本発明の別の態様は、本発明による抗体またはその機能的フラグメントの1つの生産方法であって、
a)本発明による宿主細胞を培地中、好適な培養条件で培養する工程;および
b)このようにして生産された該抗体またはその機能的フラグメントの1つを培養培地または該培養細胞から回収する工程
を含んでなることを特徴とする方法に関する。
より詳しくは、cMetの発現に関連する腫瘍形成性障害をin vitroにおいて診断するため、またはcMetの発現に関連する腫瘍形成性障害、例えば、cMetの発現に関連する癌の発症の予後をin vivoにおいて決定するための、記載されているような本発明による抗体またはその機能的フラグメントもしくは誘導体の使用が意図される。
好ましい実施形態では、診断用の抗体は、組織サンプルがホルマリン固定され、パラフィン包埋された際に標的化された受容体と結合できなければならない。
一例として、サンプルはcMet発現レベルに関して、抗体染色レベルが0〜3+のスケールでスコアを付けることができ、この場合、0は陰性であり、1+〜3+は強度が増す4つの判定量段階での陽性染色を表す。各陽性スコアはスコア0(陰性)と比べて再発および致死的疾患のリスクの有意な軽減と関連づけられるので、スコア1+〜3+は陽性と記録されるが、これらの陽性スコア間の強度の増加はさらなるリスク軽減をもたらし得る。cMetの予後値を評価するためには、従来の任意のハザード分析法が使用可能である。代表的な分析法としては、打ち切り例の存在下で、生存または時間事象データをモデル化するためのセミパラメトリック法であるコックス回帰分析(Hosmer and Lemeshow, 1999; Cox, 1972)が挙げられる。生命表またはカプラン−マイヤーなどの他の生存分析とは対照的に、コックスではモデルに予測因子変数(共変量)を含める。従来の方法、例えば、コックスを用いれば、原発腫瘍におけるcMet発現状態と、疾患の再発(無病生存時間または転移までの時間)、またはその疾患から死亡するまでの時間(全生存時間)のいずれかの発生までの時間との相関に関する仮説を検証することができる。コックス回帰分析はまた、コックス比例ハザード分析としても知られる。この方法は、患者の生存時間に対する腫瘍マーカーの予後値を検定するために標準的なものである。多変量様式で用いる場合、いくつかの共変量の効果が並行して検定されるので、独立した予後値を有する個々の共変量、すなわち、最も有用なマーカーが特定できる。腫瘍の陽性または陰性「cMet状態」[cMet(+)またはcMet(−)とも記載される]はそれぞれスコア0またはスコア1+〜3+を表す。
好ましい実施形態では、本発明によるプロセスは、スケール0〜3+の適当なスケールを表し、腫瘍細胞の膜反応性が無い場合は0、10%を超える腫瘍細胞に強い完全な反応性が見られる場合には3+のスコアが付けられる。
本発明の特定の態様では、腫瘍はスコア3+のcMet(+)である。
本発明のもう1つの特定の態様では、腫瘍はスコア2+または3+のcMet(+)である。
例えば、上述の標識の使用など、当業者により、任意の標識法を使用することができると理解される。
i)該標識抗体とc−Metの間の結合程度を検出するのに有用な試薬と、
ii)c−Met発現レベルのスコア付けに有用な陽性対照サンプルおよび陰性対照サンプル
をさらに含んでなる。
−免疫誘導工程
抗cMet抗体を作製するため、8週齢のBALB/cマウスを、その原形質膜上にcMetを発現するCHOトランスフェクト細胞系統で皮下に3〜5回(20×106細胞/用量/マウス)免疫するか、または初回免疫誘導用の完全フロインドアジュバントと追加免疫誘導用の不完全フロインドアジュバントを混合した、cMet細胞外ドメイン融合タンパク質(10〜15μg/用量/マウス)(R&D Systems、カタログ番号358MT)またはこの組換えタンパク質のフラグメントで2〜3回免疫した。マウスにCHO−cMeT細胞と組換えタンパク質の双方を施す混合プロトコールも実施した。細胞融合の3日前に、マウスに組換えタンパク質またはフラグメントでi.p.またはi.v.に追加免疫を行った。次いで、マウスの脾臓を摘出し、SP2/0−Agl4骨髄腫細胞(ATCC)と融合し、HAT選択を行った。一般に、とりわけ、ネズミ起源の、モノクローナル抗体またはそれらの機能的フラグメントの調製については、特に、手引書"Antibodies" (Harlow and Lane, Antibodies: A Laboratory Manual, Cold Spring Harbor Laboratory, Cold Spring Harbor NY, pp. 726, 1988)に記載されている技術またはKohler and Milsteinによって記載されたハイブリドーマ調製技術(Nature, 256:495-497,1975)を参照することができる。
