Description
Rは、C1−C4の直鎖または側鎖アルキル、C1−C4の直鎖または側鎖ヒドロキシアルキル、またはC1−C4の直鎖または側鎖スルホアルキルであり、好ましくは、メチル(methyl)、ヒドロキシプロピル(hydroxypropyl)、2−ヒドロキシプロピル(2−hydroxypropyl)、またはスルホブチル(sulfobutyl)であり、より好ましくは、2−ヒドロキシプロピルである。
高脂肪食餌(high fat diet)による肥満誘発マウスモデルにおける、HPBCDの自主飲水による体重変化および食餌摂取量の測定
雄C57BL/6Jのマウス(5週齢、KRIBB、Korea)を1週間順化後、高脂肪食餌(high fat diet)(60%fat kcal、Research Diets)を15日間給与して肥満を誘導した。肥満誘発動物は体重を均等に3匹ずつ群分離をした後、溶媒対照群に滅菌蒸溜水を自主飲水させ、ヒドロキシプロピル−ベータシクロデキストリン(2−hydroxypropyl−β−cyclodextrin、HPBCD)は、滅菌蒸溜水に溶解し、10、15および20%(W/V)の濃度で3日間自主飲水させた。試験期間の間、すべての動物に対して毎日1回体重変化を測定し、体重測定日に飼料の定量を給与した後、翌日に残量を測定し、1群あたりの摂取量(g/day)として算出した。
高脂肪食餌による肥満誘発マウスモデルにおける、HPBCDの自主飲水による体脂肪および肝重量変化の測定
上記<実施例1>の肥満誘発動物において、HPBCDは10、15および20%(W/V)の濃度で3日間自主飲水後、マウスの剖検を実施した。マウスはCO2ガスを用いて致死後開腹し、皮下脂肪、生殖器脂肪、内臓脂肪、肝臓を摘出してから、化学秤(chemical balance)を用いて重量を測定した後、3匹の平均重量を算出した。
高脂肪食餌マウスにおける、HPBCDの経口投与による体重変化および食餌摂取量の測定
雄C57BL/6Jのマウス(5週齢、KRIBB、Korea)を1週間順化後、動物は体重を均等に5匹ずつ群分離をした後、高脂肪食餌(high fat diet)(60% fat kcal)の給与と同時に、滅菌蒸溜水およびHPBCD10、20および40%(W/V)を、マウス20g体重あたり0.2mlずつ、5日間、1日3回(朝、昼、夕方)経口投与した。試験期間の間、すべての動物に対して毎日1回体重変化を測定し、体重測定日に飼料の定量を給与した後、翌日に残量を測定し、1匹あたりの平均摂取量(g/mouse/day)として算出した。
高脂肪食餌マウスにおける、HPBCDの経口投与による体脂肪および肝重量変化の測定
上記<実施例3>の動物順化および投与条件で、HPBCDを10、20および40%(W/V)の濃度で5日間経口投与後、マウスの剖検を実施した。マウスはCO2ガスを用いて致死後開腹し、皮下脂肪、生殖器脂肪、内臓脂肪、肝臓を摘出してから、化学秤(chemical balance)を用いて重量を測定した後、5匹の平均重量を算出した。
高脂肪食餌マウスにおける、BCDの経口投与による体重変化の測定
上記<実施例3>において、HPBCD10、20および40%(W/V)の代わりに、BCD10、20%(W/V)を用いることが異なるだけで、残りは同一の条件を用いて体重変化を測定した。BCDは、溶解度が>600mg/mLであるため、40%(W/V)の溶液は作ることができず、10、20%(W/V)のみを持って実験した。
アルファシクロデキストリンと本発明のHPBCDとの作用機序比較実験
2−ヒドロキシプロピル−ベータシクロデキストリンがアルファシクロデキストリンと類似の方式で作用するかを確認するために、オリーブ油あるいはトウモロコシ油と水との混合物を用いてエマルジョン形成能を確認した。
<1−1>高脂肪食餌による肥満誘発マウスモデルにおける、アルファシクロデキストリンの自主飲水による体重変化の測定
上記<実施例1>と同様の方法として、2−ヒドロキシプロピル−ベータシクロデキストリン(HPBCD)の代わりに、アルファシクロデキストリン(α−CD)を用いて、試験期間の間、体重変化を測定した。
上記<実施例3>と同様の方法として、HPBCDの代わりに、α−CDを用いて、試験期間の間、体重変化を測定した。
<2−1>高脂肪食餌による肥満誘発マウスモデルにおける、ベータシクロデキストリンの自主飲水による体重変化の測定
上記<実施例1>と同様の方法として、HPBCDの代わりに、ベータシクロデキストリン(β−CD)を用いて、試験期間の間、体重変化を測定した。
上記<実施例3>と同様の方法として、HPBCDの代わりに、β−CDを用いて、試験期間の間、体重変化を測定した。
経口ブドウ糖負荷試験計(Oralglucose tolerance test)でのHPBCDの血糖降下効果の測定
