Description
アレルギーは、環境(空気/水/食品)中のタンパク質に対する過敏反応であると考えられる。アレルゲンは、アトピー性患者がIgE抗体に反応し、その後アレルギー反応をもたらす抗原である。複合体または融合タンパク質における抗原は、環境アレルゲン(例えば、ハウスダスト、ダニ、樺の木花粉、草の花粉、ネコ抗原、ゴキブリ抗原)、または食品アレルゲン(例えば、ウシミルク、ピーナッツ、エビ、ダイズ)、または両方の組合せであり得る。IgE分子は、エフェクター細胞(マスト細胞、好塩基球および好酸球)活性化において機能するために重要である。最近、IgEは、B細胞および樹状細胞(DC)の抗原捕捉可能性を上流調節することによって、低親和性(CD23)および高親和性受容体(FcεRI)[1]の両方を通じて、アレルギー性疾患の誘導期において重要な役割を担い得ることが認められた。IgE抗体の負の機能は、アレルゲン特異的IgG抗体によって中和される。例えば、それらはB細胞から離れて単球およびDCに免疫応答を導くことが理由である[2]。さらに、それらは、アレルゲン結合部位に対してIgE分子と競合する。こうして、アレルゲン特異的IgG分子の誘導により、アレルギーを、処置、治療および予防する。
背景:
CD32は単球/樹状細胞およびB細胞上で強く発現する、そのため本発明の該分子は、B細胞によるアレルゲン提示を防止する意図でもって、特に樹状細胞(DC)によるアレルゲン提示を促進している間、免疫応答をこれらの重要な免疫細胞に誘導するよう設計され、後者はワクチンとして製剤され得る分子または分子複合体におけるアレルゲンに対するTh1応答の誘導を導く。最近の知識から、樹状細胞(DC)の2つの型:ミエロイド(mDC)および形質細胞様樹状細胞(pDC)[6] が存在することが示され、これはDC1およびDC2細胞の新しい2つの概念をもたらした[7]。この概念において、DC1細胞は、抗原の特異的刺激の後にTh1細胞成長の誘導を促進し、DC2細胞はTh2細胞の成長をサポートする。単球誘導性DC(またはmDC)は、一般的にDC1型であると考えられ、一方pDCはDC2型であると考えられる[6]。DCの両方の型がCD32を発現し、アレルゲン特異的T細胞応答を誘導する。しかしながら、この結果はTh1型であることを保証するものではない。実際には、アレルギー性ドナーにおいてTh2応答となりやすい[8]。重要なことには、pDCは、CpG-ODNs(非メチル化CpGモチーフを含有するオリゴデオキシヌクレオチド[ODNs])を結合するTLR9受容体を発現する。pDCにおけるこの受容体の活性化は、IFN-αおよびIL-12の非常に強力な産生をもたらし[9]、Th1誘導を促進し、こうして潜在的なDC2をDC1細胞に変換する。
・アレルゲン-特異的B-細胞を活性化しない、アレルゲン特異的Th1細胞の活性化および誘導
・アレルゲン特異的IgEまたはIgGにより、選択されたT細胞エピトープが本発明の分子または分子複合体において提示される天然のアレルゲンによって活性化されるようになり得るマスト細胞またはその他のエフェクター細胞を活性化しない、アレルゲン特異的Th1細胞の活性化および誘導。
本発明は、トル様受容体9(TLR9)およびFcγ受容体RII(CD32)と結合出来、そして少なくとも一つの抗原の少なくとも一つのエピトープを包含する分子または分子複合体を提供する。

