Description
驚くべきことに、特定のピロロキノロン誘導体が5−HT6受容体拮抗薬であることが見出された。本発明は、一般式(XIV)の化合物:

R1、R2は独立に、水素、非置換アルキル(C1−C3)基、1以上のハロゲン原子で置換されたアルキル(C1−C3)基、アルコキシ(C1−C3)基、または独立にシアノ、ニトロ、アミノ、ヒドロキシルから選択される基を表し;
Tは、CO、CH2、置換アルキル(C1−C2)基、SO、SO2を表し;
Arは、非置換アリール(5〜6員)、ビアリール(8〜10員)、N、O、Sからなる群から独立に選択される1〜3個のヘテロ原子を有するヘテロアリール(5〜6員)、ヘテロアリール(8〜10員)を表し、これらはアルキル(C1−C3)基、1以上のハロゲン原子、アルコキシ(C1−C3)基、アルケニル(C2−C4)、ハロゲン、ニトロ、ヒドロキシル、シアノ、アミノ、アルキルアミノ、カルボキサミドから選択される1以上の置換基で置換されたアルキル(C1−C3)基で置換されていてもよく;

R1、R2が独立に水素、1以上のハロゲン原子で置換されていてもよい、メチル、エチル基を表し、シアノ、ニトロ、アミノ、ヒドロキシル、メトキシルから選択される基で独立に置換され;
TがCO、CH2、置換アルキル(C1−C2)基、SO2を表し;

一般式(XIV)の化合物、もしくは互変異性体、立体異性体、N−オキシド、または前記のいずれかの薬理学的に許容される塩、水和物もしくは溶媒和物に関する。
・式(XIV)の化合物、そのプロドラッグ、薬学上許容される塩および溶媒和物と薬学上許容される担体とを含んでなる、5−HT6伝達の妨害から起こる障害を治療するための医薬組成物;
・5−HT6受容体の遮断によって治療可能な障害または病態を治療する方法であって、そのような治療を必要とする哺乳動物に式(XIV)の化合物またはその薬学上許容される塩を投与することを含んでなる方法;
・式(XIV)の化合物またはその薬学上許容される塩と薬学上許容される担体とを含んでなる、本明細書に挙げられた障害から選択される障害または病態を治療するための医薬組成物;
・5−HT6受容体に拮抗作用を及ぼす方法であって、それを必要とする対象に有効量の化合物を投与することを含んでなる方法
を含む。
a)アセトニトリル、エタノール、イソプロパノール、DMFまたはDMSOから選択される極性溶媒中、DABCO(1,4−ジアザビシクロ[2.2.2]オクタン)、キヌクリジンまたは3−ヒドロキシキヌクリジンなどの第3級アミンおよびSc(OTf)3、Yb(OTf)3、Ti(Oi−Pr)4およびCu(OTf)2から選択されるルイス酸の存在下で行われるアザ−ベイリス−ヒルマン(aza-Bayliss-Hillman)反応。このプロセスは一般に室温で24〜48時間行われる、
b)極性溶媒、好ましくは、アルコール、アセトニトリルまたはDMF中、t−BuOK、t−BuONa、K2CO3、Cs(CO3)2、TEAから選択される強塩基の存在下、臭化アリルを用いたA−1のアルキル化の際のジエン誘導体A−2の生成、
c)誘導体A−2の閉環メタセシス反応。このプロセスは一般に、ジクロロメタンまたはトルエン中、3〜10mol%のルテニウム触媒を用い、加えてマイクロ波照射により補助して行われる、
d)得られたピロリンA−3の、強塩基を用いた処理によるピロール誘導体A−4の生成。この反応は一般に、t−BuOK、t−BuONa、K2CO3、Cs(CO3)2またはTEAから選択される塩基の存在下、DMFまたはDMSOなどの好適な溶媒中で行われる、
e)ニトロ誘導体A−4のそのアミノ類似体A−5への還元。このアミノ誘導体への変換は、5〜10mol%のパラジウム/活性炭を用い、水素雰囲気下で行われる従来のプロセスである、
f)極性プロトン性溶媒、例えば、2−メトキシエタノール、イソプロパノール、n−BuOH、sec−BuOH、t−BuOH中、酸性条件での、化合物A−5のラクタムA−6への環化、
g)化合物A−6を高温下、POCl3、SOCl2、PCl5などの塩素化薬剤で処理することによる、ラクタム誘導体A−6のそのクロロ類似体A−7への変換、
h)一般構造A−8のアミン/エーテル置換ピロロキノリンの生成、このプロセスは、トルエン、ベンゼン、キシレン、テトラヒドロフランおよびジオキサンなどの非極性溶媒、またはアセトン、アセトニトリル、DMF、DMSOから選択される極性溶媒から選択される種々の溶媒を用いて行うことができる。この反応は一般に、70〜140℃で2〜24時間行われ、多くの場合、マイクロ波照射により補助される、
