフリング(米国特許第5,565,332号)によってヒト化することができる。ヒト抗体は、限定されずにファージディスプレイ方法を含む、当技術分野で公知である様々な方法によって作製することができる、齧歯動物などのトランスジェニック動物、キメラ細胞その他を用いる、米国特許第4,444,887号、第4,716,111号、第5,545,806号および第5,814,318号;ならびにWO98/46645、WO98/50433、WO98/24893、WO98/16654、WO96/34096、WO96/33735およびWO91/10741を参照。
前述のように、本発明の製剤は、抗IL−4/抗IL−13二特異的抗体およびバッファー系を含み、ここで、製剤のpHは約pH7であり、製剤は、製剤のイオン強度を低減するために低い塩濃度を有する。製剤は、場合により、非イオン性界面活性剤、糖および/または非イオン性安定化剤をさらに含むことができる。本発明の製剤は、製剤中の抗体の濃度を増加させることによる抗体の分子凝集ならびに眼に見える粒子および眼に見えない粒子の形成にしばしばつながるこれまでの抗IL−4/抗IL−13二特異的抗体製剤より、有意な改良を提供することが見出された。特に、本発明の製剤は、眼に見える粒子、眼に見えない粒子、低分子量タンパク質および高分子量タンパク質に関して優れた安定性を示す。
ある特定の実施形態では、本発明の製剤は、抗IL−4/抗IL−13二特異的抗体を含む。二特異的抗体は、IL−4および/またはIL−13またはその変異体もしくは断片に結合するかまたは特異的に結合する。IL−4および/またはIL−13分子は、任意の種からのものであってもよい。好ましくは、IL−4および/またはIL−13分子は、ヒトからのものである。IL−4およびIL−13の両方のアミノ酸配列およびタンパク質構造は、当技術分野で周知である。
バッファーは、生理的条件に近似した範囲に製剤のpHを維持するのを助ける。バッファーは、好ましくは約1mMから約50mMまでの範囲内の濃度で製剤中に存在する。本発明での使用に適するバッファーには、有機および無機の酸ならびにその塩、例えばクエン酸バッファー(例えば、クエン酸一ナトリウム−クエン酸二ナトリウム混合液、クエン酸−クエン酸三ナトリウム混合液、クエン酸−クエン酸一ナトリウム混合液など)、コハク酸バッファー(例えば、コハク酸−コハク酸一ナトリウム混合液、コハク酸−水酸化ナトリウム混合液、コハク酸−コハク酸二ナトリウム混合液など)、酒石酸バッファー(例えば、酒石酸−酒石酸ナトリウム混合液、酒石酸−酒石酸カリウム混合液、酒石酸−水酸化ナトリウム混合液など)、フマル酸バッファー(例えば、フマル酸−フマル酸一ナトリウム混合液、フマル酸−フマル酸二ナトリウム混合液、フマル酸一ナトリウム−フマル酸二ナトリウム混合液など)、グルコン酸バッファー(例えば、グルコン酸−グリコン酸ナトリウム混合液、グルコン酸−水酸化ナトリウム混合液、グルコン酸−グリコン酸カリウム混合液など)、シュウ酸バッファー(例えば、シュウ酸−シュウ酸ナトリウム混合液、シュウ酸−水酸化ナトリウム混合液、シュウ酸−シュウ酸カリウム混合液など)、乳酸バッファー(例えば、乳酸−乳酸ナトリウム混合液、乳酸−水酸化ナトリウム混合液、乳酸−乳酸カリウム混合液など)および酢酸バッファー(例えば、酢酸−酢酸ナトリウム混合液、酢酸−水酸化ナトリウム混合液など)が含まれる。リン酸バッファー、炭酸バッファー、ヒスチジンバッファー、トリメチルアミン塩、例えばTris、HEPESおよび他のそのような公知のバッファーも好適であり、使用することができる。好ましくは、本発明の製剤では、バッファーの組合せ、すなわち2つ以上のバッファーが使用される。2つ以上のバッファーの組合せは、本明細書においてバッファー系と呼ばれる。