・完全な抗体または抗体フラグメント(例えば、Fab)いずれかをコードしている構築体(PCT/EP2006/050059にしたがって)において、CD32および/またはTLR9に特異的に結合する、操作されたイムノグロブリンドメインを包含する、小分子突然変異イムノグロブリンドメイン、
・本発明の内容において、CD32への結合は単量体または多量体イムノグロブリンFc領域またはその一部によって、特にFc部分のCD32に対する親和性が増強される場合に達成され、一方TLR9結合は抗体の通常の結合部位(VH/VL)によって達成される、
・CD32に結合し、他のTLR9特異的結合モチーフと融合される、イムノグロブリンのFc-領域またはその部分
・上記抗体のFc部分は、ヒトFcγR's[15]に対する親和性を増加させるように"糖操作され"得るあらゆる種類のTLR9および/またはCD32に対する特異的に結合する修飾されたスキャッホルドを使用して、必要な場合には一緒に結合される。これらの結合スキャッホルドは、タンパク質ドメイン、フィブロネクチンIII、リポカリン、タンパク質Aまたはα-アミラーゼ阻害剤、アンキリン反復タンパク質、C2 ドメイン、A-ドメイン、EGFR様ドメイン、dab、chi-bAb、CTLA-4、γ結晶性またはその他のタンパク質、タンパク質ドメインまたはその一部であり得る。
1. CD32aとの相互作用が、受容体-結合構造複合体の内在化、受容体の細胞質尾部におけるITAMモチーフを介する抗原提示細胞の活性化および結合/融合されたT細胞エピトープの抗原提示をもたらす。
2. CD32bとの相互作用が、ITIMモチーフを介した受容体のネガティブシグナル伝達をもたらす。
3. 相互作用は、ヒトおよびサルCD32間に交差反応性を示す(対応するインビボモデルにおける有効性試験のために)
4. 相互作用は、マウスCD32との交差反応性を示す(アレルギーのための確立されたインビボモデルにおける試験のために)。
1. TLR9との相互作用はシグナル変換およびサイトカイン産生をもたらす。

抗CD32抗体に関するこの得られたVHおよびVLをCH1およびCL各々と融合する。CLは、SMID 技術(PCT/EP2006/050059)を用いて予め構築されており、下記に示したとおりTLR9と結合するようにファージディスプレイを用いて選択される。CH1をそのC-末端で選択されたT細胞エピトープをコードする配列と融合する。これらの2つの融合タンパク質コード化遺伝子を、発現ベクター中にクローニングして、2つの独立した遺伝子(または2つの独立した発現ベクター中に)を発現させ、細菌、酵母または動物細胞またはあらゆる他の適切な発現系において共発現させる。こうして、所望の特徴を有するFab、即ちCD32に結合する、TLR9に結合する、および対応するT細胞エピトープを担持するFabが産生される。
・TLR9 に対するscFvを、ファージディスプレイライブラリーからまたは存在するハイブリドーマから得る、そしてCD32結合分子を、CH2-CH4、CH3-CH4またはCH1-CH4、または小分子突然変異イムノグロブリンドメインライブラリーから得る。これらの2つのコード配列は、共に結合され、T細胞エピトープをコードする配列が結合される。次いで、融合タンパク質を、細菌、酵母または動物細胞またはその他適切な発現系において発現させる。

・抗TLR9 抗体の重鎖および軽鎖遺伝子は全体として見られる。重鎖遺伝子において、CH2(またはCH1またはCH3またはCH4)領域は、先に設計された、CD32(小分子成熟イムノグロブリンドメイン)に結合するためにファージディスプレイを用いて選択されるCH2(またはCH1またはCH3またはCH4またはCLまたはVHまたはVL)領域によって置換される。CH1、CH2、CH3またはCH4を、そのC末端でT細胞エピトープをコードする配列に融合する。これらの2つの遺伝子も、発現ベクターにクローニングして、動物細胞において発現させる。
・2つの異なる特異性(TLR9およびCD32)を有する1つの小分子成熟イムノグロブリンドメインがT細胞エピトープと組み合わされる。
本発明の分子または分子複合体の一部分である抗原は、IgEとの結合を回避するあらゆる可能な方法で処理される完全なアレルゲン、変性アレルゲンまたはあらゆる抗原であり得る。かかる処置は、Leroyら[20]によって記載された患者のIgE抗体として同一エピトープに対する高親和性IgM、IgD、IgAまたはIgG抗体を用いる抗原タンパク質のエピトープシールディングするエピトープからなり得る。また、かかる抗体は、IgE特異的エピトープ付近にも結合でき、こうして立体障害によりIgE抗体の結合を防止し得る。