−224D10についてのスクリーニング工程
図3Bに示される結果により、m224G10が、cMet標的療法に対して特に感受性が高いことが分かっているU87−MG異種移植腫瘍上のcMetを認識し得ることが立証された。予想通り、IgG1アイソタイプ対照では染色が観察されなかった(図3A)。これらの結果に基づき、224D10 Mabを利用して腫瘍上のcMetにスコアをつけることができるかどうかを判断するために、実験を計画した。
−221C9についてのスクリーニング工程
滴定曲線は図5に示している。どちらのcMet受容体形態に対しても同様の親和性が観察された。これらのELISAを行うために、ヒト二量体cMetタンパク質(R&D sytems、カタログ番号358MT)を、PBS中0.25μg/mlの濃度で、4℃で一晩コーティングする。プレート(Costar #3690)を0.5%ゼラチン溶液により37℃で2時間飽和させた後、ハイブリドーマ上清を37℃で1時間インキュベートする。一度、PBSで洗い流し、抗マウスHRP−抗体(Jackson ImmunoResearch、カタログ番号115−035−164)をELISAバッファー(PBS中、0.1%ゼラチン/0.05%Tween 20)での1/5000希釈溶液で各ウェルに加え、これらのプレートを37℃で1時間インキュベートする。PBSで3回洗浄した後、50μlのTMB基質(Uptima)を加えることによりペルオキシダーゼ(peroxydase)の活性を明らかにする。この反応を室温で5分間生じさせる。50μl/ウェルの1M H2SO4溶液を加えることにより反応を停止させ、プレートリーダーにより450nmで読み取る。単量体cMetに対しても同様のプロトコールを実施し、その場合には、タンパク質を5μg/mlでコーティングした。
上記のプロトコールを用いて、可変レベルのcMetを発現しているパラフィン包埋ヒト腫瘍組織セットを、m224D10 Mabおよびm221C9 Mabそれぞれで染色した。
m224D10 Mabについては図6に、m221C9 Mabについては図7に示される結果により、2つの腫瘍タイプでは、m224D10およびm221C9の双方とも、可変レベルのcMetを有するヒト腫瘍を識別することができることが立証された。これらの抗体を使用して、腫瘍を以下のように評価することができる:
1+:10%を超える腫瘍細胞でわずかに観察される染色、
2+:10%を超える腫瘍細胞で観察される中程度の完全な膜染色、
3+:10%を超える腫瘍細胞で強い完全な染色。
すでに上に述べたように、診断用Mabは、標的受容体のダウンレギュレーションを誘導する治療用抗体に対する「応答マーカー」としても使用することができる。その点に関して、処置した患者において、血液または生検材料の採取を行い、cMet状態について解析することができる。そのために、使用する診断用抗体は、治療用抗体によってターゲッティングされるエピトープとは異なるエピトープを認識しなければならない。治療用抗体は通常、HGFに置き換わることができるため、リガンド置換について競合しない診断用抗体の選択は、総ての治療用Mabに対する応答マーカーとして有用であり得る。
この実施例では、224D10を応答マーカーとして使用し得ることを立証するために、224D10と多くの治療用Mabとの競合実験を実施した。
診断用Mabをさらに特性評価するために、HGF競合アッセイを実施した。