本発明の製剤は、場合により、非イオン性界面活性剤をさらに含んでもよい。界面活性剤は、製剤中の生物分子および/または一般的な医薬添加剤を安定化する化学化合物である。界面活性剤は一般に、空気/溶液境界面で誘導されるストレスおよび溶液/表面で誘導されるストレスから分子および添加剤を保護し、そうでなければ分子の凝集をもたらし得る。界面活性剤は、眼に見える粒子および眼に見えない粒子形成も阻止する。
本発明の製剤は、場合により糖をさらに含むことができる。一般的に、糖は高分子量タンパク質のための安定化剤として、または凍結保護物質として、または凍結乾燥保護剤として使用される。
本発明の製剤は、場合によっては非イオン性安定化剤をさらに含むことができる。安定化剤は、増量剤から治療剤を可溶化するまたは変性もしくは容器壁への接着を阻止するのを助ける添加剤まで、機能が様々である広いカテゴリーの賦形剤を指す。安定化剤は、高分子量タンパク質の形成を最小にもする。
さらに、本発明の製剤は、場合により、限定されずに注射用水、希釈剤、可溶化剤、無痛化薬、追加のバッファー、無機または有機の塩、抗酸化剤、保存剤、増量剤、キレート化剤、張性剤その他を含む、他の賦形剤をさらに含むことができる。しかしながら、好ましくは、本発明の製剤は、上記のもの以外の他の添加剤を含まない。Remington’s Pharmaceutical Sciences、第16版、Osol,A.(編)(1980)に記載のもののような、他の薬学的に許容される担体、添加剤、または安定剤が製剤の所望の特性に有害に影響しないという条件で、それらを製剤中に含有させることができる。特定の実施形態においては、製剤は保存剤を実質的に含まないが、代替的な実施形態においては、必要に応じて保存剤を添加してもよい。例えば、凍結防止剤またはリオプロテクタントを、凍結乾燥製剤中に含有させることができる。
本発明の製剤は、液体製剤または凍結乾燥製剤であってもよい。好ましくは、製剤は液体製剤である。より好ましくは、液体製剤は、すぐに注射できる。あるいは、製剤は凍結乾燥粉末であってもよい。好ましくは、凍結乾燥粉末は、投与の直前に溶媒とすぐに混合される。
本発明の1つの例示的な実施形態では、本発明は、皮下投与に適する液体抗体製剤であって、

本発明の製剤は、2〜8℃で、少なくとも約1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27、28、29、30、31、32、33、34、35、または36カ月以上にわたって安定である。例示的実施形態においては、それらは2〜8℃で、少なくとも約6カ月以上にわたって安定である。他の例示的実施形態においては、それらは2〜8℃で、少なくとも約9カ月にわたって安定である。さらなる例示的実施形態においては、それらは2〜8℃で、少なくとも約1年以上、より好ましくは約2年、さらにより好ましくは約3年にわたって安定である。
本発明のある実施形態においては、製剤は、非経口投与、静脈内投与、筋肉内投与、皮内投与、皮下投与、またはその組合せにとって好適である。本発明の製剤は、様々な技術による送達にとって好適である。本発明の好ましい実施形態においては、製剤は皮下投与される。例えば、100mg/mLの抗IL−4/抗IL−13二特異的抗体を含有する製剤を皮下投与するのが好ましい。したがって、製剤は、好ましくは滅菌される。無菌製剤を作製するための方法は当業界で周知であり、例えば、滅菌濾過膜を通す濾過または約120℃で約30分間の抗体を除く製剤の構成成分のオートクレーブが挙げられる。
本発明の製剤の有効用量は、投与手段、標的部位、対象の生理的状態、対象がヒトまたは動物であるかどうか、投与される他の薬剤、および処置が予防的なものであるか、または治療的なものであるかなどの、多くの様々な因子に応じて変化する。通常、対象はヒトであるが、トランスジェニック哺乳動物などの非ヒト哺乳動物を処置することもできる。処置用量は安全性および効能を最適化するために滴定する必要がある。好ましくは、用量範囲は100〜200mg/バイアルである。