経口ブドウ糖負荷試験計は、マウスにまず試料を経口投与した後、ブドウ糖の摂取により誘発される急激な血糖の上昇を阻害する効果があるかどうかおよびブドウ糖の吸収/分解抑制活性などを総合的に予測する実験計である。
経口麦芽糖負荷試験計(Oral maltose tolerance test)でのHPBCDの血糖降下効果の測定
経口麦芽糖負荷試験計は、マウスにまず試料を経口投与した後、麦芽糖の摂取により誘発される急激な血糖の上昇を阻害する効果があるかどうかを予測する実験計である。
<1−1>散剤の製造
HPBCD 200mg
乳糖 20mg
前記成分を混合した後、気密布に充填して散剤を製造した。
HPBCD 500mg
トウモロコシデンプン 100mg
乳糖 100mg
ステアリン酸マグネシウム 2mg
前記成分を混合した後、通常の錠剤の製造方法により打錠して錠剤を製造した。
HPBCD 500mg
結晶性セルロース 3mg
ラクトース 14.8mg
ステアリン酸マグネシウム 0.2mg
前記成分を混合した後、通常のカプセル剤の製造方法によりゼラチンカプセルに充電してカプセル剤を製造した。
HPBCD 2g
異性化糖 10g
マンニトール 5g
精製水 適量
通常の液剤の製造方法により、精製水にそれぞれの成分を加えて溶解させ、レモンの香りを適量加えてから、前記成分を混合した後、精製水を加えて、全体として精製水を加えて、全体100mlに調整した後、褐色の瓶に充填して滅菌させ、液剤を製造する。
HPBCD 10mg/ml
希塩酸BP pH7.6になるまで
主利用塩化ナトリウBP 最大1ml
適当な容積の主利用塩化ナトリウムBP中にHPBCDを溶解させ、生成された溶液のpHを希塩酸BPを用いてpH7.6に調整し、主利用塩化ナトリウムBPを用いて容積を調整して十分に混合した。溶液を透明ガラスからなる5mlタイプIアンプル中に充電させ、ガラスを溶解させることにより、空気の上部格子下に封入させ、120℃で15分以上オートクレーブさせて殺菌し、注射液剤を製造した。
本発明のHPBCDを含む食品を、次のとおり製造した。
玄米、麦、もち米、はと麦を公知の方法でアルファ化させて乾燥させたものを焙煎した後、粉砕機で粒度60メッシュの粉末に製造した。
穀物類(玄米30重量部、はと麦15重量部、麦20重量部)、
種実類(エゴマ7重量部、黒豆8重量部、黒ゴマ7重量部)、
HPBCD(12重量部)、
レイシ(0.5重量部)、
ジオウ(0.5重量部)
HPBCD 10g
ビタミン混合物 適量
ビタミンAアセテート 70μg
ビタミンE 1.0mg
ビタミンB1 0.13mg
ビタミンB2 0.15mg
ビタミンB6 0.5mg
ビタミンB12 0.2μg
ビタミンC 10mg
ビオチン 10μg
ニコチン酸アミド 1.7mg
葉酸 50μg
パントテン酸カルシウム 0.5mg
無機質混合物 適量
硫酸第一鉄 1.75mg
酸化亜鉛 0.82mg
炭酸マグネシウム 25.3mg
第一リン酸カリウム 15mg
第二リン酸カルシウム 55mg
クエン酸カリウム 90mg
炭酸カルシウム 100mg
塩化マグネシウム 24.8mg
前記ビタミンおよびミネラル混合物の組成比は、比較的健康食品に適した成分を好ましい実施例として混合組成したが、その配合比を任意に変形実施してもよく、通常の健康食品の製造方法により前記成分を混合した後、顆粒を製造し、通常の方法により健康食品組成物の製造に用いることができる。

梅濃縮液 2mg
タウリン 100mg
精製水を加えて全体500ml
通常の健康飲料の製造方法により前記成分を混合した後、約1時間、85℃で撹拌加熱した後、作られた溶液を濾過して滅菌された1l容器に取得して密封滅菌してから、冷蔵保管後、本発明の健康飲料組成物の製造に用いる。
Claims (8)
- 下記化1の構造を有する2‐ヒドロキシプロピル‐β‐シクロデキストリンから本質的になる、肥満の予防および治療用薬学的組成物。
- 食欲抑制用組成物である、請求項1に記載の肥満の予防および治療用薬学的組成物。
- 下記化1の構造を有する2‐ヒドロキシプロピル‐β‐シクロデキストリンから本質的になる、肥満により誘発される疾患の予防および治療用薬学的組成物。
- 前記肥満により誘発される疾患は、高血圧、高脂血、動脈硬化、冠状動脈性心臓疾患、脳疾患および糖尿からなる群より選択されるいずれか1つである、請求項3に記載の肥満により誘発される疾患の予防および治療用薬学的組成物。
- 体脂肪減少用組成物である、請求項1に記載の肥満の予防および治療用薬学的組成物。
- 血糖降下用組成物である、請求項1に記載の肥満の予防および治療用薬学的組成物。
- 糖分解抑制用組成物である、請求項1に記載の肥満の予防および治療用薬学的組成物。
- 胃腸管内での糖吸収抑制用組成物である、請求項1に記載の肥満の予防および治療用薬学的組成物。