試験するMabの存在下または不在下でcMetタンパク質を含んでなる第1の反応混合物を、独立した飽和(PBS 1X中0.5%ゼラチン)プレートで調製する。ネズミ抗体(参照および研究するMab)の連続1:2希釈(12カラムで40μg/mlから出発)を行う。次いで、0.8μg/mlのrh cMet−Fcタンパク質を加える(RDSystems、参照358−MT/CF)。但し、陰性対照系統はELISA希釈液(PBS 1X中、0.1%ゼラチン、0.05%Tween 20)しか含まない。ホモジナイゼーション後、競合サンプルを、PBS中0.3μg/ml rhHGF溶液(RDSystems、参照294−HGN/CF)が入ったHGFコーティングプレート上に添加する。インキュベーションと数回の洗浄の後に、ヤギ抗ヒトIgG−HRP(Jackson、参照109−035−098)を使用して、結合しているcMetタンパク質を検出する。一度、結合させたら、TMB基質をプレートに加える。H2SO4酸性溶液を加えることにより反応を停止させ、得られた光学濃度を、マイクロプレートリーダーを使用して450nmで読み取る。
Claims (22)
- i)少なくとも次の3つのCDR、すなわち、IMGTナンバリング体系に従って定義されるCDR−H1、CDR−H2およびCDR−H3(ここで、CDR−H1は配列番号55の配列を含んでなり、CDR−H2は配列番号56の配列を含んでなり、かつ、CDR−H3は配列番号57の配列を含んでなる)を含んでなる重鎖;および/またはii)少なくとも次の3つのCDR、すなわち、IMGTナンバリング体系に従って定義されるCDR−L1、CDR−L2およびCDR−L3(ここで、CDR−L1は配列番号58の配列を含んでなり、CDR−L2は配列番号59の配列を含んでなり、かつ、CDR−L3は配列番号60の配列を含んでなる)を含んでなる軽鎖を含んでなる、c−Metタンパク質と特異的に結合することができる単離された抗体、またはその機能的フラグメントであって、かつ
a)
IMGTに従って定義される次の3つのCDR、それぞれ配列番号7の配列を有するCDR−H1、配列番号2の配列を有するCDR−H2、および配列番号8の配列を有するCDR−H3を含んでなる重鎖と、
IMGTに従って定義される次の3つのCDR、それぞれ配列番号4の配列を有するCDR−L1、配列番号5の配列を有するCDR−L2、および配列番号6の配列を有するCDR−L3を含んでなる軽鎖と
を含んでなる抗体、またはその機能的フラグメント;および
b)
IMGTに従って定義される次の3つのCDR、それぞれ配列番号29の配列を有するCDR−H1、配列番号30の配列を有するCDR−H2、および配列番号31の配列を有するCDR−H3を含んでなる重鎖と
IMGTに従って定義される次の3つのCDR、それぞれ配列番号32の配列を有するCDR−L1、配列番号33の配列を有するCDR−L2、および配列番号34の配列を有するCDR−L3を含んでなる軽鎖と
を含んでなる抗体、またはその機能的フラグメント
からなる群から選択される、抗体、またはその機能的フラグメント。 - a)配列番号13のアミノ酸配列を含んでなる配列の重鎖可変ドメインと配列番号14のアミノ酸配列を含んでなる配列の軽鎖可変ドメインとを含んでなる、抗体またはその機能的フラグメント;および
b)配列番号40のアミノ酸配列を含んでなる配列の重鎖可変ドメインと配列番号41のアミノ酸配列を含んでなる配列の軽鎖可変ドメインとを含んでなる、抗体またはその機能的フラグメント
からなる群から選択される、請求項1に記載の抗体またはその機能的フラグメント。 - ネズミ抗体としての、請求項1または2に記載の抗体またはその機能的フラグメント。
- リガンドHGFの、c−Metタンパク質への結合を遮断しない、請求項1〜3のいずれか一項に記載の抗体またはその機能的フラグメント。
- 2008年3月12日にI−3949番としてCNCM, Institut Pasteur, Paris, Franceに寄託されたハイブリドーマおよび2010年1月14日にI−4273番としてCNCM, Institut Pasteur, Paris, Franceに寄託されたハイブリドーマから選択される、請求項1〜4のいずれか一項に記載の抗体を分泌することができるネズミハイブリドーマ。
- 次の核酸:
a)請求項1〜4のいずれか一項に記載の抗体をコードする核酸、DNAまたはRNA;
b)配列番号15〜26または42〜52の配列からなる群から選択される配列を含んでなるDNA配列を含んでなる核酸;
c)配列番号27、28、53または54の配列を含んでなるDNA配列を含んでなる核酸;
d)a)、b)またはc)に定義される核酸の対応するRNA核酸;または