対象に本発明の製剤を投与することを含む、IL−4および/またはIL−13媒介性疾患または障害を処置するための方法が、本明細書でさらに提供される。ある特定の実施形態では、IL−4および/またはIL−13によって媒介される疾患は、がん、炎症、自己免疫性疾患、感染症、心血管疾患、呼吸器疾患、神経学的疾患および代謝疾患である。

100mg/mLリード抗体溶液について70mMの濃度の2つの選択されたバッファー−pH候補で塩化ナトリウム(NaCl)を試験した。
製剤開発過程での次の工程は、リード抗体製剤の最適な賦形剤を判定することであった。賦形剤はリード抗体を安定させるのに十分でなければならず、凍結乾燥に適合しなければならない。ポリソルベートおよびポロキサマーなどのいくつかの異なる界面活性剤を、様々な濃度で試験した。

・PS20 0.1%:#P12−8
上記の通り製剤開発過程での次の工程は、リード抗体製剤の最適な賦形剤を判定することであった。賦形剤はリード抗体を安定させるのに十分でなければならず、凍結乾燥に適合しなければならない。スクロース、トレハロースおよびマンニトールなどのいくつかの異なる糖を、様々な濃度で試験した。
上記の通り、製剤開発過程での次の工程は、リード抗体製剤の最適な賦形剤を判定することであった。賦形剤はリード抗体を安定させるのに十分でなければならず、凍結乾燥に適合しなければならない。マンニトール、アスパラギン酸、プロリン、グリシン、アルギニンおよびロイシンなどのいくつかの異なる安定化剤を様々な濃度で試験した。これは、下の表および上の表10〜14で見ることができる。
・アミノ酸グリシン(#P12−1およびP20−4−FDS)およびプロリン(#P20−3−FDS)は、HMW形成の最小化において最良の効果を有することが見出された(図13および図14を参照する)
・マンニトール(#P20−1−FDS)は、それに続いた(図14を参照する)
・スクロース(#P12−3)およびトレハロース(#P12−4)が後に続いたが、効果はずっと劣った(図13を参照する)
・最後に、4%の濃度でアスパラギン酸8%(#P20−2−FDS)の正の効果は確認されなかった(図14を参照する)。
・リジン(#P21−5)は、HMW形成にわずかな正の影響を及ぼすことが見出された(図15を参照する)
・マンニトール(#P21−1)は、このアッセイで最良であることが見出された。
凍結/乾燥工程のために、リード抗体濃度が20〜38mg/mLの範囲の所望のFDSプロトタイプの5〜5.5mLを、15mLバイアルに一定量注入した(例として、表13および表14を参照する)。

・凍結温度は完全固化の温度未満、すなわちガラス転移温度(Tg’)よりわずかに低く設定し、それは各FDS候補(凍結乾燥前の溶液)についてDSCによって測定した(図17を参照する)。
・一次乾燥温度は、試料の温度がTg’の近くである崩壊温度未満にとどまるように設定した(図17を参照する)。

これらの研究(実施例7〜8)の目的は、液状のリード抗体のHMW形成への高い傾向に関してFDS安定性を改善することであった。
・約6.2のpH値はリード抗体溶解性を低下させる:コハク酸塩およびヒスチジンによりゲルの形成および沈殿がそれぞれ観察された
・さらに、溶液が乳白色でわずかに熱安定性が低いので、pH6.6でヒスチジンは避けるべきである
・6.6から7.4の間のpH範囲では、HMW形成動態に及ぼす明らかなpH効果はなかった
・イオン強度の増加は、HMW形成動態に不安定化効果を及ぼした
・リン酸塩およびクエン酸塩は、それらが1.75mMなどの低い濃度であるならば、試験した最良のバッファーであった