i)ピロロキノリンA−8の、t−BuOK、t−BuONa、NaOH、NaH、TEA、DIEAまたはホスファゼンから選択される強塩基またはBTPPなどのホスファゼン塩基の存在下での各種置換アリールスルホニルハライド、アリールアシルハライドまたはアリールアルキルハライド誘導体を用いた処理による、最終生成物XA−9の生成、Boc保護アミンの場合は、この最終生成物は酸性条件下で脱保護した。
本明細書に開示される化合物の既述に使用される一般用語は、それらの通常の意味を有する。用語アルキルは、本明細書で使用する場合、一価飽和分岐または直鎖炭化水素鎖を表す。特に断りのない限り、このような鎖は1〜3個の炭素原子を含み得る。このようなアルキル基の代表例はメチル、エチル、プロピル、イソプロピルなどである。同じ炭素含有量が本特許の用語「アルカン」、および「アルコキシ」などの派生語にも当てはまる。種々の炭化水素含有部分の炭素含有量は、その部分の炭素原子の最小数と最大数を表す接頭辞により示され、すなわち、接頭辞Cx−Cyは、整数「x」から整数「y」(含む)まで存在する炭素原子数を定義する。例えば、「アルキル(C1−3)」はメチル、エチル、n−プロピルまたはイソプロピルを意味する。
合成は、特に断りのない限り、周囲温度で行った。有機溶媒(Aldrich and Chempurから入手)は試薬級のものであり、精製せずに使用した。試薬はAldrich、Chembridge、Fluorochemから入手した。

エーテル誘導体A−8(w,x)の一般製造手順
化合物A−7(400mg、2mmol、1当量)をDMFに溶かし、Cs2CO3(775mg、2.38mmol、1.2当量)を加えた。臭化ベンジル(270μl、2.18mmol、1.1当量)を滴下した。この反応を室温で30分間行った。次に、この混合物をAcOEtで希釈し、水(3×)およびブライン(1×)で洗浄し、Na2SO4で乾燥させ、濾過し、減圧下で濃縮した。残った粗物質をクロマトグラフィーカラムにて、展開溶媒としてAcOEt/Hex 2/8(v/v)を用いて精製した。得られた化合物A−8u(500mg、1.71mmol、1当量)をPd2(dba)3(31mg、0.03mmol、0.02当量)、BINAP(42mg、0.07mmol、0.04当量)およびt−BuOK(268mg、2.4mmol、1.4当量)とともに混合した。この混合物をトルエン(10ml)に懸濁させ、1−Boc−3−ヒドロキシピロリジン(382mg、2.00mmol、1.2当量)を加えた。この反応物に115℃で1時間マイクロ波照射を行った。得られた混合物を濃縮し、シリカゲルにて、展開溶媒としてAcOEt/Hex 3/7(v/v)を用いて精製した。化合物A−8w(620mg、1.40mmol、1当量)をDMSOに懸濁させ、t−BuOK(1.25g、11.2mmol、8当量)を固体として加えた。このフラスコを油浴中に置き、混合物に空気をバブリングさせた。反応は70℃で30分間行い、脱保護エーテル誘導体A−9xを得た。次に、この混合物を水で希釈し、AcOEt(3×40mL)で抽出した。MgSO4で乾燥させた後、これを濃縮し、シリカにて、展開溶媒としてAcOEt/Hex 4/6(v/v)を用いて精製した。
一般構造A−9のピロロ[3,2−c]キノリン誘導体の一般製造手順
化合物A−8(0.28mmol、1当量)をDCM(5ml)に溶かした。この混合物を氷浴に置き、塩化スルホニルまたは塩化アリールアシルまたはアリールアルキルハライド(0.50mmol、1.8当量)を少量ずつ加えた。反応はカリウムtert−ブトキシド、水素化ナトリウム、炭酸セシウムまたはBTPPから選択される強塩基の存在下で行い、3時間実施した。次に、この混合物を蒸発させ、残った黄色粗物質をシリカゲルで精製した。一般構造A−9の1H−ピロロ[3,2−c]キノリンのBoc保護アリールスルホンアミド、アリールアミドおよびアリールアルキル誘導体の総てをLC−MS法および1H NMR法により同定した(代表的化合物5’、21’、27’、29’、43’、46’、48’に関してデータを示す)。それらをジオキサン中4NのHCl溶液を用いてさらに脱保護して、最終的なアミン/エーテル−置換ピロロキノリンA−9のアリールスルホンアミド誘導体を得た。
