モル収率97%、化学的純度99.66% HPLC(A%)。
2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン ハイドロクロライド(D)の水素化。
化学的純度(面積、HPLC):塩酸リトドリン98.1%、スレオ異性体(threo isomers)1.3%。
2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン ハイドロクロライド(D)の水素化。
化学的純度(面積、HPLC):塩酸リトドリン98.1%、スレオ異性体1.2%。
4−((1RS,2SR)−1−ヒドロキシ−2−{[2−(4−ヒドロキシフェニル)エチル]アミノ}プロピル)フェノール ハイドロクロライド(塩酸リトドリン)Form I。
アセトンで溶媒和された−4−((1RS,2SR)−1−ヒドロキシ−2−{[2−(4−ヒドロキシフェニル)エチル]アミノ}プロピル)フェノール ハイドロクロライド(塩酸リトドリン)。
酸リトドリンを得た。
化学的純度:99.84% HPLC(A%)。そのX線回折図形は、表3に示した、アセトンで溶媒和された塩酸リトドリンのX線回折図形に相当する。
エチルアセトンで溶媒和された−4−((1RS,2SR)−1−ヒドロキシ−2−{[2−(4−ヒドロキシフェニル)エチル]アミノ}プロピル)フェノール ハイドロクロライド(塩酸リトドリン)。
14.5gの塩酸リトドリンを確保した。
化学的純度:99.25% HPLC(A%)。そのX線回折図形は、表4に示した、エチルアセトンで溶媒和された塩酸リトドリンのX線回折図形に相当する。
4−((1RS,2SR)−1−ヒドロキシ−2−{[2−(4−ヒドロキシフェニル)エチル]アミノ}プロピル)フェノール ハイドロクロライド(塩酸リトドリン)モノハイドレート。
そのX線回折図形は、表5に示した塩酸リトドリンモノハイドレートのX線回折図形と同じである。
アセトンで溶媒和された塩酸リトドリンの塩酸リトドリンForm Iへの変態。
アセトンで溶媒和された塩酸リトドリンの塩酸リトドリンForm Iへの変態。
塩酸リトドリンモノハイドレートの塩酸リトドリンForm Iへの変態。
エチルアセテートで溶媒和させた塩酸リトドリンの塩酸リトドリンForm Iへの変態。
塩酸リトドリンForm IIの調製。
塩酸リトドリンForm IIの塩酸リトドリンForm Iへの変態。
塩酸リトドリンの再結晶化。
化学的純度:99.9% HPLC(A%)。
塩酸リトドリンForm Iの安定度。
塩酸リトドリンForm IIの安定度。
Claims (9)
- 塩酸リトドリンを調製するための方法であって、
(a)4−(2−ブロモプロピオニル)フェニルメタンスルホネートを、ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミンと反応させて、4−(2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−プロピオニル)−フェニルメタンスルホネートを得る工程と、 (b)4−(2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−プロピオニル)−フェニルメタンスルホネートを加水分解して、2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オンを得る工程と、 (c)2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オンを塩化する工程と、 (d)2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン ハイドロクロライドを接触水素化させて、98:2のエリトロ/トレオ混合物として塩酸リトドリンを確保する工程とを有する、塩酸リトドリンを調製するための方法。 - 前記工程(b)における加水分解を塩基によって実施し、該塩基が、アルカリ金属及び又はアルカリ土類金属の水酸化物,アセトン水とメチルエチルケトン水との混合物によって作られる溶媒中の水酸化カリウム又は水酸化ナトリウム、又は、それらの混合物である、請求項1に記載の方法。
- 2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オンが、結晶化によって、アセトン、メチルエチルケトン、アセトン水とメチルエチルケトン水との混合物、又は、それらの混合物から単離される、請求項2に記載の方法。
- 前記塩化が、有機溶媒中でガス状になっている又は溶解している塩酸を用いることにより起こり、該有機溶媒が、トルエン、又は、エタノール、又は、メタノール、又は、水、又は、アセトン、又は、メチルエチルケトン、又は、エチルアセテート、又は、それらの混合物である、請求項1に記載の方法。
- 2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]アミノ}−1−(4−オキシ−フェニル)−プロパン−1−オン メタンスルホネートを、2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン ハイドロクロライド塩に直接変態させる、請求項1に記載の方法。
- 2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン ハイドロクロライドの接触水素化が、2バールと12バールとの間の圧力で且つ20℃と45℃の間の温度の水素雰囲気中で、アルコール、又は、水と混合されたアルコールである有機溶媒中のパラジウム炭素(Pd/C)によって構成される触媒を用いることにより生じる、請求項1に記載の方法。
- 2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン、又は、それの塩である化合物。
- 化学式が、2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−オキシ−フェニル)−プロパン−1−オン メタンスルホネートである、請求項7に記載の化合物。
- 化学式が、2−{ベンジル−[2−(4−ベンジルオキシフェニル)−エチル]−アミノ}−1−(4−ヒドロキシ−フェニル)−プロパン−1−オン ハイドロクロライドである、請求項7に記載の化合物。