・試験した全ての賦形剤の中で、最良の安定化効果は、グリシン10および72mMならびにスクロース2.4%で得られた。しかし、この安定化効果はHMW形成を有意に遅くすることを可能にしなかった
リード抗体は、サイトカインIL−4およびIL−13を標的にする操作されたヒト化二特異的抗体(BsAb)である。質量分析によって判定されるその分子量は198kDaであり、IEFによって判定されるそのIpは5.8から6.2の間である。

・バッファー系リン酸塩/TrispH.0と一緒にポリソルベート80を0.2%の濃度で用いることによって、眼に見える粒子/眼に見えない粒子は有意に低減された
・HMW形成動態はリード抗体濃度と一緒に増加し、試験した製剤は、100mg/mLにおいて液体製剤の満足な有効期間が予想されるほど十分にHMW形成を阻止しなかった。
・液体状態でのHMW形成は、35mg/mLのFDSから100mg/mLを達成するために初期容量(凍結乾燥前)の1/3よりわずかに少ない量で再構成される凍結乾燥DPの方面に開発を推進した。凍結保護物質および凍結乾燥保護剤が選択された:5%濃度のスクロース。
・相IおよびIIaのために選択された製剤は、以下の通りであった:リン酸塩6.5mM/Tris3.7mM、pH7.0、PS80 0.2%(w/v)、スクロース5%(w/v)、プロリン3%(w/v)。
・わずかな正の効果を有すると同定された賦形剤、すなわち、スクロース、マンニトールならびにグリシンおよびプロリンなどのアミノ酸を組み合わせることによって、この製剤は、HMW形成に関して最適化することができるかもしれない。
・RTで35mg/mLのFDS:ΔHMW=7時間で+1.6%、24時間で+4.1%
・RTで100mg/mLのDP:ΔHMW=1時間で+0.6%、5時間で+3.5%
この研究(実施例7〜8)の目的は、HMW形成に関して液体状態のリード抗体の安定性を増加させることであった。この研究の定量的目標を設定するために、以下の値が提案された:
・使用中の安定性のためのDP再構成からRTで5時間:5時間でΔHMW<+1%、
・DP製造を容易にするためのFDSの12時間のTOR:12時間でΔHMW<+1%。
この研究(実施例7〜8)のために提案されたアプローチは、35mg/mLのリード抗体濃度で広範囲の製剤にわたって組み合わせた賦形剤をスクリーニングすることであった。
・それらの濃度でHMW形成に負の影響は実証されず、製造工程および/または再構成への強い正の影響が観察されたので、製剤相IのPS80およびスクロースの濃度がスクリーニングで保たれた。
・以前の研究は約pH7にpHを保つ利点を示していたので、pH範囲は6.2から7.4の間に引き締められた
・このpH範囲内の5つの注射可能なバッファーを、単独でまたはいくつかの賦形剤と一緒にスクリーニングした:他のバッファー系および/または添加剤(主にアミノ酸、スクロース(相I製剤に既に含有されたものに加えて)および塩)。
この研究(実施例7〜8)で使用したDSは、表25に記載する。
この研究(実施例7〜8)で使用したDPは、製剤相I/IIa(リン酸塩6.5mM/Tris3.7mM、pH7.0、PS80 0.2%、スクロース5%、プロリン3%)で製剤化した(表26を参照する)。
実施例7〜8の各アッセイのための処方は、下の表27に掲載する。
実施例7〜8の製剤は、スクロース2.1%に42mg/mLの濃度でUF/DFによって得られたリード抗体の濃縮溶液で製造した。このリード抗体溶液は、賦形剤の濃縮保存溶液で希釈した。
UF/DF工程(実施例7〜8)の間、35mg/mLの初期タンパク質溶液を40mg/mLに先ず濃縮し、次にバッファーを交換した。ダイアフィルトレーションの後、タンパク質溶液を目標の最終濃度であった42mg/mLを超えて濃縮した(表28を参照する)。