合成された化合物の、クローニングされたセロトニン:5−HT1A、5−HT2A、5−HT6、5−HT7およびドーパミンD2L受容体に対する親和性および選択性の特性を決定するために放射性リガンド結合アッセイを用いた。これはクローニングされたヒト受容体(総てHEK293細胞で安定に発現する)からの各放射性リガンドの置換により達成された:5−HT1ARに対して[3H]−8−OH−DPAT、5−HT2ARに対して[3H]−ケタンセリン、および5−HT6Rに対して[3H]−LSD、5−HT7Rに対して[3H]−5−CT、およびD2Rに対して[3H]−ラクロプリド。
細胞培養および細胞膜の調製
ヒトセロトニン5−HT1AR、5−HT2A,5−HT6、5−HT7bRまたはドーパミンD2LRを安定発現するHEK293細胞(総てリポフェクタミン2000の使用により作製)を、5%CO2を含む加湿雰囲気中、37℃で維持し、10%透析済みウシ胎仔血清および500mg/ml G418スルフェートを含有するダルベッコの改変イーグル培地中で増殖させた。膜調製のために、細胞を直径10cmのディッシュで継代培養し、90%の集密度まで増殖させ、37℃に予温したリン酸緩衝生理食塩水(PBS)で2洗浄し、0.1mM EDTAおよび1mMジチオトレイトールを含有するPBS中での遠心分離(200g)によりペレットとした。膜調製まで、ペレットを−80℃℃で保存した。
細胞ペレットを解凍し、Ultra Turrax組織ホモジナイザーを用いて20倍容量のアッセイバッファー中でホモジナイズし、4℃、35000gで20分間2回(間に37℃で15分間のインキュベーション)遠心分離した。アッセイバッファーの組成は次の通りであった:5−HT1ARの場合:50mM Tris−HCl、0.1mM EDTA、4mM MgCl2、10mMパルギリンおよび0.1%アスコルビン酸;5−HT2ARの場合:50mM Tris−HCl、0.1mM EDTA、4mM MgCl2および0.1%アスコルビン酸;5−HT6Rの場合:50mM Tris−HCl、0.5mM EDTAおよび4mM MgCl2;5−HT7bRの場合:50mM Tris−HCl、4mM MgCl2、10mMパルギリンおよび0.1%アスコルビン酸;ドーパミンD2LRの場合:50mM Tris−HCl、1mM EDTA、4mM MgCl2、120mM NaCl、5mM KCl、1.5mM CaCl2および0.1%アスコルビン酸。
ラット大脳皮質におけるα1受容体(Greengrass et al., 1979)、HEK−293細胞で発現させたH1受容体(Smit et al., 1996)および5−HT2C受容体(Stam et al.)、ならびにCHO細胞.、1994)で発現させたM1受容体(Dorje et al., 1991)およびD3受容体(Mackenzie et al., 1994)に対する、選択された化合物の親和性を決定するために、放射性リガンド結合アッセイを用いた。これは、各放射性リガンドの置換により達成された:α1Rに対して[3H]プラゾシン、H1Rに対して[3H]ピリラミン、M1Rに対して[3H]ピレンゼピン、5−HT2CRに対して[3H]メスレルギンおよびD3Rに対して[3H]メチル−スピペロン。
CHO−ヒト−5HT6−エクオリンアッセイはEuroscreen、Brussels(Euroscreen、Technical dossier、ヒト組換えセロトニン5−HT6−A1受容体、DNAクローンおよびCHO AequoScreen(商標)組換え細胞株、カタログ番号:ES−316−A、2003年2月)から購入した。ヒト−5HT6−エクオリン細胞は、ミトコンドリア標的アポ−エクオリンを発現する。活性エクオリンを再構成するためには、細胞にセレンテラジンを付加しなければならない。作動薬がヒト5−HT6受容体に結合した後、細胞内カルシウム濃度は上昇し、アポ−エクオリン/セレンテラジンへのカルシウムの結合は、セレンテラジンの酸化反応をもたらし、その結果、アポ−エクオリン、セレンテラジン、CO2および光(λmax 469nm)が生じる。この発光応答は作動薬濃度に依存する。発光はMicroBeta Jet(Perkin Elmer)を用いて測定する。化合物の促進効果はpEC50として表される。化合物の拮抗効果は10−6Mのα−メチルセロトニンにより誘導される発光の阻害として求め、pA2はチェン−プルソフ(Cheng-Preushoff)式に従って計算した。化合物は2回の独立した実験で試験し、2反復で行った。
腹腔内(i.p.)投与について:ガラスビーカー中の所望の量の固体化合物に必要容量の無菌注射水を加え、化合物が完全に溶解するまでガラム棒で撹拌した。
