Description
アルビグルチドの試料を、表Iに記載される、分析用調製マトリクス(analytical preparation matrix)に従って凍結乾燥用に調製した。凍結乾燥物(lyophile)を、適切な濃度のt−ブタノール、エタノール、およびアセトニトリル(1、5、および10%w/v);ならびに適切な濃度の酢酸NH4およびNH4HCO3(100mMおよび250mM)を含有するバイアル(2mL 13mm)中に、117mMトレハロース、153mMマンニトール、0.01%(w/v)ポリソルベート−80と共に、10mMリン酸ナトリウム(pH7.2)中の50mgのアルビグルチドを含有する0.750mLの溶液を凍結乾燥することによって、調製した。
凍結乾燥物を、凍結乾燥させたバイアルストッパー(ツベルクリン注射器、ベクトン・ディッキンソン社製品番号309626)を通し、1mL注射器および25G(5/8”)針を介して、0.675mLの注射用水(WFI)で再構成した。0.675mLの体積で、乾燥成分の体積を補正し、0.75mLの最終体積を得た。「振盪法(shaking method)」または「静置法(undisturbed method)」のいずれかを用いて、再構成を行った。振盪法については、試料を10秒間振盪し、その後15秒間静置した。静置法については、試料を5秒間前後に揺すり、その後静置した。バイアル中に粒子が見えなくなったとき、試料が再構成されたとみなした。両方の再構成法の結果を、表1と、さらに図1および図2に示す。
アルビグルチドの試料を、表Iに記載の分析用調製マトリクスに従って、凍結乾燥用に調製した。凍結乾燥物を、適切な濃度のt−ブタノール(34mM〜202mM、または0.25〜2%w/v)を含有するバイアル(2mL 13mm)に、117mMトレハロース、153mMマンニトール、0.01%(w/v)ポリソルベート−80と共に、10mMリン酸ナトリウム(pH7.2)中の50mgのアルビグルチドを含んでなる、0.750mLの溶液を、凍結乾燥することによって調製した。
凍結乾燥物を、凍結乾燥させたバイアルストッパー(ツベルクリン注射器、ベクトン・ディッキンソン社製品番号309626)を通し、1mL注射器および25G(5/8」)針を介して、0.675mLの注射用蒸留水(WFI)で再構成した。0.675mLの体積で、乾燥成分の体積を補正し、0.75mLの最終体積を得た。「静置法」(水を添加した後すぐに軽く混合し、その後バイアルを放置した)を用いて、再構成を行った。水の添加は、分析者(再構成工程を実行する異なる3人)に基づく4つのバリエーションおよびピペッティング法で行った:
分析者1:ストッパーを通じた注射器による再構成
分析者2:ストッパーを通じた注射器による再構成
分析者3:ストッパーを通じた注射器による再構成。
分析者1:従来のP1000 Raininピペッティング法による再構成。
抗NOGOmAb(配列番号3および4)、抗TNFR1dAb(配列番号5)、IL18(配列番号6)、抗IL5(配列番号7および8)、および抗CD20(可変領域の配列番号:9および10)を、約0.75mLの体積で、26mMヒスチジン、150mMトレハロース、0.02%ポリソルベート80(PS80)を含んでなる製剤(pH6.0)中で、以下に示される濃度に濃縮し、次に2mLバイアル中で凍結乾燥した。

29.1mgの抗TNFR1dAb

Claims (21)
- ポリペプチド組成物の製造方法であって、ポリペプチドと揮発性添加剤とを混合して液体混合物を形成し、前記液体混合物を凍結乾燥して凍結乾燥ポリペプチド組成物を得ることを含んでなり、前記液体混合物が約0.25体積%〜約5体積%の揮発性添加剤を含んでなりかつ該揮発性添加剤がt−ブタノールである、方法。
- 前記ポリペプチドが抗原結合性ポリペプチドである、請求項1に記載の方法。
- 前記ポリペプチドが、ヒト血清アルブミンのN末端に遺伝子操作により融合された少なくとも2つのGLP−1ポリペプチドを含んでなる、請求項1に記載の方法。