UF/DF後リード抗体溶液(実施例7〜8)を、所望の最終製剤まで適当な量の濃縮保存溶液で希釈した。この研究のために製造された濃縮溶液の参照および組成を、表29に要約する。
熱ストレス
製剤アッセイに従って実施例7〜8で実施した熱ストレス条件は、表30に掲載する。

・変性温度のためのDSC:
−異なる製剤での抗体の熱安定性を推定するために示差走査熱量測定(DSC)を使用した。
−熱量測定は、1℃/分の加熱速度により、25℃から100℃のVP−毛細管DSCで実施した。
−能力曲線は、変性温度Td(℃)に関する情報を与えた(ピーク最大値)。
・SEC:差がHMWの0.5%以下であったとき、結果は同等であると考えた。
・DSC:Td1の差が0.4℃以下であったとき、結果は同等であると考えた。
第1のスクリーニング(アッセイ#H04−150から172)は、相Iのために開発されたHPLC方法で実施されていた。多数の試料を続けて分析したときに、HMWレベル判定に影響を与えたベースラインの変動に関する問題が観察されていた。
経時的HMW発生に関して異なる系列を比較するために、製剤相Iを参照として使用した:
・第1のスクリーニング(アッセイ#H04−150から172)については、35mg/mLのDP凍結乾燥物#H04−016でWFIによる再構成によって、3つの系列の各々についてFDS相Iを得た。
・第2のスクリーニング(アッセイ#H04−185から190)については、以前のFDS相I(H04−016)に加えて、3つの系列の各々について試験製剤と一緒に新たに製造された別のFDS相Iを使用した。さらに、第3のFDS相I参照(#LT−10006− DS)を使用した:FDSは凍結乾燥されずに解凍されるだけであった。
実施例7〜8で製造された全ての製剤は、少なくとも1.75%(w/v)のスクロースおよび約0.07%(w/v)のPS80を含有した。これらの濃度は公称値であり、リード抗体出発溶液に適用される希釈係数に基づく。PS80濃度に関して、UF/DFの間のPS80吸着は無視できると仮定された。これらの賦形剤濃度は、製剤相Iと同じだった。
この実施例でのpHは6.2から7.4でスクリーニングされ、この範囲の中で以下の全ての注射可能なバッファーが試験された:コハク酸塩、ヒスチジン、クエン酸塩、リン酸塩およびTris。
第1のスクリーニング(アッセイ#H04−150から172)では、実施例8−添加剤スクリーニング−でさらに詳述されるいくつかの賦形剤と一緒に、上記のバッファーを10mMの濃度で試験した。バッファー濃度は10mMに固定され、HMW形成へのその影響がバッファー濃度に関するセクションで見られることに留意する(実施例7)。
DSC結果は、熱安定性がわずかに低かった(Td1=64.7℃から65.1℃)ヒスチジン製剤を除いて、全ての製剤が参照と同等である(Td1=65.5℃)ことを示した(図26を参照する)。
pH6.2の製剤およびpH6.6のヒスチジン製剤を除いて、全ての製剤は透明で参照と同等だった。
・ヒスチジン製剤はRTで沈殿する(図27を参照する)
・RTでコハク酸塩製剤はわずかに乳白色であり、5℃でゲルの形成を観察することができ(図27を参照する)、それは、溶液をRTに戻して静かに振盪したときに可逆的であった。コハク酸塩は、キレート特性を有することができる。
RTにおいて、pH6.6のヒスチジン製剤は、参照よりわずかに乳白色であった。
ここで示す結果は3つの異なる系列からであったので、HMW発生を直接に比較することができず、参照と比較することができただけである。
・ヒスチジンpH6.6はRTで参照と同等で、5℃でわずかにより優れていた(ヒスチジン単独では+2.4%、参照については144時間で+3.2%)。
・クエン酸塩pH6.6はRTで参照よりわずかに優れ(クエン酸塩単独では+4.6%、参照については24時間で+5.2%)、5℃で参照と同等であった。