動物 体重220〜250gの雄ウィスターラットを試験に用いた。これらの動物をポリカーボネートMakrolonタイプ3ケージ(寸法26.5×15×42cm)にて4群で飼育した。総ての動物を環境制御室(周囲温度22±2℃;相対湿度50〜60%;12:12明:暗周期、8:00に点灯)で維持した。動物を試験開始前に1週間環境に順応させた。標準実験食(Ssniff M−Z)および濾過水を自由に摂らせた。試験は明期の09.00〜14.00時の間に行った。試験開始の1時間前に、順応のためにラットを試験室に移した。

の化合物から選択される、請求項1または2に記載の一般式(XIV)の化合物。 - 光学的に活性な鏡像異性体である、請求項1〜3のいずれか一項に記載の化合物。
- 式(XIV)および式(XIX)(式中、総ての記号は請求項1およびスキーム1で定義された内容と同義である)の化合物の製造方法であって、
a)極性溶媒中、DABCO(1,4−ジアザビシクロ[2.2.2]オクタン)、キヌクリジンまたは3−ヒドロキシキヌクリジンなどの第3級アミンおよびSc(OTf)3、Yb(OTf)3、Ti(Oi−Pr)4およびCu(OTf)2から選択されるルイス酸の存在下で行われるアザ−ベイリス−ヒルマン反応、
b)極性溶媒中、t−BuOK、t−BuONa、K2CO3、Cs(CO3)2、TEAから選択される強塩基の存在下、臭化アリルを用いた式A−1の化合物:
c)場合によりマイクロ波照射により補助される、ジクロロメタンまたはトルエン中、3〜10mol%のルテニウム触媒を用いた式A−2の誘導体の閉環メタセシス反応、
d)塩基の存在下、DMFまたはDMSOなどの好適な溶媒中での強塩基を用いた、生成した式A−3のピロリン:
e)式A−4のニトロ誘導体の式A−5のそのアミノ類似体:
f)極性プロトン性溶媒中、酸性条件での、式A−5の化合物の式A−6のラクタム:
g)式A−6の化合物を高温下、POCl3、SOCl2、PCl5などの塩素化薬剤で処理することによる、式A−6のラクタム誘導体の式A−7のそのクロロ類似体:
h)場合によりマイクロ波照射により補助される、非極性溶媒または極性溶媒から選択される溶媒を用いた、式A−7の化合物からの一般構造XIX:
i)ピロロキノリンXIXの、強塩基またはホスファゼン塩基の存在下での各種置換アリールスルホニルハライド、アリールアシルハライドまたはアリールアルキルハライド誘導体を用いた処理による、最終生成物XIV:
の工程を含んでなる、製造方法。 - 請求項1〜4のいずれか一項に記載の式XIVの化合物もしくは一般式(XIV)の化合物の互変異性体、立体異性体、N−オキシド、もしくは同位体標識類似体、または前記のいずれかの薬理学的に許容される塩、水和物もしくは溶媒和物を含んでなる医薬組成物。
- 薬学上許容される担体または希釈剤をさらに含んでなる、請求項6に記載の医薬組成物。
- 5−HT6伝達の妨害から起こる疾患、障害または病態の治療または予防に用いるための、請求項6または7に記載の医薬組成物。
- 疾患、障害または病態が、統合失調症、不安、鬱病、躁鬱病、癲癇、強迫性障害、気分障害、片頭痛、アルツハイマー病、加齢性認知機能低下、軽度認知障害、睡眠障害、摂食障害、食欲不振症、過食症、パニック発作、注意欠陥多動性障害、注意欠陥障害、パーキンソン病、ハンチントン病、コカイン、エタノール、ニコチンまたはベンゾジアゼピン類の乱用からの離脱症状、疼痛、肥満および2型糖尿病、機能性腸障害および過敏性腸症候群からなる群から選択される、請求項8に記載の医薬組成物。
- 少なくとも1つの追加の治療薬を組み合わせてなる、請求項6〜9のいずれか一項に記載の医薬組成物。
- 追加の治療薬が、5−HT6伝達の妨害から起こる疾患、障害または病態の治療または予防に用いるための別の医薬である、請求項10に記載の医薬組成物。
- 疾患、障害または病態が、統合失調症、不安、鬱病、躁鬱病、癲癇、強迫性障害、気分障害、片頭痛、アルツハイマー病、加齢性認知機能低下、軽度認知障害、睡眠障害、摂食障害、食欲不振症、過食症、パニック発作、注意欠陥多動性障害、注意欠陥障害、パーキンソン病、ハンチントン病、コカイン、エタノール、ニコチンまたはベンゾジアゼピン類の乱用からの離脱症状、疼痛、肥満および2型糖尿病、機能性腸障害および過敏性腸症候群からなる群から選択される、請求項11に記載の医薬組成物。
- 光学的に活性な鏡像異性体である、請求項13に記載の化合物。