本発明の融合タンパク質は利用可能な許容し得る医薬的製剤のいずれかにおいて製剤され得るが、好ましくはワクチンとして製剤される。本発明の融合タンパク質のCD32結合部は、関連細胞を選択する。これらの細胞上のCD32のトリガーは、受容体に加えて接合融合タンパク質の内在化を積極的に誘導し、そうすることでTLR9と融合タンパク質のTLR9結合部の相互作用を促進し、関連抗原提示細胞[10;11]の細胞質内で発現される。
下記例示はより詳細に本発明を説明するものであるが、それらを限定するものではない。
1st パンニングラウンド: 1 mg/ml TLR-9 ペプチド
2nd パンニングラウンド: 500 μg/ml TLR-9 ペプチド
3rd パンニングラウンド: 100 μg/ml TLR-9 ペプチド
可溶性発現のためのTLR-9に対して選択されたヒトCL変異株の選択されたクローンのクローニング
3つのパンニング・ラウンドから選択されたファージ由来のファージミドDNAを、mid-prepを用いて単離した。DNAコード突然変異CL-領域を、PCRによりバッチ増幅させ、NcoI-NotIを、クローニングを促進させるために挿入されたNotI 制限部位を有するE. coli発現ベクターpBAD/Myc-His (Invitrogen)であるベクターpNOTBAD/Myc-Hisにクローニングした。連結した構築体を、エレクトロポーレーションによりE. coli LMG194 細胞(Invitrogen)に形質転換し、30℃にて1%グルコースおよびアンピシリンを含むTYE培地で一晩増殖させた。選択されたクローンを、アンピシリンを含む2xYT 培地(200 μl)中に接種し、一晩30℃で増殖させて、L-アラビノースを0.1%.の終濃度まで添加することによって誘導した。16℃で一晩発現後に、該細胞が遠心分離により収集し、4℃にて一晩、ペリプラスム抽出物の調製のためにNa-ホウ酸塩緩衝液(pH 8.0)(100μl)により処理した。ペリプラスム抽出物(50 μl)をELISAに使用した(下記を参照されたい)。
選択したクローンを、ELISAによりTLR-9ペプチドに対する特異的結合を評価した。

軽鎖カセットを、pPICLA23 (4235 bp)からBglII(pos.1)およびBamHI (pos. 2319)を用いて切り出し、2319 bp フラグメントを、分取ゲル・エレクトロポーレーションにより精製した。1916 bp フラグメントを除いた。ベクター pPICHA23 (4219 bp)をBamHIで消化し、pPICLA23由来の予め精製した2319 bpフラグメントを挿入した。2つの発現カセット、VL-CH3 融合タンパク質に対するものおよびVH-CH3 融合タンパク質に対するものを担持する得られたPichia pastoris発現ベクターを、両方の挿入物が転写が同じ方向を持ち得るようにスクリーニングされる。次いでベクターpPICHLA23 (6537 bp)を直線化して、Pichia pastoris中に形質転換する、例えばBamHIまたはBssSIを用いるエレクトロポーレーションにより Pichia pastoris中に形質転換し、そして陽性形質転換体をZeocinにより選択した。いくつかのクローン組換えタンパク質の発現のためにスクリーニングした。次いで、クローンをより大きな規模の産生のために選択し、組換え体融合タンパク質を、標準的方法を用いて固定化金属親和性クロマトグラフィーにより精製した。全てのPichiaの操作、培養および発現等は、下記標準的プロトコール(Invitrogen)により為される。