・リン酸塩pH6.6はRTで参照よりわずかに優れ(リン酸塩単独では+8.2%、参照については48時間で+9.0%)、5℃で参照と同等であった。
・リン酸塩pH7は、RTおよび5℃で参照と同等であった。
・TrispH7はRTで参照よりわずかに悪く(Tris単独では+7.8%、参照については48時間で+7.0%)、5℃で参照と同等であった。
・TrispH7.4は、RTおよび5℃で参照と同等であった。

10mMの濃度でのバッファー系スクリーニングに関して、以下が結論づけられる:
・約6.2(pIの近く)のpH値は、リード抗体の溶解性を低下させた:
−ゲル形成は、pH6.2のコハク酸塩で観察された
・さらに、溶液が乳白色でわずかに熱安定性が低いので、pH6.6でヒスチジンは避けるべきである
・試験したバッファー:ヒスチジン、クエン酸塩、リン酸塩およびTrisに関して、HMW形成に及ぼす6.6から7.4の範囲内のpHの影響に明らかな傾向はなかった
・クエン酸塩およびリン酸塩は10mMで最良のバッファーであるようだったが、HMW形成に関してバッファー効果は非常に弱かった
HMW形成に及ぼすpHの影響を同じ操作で判定するために、相I製剤(アッセイ#H04−187)でpHを6.7から7.2の範囲にわたってスクリーニングした。
DSC結果は、全ての製剤が参照と同等であることを示した(pH7.0)(図29を参照する)。
pHを6.7から7.2まで上昇させたとき、HMW形成のわずかな減少傾向があった(図30を参照する)が、これはRTで24時間後に有意になっただけである(pH6.7では+4.1%、pH7.2では+3.4%)。
バッファーリン酸塩2.2mM/Tris1.3mM(製剤相I)では、RTで24時間後に6.7から7.2の範囲のpHのわずかな影響があった。しかし、この効果は最初の16時間の間有意でなかったので弱かった。このことは、このpH範囲でのHMW形成へのpHの弱い効果を確認した。
HMW形成に及ぼすバッファー濃度の影響を調査するために、クエン酸塩およびリン酸塩を10mMより低い濃度:0、1.75および5.25mMで試験した(アッセイ#H04−185)。pHは、試験した範囲の中間点:pH=6.8に設定した。全ての製剤は、オスモル濃度を調整するために72mMのグリシン、ならびに結果および考察に関するセクションの最初で記載した通り、1.75%のスクロースおよび約0.07%のPS80を含有した。
DSC結果は、全ての製剤が参照(製剤相I)と同等であることを示した(図31を参照する)。バッファー無しのグリシン(185A2)は、他の試験製剤よりわずかに安定性が低かったことに留意する(Td1で約1℃の差)。
試験した両バッファーについて、バッファー濃度を低下させたときにHMW形成のわずかな減少傾向があり、最良の結果はバッファー無しおよびバッファーに1.75mMで得られた(例えば、RTの48時間でクエン酸塩10mMについては+7.8%およびバッファー無しでは+7.0%)(図32を参照する)。1.75mMのクエン酸塩およびバッファー無しは、参照と同等だった(例えば、RTの48時間でクエン酸塩については+7.1%およびバッファー無しでは+7.0%であったが、参照については+6.8%であった)。
バッファー濃度を低下させることは、HMW形成動態にわずかに安定させる効果を及ぼした。この効果は、おそらくイオン強度に関係があった。HMW形成の点でバッファー無しおよびバッファークエン酸塩1.75mMが最良の候補であり、わずかに劣ったリン酸塩1.75mMが続いた。グリシン72mMで試験したそれらのバッファーは、製剤相I/IIaと同等だった。
