Description
[1]β−シクロデキストリン又はその誘導体を有効成分とするGLP-1分泌促進剤。
[2]前記誘導体がヒドロキシブチル−β−シクロデキストリンである、[1]に記載のGLP-1分泌促進剤。
[3]前記ヒドロキシブチル−β−シクロデキストリンの置換度が1〜9である、[2]に記載のGLP-1分泌促進剤。
[4]糖尿病、肥満、食後高血糖又は神経変性疾患の予防又は改善、糖尿病に伴う神経障害の改善、グルカゴン分泌抑制、胃排泄や胃酸分泌の抑制、食欲や摂食の抑制、又は膵β細胞の増殖促進のために用いられる、[1]〜[3]のいずれか一項に記載のGLP-1分泌促進剤。
[5][1]〜[3]のいずれか一項に記載のGLP-1分泌促進剤を含有する、糖尿病又は肥満の予防又は改善用組成物。
[6]医薬、医薬部外品又は食品である、[5]に記載の組成物。
1.方法
マウス大腸由来のGLP-1産生細胞株GLUTag細胞を、10% FBS含有DMEM(Dulbecco’s modified eagle’s medium, high glucose、SIGMA)を用い、CO2インキュベーター内(37℃、CO2 濃度5%)で培養した。80%コンフルエントになったところで、各種供与試料を最終濃度として10mg/mLになるように培地を添加し2時間培養後、培養上清中に分泌されたGLP-1量をELISAキットにより測定した。キット添付のプロトコールに従い測定した。結果は非投与群(コントロール群)の値を1とした時の相対的な分泌量(Fold of Control)として表した。統計処理は、Dunnet’s testを用いた。コントロール群と試料投与群間においてP<0.05を有意差ありとした。検討した供与試料は以下の通りである。
(1)シクロデキストリン類
セルデックスA-100(製品名、日本食品化工株式会社。α−シクロデキストリンの含有量98.5%以上)、セルデックスB-100(製品名、日本食品化工株式会社。β−シクロデキストリンの含有量98.0%以上)、セルデックスG-100(製品名、日本食品化工株式会社。γ−シクロデキストリンの含有量98.5%以上)
(2)シクロデキストリン誘導体類
セルデックスHB-β-CD(製品名、日本食品化工株式会社。β−シクロデキストリンの水酸基をヒドロキシブチル基で修飾した誘導体。平均するとβ−シクロデキストリン1分子中に約5個のヒドロキシブチル基が導入されている。)
実験結果を図1及び2に示す。コントロールと比較してβ−シクロデキストリン(B-100)は有意にGLP-1分泌を促進させた。ヒドロキシブチル−β−シクロデキストリン(HB-β-CD)のGLP-1分泌促進効果は一層高く、β−シクロデキストリンの水酸基にヒドロキシブチル基が導入されるとGLP-1分泌促進作用が高まることがわかる。
Claims (5)
- ヒドロキシブチル−β−シクロデキストリンを有効成分とするGLP-1分泌促進剤。
- 前記ヒドロキシブチル−β−シクロデキストリンの置換度が1〜9である、請求項1に記載のGLP-1分泌促進剤。
- 糖尿病、肥満又は食後高血糖の予防又は改善、グルカゴン分泌抑制、胃排泄や胃酸分泌の抑制、食欲や摂食の抑制、又は膵β細胞の増殖促進のために用いられる、請求項1又は2のいずれか一項に記載のGLP-1分泌促進剤。
- 請求項1又は2に記載のGLP-1分泌促進剤を含有する、糖尿病又は肥満の予防又は改善用組成物。
- 医薬、医薬部外品又は食品である、請求項4に記載の組成物。
