感光性樹脂組成物(PHOTEC RD−1225:厚さ25μm、日立化成株式会社製)を250mm角の基板(MCL−E679F:MCLの表面にピーラブル銅箔を貼り合わせた基板、日立化成株式会社製)の片面にラミネートした。ラミネート条件はロール温度90℃、圧力0.3MPa、コンベア速度2.0m/分で行った。
電気Niめっきの代わりに電気銀めっきを用いた以外は、実施例1と同様に表面に生体適合性高分子を有する金めっきフィルターを作製した。銀めっき液はシルブレックス400(日本エレクトロプレイティング・エンジニヤース株式会社:商品名)を用いた。めっき温度は25℃、電流密度1.5A/dm2の条件でめっきを行い、約1μm/分の条件で約20μmとなるように電気めっきを行った以外は実施例1と同様の条件でめっきを行った。
電気Niめっきの代わりに電気パラジウムめっきを用いた以外は、実施例1と同様に表面に生体適合性高分子を有する金めっきフィルターを作製した。電気パラジウムめっき液はパラディックスLF−5(日本エレクトロプレイティング・エンジニヤース株式会社:商品名)を用いた。めっき温度は50℃、電流密度1A/dm2の条件でめっきを行い、約4.2分/μmの条件で約20μmとなるように電気めっきを行った以外は実施例1と同様の条件でめっきを行った。
MCLのピーラブル銅箔の代わりにピーラブルニッケル箔を用いた(電気めっき後はニッケル箔を除去)。さらに電気Niめっきの代わりに電気銅めっきを用いた以外は、実施例1と同様に表面に生体適合性高分子を有する金めっきフィルターを作製した。電気銅めっき液はミクロファブCu200(日本エレクトロプレイティング・エンジニヤース株式会社:商品名)を用いた。めっき温度は25℃、電流密度3A/dm2の条件でめっきを行い、約1.5分/μmの条件で約20μmとなるように電気めっきを行った以外は実施例1と同様の条件でめっきを行った。
置換金めっき後に還元金めっきを行わなかったこと以外は、実施例1と同様に表面に生体適合性高分子を有する金めっきフィルターを作製した。尚、金めっき厚は0.05μmであった。
置換金めっき後に還元金めっきを行い、表面処理を行わなかったこと以外は実施例1と同様に金めっきフィルターを作製した。
置換金工程以外は、実施例1と同様に表面に生体適合性高分子を有する金めっきフィルターを作製した。
金めっきを行わなかったこと以外は、実施例1と同様に表面に生体適合性高分子を有するフィルターを作製した。
小細胞癌細胞株であるNCI−H358細胞を10%ウシ胎児血清(FBS)を含むRPMI−1640培地にて、37℃、5%CO2条件下にて静置培養した。トリプシン処理により培養皿から細胞を剥離させて回収し、リン酸緩衝液(Phosphate buffered saline、PBS)を用いて洗浄した後に、10μM CellTracker Red CMTPX(ライフテクノロジーズジャパン株式会社)にて37℃、30分間静置させることで、NCI−H358細胞を染色した。その後、PBSにて洗浄し、トリプシン処理にて37℃にて3分間静置させることで、細胞塊を解離させた。その後、培地にてトリプシン処理を停止させ、PBSにて洗浄後、2mMEDTA及び0.5%ウシ血清アルブミン(BSA)を含むPBS(以下2mM EDTA−0.5% BSA−PBSという。)に懸濁した。尚、PBSはリン酸緩衝生理食塩水であり和光純薬工業製、製品コード166−23555を用いた。EDTAは2Na(エチレンジアミン−N,N,N’,N’−4酢酸二ナトリウム塩二水和物)(和光純薬工業製、製品コード345−01865)を用いた。
実施例又は比較例のフィルターをセットしたCTC回収装置:CTC SEPARATOR(日立化成株式会社製、仮の商品名)を用いて実験を行った。CTC回収装置は血液サンプル又は試薬を導入するための流路を備えており、流路の入り口は、シリンジを加工して作製したリザーバーに接続した。このリザーバーに、血液サンプル及び試薬を順次投入していくことで、CTCの捕捉、染色、洗浄などの操作を連続的に容易に行えるようにした。
先に述べたように、多くの金属イオンは細胞毒がある為、回収した癌細胞の解析を行う際、障害になる。そこで、以下の条件で金属イオンの測定を行った。
溶出の割合(ppm)=全溶出イオン/フィルター質量
ドロップマスター500(協和界面科学製、商品名)を用いてフィルターの開口部以外の接触角の測定を行った。
結果を表2に示す。実施例1に示すように、非シアン系の置換めっきと還元めっきにポリマー処理を行うことで金属イオンの溶出を抑制できる。親水性の高分子を処理しているので、接触角が下がり、濡れ性が向上し、貫通孔周辺の気泡発生を抑制できる。結果的にCTCの回収率、白血球の残存率が良好である。実施例4はNiの代わりにCuを用いているので、金属の溶出を低減できる。実施例2と3はNiの代わりに貴金属を用いているので、金属イオンが溶出しない。実施例5は無電解金めっきを施していない比較例1に比べて、フィルターの変色が抑えられ、Niイオンの溶出も少ない。しかし、実施例5は還元型の金めっきを行わなかったので、金めっきの厚みが0.05μmと薄い。従って、フィルターがやや変色し、実施例1に比べてNiイオンの溶出が増える結果となった。実施例6は生体適合性の高分子処理を行っていないので、フィルター表面が撥水性であり、気泡が発生しやすい。Niイオンの溶出も多くなる傾向がある。実施例7はシアン系の置換金を用いている。実施例7は比較例1に比べて、Niイオンの溶出は少ない。シアンは金属Niを溶解する効果が高く、金めっきの被覆率が悪くなる傾向がある。従ってNi溶出が多くなるため、非シアンを用いている実施例1に比べて劣る傾向がある。この点で、シアン金よりも非シアン金のほうが優れていることが分かった。比較例1は金めっきを行っていない。Niの溶出が大きい上、外観が大幅に劣化する。さらにCTCの濃縮率及び回収率も低下する。さらに、Niは酸化被膜が厚い為、Auに比べて表面処理が困難である。従って接触角が高い値を示している。

- 生体物質捕獲用のフィルターの貫通孔の開口形状が長方形及び角丸長方形からなる群から選択される一つ以上の形状を含み、その短辺の長さが5μm以上15μm以下であることを特徴とする、請求項1〜10のいずれか一項に記載の生体物質捕獲用のフィルター。
- 生体物質捕獲用のフィルターの膜厚が3μm以上50μm以下であることを特徴とする、請求項1〜11のいずれか一項に記載の生体物質捕獲用のフィルター。