賦形剤の間の潜在的相乗効果を判定するために、添加剤グリシン、プロリン、ヒスチジンおよびTrisのスクリーニングは、バッファースクリーニングと組み合わせて行った(第1のスクリーニング:アッセイ#H04−150から172)。添加剤塩化ナトリウム、安息香酸ナトリウム、グリシンおよびスクロースのスクリーニング(第2のスクリーニング:アッセイ#H04−185から190)は、第1のスクリーニングから選択された最良のバッファーリン酸塩またはクエン酸塩で行った(実施例7)。
HMW形成に及ぼすイオン強度および疎水性相互作用の効果をそれぞれ評価するために、塩化ナトリウムおよび安息香酸ナトリウムを試験した。製剤相Iオスモル濃度(FDSで165mOsm/kg)を超えない濃度で、これらの添加剤をリン酸塩1.75mM pH6.8で試験した。
DSC結果は、NaCl製剤は参照と同等である(NaClでTd1=65.7℃、参照で65.6℃)が、安息香酸ナトリウム製剤は熱安定性がわずかに低い(Td1=65.0℃)ことを示した。
試験した両添加剤に関して、参照と比較してHMW形成に明らかな負の効果があった(例えば、RTの24時間で、38.5mMのNaClでは+5.2%、37.8mMの安息香酸ナトリウムでは+5.2%、および参照では+3.2%)。発生は両賦形剤で類似しているので、NaClの結果だけをここで示す(図33を参照する)。
バッファー濃度について見られる通り、NaClまたは安息香酸ナトリウムでイオン強度を増加させることは、HMW形成動態に明らかな負の効果を及ぼした。さらに、安息香酸ナトリウムの添加によるHMW動態に及ぼす疎水性相互作用の効果は、観察されなかった。
これらの添加剤(グリシン、プロリン、ヒスチジンおよびTris)は、10mMの濃度で試験した(第1のスクリーニング:アッセイ#H04−150から172)。
DSC結果は、上記の添加剤を有する全ての製剤がそれらのない製剤(バッファーだけ)と同等であることを示した。
製剤間の差は弱かったが、RTで48時間後および5℃で6日後に傾向を見ることができた:

HMW形成に関して、これらの添加剤の効果は弱かったが、傾向を見ることができた:
・10mMのグリシンは、わずかな正の効果を有した
・10mMのヒスチジンおよびプロリンは、効果を有しないようだった
・10mMのTrisは、安定化効果を有さず、わずかな不安定効果を有することさえできた
第2のスクリーニング(アッセイ#H04−185)では、2.4%のスクロースの追加の量(合計4.15%(w/v)のスクロースを与えた、全ての製剤に含有された1.75%の既存のスクロースに加えて)および72mMの濃度のグリシンを、選択された最良のバッファーで試験した(バッファースクリーニングに関するセクションを参照する)。製剤相Iのオスモル濃度(FDSで165mOsm/kg)を超えないように、これらの濃度を最大にした。
DSC結果は、全てのスクロース2.4%およびグリシン72mM製剤が参照と同等であったことを示した(例えば、スクロースの場合、リン酸塩1.75mMではTd1=65.7℃、参照では65.6℃)。
クエン酸塩1.75mMおよびバッファー無しについてスクロースを含有する製剤と比較したときだけ、グリシンのわずかな正の効果が確認された(図34を参照する)。しかし、この正の効果は、RTで48時間後に有意なだけだった。
HMW形成に関しては、対象のタイムスケール(RTで<24時間)で、最良のバッファー候補で試験したスクロースおよびグリシン製剤は、製剤相Iと同等であった。
この研究のために、約50個の製剤を製造して、現行の相I/IIa製剤と並べて比較した。pHスクリーニングは6.2から7.4にわたり、賦形剤の間の潜在的相乗効果を評価するために、単独、またはいくつかの賦形剤(全てGRAS)と組み合わせたこのpH範囲内の全ての注射可能なバッファーを含んだ。
・約6.2のpH値は、リード抗体溶解性を低下させる:コハク酸塩およびヒスチジンによりゲルの形成および沈殿がそれぞれ観察された;
・さらに、溶液が乳白色でわずかに熱安定性が低いので、pH6.6でヒスチジンは避けるべきである;