e)a)、b)およびc)に定義される核酸の相補的核酸。 - cMetの発現に関連する腫瘍形成性障害をin vitroにおいて診断するのを補助するため、またはcMetの発現に関連する腫瘍形成性障害の発症の予後をin vitroにおいて判定するのを補助するための、請求項1〜4のいずれか一項に記載の抗体またはその機能的フラグメントの使用。
- 被験体からのcMet発現腫瘍の存在および/または場所をin vitroにおいて検出する方法であって、(a)被験体からのサンプルを請求項1〜4のいずれか一項に記載の抗体またはその機能的フラグメントと接触させる工程、および(b)該抗体とサンプルとの結合を検出する工程を含んでなる、方法。
- 被験体からのcMet発現腫瘍におけるcMetの発現レベルをin vitroで検出する方法であって、(a’)被験体からのサンプルと請求項1〜4のいずれか一項に記載の抗体またはその機能的フラグメントを接触させる工程、および(b’)該サンプルにおいてcMetと結合する抗体のレベルを定量する工程を含んでなる、方法。
- c−Met発現レベルが、免疫組織化学(IHC)によって測定される、請求項9に記載の方法。
- cMet発現腫瘍をin vitroにおいて診断するのを補助する、または被験体におけるcMet発現腫瘍の発症に関して予後をin vitroにおいて判定するのを補助する方法であって、(i)請求項9に従ってcMetの発現レベルを判定する工程、および(ii)工程(i)の発現レベルを、正常組織のcMetの参照発現レベルと比較する工程を含んでなる、方法。
- 被験体の腫瘍のc−Met状態をin vitroで判定する方法であって、(1)請求項9に従ってc−Metの発現レベルを判定する工程、(2)該腫瘍のc−Met発現レベルのスコアを付ける工程、および(3)該スコアを対照サンプルから得られたものと比較する工程を含んでなる、方法。
- 前記スコア付けが、染色強度および陽性細胞のパーセンテージである2つのパラメーターに基づく適当なスケールの使用を含んでなる、請求項12に記載の方法。
- 前記適当なスケールが、0〜3+のスケールであり、腫瘍細胞に膜反応性がない場合は0、および10%を超える腫瘍細胞に強い完全な反応性が見られる場合は3+のスコアが付けられる、請求項13に記載の方法。
- 前記適当なスケールが、0〜3 + のスケールであり、腫瘍細胞に膜反応性がない場合は0;10%を超える腫瘍細胞にわずかな認知可能な膜反応性が見られる場合には1+;10%を超える腫瘍細胞に弱い〜中程度の完全な膜反応性が見られる場合には2+;および10%を超える腫瘍細胞に強い完全な反応性が見られる場合には3+のスコアが付けられる、請求項14に記載の方法。
- 腫瘍が、スコア2+または3+のc−Met(+)である、請求項14または15に記載の方法。
- 腫瘍形成性障害が、抗c−Met抗体またはそのフラグメントによる処置に感受性があるかどうかを判定する方法であって、(a)請求項16に従って被験体の腫瘍のc−Met状態をin vitroにおいて判定する工程、および(b)その状態がc−Met(+)であれば、その腫瘍形成性障害は抗cMet抗体またはそのフラグメントによる処置に感受性があると判定する工程を含んでなる、方法。
- 請求項1〜4のいずれか一項に記載の抗c−Met抗体またはその機能的フラグメントを少なくとも含んでなる、キット。
- 該抗c−Met抗体とc−Metの間の結合程度を検出するのに有用な試薬をさらに含んでなる、被験体においてc−Met発現腫瘍の存在および/または場所をin vitroで検出するための請求項18に記載のキット。
- 該抗c−Met抗体とc−Metの間の結合レベルを定量するのに有用な試薬をさらに含んでなる、c−Met発現腫瘍におけるc−Metの発現レベルをin vitroで判定するための請求項18に記載のキット。
- i)該抗c−Met抗体とc−Metの間の結合程度を検出するのに有用な試薬と、
ii)c−Met発現レベルのスコア付けに有用な陽性対照サンプルおよび陰性対照サンプル
をさらに含んでなる、腫瘍のc−Met状態をin vitroで判定するための請求項18または19に記載のキット。 - ネズミ抗体に特異的なポリクローナル抗体をさらに含んでなる、腫瘍のc−Met状態をin vitroで判定するための請求項21に記載のキット。