- 前記抗原結合性ポリペプチドが、可溶性受容体、抗体、抗体断片、免疫グロブリン単一可変領域、Fab、F(ab’)2、Fv、ジスルフィド連結Fv、scFv、閉構造多特異的抗体、ジスルフィド連結scFv、および二重特異性抗体からなる群から選択される、請求項2に記載の方法。
- 液体ポリペプチド組成物の製造方法であって、請求項1に記載の方法によって製造された凍結乾燥ポリペプチドを得、該凍結乾燥ポリペプチドを、十分な量の薬学的に許容可能な分散剤で再構成して液体ポリペプチド組成物を得ることを含んでなる、方法。
- 前記ポリペプチドが抗原結合性ポリペプチドである、請求項5に記載の方法。
- 前記ポリペプチドがヒト血清アルブミンのN末端に遺伝子操作により融合された少なくとも2つのGLP−1ポリペプチドを含んでなる、請求項5に記載の方法。
- 前記抗原結合性ポリペプチドが、可溶性受容体、抗体、抗体断片、免疫グロブリン単一可変領域、Fab、F(ab’)2、Fv、ジスルフィド連結Fv、scFv、閉構造多特異的抗体、ジスルフィド連結scFv、および二重特異性抗体からなる群から選択される、請求項6に記載の方法。
- 凍結乾燥ポリペプチド組成物の再構成時間を減少させる方法であって、
a)前記ポリペプチドを含んでなる液体混合物を凍結乾燥し、ここで、前記液体混合物は約0.25体積%〜約5体積%の揮発性添加剤を含んでなりかつ該揮発性添加剤はt−ブタノールであり、かつ、
b)前記凍結乾燥ポリペプチドを、前記凍結乾燥ポリペプチド組成物に対し十分な量の薬学的に許容可能な分散剤で再構成して液体ポリペプチド組成物を得ることを含んでなり、 ここで、揮発性添加剤の存在下で凍結乾燥されたポリペプチドを再構成させる時間が、揮発性添加剤の非存在下で凍結乾燥された同一のポリペプチドを再構成させる時間より短い、方法。 - 前記ポリペプチドが抗原結合性ポリペプチドである、請求項9に記載の方法。
- 前記ポリペプチドがヒト血清アルブミンのN末端に遺伝子操作により融合された少なくとも2つのGLP−1ポリペプチドを含んでなる、請求項9に記載の方法。
- 前記抗原結合性ポリペプチドが、可溶性受容体、抗体、抗体断片、免疫グロブリン単一可変領域、Fab、F(ab’)2、Fv、ジスルフィド連結Fv、scFv、閉構造多特異的抗体、ジスルフィド連結scFv、および二重特異性抗体からなる群から選択される、請求項10に記載の方法。
- 治療用ポリペプチド、緩衝液、界面活性剤、少なくとも1つの賦形剤、および少なくとも1つの揮発性添加剤を含んでなり、前記揮発性添加剤がt−ブタノールでありかつ該t−ブタノールが約0.25体積%〜約5体積%で存在する、組成物。
- 前記治療用ポリペプチドがGLP−1またはその断片および/もしくは変異体を含んでなる、請求項13に記載の組成物。
- 前記治療用ポリペプチドが、ヒト血清アルブミンのN末端に遺伝子操作により融合された少なくとも2つのGLP−1(7−36(A8G))ポリペプチドを含んでなる、請求項13に記載の組成物。
- 前記治療用ポリペプチドが配列番号1を含んでなる、請求項13〜15のいずれか一項に記載の組成物。
- 前記緩衝液がリン酸ナトリウムである、13または16のいずれか一項に記載の組成物。
- 前記界面活性剤がポリソルベート−80である、請求項13〜17のいずれか一項に記載の組成物。
- 前記少なくとも1つの賦形剤が、トレハロース、マルトース、スクロース、マンノース、ラクトース、マンニトール、ソルビトール、グリセロールおよびデキストロースから選択される、請求項13〜18のいずれか一項に記載の組成物。
- 前記賦形剤がトレハロースおよびマンニトールを含んでなる、請求項13〜19のいずれか一項に記載の組成物。
- 配列番号1を含んでなるポリペプチド、リン酸ナトリウム、トレハロース、マンニトール、ポリソルベート−80およびt−ブタノールを含んでなり、該t−ブタノールが約0.25体積%〜約5体積%で存在する、組成物。
